Traces of mercury ions in environmental water can harm humans and animals. Paper-based visual detection methods have been widely developed for the rapid detection of mercury ions; however, existing methods are not sensitive enough to be used in real environments. Here, we developed a novel, simple and effective visual fluorescent sensing paper-based chip for the ultrasensitive detection of mercury ions in environmental water. CdTe-quantum-dots-modified silica nanospheres were firmly absorbed by and anchored to the fiber interspaces on the paper’s surface to effectively avoid the unevenness caused by liquid evaporation. The fluorescence of quantum dots emitted at 525 nm can be selectively and efficiently quenched with mercury ions, and the ultrasensitive visual fluorescence sensing results attained using this principle can be captured using a smartphone camera. This method has a detection limit of 2.83 µg/L and a fast response time (90 s). We successfully achieved the trace spiking detection of seawater (from three regions), lake water, river water and tap water with recoveries in the range of 96.8–105.4% using this method. This method is effective, low-cost, user-friendly and has good prospects for commercial application. Additionally, the work is expected to be utilized in the automated big data collection of large numbers of environmental samples.

With the emergence of numerous food safety problems, rapid and accurate detection of histamine in food spoilage remains a challenge. To this end, we developed a simple design and easy synthesis of fluorescein-based probe FCHO to achieve specific and rapid (<1 s) quantitative detection of histamine through “imine formation” reaction. Significant enhanced fluorescence signal in response to histamine enabled our probe with high sensitivity as low as 51 nM. Utilizing the visualized fluorescence color changes of the probe as histamine increasing, we combined it with paper-based test chip to construct a color-resolved and highly selective recognition system. In addition, our proposed probe has been successfully used to visually imaging histamine changes in fish samples. Finally, for the first time, we have proved it possesses reliable ability to directly in situ imaging the distribution of histamine in whole spoiled fish. Thus, our strategy will provide great potential for monitoring food spoilage.

Background Fluoroquinolones (FQs) are widely used for their excellent antimicrobial properties, yet their release into aquatic environments pose risks to ecosystems and public health. The accurate monitoring and analysis of FQs present challenges due to their low concentrations and the complex matrices found in actual environmental samples. To address the need for auto-pretreatment and on-line instrumental analysis, developing new microextraction materials and protocols is crucial. Such advancements will provide better analytical assurance for the effective extraction and determination of FQs at trace levels, which is of great significance to environmental protection and human health. Results In this work, we presented a Co 2+ mediated paper-based molecularly imprinted polymer chip (CMC@Co-MIP), combined with UPLC analysis, to develop an effective analytical method for identifying and quantifying trace amounts of ciprofloxacin (CIP) and enrofloxacin (ENR) in water samples. Notably, the addition of Co 2+ in CMC@Co-MIP helped to capture the template molecule CIP through coordination before imprinting, which significantly improved the ordering of the imprinted cavities. CMC@Co-MIP exhibited a maximum adsorption capacity up to 500.20 mg g −1 with an imprinting factor of 4.12, surpassing previous reports by a significant margin. Furthermore, the enrichment mechanism was extensively analyzed by various characterization techniques. The developed method showed excellent repeatability and reproducibility (RSD < 13.0 %) with detection limits ranging from 0.15 to 0.21 μg L −1 and recoveries ranging from 64.9 % to 102.3 % in real spiked water samples. Significance We developed a novel microextraction paper-based chip based on Co 2+ mediation, which effectively improved the selectivity and convenience of extracting FQs. This breakthrough allowed the chip to have a high enrichment efficiency as well as provide a robust on-line instrumental program. It also confirms that the imprinting scheme based on metal ion coordination is a high-performance strategy.

In this work, zinc-doped carbon dots (Zn-CDs) were anchored on a three-dimensional wheel type paper-based microfluidic chip, and were decorated with 6-mercaptonicotinic acid (MNA) and L-cysteine (L-Cys) for highly sensitive and rapid fluorescence detection of Cu2+. Zn-CDs were first anchored on paper through the amide bonds between the carboxyl groups of the Zn-CDs and the amino groups of the paper. Afterwards, Zn-CDs were decorated with MNA and L-Cys, effectively preventing the Zn-CDs from aggregation. The nitrogen atom on the pyridine ring and the carboxylic acid groups in MNA and L-Cys coordinated with Cu2+ to form a nonfluorescent ground-state complex, causing the fluorescence quenching of the Zn-CDs. The three-dimensional rotary design could simplify the operation process and achieve simultaneous analysis of multiple samples with different concentrations. Under optimal conditions, the fluorescent sensor exhibits linear response for the determination of Cu2+ in the range from 0.1 to 60 μg L−1 with the detection limit (LOD) of 0.018 μg L−1. The proposed strategy provides a novel way for the highly sensitive detection of Cu2+ in a complex water environment.

