A dual-usage near-infrared (NIR) cell membrane targeting chimeric peptide for cancer cell membrane imaging and photothermal ablation

Chen Pei-Ling,Shi Qun-Ying,Chen Tian,Wang Ping,Liu Yun,Liu Li-Han

Due to the low background and deep tissue penetration of near-infrared light, near-infrared fluorescence (NIRF) probes have received much attention in recent years. Cell membrane targeting probes can be used in in vitro and in vivo for living cell imaging, disease diagnosis and tumor targeted therapy. However, most of the currently available cell membrane probes are blue or green emissive fluorophores, NIRF probes are relatively rare. Herein, we designed and prepared a NIRF chimeric peptide probe named C 16 -CARK, which could target the tumor cell membrane with longtime retention (more than 4 h). Moreover, the C 16 -CARK can heat and disrupt the cell membrane upon high irradiation dose, revealing its dual roles as a cell membrane imaging probe and a photothermal agent. This kind of near-infrared fluorescence probe with cell membrane targeting and retention property provides a well design paradigm for the field of cancer imaging and treatment.

Cancer cell membrane as gate keeper of mesoporous silica nanoparticles and photothermal-triggered membrane fusion to release the encapsulated anticancer drug

Ding Yi,Zhu Yingmei,Wei Shaohua,Zhou Jiahong,Shen Jian

The cancer cell membrane (CCM) is widely used for nanomedicine encapsulation to improve their tumor-targeting ability. However, few researches are focused on drug release process and mechanism of these composite. Cell membrane fusion (two separate lipid membranes merge into a single continuous bilayer), one of the most fundamental processes in biology could be promoted by heating. Thus, in this manuscript, CCM was exploited as the gate keeper and tumor-targeting moiety of anticancer drug-loaded mesoporous silica nanoparticles (MSN). In addition, the drug release process and mechanism based on cell membrane fusion were studied. The loaded drugs included indocyanine green (ICG), a FDA-approved near-infrared photothermal agent for clinical applications, and doxorubicin (DOX), a chemotherapy drug. By coating CCM on drug-loaded MSN, the keeper could prevent unexpected drug leaking during the circulation and provide tumor targeting based on its specific recognition function. After arriving tumor tissue post-tail vein injection, the tumor position is irradiated by 808 nm light to trigger light–thermal conversion process of ICG to heat the composite (DOX-ICG@MSN@CCM) and promote cell membrane fusion between the encapsulated CCM and targeted tumor cell membrane. Such process can enhance the uptake efficacy of DOX-ICG@MSN@CCM to tumor cells. Furthermore, ICG could provide a synergistic photothermal therapy activity with DOX.

In Situ Time-Controllable Chemical Plasma Membrane Injury by Microfluidic Probe Reveals Self-Repair Ability of Single Cells

Yang Song,Qiang Zhang,Yuxuan Li,Jiaxu Lin,Jin-Ming Lin

The loss of plasma membrane integrity poses a serious threat to cells, and the plasma membrane repair mechanism is vital to cell survival. To investigate the plasma membrane repair mechanism, a method for causing controllable plasma membrane injury on single cells and the visualization of the plasma membrane repair process are proposed. A microregion with harmful chemicals is created by open microfluidic probe, which is applied to target single cells for injury. The injury time is precisely controlled by simply moving the target cells in and out of the microregion. Wound-repair process caused by injury is analyzed by fluorescence imaging in real time, revealing the repair kinetics at the single cell level. Using this method, the effect of external conditions (Ca 2+ , Mg 2+ , glucose) on plasma membrane repair in single cells is studied and the dynamics of intracellular species (Ca 2+ ) is monitored. Meanwhile, cellular heterogeneity of plasma membrane repair is revealed. This method provides a precise way to induce chemical plasma membrane injury at the single-cell level, which is a promising tool to study the mechanism of plasma membrane repair.

