Fluorescent paper–based sensor based on carbon dots for detection of folic acid

Li Wang,Zhang Xiaoyue,Miao Chunyue,Li Ruijun,Ji Yibing

Paper-based devices have been very much in the foreground of analytical science recently. This work innovatively proposed a fluorescent paper–based sensor (FPS) constructed on a hybrid polydimethylsiloxane (PDMS)/paper platform where cellulose papers functionalized with carbon dots (CDs) as fluorophores by Schiff base chemistry were loaded on the grooves array of a designed PDMS plate. As a proof of concept, the performance of FPS was investigated with folic acid (FA) as the target analyte. Under optimal conditions, FPS enabled a rapid fluorescence quenching response to FA via inner filter effect in a wide range of 1–300 μmol L −1 with the limit of detection of 0.28 μmol L −1 . The feasibility of FPS was further verified by the detection of FA in orange juice and urine samples with satisfactory results. The covalent modification of CDs on paper endowed the FPS with good assay reproducibility and stability. Interestingly, FPS achieved a more sensitive assay of FA than the conventional strategy, by which the same CDs were directly used to detect FA in a solution-based system. The FPS illuminated a novel strategy for construction of reliable and sensitive assays based on paper-based devices. It is of paramount importance for its practical application in biosensing and clinical diagnosis. Graphical abstract

Anti-fouling array paper-based device for rapid and accurate discrimination of foodborne pathogens in real samples

Kuiyu Wang,Yayue Huang,Yu Wan,Wang Chen,Jianwei Liang,Liangke Gong,Zebao Rui,Tao Yang

Foodborne pathogens contamination can cause serious food safety incidents. Early and rapid detection of foodborne pathogens is very important. Paper-based devices have been applied for on-site rapid detection of foodborne pathogens due to their cheapness and portability. However, the paper-based foodborne pathogen detection devices for real samples often receive interference, resulting in inaccurate detection results. In this study, bovine serum albumin (BSA) was modified on the surface of nitrocellulose membrane to construct anti-fouling sensing interface. Through array design, the anti-fouling array paper-based device (APAD) with multiple detection channels was constructed. The naked eye cut-off limits of the APAD were 10 2 colony forming units (cfu)/mL, 10 2  cfu/mL, and 10 3  cfu/mL for Staphylococcus aureus in orange juice, Escherichia coli O157: H7 in milk and orange juice, and Escherichia coli O157:H7 in soy sauce, respectively. The above results indicate that the APAD has good practical application potential in food safety.

Quantum-dot-functionalized paper-based device for simultaneous visual detection of Cu(II), Mn(II), and Hg(II)

Jinping Yue,Qing Lv,Wan Wang,Qing Zhang

In this work, a new paper-based device (PAD) was developed for the colorimetric detection of multiple heavy metals. The sensing principle of this PAD is based on the analyte-regulated photocatalytic activity of quantum dots (QDs) to certain substrates. The PAD consisted of three filter paper layers, defining a sample area, three channels, and three testing areas. Three photocatalytic reactions between different QDs and their corresponding substrates could be performed in one device, achieving the simultaneous detection of Cu(II), Mn(II), and Hg(II). The detection limits are 7.3 nM for Cu(II), 5.4 nM for Mn(II), and 8.1 nM for Hg(II), respectively. This PAD was applied to analyze real samples and the results were consistent with those obtained by inductively coupled plasma-optical emission spectrometer. This approach may provide a promising colorimetric sensing platform for multi-target analysis.

