Magnetically Actuated Heterogeneous Microcapsule-Robot for the Construction of 3D Bioartificial Architectures

Yang Liu,Gen Li,Haojian Lu,Yuanyuan Yang,Zeyang Liu,Wanfeng Shang,Yajing Shen

Core–shell microcapsules as one type of the most attractive carriers and reactors have been widely applied in the fields of drug screening and tissue engineering owing to their excellent biocompatibility and semi-permeability. Yet, the spatial organization of microcapsules with specific shapes into three-dimensional (3D) ordered architectures still remains a big challenge. Here, we present a method to assemble shape-controllable core–shell microcapsules using an untethered magnetic microcapsule-robot. The microcapsule-robot with the shape-matching design can grab the building components tightly during the transportation and assembly processes. The core–shell feature of the microcapsule effectively prevents the magnetic nanoparticles from interacting with bioactive materials. The assembly results of cell-loaded heterogeneous microcapsules reveal that this strategy not only allows the magnetic microcapsule-robot to work in different workspaces in vitro for the creation of 3D constructions but also offers a noninvasive and dynamical manipulation platform by remotely controlling the position and orientation of the soft and liquid-like microcapsule components individually.

Self-powered electrostatic tweezer for adaptive object manipulation

Cong Liu,Fang Wang,Xuemin Du

Manipulating objects using physical tweezers is crucial in physics, chemistry, and biology. Despite significant advancements, however, these tweezers are still limited by the need for complex electrode arrays, external power sources, and restricted charge-generation capability. Here, we report an innovative self-powered electrostatic tweezer (SET) that harnesses the unique capability of accumulation and tunability of triboelectric charges of the poly(vinylidene fluoride-trifluoroethylene) (P(VDF-TrFE))-based self-powered electrode during sliding triboelectrification, imparting unprecedented flexibility and adaptivity for object manipulation. The SET enables the precise manipulation of objects with diverse materials, including bubbles, solid balls, and liquid droplets, with a superior high velocity of 353 mm s −1 . Notably, the SET is adaptive to open-to-closed platforms, single-to-multiple objects, and two-dimensional (2D)-to-3D surfaces, thereby extending the SET to various applications including droplet robots, cell assembly, and pump-free microfluidics. By eliminating the need for complex electrode arrays, external power sources, and heat generation, the SET holds great potential for next-generation tweezers and microfluidics.

Rapid, Large-scale, and Precise Cell Assembly for Tumor Model Fabrication via Template-modulated Flow-driven Strategy

Yiqing Chen,Yujiao Wang,Shangze Jiang,Xiaolei Guo,Xuanhe Zhang,Zhenwei Liang,Huayu Yang,Yuan Ma,Jiadao Wang

Biomimetic performance of organoids heavily relies on rapid, precise, and programmable cell patterning at large scales. However, current methods, such as 3D printing, external field-driven cell assembly, and template-assisted cell assembly, face challenges in achieving high precision, rapid processing, and large-scale production simultaneously. To overcome these limitations, a flow-driven cell assembly (FDCA) strategy is introduced. Utilizing directional drag force in a template-modulated flow field, cells can be patterned on a micropore membrane, subsequently imprinted onto a substrate. Following the principle of layer-by-layer fabrication, the slices can be stacked to compose a 3D architecture. This approach enables fast, large-scale fabrication with precise cell patterning. To validate the platform's effectiveness, epithelial-mesenchymal transition (EMT) process of the assembled A549 cells is examined. Compared to cells cultured in 2D well plates or 3D spheroids, after treatment with TGF-β, cells patterned via the FDCA method exhibited more realistic variations in cytoskeletal structure and the expression levels of metastasis-related proteins, as well as an improvement in the capability of cell invasion. This strategy is believed offers a novel approach for rapid fabrication of large, precise organoids as well as in vitro cancer models, with promising applications in physiological research, drug screening, and organ transplantation.

Long-span delivery of differentiable hybrid robots for restoration of neural connections

Jie Shen,Yun Wang,Min Yao,Shubo Liu,Zhiguang Guo,Li Zhang,Ben Wang

Lumbar puncture is a minimally invasive technique for delivering drugs into cerebrospinal fluid (CSF). However, its effectiveness is limited by the spine’s extensive length and the narrow, complex CSF spaces, which hinder efficient passive diffusion of therapeutic agents. Here, we have engineered a differentiable stem cell-assembled soft robot (SCASR) by using a three-dimensional self-assembly process in liquid marbles. These biologically viable robots can interface with the tissues inside the CSF, adapting to the complex surroundings while navigating. With X-ray imaging and magnetic actuation, the SCASRs can be guided toward targeted regions. The SCASRs autonomously disassemble into individual cells, which then differentiate into neural cells to facilitate the restoration of neural connections, thereby aiding the recovery of paralyzed lower limbs. The proposed strategy provides an imaging-based therapeutic system for nerve injury, enabling accessibility of hard-to-reach spinal regions and facilitating efficient therapy in minimally invasive manner, by means of soft microrobots.

