Facile preparation of highly sensitive and selective fluorescent paper sensor for the visual and cyclic detection of Cu2+ and Hg2+

Liying Yu,Yaoyao Qu,Fang Chai,Lihua Chen

The highly luminescent blue fluorescent carbon quantum dots (CQDs) were prepared via the one-step hydrothermal method using L-ascorbic acid (LAA) as a carbon source. The obtained carbon quantum dots (LAA-CQDs) demonstrated high sensitivity and selectivity for detecting Cu2+ and Hg2+ based on the efficient fluorescence quenching effect, with the lowest limits of 7.59 nM and 2.29 nM, respectively. Moreover, LAA-CQDs based paper sensors were fabricated, which showed a clear fluorescence response after being exposed to Cu2+ and Hg2+, with visual detection limits of 5 μM and 3 μM, respectively. The cyclic utilization of the paper sensors was realized by restoring their fluorescence after soaking them in saturated ethylene diamine tetraacetic acid solution. The sensors can be recycled to detect Cu2+ and Hg2+ ions for at least four turns, which was conducive to environmental protection and saving resources. The LAA-CQDs with good environmental applicability have been successfully applied for the detection of Cu2+ and Hg2+ ions in real environmental water samples. In addition, we verified the low toxicity and good biocompatibility of LAA-CQDs via cytotoxicity experiments and cell imaging.

Green synthesis of carbon dots by celery leaves for use as fluorescent paper sensors for the detection of nitrophenols

Yaoyao Qu,Liying Yu,Baoya Zhu,Fang Chai,Zhongmin Su

Herein, natural celery leaves and glutathione were used as the precursors to prepare carbon dots (CDs) as novel fluorescent paper sensors, which exhibited high sensitivity and selectivity for the detection of explosive nitrophenols (including 2-nitrophenol (2-NP), 3-nitrophenol (3-NP) and 4-nitrophenol (4-NP)). The prepared CDs featured blue fluorescence emission under irradiation using an UV lamp, and the fluorescence quantum yield was as high as 53%. As fluorescent probes, the CDs showed a particular response to 2-NP, 3-NP and 4-NP, and the detection limits of 2-NP, 3-NP and 4-NP were determined to be 39 nM, 43 nM and 26 nM, respectively. Due to the multiple advantages including good water solubility, low toxicity and excellent stability, CDs are expected to be widely used. Meanwhile, a CD-based test paper was exploited and it displayed a distinctive quenching response when exposed to 3-NP and 4-NP, with the visual detection limits of 0.5 μM and 0.1 μM, respectively. Furthermore, the low toxicity and biocompatibility of CDs were also demonstrated by evaluating cell culture experiments, and CDs showed good potential applications in imaging. In summary, the prepared CDs are promising for fluorescent paper sensors and bioimaging.

Yunqi Hao,Liying Yu,Tingting Li,Lihua Chen,Xu Han,Fang Chai

In this work, the blue emission carbon dots (FA-CDs) are synthesized by one-pot solvothermal method by using folic acid as precursor. The FA-CDs emitted bright emission at 445 nm when excited at 360 nm with the QY of 31.2 %. The FA-CDs exhibit sensitive quenching response to Hg 2+ with variable concentrations systematically, which determined FA-CDs can be employed as fluorescent probe, with a reliable linear relationship between fluorescence intensity and Hg 2+ concentration, and a limit of detection (LOD) of 1.29 nM. Notably, the quenched FA-CDs can be recovered by using EDTA saturated solution with the emission comparable to initial in succession. The FA-CDs based paper sensor can be explored with similar detection performance, and it can also be restored by EDTA saturated solution. Both the restored CDs and paper sensor can be reused in the next turn for detecting Hg 2+ , which allowed the FA-CDs and their paper sensor can be serviced as sustainable probe for Hg 2+ detection. The visual LOD of paper sensor can be determined at 0.1 μM, notably, the paper sensor can be reused at least 3 times with good performance, which is beneficial to environmental protection and saving resources. Possess excellent water solubility and non-toxic properties, the cellular imaging of FA-CDs was evaluated with excellent quality fluorescent image results. The FA-CDs provide a promising convenient fluorescent probe for multi-application in detection and imaging.