Yuanyuan Li,Huinan Chen,Rong Huang,Dongmei Deng,Xiaoxia Yan,Liqiang Luo

Backgroud The global prevalence of diabetes mellitus, a serious chronic disease with fatal consequences for millions annually, is of utmost concern. The development of efficient and simple devices for monitoring glucose levels is of utmost significance in managing diabetes. The advancement of nanotechnology has resulted in the indispensable utilization of advanced nanomaterials in high-performance glucose sensors. Modulating the morphology and intricate composition of transition metals represents a viable approach to exploit their structure/function correlation, thereby achieving optimal electrocatalytic performance of the synthesized catalysts. Results Herein, a sensitive and rapid Cu-encapsulated Cu 2 S@nitrogen-doped carbon (Cu@Cu 2 S@N–C) hollow nanocubes-functionalized microfluidic paper-based analytical device (μ-PAD) was fabricated. Through a delicate sacrificial template/interface technique and thermal decomposition, inter-connected hollow networks were formed to boost the active sites, and the carbon shell was coated to protect Cu from being oxidation . For application, the constructed μ-PAD is used for glucose sensing utilizing an origami automated sample pretreatment system enabled by a simple application of strong alkaline solution on wax paper. Under optimal circumstances, the Cu@Cu 2 S@N–C electrochemical biosensor exhibits broad detection range of 2–7500 μM (R 2  = 0.996) with low detection limit of 0.16 μM (S/N = 3) and high sensitivity of 1996 μA mM −1  cm −2 . Additionally, the constructed μ-PAD also exhibited excellent selectivity, stability, and reproducibility. Significance By rationally designing the double-shell hollow nanostructure and introducing Cu-encapsulated inner layer, the synthesized Cu@Cu 2 S@N–C hollow nanocubes show large specific surface area , short diffusion channels, and high stability. The proposed origami μ-PAD has been successfully applied to serum samples without any additional sample preparation steps for glucose determination, offering a new perspective for early nonenzymatic glucose diagnosis.

Integrated Sandwich-Paper 3D Cell Sensing Device to In Situ Wirelessly Monitor H2O2 Released from Living Cells

Zhuanzhuan Shi,Yunpeng Li,Xiaoshuai Wu,Bo Chen,Wei Sun,Chunxian Guo,Chang Ming Li

Point-of-care testing (POCT) has attracted great interest because of its prominent advantages of rapidness, precision, portability, and real-time monitoring, thus becoming a powerful biomedical device in early clinical diagnosis and convenient medical treatments. However, its complicated manufacturing process and high expense severely impede mass production and broad applications. Herein, an innovative but inexpensive integrated sandwich-paper three-dimensional (3D) cell sensing device is fabricated to in situ wirelessly detect H2O2 released from living cells. The paper-based electrochemical sensing device was constructed by a sealed sandwiched bottom plastic film/fiber paper/top hole-centered plastic film that was printed with patterned electrodes. A new (Fe, Mn)3(PO4)2/N-doped carbon nanorod was developed and immobilized on the sensing carbon electrode while cell culture solution filled the exposed fiber paper, allowing living cells to grow on the fiber paper surrounding the electrode. Due to the significantly shortening diffusion distance to access the sensing sites by such a unique device and a rationally tuned ratio of Fe2+/Mn2+, the device exhibits a fast response time (0.2 s), a low detection limit (0.4 μM), and a wide detection range (2–3200 μM). This work offers great promise for a low-cost and highly sensitive POCT device for practical clinic diagnosis and broad POCT biomedical applications.

Single-Layered Paper-Based Microfluidic Devices Made by Paint-Spraying Technique with Great Barrier Resistance for Colorimetric Assays

Nie Beixi,Zhao Shaoyu,Deng Muhan,Yu Peng,Yang Yi,Lei Weixin,Yin Lu

The single-layered paint-spraying paper based microfluidic devices (sp-μPADs) have been for the first time integrated with the colorimetric assay. The polymethyl methacrylate (PMMA) board was applied as the mask instead of the iron mask, reducing the power requirement of the cutting machine. The paint was sprayed into the paper, which blocked the pores of the paper and constructed the hydrophobic barrier on the paper. The paint barrier was highly resistant to the various kind of chemicals and the minimum channel dimensions was 1.4 mm. By using the PMMA masks with different designs, the sp-μPADs with different designs have been fabricated and successfully applied to the single analyte assay of iron or bovine serum albumin (BSA) and the simultaneous assay of copper and nickel. Moreover, the feasibility of the multiplexed assay of eight analytes was also demonstrated. The analytical performances indicated that the fabricated sp-μPADs offered a promising platform in the colorimetric assays of either single or multiple analytes.