The interstitial fluid (ISF) contains rich bioinformation for disease diagnosis and healthcare monitoring. However, the efficient sampling and detection of the biomolecules in ISF is still challenging. Herein, we develop a facile but versatile ISF analysis platform by combining controllable hollow microneedles (HMNs) and elaborate microfluidic paper-based analytical devices (μPADs). The HMNs and μPADs was fixed in a bottom PDMS layer. A top PDMS layer containing a cylindrical cavity to produce negative pressure for sampling was packaged on the bottom PDMS layer. The HMNs enable efficient and swift sampling of sufficient ISF to the μPADs through one-touch finger operation without extra manipulations. The μPADs realized to simultaneously detect glucose and lactic acid in the detection area to produce chromogenic agents and analyzed by the self-programed RGB application (APP) in smartphones. The HMN microfluidic paper-based chip provides a point-of-care platform for accurate detection of biomolecules in ISF, holding great promise in the development of wearable device.

A simple one-pot method is developed to prepare positively charged aggregated silver nanoparticles (a-AgNPs). The obtained a-AgNPs show strong localized surface plasmon resonance (LSPR) absorption, whose wavelength can be easily tuned to match the commonly used lasers in surface enhanced Raman scattering (SERS). Furthermore, the obtained a-AgNPs can be easily fabricated into paper-based SERS chips by filtering against a negatively charged filter membrane. On the basis, a convenient SERS sensor has been developed for the detection of thiram using a 785 nm handheld Raman spectrometer.

A high-performance lateral flow and paper-based microfluidic device ( μ PAD) was constructed to develop a rapid read-out analytical platform for heavy metal elements (Pb (II)) in green tea with complex matrices. The entire analytical device was integrated with several functional parts, including a liquid flow channel and an enrichment visualization zone prepared by the molecularly imprinted polymer (MIP) protocol. Under the optimal conditions, the Pb (II) content in tea products can be sensitively and specifically analyzed using an in situ visualization readout. Herein, a rapid visual read-out analysis of the lead level can be achieved by overcoming the other co-existing interfering elements in the traditional green tea sample. The ionic imprinted paper chip can effectively capture the Pb (II) of the digested sample solution, and a visualization sensing process occurred between the metal elements and a chelating visual reagent with high sensitivity and selectivity in situ . Owing to the stable visualization performance, the constructed chip demonstrated good contrast, with a low background and easily recognizable colored spots. The linear range was 2.5–100 mg kg −1 , and the limit of detection for Pb (II) in traditional green tea was 1.375 mg kg −1 . In addition, other co-existing metal elements did not interfere with the analysis because of the appropriate pretreatment and selectivity of the paper-based chip. Furthermore, the analytical results for tea samples were verified by atomic absorption spectrophotometer (AAS) tests, which demonstrated the robustness and reliability of the proposed method. Therefore, the high-performance visualization lateral flow- μ PAD is a promising platform for the rapid analysis of metal element levels in food products.

In recent years, the widespread use of sulfonamide antibiotics has led to increasing concerns over antibiotic residues. To address the challenges, this study developed a rapid and highly sensitive detection method based on ratiometric fluorescence. By employing nitrogen-doped carbon quantum dots (N-BCDs) and biomass-derived carbon quantum dots (RCDs), single-channel and ratiometric fluorescence sensors were constructed. The incorporation of molecularly imprinted polymers (MIPs) enhanced the specificity and sensitivity of target recognition. Experimental results demonstrated that the ratiometric fluorescence sensor exhibited superior anti-interference capability and detection stability compared to the single-channel sensor, achieving a detection limit as low as 10.41 nM, significantly outperforming the single-channel sensor's limit of 25.94 nM. To enable rapid on-site detection, the ratiometric fluorescence sensor was integrated with a microfluidic paper-based chip, resulting in a visual detection platform. Moreover, a microfluidic paper-based chip with a spatially rotational structure was developed, integrating smartphone photo-recognition functionality to convert fluorescence ratio changes into RGB values, enabling rapid visual detection of SDZ. In practical applications involving milk and water samples, the sensor achieved recovery rates between 98.70 % and 108.70 %, demonstrating excellent performance. This ratiometric fluorescence microfluidic paper-based chip sensor offers simplicity, high sensitivity, and strong selectivity, making it suitable for non-professional users to perform rapid on-site detection. It shows great potential for applications in food safety and environmental monitoring.