Effect of Plasma-Activated Solution Treatment on Cell Biology of Staphylococcus aureus and Quality of Fresh Lettuces

Jianying Zhao,Jing Qian,Hong Zhuang,Ji Luo,Mingming Huang,Wenjing Yan,Jianhao Zhang

This study aimed to investigate effects of plasma-activated solution (PAS) on the cell biology ofStaphylococcus aureusand qualities of fresh lettuce leaves. PAS was prepared by dielectric barrier discharge plasma and incubated withS. aureusfor 10–30 min or with lettuces for 10 min. Effects on cell biology were evaluated with microscopic images, cell integrity, and chemical modification of cellular components. Effects on lettuce quality were estimated with the viable microbial counts, color, contents of vitamin C and chlorophyll, and surface integrity. PAS reducedS. aureuspopulation by 4.95-log and resulted in increased cell membrane leakage. It also resulted in increased contents of reactive oxygen species in cells, C=O bonds in peptidoglycan, and 8-hydroxydeoxyguanosine content in cellular DNA, and reduced ratios of unsaturated/saturated fatty acids in the cell membrane. PAS treatment reduced bacterial load on fresh lettuce and had no negative effects on the quality. Data suggest that PAS can be used for the disinfection of ready-to-eat fresh vegetables. Keywords:reactive oxygen species;peptidoglycan;cell membrane;DNA;lettuces

Inhibitory effect of nerol against Aspergillus niger on grapes through a membrane lesion mechanism

Yanzhen Wang,Xiaobin Zeng,Zhengkun Zhou,Ke Xing,Akalate Tessema,Hong Zeng,Jun Tian

The antifungal activity of Nerol (NEL) was evaluated against Aspergillus niger , a known cause of grape spoilage, and possible modes of action were explored. The antifungal efficacy of NEL against A. niger has been proven in a dose-dependent manner through in vitro (mycelial growth and spore germination) and in vivo (grapes) tests. An obvious increase in the membrane permeability was detected through the determination of extracellular pH and conductivity. The damage in the cell membrane was measured by monitoring the influx of Propidium iodide (PI) using flow cytometry and was further verified through the inhibition of ergosterol synthesis in the cell membrane. NEL induced morphological changes, and the integrity of the A. niger fungal cells decreased with increasing NEL concentrations. A dose-dependent decrease in ergosterol production was also seen in A. niger cell membranes upon exposure to NEL. The mechanism-of-action study revealed that NEL activated a membrane-active mechanism that inhibits ergosterol synthesis and consequently compromised the membrane integrity of A. niger . As a result, the membrane permeability changes and causes cell death. The membrane lesion mechanism makes NEL a natural alternative to commercial fungicide for control of stored grape spoilage.

NINJ1 regulates plasma membrane fragility under mechanical strain

Zhu Yunfeng,Xiao Fang,Wang Yiling,Wang Yufang,Li Jialin,Zhong Dongmei,Huang Zhilei,Yu Miao,Wang Zhirong,Barbara Joshua,Plunkett Christopher,Zeng Mengxue,Song Yiyan,Tan Tian,Zhang Ruibin,Xu Kezhen,Wang Zhongxing,Cai Changjie,Guan Xiangdong,Hammack Scott,Zhang Liang,Shi Zheng,Xiang Fu-li,Shao Feng,Xu Jie

Plasma membrane integrity is vital for nearly all aspects of cell functioning 1 . Mechanical forces can cause plasma membrane damage 2 , but it is not known whether there are large molecules that regulate plasma membrane integrity under mechanical strain. Here we constructed a 384-well cellular stretch system that delivers precise, reproducible strain to cultured cells. Using the system, we screened 10,843 siRNAs targeting 2,726 multi-pass transmembrane proteins for strain-induced membrane permeability changes. The screen identified NINJ1, a protein recently proposed to regulate pyroptosis and other lytic cell death 3,4 , as the top hit. We demonstrate that NINJ1 is a critical regulator for mechanical strain-induced plasma membrane rupture (PMR), without the need of stimulating any cell death programs. NINJ1 level on the plasma membrane is inversely correlated to the amount of force required to rupture the membrane. In the pyroptosis context, NINJ1 on its own is not sufficient to fully rupture the membrane, and additional mechanical force is required for full PMR. Our work establishes that NINJ1 functions as a bona fide determinant of membrane biomechanical properties. Our study also suggests that PMR across tissues of distinct mechanical microenvironments is subjected to fine tuning by differences in NINJ1 expression and external forces.