Programing Cell Assembly via Ink-Free, Label-Free Magneto-Archimedes Based Strategy

Tanchen Ren,Miribani Maitusong,Xuhao Zhou,Xiaoqian Hong,Si Cheng,Yin Lin,Junhui Xue,Dilin Xu,Jinyong Chen,Yi Qian,Yuwen Lu,Xianbao Liu,Yang Zhu,Jian’an Wang

Tissue engineering raised a high requirement to control cell distribution in defined materials and structures. In “ink”-based bioprintings, such as 3D printing and photolithography, cells were associated with inks for spatial orientation; the conditions suitable for one ink are hard to apply on other inks, which increases the obstacle in their universalization. The Magneto-Archimedes effect based (Mag-Arch) strategy can modulate cell locomotion directly without impelling inks. In a paramagnetic medium, cells were repelled from high magnetic strength zones due to their innate diamagnetism, which is independent of substrate properties. However, Mag-Arch has not been developed into a powerful bioprinting strategy as its precision, complexity, and throughput are limited by magnetic field distribution. By controlling the paramagnetic reagent concentration in the medium and the gaps between magnets, which decide the cell repelling scope of magnets, we created simultaneously more than a hundred micrometer scale identical assemblies into designed patterns (such as alphabets) with single/multiple cell types. Cell patterning models for cell migration and immune cell adhesion studies were conveniently created by Mag-Arch. As a proof of concept, we patterned a tumor/endothelial coculture model within a covered microfluidic channel to mimic epithelial-mesenchymal transition (EMT) under shear stress in a cancer pathological environment, which gave a potential solution to pattern multiple cell types in a confined space without any premodification. Overall, our Mag-Arch patterning presents an alternative strategy for the biofabrication and biohybrid assembly of cells with biomaterials featured in controlled distribution and organization, which can be broadly employed in tissue engineering, regenerative medicine, and cell biology research.

Polyphenol-Scaffolded Modular Assembly of Heterotypic Cell–Cell Assemblies for Potentiating Cancer Immunotherapy

Guidong Gong,Qimin Yin,Qinling Liu,Yajing Zhang,Jinyuan Liu,Mengyuan Dai,Jiawen Li,Xianglian He,Jiezhou Pan,Yunxiang He,Bo Wang,Jiyu Tong,Junling Guo

Cells existing in the form of clusters often exhibit distinct biological functions from their single-cell counterparts. However, the ability to modulate cell–cell interactions among multiple cell types through molecular scaffolds remains an ongoing challenge. Here, a supramolecular phenolic network on surfaces of live cells designed is engineered to act as modular scaffolds that promote intercellular interactions, presenting a universal platform for the construction of cell–cell assemblies (CCAs). Utilizing the inherent cell surface affinity of supramolecular phenolic network modules, a series of heterotypic CCAs is efficiently established by assembling five distinct microorganisms and five types of mammalian cells. A specific application of CCAs with the combination of L. casei and macrophages ( L. casei @Mφ) is highlighted for enhanced cancer immunotherapy. Upon intravenous administration, L. casei @Mφ allows the chemotactic Mφ to facilitate the accumulation of L. casei at the tumor. The accumulated L. casei enables the polarization of tumor-associated Mφ toward a pro-inflammatory phenotype, markedly improving the immunotherapeutic response, slowing tumor progression, and mitigating lung metastasis. This work demonstrates the potential of polyphenol-scaffolded modular assembly in manipulating cell–cell interactions to enhance multicellular cooperation, offering a general approach to controlling complex cellular behaviors.

Photopyroelectric tweezers for versatile manipulation

Fang Wang,Cong Liu,Zhengjin Dai,Weizhong Xu,Xinyue Ma,Yufeng Gao,Xuewu Ge,Wei Zheng,Xuemin Du

Optical tweezers and related techniques offer extraordinary opportunities for research and applications in physical, biological, and medical fields. However, certain critical requirements, such as high-intensity laser beams, sophisticated electrode designs, additional electric sources, or low-conductive media, significantly impede their flexibility and adaptability, thus hindering their practical applications. Here, we report innovative photopyroelectric tweezers (PPT) that combine the advantages of light and electric field by utilizing a rationally designed photopyroelectric substrate with efficient and durable photo-induced surface charge-generation capability, enabling diverse manipulation in various working scenarios. These PPTs allow for remote and programmable manipulation of objects with diverse materials (polymer, inorganic, and metal), different phases (bubble, liquid, and solid), and various geometries (sphere, cuboid, and wire). Furthermore, the PPT is not only adaptable to high-conductivity media but also applicable to both portable macroscopic manipulation platforms and microscopic manipulation systems, enabling cross-scale manipulations for solid objects, liquid droplets, and biological samples. The high-level flexibility and adaptability of the PPT extend to broad applications in manipulating hydrogel robots, sorting particles, assembling cells, and stimulating cells. By surpassing the limitations of conventional tweezers, the PPT bridges the gap between macroscopic and microscopic manipulations, offering a revolutionary tool in robotics, colloidal science, biomedical fields, and beyond.