Herbal allies in antibiotic warfare: Unveiling potent β-lactamase inhibitors with innovative cellulose-based colorimetric sensors

Yue Zhang,Weibiao Wang,Mei Wang,Lijuan Ma,Weiman Zhang,Shuxian Zhang,Gidion Wilson,Zhigang Yang,Yuping Sa,Fen Ma,Xinmin He,Tao Gao,Hui Yuan,Guoning Chen,Xueqin Ma

β-Lactamases (β-LS) are a significant threat to public health due to their role in conferring antibiotic resistance against nearly all clinically used β-lactam antibiotics. Consequently, the search for β-lactamase inhibitors (β-LSI), which can reversibly bind to and inactivate β-LS, has intensified. Here, a sensitive and rapid cellulose-based colorimetric sensor integrated with affinity identification model was designed for high-throughput screening of β-LSI. The innovative concept of a nitrocefin-based colorimetric paper sensor strategy was effective in rapidly detecting the inhibitory effects of herbals on β-LS. Then, potential β-LSI present in the screened herbals were captured, separated, and identified utilizing affinity identification model. The efficacy of the developed method was validated with both negative and positive drugs and was subsequently applied to the screening of β-LSI in 7 different herbal samples. Notably, Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H. F. Chou exhibited significant β-LS inhibitory activity. Further affinity separation and LC-MS analyses identified 2 bioactive compounds, spinosin, and magnoflorine, as the agents responsible for this inhibitory effect, with IC 50 values of 143.53 μM and 294.66 μM, respectively. The inhibitory effects and mechanisms were validated through antibacterial activity testing, molecular docking, and dynamic simulation. Notably, spinosin combined with aztreonam showed greater antibacterial effectiveness than avibactam. This study not only establishes a platform for the effective preliminary screening of β-LSI as a potential antimicrobial resistance-reversing agent for future clinical applications, but also inspires the further development of other cellulose-based colorimetric sensors for the rapid and high-throughput discovery of various enzyme inhibitors in complex natural medicines.

Rod-Shaped NanoZnTPyP Paper-Based Sensor for Visual Detection of Dopamine in Human Plasma

Linlin Yin,Yuyu Du,Miaohua Ge,Xiang Zhang,Xinyi Du,Xiaoqiong Wu

Dopamine (DA) is a catecholamine neurotransmitter secreted by the human adrenal medulla and is related to many medical diseases. The rapid and sensitive detection of DA levels in physiological media is attracting attention. This paper has developed a fluorescence paper-based sensor using CdTe quantum dots (QDs)-rod nanozinc 5, 10, 15, 20-tetra (4-pyridyl)−21H-23H-porphine (nanoZnTPyP) for sensitive and visual detection of DA. After adding DA, the original quenching fluorescence of the CdTe QDs-rod nanoZnTPyP sensor was effectively restored. The detection mechanism may be that the oxidation of DA to the alkaline CdTe QDs-rod nanoZnTPyP solution produced DA-quinine, and the recovery of fluorescence was caused by the electronic effect of DA-quinine and rod-shaped nanoZnTPyP. The detection range is 0.5∼10 nmol/L, and the limit of detection (LOD) is 0.38 nmol/L (S/N = 3). The sensor system was used on paper device to detect significant changes in the fluorescent color of DA at different concentrations. In addition, this method has been successfully used for the determination of DA in human plasma. The sensor system is simple, easy to operate, and has high selectivity for possible DA interfering substances, which provided new ideas for detecting DA and Parkinson’s disease, Alzheimer’s disease, and other DA-related diseases.