Dynamic membrane changes and osmotic effects by sugar alcohols

Chen Lichun,Yao Feng,Xue Songwen,Yao Kuang,Huang Yun,Zhao Huimin,Shao Qiong

Sugar alcohols are natural sweeteners with various physiological functions, commonly used in low-calorie foods and pharmaceuticals. Current research primarily focuses on their sweetening properties and metabolic effects, often overlooking their interactions with cell membranes. This study built a giant phospholipid vesicle model to examine vesicle deformation in erythritol (Ery) and xylitol (Xyl) environments. The permeation of these sugar alcohols through real cell membranes was also investigated. Fluorescence microscopy and zeta potential measurements showed that osmotic stress from concentration gradients disrupted vesicle membrane structure. Ery and Xyl reduced ROS levels in HEK-293 cells and influenced membrane permeability. Notably, Xyl increased vesicle adsorption on the cell membrane at the same concentration. The findings indicate that sugar alcohols interact with membrane lipids through hydrogen bonds or other non-covalent interactions, modifying cell membrane structure and properties, thus providing a theoretical foundation for understanding their role in physiological environments.

Role of chemical groups in regulating membrane tension of mBMSCs under stretch stimulation

Yunqing Ye,Haoyang Huang,Hong Li,Gang Wu

As a crucial mechanobiological regulator, the tension of the cell membrane plays a vital role in governing cellular adhesion, proliferation, and differentiation processes. Additionally, it displayed a dynamic response to mechanical microenvironmental changes. This research systematically examines the mechanoresponsive behaviors of mouse bone marrow mesenchymal stem cells (mBMSCs) that are cultured on poly(dimethylsiloxane) (PDMS) substrates which are functionalized with methyl (-CH 3 ), amino (-NH 2 ), and carboxyl (-COOH) groups. Under both static and stretching conditions, it is found that compared with the -CH 3 surface, static culture on the -NH 2 and -COOH functionalized surfaces significantly promotes the proliferation of mBMSCs and upregulates the expression of extracellular matrix adhesion-related genes, especially focal adhesion kinase (FAK) and integrin β1. Morphometric analysis reveals that there are concomitant increases in the cell spreading area and the number of pseudopods on these modified surfaces. Mechanical stretching stimulation not only amplifies these cellular responses but also leads to more uniform FAK distribution. The assessment by atomic force microscopy (AFM) shows that both chemical functionalization (-NH 2 /-COOH) and stretch stimulation reduce the deformability of the cell membrane, and the -NH 2 modification exhibits a greater membrane-stiffening effect than -COOH.

Synthesis of Amphiphilic and Red Emissive Carbon Dots as Nanoprobes for Targeted Stimulated Emission Depletion Imaging of Live Cell Plasma Membranes

Taihe Zhang,Lu Gao,Hong Du,Xiaoyu Huang,Wei Ge,Haisen Ta,Fu Wang

The cell plasma membrane, serving as the fundamental barrier separating cells from the extracellular environment, is a complex and dynamic structure that is crucial to cell biology. The super-resolution observation of plasma membrane morphology and dynamics in living cells, facilitated by stimulated emission depletion (STED) microscopy, holds significant importance for advancing the understanding of plasma membrane functions. Due to the photobleaching and unclear target, the development of plasma membrane nanoprobes suitable for STED imaging remains challenging. In this study, we present red fluorescent carbon dots (R-CDs) characterized by amphiphilicity and photostability. As carbon-based nanomaterials, R-CDs exhibit the targeting ability of the cell plasma membrane, resulting in a superior imaging signal-to-noise ratio. Furthermore, R-CDs enable STED imaging of plasma membranes in living cells, achieving a resolution enhancement of at least 3-fold compared to confocal imaging. Leveraging the excellent performance of R-CDs, the dynamic behavior of microstructures such as vesicles on the plasma membrane was successfully observed through time-lapse STED imaging under nanoscale. Hence, R-CDs, as fluorescent nanoprobes with good photostability and biocompatibility, demonstrate significant potential for long-term STED imaging of live-cell plasma membranes and the exploration of plasma membrane dynamics.

Red blood cell membrane-derived phototherapeutic nanodiscs: A new platform for enhanced phototherapy

Zhuge Deli,Li Li,Sun Xueying,Liang Hui,Jin Chenjie,Lu Ailing,Gao Wenli,Zhong Yutong,Li Wenlu,Chen Sihao,Wang Fang,Yan Linzhi,Meng Weiyang,Lin Xiaoji,Zhang Xufei,Liu Yong,Li Yuanfeng,Zhao Yingzheng,Zhang Xiang,Chen Mengchun,Chen Yijie

