Integrating a Copper–Histidine Brace in a Mimetic Nanozyme Streamlines the Tyrosinase Recognition Moiety to Achieve Chiral Differentiation

Lingyu Nie,Hui Zhang,Weiheng Kong,Rong-Mei Kong,En-Sheng Zhang,Jin Li,Yan Zhao,Fengli Qu

Designing artificial mimetic enzymes with high activity/selectivity to replace chiral bioenzymes is of great interest in the development of chiral materials consisting of molecules, enantiomers, that exist in two forms as mirror images of one another but cannot be superimposed. In this study, the chiral catalytic structural unit was streamlined from tyrosinase to integrate a mimetic nanozyme. The chiral amino acid l-histidine, as the chiral binding/recognition site, and the active metal site Cu were coupled (Cu@l-His) to create a copper–histidine brace with enantioselective catalytic ability to tyrosinol enantiomers. Results of kinetic parameters and activation energies confirmed the excellent peroxidase-like activity with a preference of Cu@l-His to l-tyrosinol. Such a preference could be attributed to the structurally oriented copper–histidine brace with a stronger affinity and catalytic activity to l-tyrosinol. By accurately evaluating chiral recognition units derived from bioenzymes, stable and superior chiral mimetic nanoenzymes could be constructed in a more straightforward and simplified manner, and they could also be extended to the reconstruction of diverse chiral enzymes.

Spiropyran-modified upconversion nanocomposite as a fluorescent sensor for diagnosis of histidinemia

Jian Su,Yiwei Li,Wen Gu,Xin Liu

Histidinemia is a congenital metabolic disorder where the histidine (His) metabolism is blocked, resulting in increased concentrations of His in blood and urine. The disease causes an abnormal development of the patient's nervous system, which leads to many serious illnesses. Therefore, it is very important to diagnose early. In this study, we developed a novel fluorescent nanosensor NaGdF4:Yb3+, Er3+@SiO2–spiropyran (UCNP@SiO2–SP). The nanosensor displayed a “turn-off” fluorescence response towards His. When His was mixed with UCNP@SiO2–SP, His could specifically bind to SP, which could cause the isomerization of SP. The structure of SP was changed from spiroform into merocyanine form. The luminescence of the sensor was overlapped with the absorption of the merocyanine form. As a result, His will lead to fluorescence quenching of the sensor based on inner filter effects (IFE), which can be used to detect His. Importantly, as the first report of a UCNP@SiO2–SP nanosensor for detecting His, this method exhibits good selectivity and anti-interference capability. The detection limit is 4.4 μM. In addition, the amount of His in urine was also measured, suggesting the applicability of this sensor for histidinemia diagnosis.

Formation of urocanic acid versus histamine from histidine in chub mackerel (Scomber japonicus) fillets as determined by a mixed-mode HPLC method

Lingling Zhao,Xinqi Pan,Jie Shi,Hua Ye,Huanan Guan,Yuanxin Guo,Jian-Jun Zhong

Urocanic acid, like histamine, is a degradation product of histidine that may accumulate over the storage of histidine-rich pelagic fish. Recently evidence from the dermatological and related fields has directed the attention of food scientists toward the adverse health effects of urocanic acid, mostly as a possible “missing factor” responsible for scombrotoxicosis besides histamine. However, experimental effort to study the hypothesis remains scarce, especially regarding the formation of urocanic acid in comparison to the well-documented histamine. Here we report a mixed-mode HPLC method for simultaneously analyzing urocanic acid (both the trans - and cis -isomers), histamine, and histidine in chub mackerel ( Scomber japonicus ) fillets and the use of the method to monitor their changes in concentration at typical refrigeration and abuse temperatures. The method features a simple, reliable chromatographic analysis in cation-exchange/reversed-phase/hydrophilic-interaction triple mode of retention, without the need of precolumn derivatization or addition of ion-pairing agents. At both 4 and 20 °C slow but consistent formation of trans -urocanic acid was observed over time with a relatively faster rate at 20 °C. Histamine formation, by contrast, occurred at 20 °C but ceased completely at 4 °C. In addition, we demonstrate the occurrence of trans -to- cis isomerization of UCA in chub mackerel fillets upon direct sunlight exposure: under our conditions, an isomerization yield around 13 % was achieved within hours on a sunny winter day. Overall, this study provides a chemical analysis method and relevant data for exploring the possible role of urocanic acid in the aetiology of scombrotoxicosis.