Colorimetric Measurement of Deltamethrin Pesticide Using a Paper Sensor Based on Aggregation of Gold Nanoparticles

Jingyang Zhu,Lifeng Yin,Weiyi Zhang,Meilian Chen,Dongsheng Feng,Yong Zhao,Yongheng Zhu

Deltamethrin (DEL) is one of the most commonly used pyrethroid pesticides that can cause serious harms to the ecological environment and human health. Herein, we have developed a paper-based colorimetric sensor impregnated with gold nanoparticles (AuNPs) for on-site determination of DEL pesticide. AuNPs show obvious color change on paper device with the presence of DEL. Measuring the gray intensity of the AuNPs on the reaction zone of the paper sensor allows accurate quantitative analysis. The detection mechanism of DEL on paper sensor was confirmed by UV-Vis spectrophotometry (UV-Vis), Fourier transform infrared spectroscopy (FT-IR), and transmission electron microscope (TEM). Under optimal conditions, the colorimetric sensor exhibited high sensitivity, rapid detection, and low detection limit within the values stipulated by Chinese detection standards (LOD = 0.584 mg/L). Besides, detecting DEL in vegetable and fruit samples also gave satisfying results, which were much consistent with those obtained by spectrophotometry. Overall, this work provided a user-friendly, cost-effective and visualized detection platform, which could be applied to rapidly detect DEL pesticides in the food safety field. Keywords:colorimetric signal;deltamethrin pesticide;gold nanoparticles;paper sensor;rapid detection

Preparation of SERS active filter paper for filtration and detection of pesticides residue from complex sample

Min Hou,Ning Li,Xiaoran Tian,Qian Yu,Juan-P. Hinestroza,Xianming Kong

The selectivity is needed mostly in SERS sensing because analytes of interest are commonly present in a complex mixture containing particles and impurities, which hinder the interactions between the laser and analyte being detected. In this manuscript, we describe our efforts developing a simple and instant. method to prepare a filter paper SERS sensor. Colloidal Ag nanoparticles were immobilized on one side of filter paper via an in-situ growth method. The fabrication process of the sensor could be finished in several minutes, and no special facility needed. The filter paper SERS sensor demonstrated a spectra uniformity with a 7.0 % point-by-point signal deviation. And the filter function of the sensor could effectively filter out interferences from samples in 1 min, that allowing the direct detection of thiram in ketchup by SERS with detection limit of 93 ppb. Furthermore, we used a Quick Easy Cheap Effective Rugged and Safe (QuEChERS) sample preparation method to detect malachite green (MG) in soil with a sensitivity as low as 0.01 ppm without any sample pre-treatment or purification. A SERS filter paper may open a new avenue for rapid testing of food quality during manufacturing as well as fast detection of potential contaminants in a myriad of substrates.

Construction of chiral nanozymes with high enantioselectivity for visual detection via smartphone-based paper sensors

Qing Li,Congnan Wu,Jiaxing Zhang,Rongxin Su,Yuefei Wang,Wei Qi

The development of enantioselective nanozymes holds profound significance in synthesizing chiral compounds and elevating analytical precision. Herein, we have synthesized chiral gold/iron hierarchically organized particles (Cys@Au/Fe Hops) possessing peroxidase-like catalytic activity, through a one-pot method using cysteine as a chiral inducer. The nanozymes exhibited remarkable enantioselective catalytic activity to 3,4-dihydroxy-phenylalanine (DOPA) enantiomer. Notably, L -Cys@Au/Fe Hops, comprising right-handed helical nanopetals, exhibited elevated catalytic efficiency for L -DOPA over D -DOPA, whereas the reverse held for D -Cys@Au/Fe Hops. The molecular dynamics simulations demonstrated hydrogen bonding interactions between the active site and substrate, thereby revealing how steric effects affected the enantioselective catalysis of chiral DOPA by L/D -Cys@Au/Fe Hops. Moreover, the chiral nanozymes were developed into a test paper sensor due to their exceptional stability, which can achieve the visual detection of DOPA enantiomer combined with a smartphone platform. This work is significant in advancing the design and practical utility of highly selective enzyme mimics.

