Rapid quantitative analysis of hormones in serum by multilayer paper spray MS: Free MS from HPLC

Tianhao Wu,Gongwei Sun,Mingying Ma,Xingyu Pan,Sichun Zhang,Xinrong Zhang

Developing rapid and reliable method for simultaneous hormones quantitation is of great significant because of important roles of hormones in metabolism. However, current methods are faced with problems of low throughput or complicated operation procedure to remove matrices from serum samples in routine clinical diagnosis. In the present work, a multilayer PS-MS method was developed for rapid and simple detection of hormones. In the strategy, multilayer filter paper acted as the Liquid Chromatography in LC-MS/MS for separation of hormones and biological matrices. Qualitative and quantitative analysis of three hormones, testosterone (T), androsterone (ADT) and androstenedione (4-AD) were realized through MS/MS spectra. The method exhibited linearity in the range of 0.02–2 μg/L and the results of recovery and repeatability were satisfactory for standard samples and spiked serum. The time-cost of a whole detection process was less than 3 min. The established multilayer PS-MS realized rapid, simple and reliable quantitative analysis of various hormones and provided broad prospect for clinical analysis of small molecules in different biological samples. Moreover, it provides a novel MS approach with high through-put and free HPLC, meeting the requirements of point-of-care testing (POCT).

Rapid analysis of Aurantii Fructus Immaturus (Zhishi) using paper spray ionization mass spectrometry

Xuemei Liu,Zhixin Gu,Yuan Guo,Jingjing Liu,Ming Ma,Bo Chen,Liping Wang

Paper spray-mass spectrometry (PS-MS) is a rapid, solvent-efficient, and high-throughput analytical method for analyzing complex samples. In this study, a PS-MS method was developed to obtain MS profiles of Aurantii Fructus Immaturus ( aka Zhishi in Chinese) in positive and negative ion modes. In combination with multivariate analyses, including principal component analysis and cluster analysis, the PS-MS profiles of 25 batches of Zhishi were discriminated in 25 batches of Citri Reticulatae Pericarpium Viride ( aka Qingpi in Chinese; an adulterant of Zhishi). Moreover, a rapid quantitative analysis of synephrine, a prescriptive quality control component of Zhishi listed in the Chinese Pharmacopoeia, was conducted with PS-MS using synephrine-d2 as an internal standard (IS). The linearity range was 1.68–16.8   μg/mL (R 2   =   0.9985), the limit of quantitation was 0.5   μg/mL. Relative standard deviations in the intra- and inter-day precision of the MS were 4.87 and 4.90%, respectively. Compared with HPLC results, there was no significant difference in the quantitation of synephrine. This study demonstrated that the PS-MS method is useful for the rapid discrimination and quality control of Zhishi samples.

Boronate affinity paper spray mass spectrometry for determination of elevated levels of catecholamines in urine

Wei Luo,Teris A. van Beek,Bo Chen,Han Zuilhof,Gert IJ. Salentijn

The analysis of catecholamines, such as dopamine, epinephrine and norepinephrine in urine can be used in the diagnosis of certain pathologies, such as hormone-producing tumors. Here, a fast and simple quantitative boronate affinity paper spray tandem mass spectrometric (PS-MS/MS) method is established, which can improve selectivity and reduce ion suppression without needing any instrumental chromatography. We use here the property of boronic acids, which can selectively bind ortho -diol-containing compounds under alkaline conditions. Paper tip modification and catechol enrichment protocols were developed to selectively bind, clean up and subsequently desorb such catecholamines. Standard catecholamine solutions, as well as human urine samples were analyzed with the PS-MS(/MS) method, which is fast, cheap and easy-to-operate compared to HPLC-MS/MS. Despite its high simplicity, boronate affinity PS-MS/MS exhibits good performance compared to HPLC-MS/MS in human urine analysis in terms of precision (2.1%–7.2% vs. 1.1%–2.9%) and accuracy (−10.2%–9.3% vs. −4.8%–5.1%), and a physiologically relevant limit of detection (0.027–0.12 μg mL −1 ). The boronate affinity PS-MS/MS clearly achieved the detection limits that would allow the fast analysis of urine samples for clinical purposes, such as screening for pheochromocytoma (exceeding 0.5 μg mL −1 ).

Rapid detection of monoamine neurotransmitters by carboxyl ionic organic crystal-functionalized paper spray mass spectrometry

Zhao Wang,Jing He,Xin Li,Jia Chen,Mingxia Sun,Hongdeng Qiu

The development of a mass spectrometry-based workflows for monoamine neurotransmitters (NTs) detection in biological samples is crucial for the accurate diagnosis of adrenal tumor. In this study, a carboxyl ionic organic crystal (CIOC) was synthesized via ion self-assembly of cationic and anionic monomers, featuring abundant carboxyl active sites for selective monoamine NTs capture. The CIOC-functionalized paper strips significantly enhance the sensitivity of direct capillary spray miniature mass spectrometry (DCS-mini MS) for monoamine NTs analysis. For biofluid analysis, the CIOC-functionalized paper strips were immersed in samples, washed with ACN to remove interferents, and analyzed by DCS-mini MS. Compared to unmodified paper strips, the CIOC-functionalized paper strips exhibited significantly enhanced sensitivity, with signal amplification factors of 7.6-fold for dopamine, 3.4-fold for norepinephrine, 4.6-fold for 5-hydroxytryptamine, and 5.6-fold for epinephrine in plasma, and corresponding increases of 10.9-fold, 4.0-fold, 6.3-fold, and 7.5-fold in urine. This is attributed to the synergistic effects of electrostatic interactions and hydrogen bonding between CIOC materials and the monoamine NTs. The method showed excellent linearity (R² > 0.9981), with LOD ranging from 0.51 to 2.11 ng mL −1 in urine and 1.53–4.51 ng mL −1 in plasma, and LOQ ranging from 1.7 to 7.01 ng mL −1 in urine and 5.1–15.03 ng mL −1 in plasma. The robustness of the method was confirmed through validation of precision (<10 % RSD) and recoveries (80.1–97.3 %). The CIOC@DCS-mini MS platform enables rapid (3 min), label-free clinical monoamine NTs monitoring, offering a precise solution for diagnosing monoamine NTs-related disorders.