Nanofibers comprising polystyrene and zinc acetate for extraction of salivary histidine prior to its determination by HPLC–DAD

Wu Xiaoxiao,Huang Jingjing,Zhang Chi,Kang Xuejun

A novel nanofiber-based solid phase extraction (NFSPE) coupled with HPLC–DAD was developed for the determination of salivary histidine, a new potential index of physiological events. The treatment of the sample was based on a coupling reaction of histidine with the diazotization products of sulfanilic acid and sodium nitrite in a slightly alkaline medium, followed by NFSPE. Nanofibers comprising polystyrene and zinc acetate were used as a novel solid-phase sorbent to extract derived histidine via the chelation of Zn 2+  with the imidazole group in the analyte. The eluted target compound was analyzed by HPLC–DAD. Good linearity was achieved with the concentrations of histidine ranging from 0.645 to 64.5 µM; the detection limit was 0.193 µM. The intra-day and inter-day coefficients of variation were less than 8.0%. The method was successfully applied to the determination of salivary histidine in the subjects exposed to a public speaking stress. Identification of Binding Sites in Copper(II)-Peptide Complexes Using Infrared Spectroscopy

Zhenghangcheng Li,Xueyu Feng,Kaijun Yuan,Xin-Xing Zhang

Complex formation of the copper(II) ion (CuII) with histidine (H) and H-containing peptides plays a crucial role in various metallo-enzymatic reactions. To elucidate the nature of coordinate bonding in CuII complexes, Fourier-transform infrared spectroscopy and 2D IR spectroscopy were employed to investigate the coordination geometries of CuII with diglycine, l-histidylglycine (HG), glycyl-l-histidine (GH), and glycylglycyl-l-histidine. The coordination of CuII to different peptide groups, including the peptide N- and C-termini, the amide group, and the imidazole of the H side chain, exhibits distinct spectral features. The derived molecular structure of the CuII–HG complex based on these spectral features significantly differs from that of CuII–GH, suggesting a preference of the N-terminus and the steric hindrance of the H side chain in CuII chelation.

Histidine-inspired polyacrylonitrile-based adsorbent with excellent hemocompatibility for the simultaneous removal of bacteria and endotoxin in septic blood

Ziyue Ling,Chunji Jiang,Xianda Liu,Yupei Li,Weifeng Zhao,Changsheng Zhao

Endotoxins (LPS) are components of the cell wall of Gram-negative bacteria that are released when bacterial cell membrane is destroyed, the bacteria and the released LPS have led to serious diseases such as sepsis and septic shock. Histidine functionalized adsorbents have been used to remove LPS, however, the present hemoperfusion adsorbents have drawbacks such as inadequate LPS adsorption capability, high cost of raw material, and so on. Inspired by the structure of histidine and incorporated antibacterial molecules, we synthesize allylamine-modified polyacrylonitrile (PAD)/poly (imidazolium bromide) (PMIBr) composite (PAI) microspheres as hemoperfusion adsorbents. The PAI microspheres show excellent antibacterial ability with high killing efficiency of 98.8% against S. aureus and 95.4% against E. coli . In addition, the adsorption capacity of the microspheres for LPS in PBS is 433.5 EU/g within 3 h, and the LPS concentration could also be reduced from 7.34 EU/mL to below 0.5 EU/mL in the plasma environment. The PAI microspheres exhibit outstanding blood compatibility, antibacterial property and endotoxin removal capability, which have great potential in the treatment of sepsis patients.

Inhibitory mechanism of cell-free supernatants of Lactobacillus plantarum on Proteus mirabilis and influence of the expression of histamine synthesis-related genes

Yilun Wang,Huijie Pei,Yuxuan Liu,Xiaohong Huang,Lin Deng,Qinjie Lan,Shujuan Chen,Li He,Aiping Liu,Xiaolin Ao,Shuliang Liu,Likou Zou,Yong Yang

The purpose of this study was to evaluate the inhibitory mechanism of cell-free supernatants of Lactobacillus plantarum (LCFS) on Proteus mirabilis and estimate the expression of histamine synthesis-related genes. The minimum inhibitory concentration (MIC) of the LCFS and antibacterial curve assay was determined by the tube double-dilution method. The antibacterial mechanism was explored by cell integrity, morphology, and structure of P. mirabilis . Further, the expression of the histidine decarboxylase ( hdcA ) gene and histidine/histamine antiporter ( hdcP ) gene of P. mirabilis and the intracellular and extracellular contents of histidine and histamine were determined. The MIC of the LCFS was 12.5 mg/mL, and the antibacterial curve assay showed an inhibited growth of P. mirabilis . The increase in the Alkaline Phosphatase (AKP) content, membrane permeability, and potential indicated the destruction of the cytoplasm, cell membrane, and cell wall of P. mirabilis . The results of SEM and TEM revealed that the LCFS disrupted the normal morphology and structure of the cell. Moreover, differences in the extracellular and intracellular histidine and histamine content between the MIC group and the control group was consistent with the decreased expression of hdcA and hdcP genes in the MIC group observed in the previous antimicrobial studies. Hence, the inhibitory mechanism of LCFS on histamine formation was clarified at cellular and molecular levels.