Programmable-Printing Paper-Based Device with a MoS2 NP and Gmp/Eu-Cit Fluorescence Couple for Ratiometric Tetracycline Analysis in Various Natural Samples

Zongbao Sun,Yunlong Gao,Zeng Niu,Haodong Pan,Xuechao Xu,Wen Zhang,Xiaobo Zou

Paper-based fluorescence devices, with smartphone aids, bring considerable operation convenience for tetracycline (TC) sensing. Nevertheless, they must meet the challenge in real determination against complicated backgrounds. Considering that, we present a programmable-printing paper-based device and then apply it to TC determination for various natural samples. MoS2 NPs and Gmp/Eu-Cit are synthetized as composite probes. A static quenching process is found with MoS2 NP fluorescence at 430 nm, while significant magnification of Gmp/Eu-Cit emission is obtained at 617 nm, establishing a valuable ratiometric indicator. Remarkably, two-stage programmable printing maximizes the proposed sensing capability. A transitive device, containing a gradually changing amount of a certain probe, is prepared to sense TC. With a homemade smartphone application and 3D-printed measurement chamber, the corresponding signals are examined to explore optimal setups. These setups are automatically processed to prepare the final-version device, not requiring manual operations. Benefitting from this interesting feature, the proposed device gains many rewards in performances. It effectively senses TC in a wide range from 12.7 nM to 80 μM and simultaneously provides naked eye-legible signals and smartphone-based readouts with confident selectivity and stability. This device is consequently applied for various samples of soil, river water, milk, and serum and meets well with HPLC–MS and recovery tests.

Paper-based analytical device for fast colorimetric detection of total hemoglobin and free hemoglobin from human blood sample

Wan-qi Ren,Bin Ji,Zhi Kuang,Fang Fang,Xiao-Lin Guo,Zhi-Yong Wu

Total hemoglobin (THb) and free hemoglobin (FHb) are biomarkers of anemia and hemolysis, playing a critical role in human health status. This study reports a paper-based device by field amplified sample stacking (FASS) using colorimetric method to quantify both THb and FHb. The detection of THb was based on the color intensity of the Hb stacking band, a limit of detection (LOD) of 11 g∙L −1 and a linear working range of 16 ∼ 140 g∙L −1 were achieved. With selective chromatic reaction of the stacking band with o-tolidine, the sensitivity of Hb detection was further enhanced by 2000 times, a LOD of 25.8 mg∙L −1 and a linear range of 30 ∼ 1000 mg∙L −1 was obtained for FHb. This paper-based device was successfully used for detection of high concentration THb and low concentration FHb from the same blood sample, and both meeting the requirements of clinical diagnosis. In addition, the feasibility of on-site plasma preparation and online integration with the device was also preliminarily investigated with a blood separation membrane. This paper-based device could be used as a new platform for developing fast and low-cost Hb point-of-care testing devices.

A simple paper-based ratiometric luminescent sensor for tetracyclines using copper nanocluster-europium hybrid nanoprobes

Mengjiao Mo,Xueshen Wang,Lingyue Ye,Yaoquan Su,Yang Zhong,Lingzhi Zhao,Yunyun Zhou,Juanjuan Peng

Tetracyclines (TCs), as one of the broad-spectrum antibiotics, are widely used to treat bacterial infections. The residues of TCs in animal-origin foods and drinking water have raised safety concerns and affected the public health. Thus, there is a high demand to develop a simple and rapid method for the detection of TCs. In this work, we developed a ratiometric luminescence probe for the sensitive and visualized detection of TCs. Specifically, tannic acid-stabilized copper nanoclusters (TA-CuNCs) with blue emission at 433 nm were synthesized. The luminescence of TA-CuNCs attenuated partially by the europium ions (Eu 3+ ) due to the aggregation-induced quenching. When TCs were added to the TA-CuNCs-Eu 3+ system, the luminescence of TA-CuNCs at 433 nm can be further quenched by the inner-filter effect, and the characteristic luminescence of Eu 3+ at 617 nm emerged due to the formation of Eu 3+ -TCs complex. The ratio of the luminescence at 617 nm–433 nm increased linearly to the concentration of TCs. Additionally, we demonstrated the detection of oxytetracycline in real samples such as tap and lake water, milk, pharmaceutical industry wastewater, honey and soil extract with high recovery rate (97.25%–103.44%). Furthermore, a portable paper device is fabricated by the luminescent probe to conduct the on-site analysis of TCs.