Phototherapy holds great potential for treating cancer and infections but faces limitations related to photosensitizer accumulation, tissue penetration, and diminished photo-conversion efficiency, particularly in deep-seated tumors and infections. Here, a biomimetic phototherapeutic nanodisc platform consisting of erythrocyte membranes and the photosensitizer IR780 is developed. With the advantages of the ultra-small size and exceptional biosafety of cell membrane-derived nanodiscs, this platform facilitates efficient accumulation and deep tissue penetration at disease sites. Upon near-infrared (NIR) irradiation, IR780 delivered via the nanodisc exhibits enhanced photothermal conversion efficiency, markedly inhibiting tumor growth in an orthotopic 4T1 breast cancer model and reducing bacterial load in a methicillin-resistant Staphylococcus aureus (MRSA) skin infection model. Furthermore, the nanodisc platform demonstrates outstanding biocompatibility in mice. In conclusion, this nanodisc system significantly extends the functional potential of cellular nanodiscs, presenting a promising strategy to address the challenges of photosensitizer delivery and biosafety in phototherapeutic applications.

Erythrocyte membrane-coated gold nanocages for targeted photothermal and chemical cancer therapy

Dao-Ming Zhu,Wei Xie,Yu-Sha Xiao,Meng Suo,Ming-Hui Zan,Qing-Quan Liao,Xue-Jia Hu,Li-Ben Chen,Bei Chen,Wen-Tao Wu,Li-Wei Ji,Hui-Ming Huang,Shi-Shang Guo,Xing-Zhong Zhao,Quan-Yan Liu,Wei Liu

Recently, red blood cell (RBC) membrane-coated nanoparticles have attracted much attention because of their excellent immune escapability; meanwhile, gold nanocages (AuNs) have been extensively used for cancer therapy due to their photothermal effect and drug delivery capability. The combination of the RBC membrane coating and AuNs may provide an effective approach for targeted cancer therapy. However, few reports have shown the utilization of combining these two technologies. Here, we design erythrocyte membrane-coated gold nanocages for targeted photothermal and chemical cancer therapy. First, anti-EpCam antibodies were used to modify the RBC membranes to target 4T1 cancer cells. Second, the antitumor drug paclitaxel (PTX) was encapsulated into AuNs. Then, the AuNs were coated with the modified RBC membranes. These new nanoparticles were termed EpCam–RPAuNs. We characterized the capability of the EpCam–RPAuNs for selective tumor targeting via exposure to near-infrared irradiation. The experimental results demonstrate that EpCam–RPAuNs can effectively generate hyperthermia and precisely deliver the antitumor drug PTX to targeted cells. We also validated the biocompatibility of the EpCam–RAuNs in vitro. By combining the molecularly modified targeting RBC membrane and AuNs, our approach provides a new way to design biomimetic nanoparticles to enhance the surface functionality of nanoparticles. We believe that EpCam–RPAuNs can be potentially applied for cancer diagnoses and therapies.

A “Glue” Strategy to Engineer Cancer Cell Membrane-Camouflaged Prodrug Nanoassemblies for Efficient Targeted Chemotherapy

Lingxiao Li,Xin Wang,Yafan Xiao,Minglong Huang,Shiyi Zuo,Yingjie Zhao,Xiaoyuan Fan,Qi Lu,Xin Li,Yuki Takahashi,Zhonggui He,Yoshinobu Takakura,Bingjun Sun,Jin Sun

Homodimeric prodrug nanoassemblies (HDPNs) offer a unique approach to improving the selectivity of chemo-drugs but demonstrate suboptimal tumor targeting efficiency. Cell membrane-camouflaged nanoparticles, particularly those using homologous cancer cell membranes (CM), can achieve excellent tumor accumulation. However, it is challenging to prepare CM-camouflaged HDPNs due to the poor affinity between hydrophobic HDPNs and hydrophilic CM. In addition, it is unclear whether the amount of CM would affect the performance of HDPNs. Herein, the amphiphilic molecule DSPE-PEG 2k is utilized as a “glue” to enhance the affinity between redox-responsive paclitaxel (PTX) HDPNs and CM. The addition of 20% DSPE-PEG 2k is found to effectively improve this affinity. Stable CM-HDPNs are successfully prepared by fine-tuning CM amount to a drug-to-membrane weight ratio of 1:0.25. Benefiting from the homologous targeting, CM-HDPNs demonstrated high tumor accumulation followed by rapid internalization by tumor cells. In response to the redox tumor microenvironment, PTX is rapidly released, exerting efficient antitumor effects without obvious toxic effects. The “glue” strategy will provide new insights for the preparation of CM-camouflaged nano-formulations in the case of target-selective chemotherapy.