A stable and specific hydrogel-based plasmonic nanosensor for histamine detection

Fangfang Wang,Yawen Liu,Wenping Liu,Huabing Yang,Haiming Hu,Zhigang Zhang,Songlin Liu,Junping Zheng,Hongtao Liu

Plasmonic nanosensors have garnered increasing attention due to their naked-eye visibility and facile preparation. However, the inadequate stability and specificity significantly challenged their widespread application. Here, we hypothesized that capping with double-charged acids and loading in the hydrogel should bring gold nanoparticles (AuNPs) more stability and specificity for sensing small molecules. In the plasmonic nanosensor, aspartic acid-capped AuNPs (Asp-AuNPs) served as both the sensing material and colorant. Compared with conventional AuNPs, Asp-AuNPs exhibited a lower Zeta potential and enhanced selectivity for histamine. Moreover, Asp-AuNPs rapidly responded to histamine in less than 2 min, even after seven weeks of storage. The isothermal titration calorimetry analysis further supported the affinity of Asp-AuNPs for histamine. Using the alginate hydrogels as a matrix for Asp-AuNPs, the hydrogel-based plasmonic nanosensor could detect histamine on the surface of and in the atmosphere around expired fish. In summary, we have developed a novel, durable, and stable hydrogel-based plasmonic nanosensor for contact and contactless freshness detection.

Fluorescence Sensors for the Detection of L-Histidine Based on Silver Nanoclusters Modulated by Copper Ions

Yuxia Li,Min Li,Liuzhi Hu,Baozhu Zhang

In this study, Cu2+modulated silver nanoclusters were constructed for the turn-on, label-free detection of L-histidine. Six Ag NCs protected by oligonucleotides (DNA-Ag NCs) were tested in a series of experiments. Finally, A-DAN-Ag NCs were chosen as the best candidate due to their excellent fluorescent properties. The fluorescence of A-DAN-Ag NCs was quenched using Cu2+through energy or electron transfer. However, quenched fluorescence could be restored dramatically in the presence of L-histidine due to Cu2+liberation from A-DAN-Ag NCs and because of the chelation between the imidazole group of L-histidine and Cu2+. The proposed sensor exhibited high selectivity towards L-histidine over other amino acids, with a limit of detection (LOD) of 0.096 μM ranging from 0 to 8 μM. The proposed sensor succeeded in detecting L-histidine in diluted human urine. Therefore, the sensor has promising practical applications in biological systems.

Rapid synthesis of multifunctional copper nanoclusters for selective and sensitive detection of hexavalent chromium and anti-counterfeiting

Yidan Liang,Menglei Sun,Mao Wang,Haoting Hou,Shujie Pang,Zhe Zhang,Xudong Yang

In this work, a copper nanocluster (His-Cu NCs) with green fluorescence, which uses histidine (His) as the template agent, was developed. This nanomaterial was characterized using methods such as transmission electron microscopy (TEM). Based on this nanocluster, a fluorescence sensing platform was developed for the linear detection of Cr(VI) ions in solution. The surface of His-Cu NCs contains a large number of oxygen-containing and nitrogen-containing functional groups, which can react with Cr(VI), leading to charge transfer between ligands and metal ions, thereby quenching the fluorescence of His-Cu NCs. The sensor exhibits excellent sensitivity and selectivity for detecting Cr(VI), with a 0.31 μM detection limit for Cr(VI). Additionally, a fluorescent filter paper was created to detect Cr(VI). The fluorescence color of the fluorescent filter paper changed from green to blue with the increase in Cr(VI) concentration under the irradiation of a UV lamp at 365 nm. Furthermore, this His-Cu NCs’ fluorescence exhibits good stability, making it possible to employ the NCs as fluorescent ink for writing and drawing anti-counterfeiting icons on paper.

Copper nanoclusters-modified with papaya juice for fluorescence turn-on detection of serum l-histidine Today four files of proofs was sent to m.saksena@elsevier.com Please check the files

Qiqi Tan,Juan Qiao,Rongyue Zhang,Li Qi

Papaya juice protected copper nanoclusters was depicted for detection of serum l -histidine. The fluorescent probe was synthesized by using a simple “one pot” hydrothermal preparation strategy with papaya juice playing dual roles both as a green reductant and stabilizer. The excitation and emission peaks of the copper nanoclusters were at 400 nm and 490 nm, respectively. The fluorescence was turned-on in addition of l -histidine due to the enhanced surface electron density. The fluorescent intensity increased linearly with l -histidine concentration ranging from 2.0 to 50.0 µM. The proposed copper nanoclusters was further applied to determine l -histidine in rat serum after intraperitoneal injection.

Solubilisation of myosin in a solution of low ionic strength l-histidine: Significance of the imidazole ring

Xing Chen,Yufeng Zou,Minyi Han,Lihua Pan,Tong Xing,Xinglian Xu,Guanghong Zhou

Myosin, a major muscle protein, can be solubilised in a low ionic strength solution containing l -histidine (His). To elucidate which chemical constituents in His are responsible for this solubilisation, we investigated the effects of 5   mM His, imidazole (Imi), l -α-alanine (Ala), 1-methyl- l -histidine (M-his) and l -carnosine (Car) on particle properties of myosin suspensions and conformational characteristics of soluble myosin at low ionic strength (1   mM KCl, pH 7.5). His, Imi and Car, each containing an imidazole ring, were able to induce a myosin suspension, which had small particle size species and high absolute zeta potential, thus increasing the solubility of myosin. His, Imi and Car affected the tertiary structure and decreased the α-helix content of soluble myosin. Therefore, the imidazole ring of His appeared to be the significant chemical constituent in solubilising myosin at low ionic strength solution, presumably by affecting its secondary structure.