Detecting N-Phenyl-2-Naphthylamine, L-Arabinose, D-Mannose, L-Phenylalanine, L-Methionine, and D-Trehalose via Photocurrent Measurement

Feng Li,Ruoxi Yang,Jian Xu,Guohai Xu,Ye Wu

The concentration of small molecules reflects the normality of physiological processes in the human body, making the development of simple and efficient detection equipment essential. In this work, inspired by a facile strategy in point-of-care detection, two devices were fabricated to detect small molecules via photocurrent measurement. A linear response of the photocurrent against the concentration of the small molecules was found. In the first device, metal ions were introduced into gel substrates made by xanthan gum to enhance photocurrent response. N-phenyl-2-naphthylamine was measured when iron or manganese ions were used. L-Phenylalanine was measured when the gel was modified by samarium, iron, cerium, or ytterbium ions. L-(+)-Arabinose was detected via the gels modified by iron or holmium ions. D-(+)-Mannose was detected when the gel was modified by ytterbium, manganese, chromium, or sodium ions. L-Methionine was detected in the gels modified by samarium, zinc, or chromium ions. The second device was based on a paper sheet. A sugar-like molecule was first synthesized, which was then used to modify the paper. The detection was possible since the photocurrent showed a linear trend against the concentration of D-Trehalose. A linear fit was conducted to derive the sensitivity, whose value was found to be 5542.4. This work offers a novel, simple, and environmentally sustainable platform that is potentially useful in remote areas lacking medical devices.

Paper-based lipid bilayer magnetic interface for ultrasensitive assay of tumor-derived extracellular vesicles PD-L1

Mingli Ye,Qi Niu,Lu Yan,Jianzhou Feng,Lingling Wu,Xiaoya Hu,Qin Xu,Yun Shu

Programmed death ligand 1 (PD-L1) of tumor-derived extracellular vesicles (T-EVs) is an important biomarker to predict immunotherapy response. The traditional bioassays of T-EVs PD-L1 are limited by the detection specificity because of the distribution of the signals by PD-L1 from nontumor-derived EVs, which leads to unsatisfactory predictive accuracy. In this work, we developed a simple and ultrasensitive method for the quantification of T-EVs PD-L1 by using the lipid bilayer magnetic interface as the isolation and enrichment carrier along with hybridization chain reaction (HCR) for signal amplification in a paper-based device. First, magnetic beads were covered by liposome bilayers and then anchored with the cholesterol modified epithelial cell adhesion molecule (EpCAM) aptamer to construct a fluid multivalent magnetic interface and realize the high-performance isolation of T-EVs. Moreover, the biosensor relied on the PD-L1 aptamer to initiate the HCR of EVs. Benefiting from the high isolation ability of the lipid bilayer magnetic interface and the high signal amplification ability of HCR, the dual-aptamer-based biosensor is capable of highly sensitive detection of T-EVs with a detection limit as low as 5.0 EVs·µL −1 in a paper-based device. Overall, the paper-based lipid bilayer magnetic interface biosensor not only can serve as an alternative tool for predicting the immunotherapy response but also displays great potential in point-of-care disease diagnosis.

Fluorescent paper sensor based on green emissive carbon nanodots for visual detection of acid orange II in food samples

Qin Hu,Wenqian Sun,Lixia Xiao,Jie Han,Tianzu Guan,Wenyuan Zhou,Xiaojuan Gong,Zhen-quan Yang

This work for the first time presents a visual fluorescence paper sensor for accurate and on-site detection of acid orange II (AO 2 ) in food matrix, where green-emissive carbon nanodots (G-CDs) were exploited. The as-synthesized G-CDs had an average particle size of 3.9 ± 0.7 nm and displayed green emission at 511 nm upon excitation at 450 nm. When there existed AO 2 , the G-CDs fluorescence was effectively suppressed via inner filter effect (IFE) and dynamic interaction. The as-prepared G-CDs probe achieved a 0.01–25.0 μM linear scope and a 36.5 nM limit of detection. Particularly, owing to a distinguishable reduction in G-CDs emission intensity by a smartphone application , a visual probing platform along with a nanopaper-based device were be developed for recognizing AO 2 . The corresponding detection limits were severally 0.61 μM and 1.39 μM. Success was achieved in applying our fluorescence approaches for detecting AO 2 in Yuba, chili powder, sunflower seeds, tomato paste and orange juice samples, attaining desirable results. The present work underlines the potential of smartphone-integrated nanopaper based on CDs as an encouraging candidate for on-site AO 2 recognition in food matrix.