In this work, a new chemiluminescence (CL) system of Co-Fe prussian blue analogs nanocubes (Co-Fe PBA NCs) that can catalyze luminol to produce strong CL in the absence of H2O2 was established. Co-Fe PBA NCs have the property of oxidase-like activity, and it can catalyze the generation of active oxygen radicals in a dissolved oxygen system. Since caffeic acid (CA) can remove reactive oxygen species in the system, a sensitive detection method for CA on a paper-based chip was developed. Under the optimal conditions, this method showed a good linear response to CA in the range of 10 – 800 ng mL−1 with a limit of 3 ng mL−1. The proposed method had been used for the determination of CA in tea samples. The results may open a new avenue for the catalytic property on luminol CL system without extra oxidants.

High-speed and accuracy identification of pathogens has become increasingly critical in both individual patient care and public health. Artificial intelligence (AI)-assisted surface-enhanced Raman scattering (SERS) biosensors enable simultaneous identification of multiple pathogens. However, there are still problems such as low accuracy and limited diversity in bacterial fingerprints. To this end, we present a novel multi-branch adaptive attention convolutional neural network (MBAA-CNN)-assisted paper-based SERS chip for prompt and reliable pathogen discrimination. In the approach, we employed a dual-function molecule 4-mercaptophenylboronic acid (4-MPBA) to capture bacteria and enhance Raman spectra diversity, referring as 4-MPBA labeled mode (label mode). Meanwhile, we utilized the K -means algorithm to identify pathogens in the label mode, producing much higher accuracy compared to label-free mode ( n = 2000). Furthermore, we acquired 98.6 % accuracy at all pathogen species and 99.5 % accuracy at the antibiotic-resistant and sensitive strains ( n = 10,000) using MBAA-CNN. The superior performance of MBAA-CNN was further validated through comparisons with traditional machine learning models, particularly in terms of loss value, speed and accuracy. We envision the developed approach has potential for early culture-free diagnosis of pathogens and real-time monitoring of microbial contamination in water environment.

The development of high-performance specific sensors is promising for the rapid detection of harmful residues in animal-derived foods. Recently, luminescent metal-organic framework/molecularly imprinted polymer (LMOF/MIP) materials have been developed as ideal candidates for the analysis of harmful residues. Here, we reported a simple fabrication protocol of paper-based chip through in-situ growth of LMOF on a negatively charged modified filter paper, a paper-based molecularly imprinting layer (FP@BA-Eu@MIP) was thereafter successfully prepared via the boronate affinity-based controllable oriented surface imprinting strategy. The paper-based chips obtained were used to construct a rapid test strip of tetracycline (TC). After addition of TC, significant fluorescence changes on the surface of the FP@BA-Eu@MIP paper-based chip could be observed from blue to red via inner filter effect and photo-induced electron transfer under the excitation of 360 nm. The adsorption kinetics was explored in detail. The presented strip exhibited satisfied selectiveness and sensitivity with a limit of detection of 8.47 μg L −1 for TC. It was confirmed that LMOF/MIP as a biomimetic recognition module can play a crucial role in enrichment and fluorescence response. This study provided a real application case for an in-situ fabricated fluorescence paper-based chip in rapidly detecting harmful residues.

Antibiotic resistance, the ability of some bacteria to resist antibiotic drugs, has been a major global health burden due to the extensive use of antibiotic agents. Antibiotic resistance is encoded via particular genes; hence the specific detection of these genes is necessary for diagnosis and treatment of antibiotic resistant cases. Conventional methods for monitoring antibiotic resistance genes require the sample to be transported to a central laboratory for tedious and sophisticated tests, which is grueling and time-consuming. We developed a paper-based chip, integrated with loop-mediated isothermal amplification (LAMP) and the “light switch” molecule [Ru(phen)2dppz]2+, to conduct turn-on fluorescent detection of antibiotic resistance genes. In this assay, the amplification reagents can be embedded into test spots of the chip in advance, thus simplifying the detection procedure. [Ru(phen)2dppz]2+ was applied to intercalate into amplicons for product analysis, enabling this assay to be operated in a wash-free format. The paper-based detection device exhibited a limit of detection (LOD) as few as 100 copies for antibiotic resistance genes. Meanwhile, it could detect antibiotic resistance genes from various bacteria. Noticeably, the approach can be applied to other genes besides antibiotic resistance genes by simply changing the LAMP primers. Therefore, this paper-based chip has the potential for point-of-care (POC) applications to detect various gene samples, especially in resource-limited conditions.