Sulfur quantum dot-based portable paper sensors for fluorometric and colorimetric dual-channel detection of cobalt

Li Lu,Yang Chao,Li Yong,Nie Yulun,Tian Xike

Abstract Sulfur quantum dots are promising alternatives for traditional heavy metal-containing quantum dots due to their benign chemical properties and low cytotoxicity. Herein, cysteine-decorated sulfur dots were prepared from facile modification of pristine sulfur dots and can be acted as a chemosensor for the fluorometric and colorimetric dual-channel detection of cobalt (Co 2+ ) with high sensitivity and selectivity. Visual colors of the as-prepared chemosensor in the presence of Co 2+ changed from blue to colorless under UV light and also can transform from colorless to yellow under sunlight based on the photoinduced electron transfer effect. The detection limit of cysteine-decorated sulfur dots toward Co 2+ was determined as low as 0.16 μM with a wide detection range, which is lower than the permitted guideline level by Department of Environmental Protection for drinking water (1.7 μM). Furthermore, portable paper sensor-based cysteine-decorated sulfur dots were fabricated for Co 2+ detection and showed superior detection ability. Aided by a common smartphone as detector, the rapid, on-site and accurate quantification of Co 2+ in real water samples can be accomplished. Our research has provided a novel chemosensor based on sulfur dots for dual-channel detection of Co 2+ , which would expand applications of sulfur dots in environmental monitoring, diseases diagnosis, cell imaging, light-emitting diodes, etc. Graphical Abstract Cysteine-decorated sulfur dots were acted as a chemosensor for fluorometric and colorimetric dual channel detection of Co2+ based on the photoinduced electron transfer effect.

Flexible paper sensor fabricated via in situ growth of Cu nanoflower on RGO sheets towards amperometrically non-enzymatic detection of glucose

Bin Wang,Yuanya Wu,Yanfen Chen,Bo Weng,Changming Li

A flexible free-standing paper sensor was fabricated for the first time via in situ growth of Cu nanoflower on reduced graphene oxide (RGO) sheets towards amperometrically non-enzymatic detection of glucose. The low cost of copper (Cu) and graphene made the prepared sensor inexpensive in comparison with sensors made from noble metal catalysts . This Cu-RGO paper sensor exhibited high sensitivity and good selectivity when utilised in glucose sensing attributed to the synergistic effects of graphene paper and Cu nanoflowers. The combination of excellent catalytic activity , largely reactive surface area of Cu nanoflower and good conductivity of graphene paper enabled the sensor to perform well in glucose detection. The prepared glucose sensor holds a great promise for inexpensive flexible electrochemical non-enzymatic glucose sensors.

Fluorescence paper sensor meets magnetic affinity chromatography: discovering potent neuraminidase inhibitors in herbal medicines

Ma Fen,Wang Weibiao,Wang Mei,Zhang Weiman,Zhang Shuxian,Wilson Gidion,Sa Yuping,Zhang Yue,Chen Guoning,Ma Xueqin

Given the inherent complexity of natural medicines, finding a straightforward and efficient method for identifying active ingredients remains a significant challenge, yet it is of paramount importance. Influenza virus neuraminidase (NA), a primary target for anti-influenza drug development, plays a crucial role in the infection process, making it essential to develop rapid and facile methods for screening NA inhibitors. Herein, we developed a novel and efficient analytical technique for the identification of NA inhibitors from complex herbal medicines by integrating dual sensing with affinity chromatography. This approach simplifies the experimental process and highlights the benefits of being quicker, more sensitive, and cost-effective. Regarding the biosensing section, the innovative concept of a 4-methylumbelliferyl-N-acetylneuraminic acid-NA-based fluorescence paper sensor strategy enables the rapid detection of NA inhibitors in complex herbal samples. In affinity chromatography, bioactive compounds were precisely captured, separated, and identified. The efficacy and reliability of the developed method were confirmed using both negative and positive controls. Then, the method was applied to screen for NA inhibitors in 20 different herbal medicines. The results revealed that Bupleurum chinense DC. exhibited the most pronounced inhibitory effect on NA. Subsequent analysis utilizing affinity chromatography identified three bioactive compounds, namely saikosaponin a, saikosaponin d, and baicalin, as the active agents responsible for this inhibitory effect, with IC 50 values of 177.3 μM, 262.9 μM, and 241.4 μM, respectively. Molecular docking studies further indicated that these three bioactive compounds exhibit a strong binding affinity with NA. This research provides novel insights into the screening of enzyme inhibitors within herbal medicines. Graphical abstract
