Natural bacteria and bacterial bionics offer significant advantages in the synergistic approach to cancer treatments, while the development of such platforms remains challenging. Herein, bacterial membrane-coated chlorin e6-conjugated bacteriform manganese dioxide platform (BCBM) has been developed for amplifying phototherapeutic efficacy of tumors. The BCBM platform has been prepared by the in-situ growth of manganese dioxide on the surface of silica nanorods and subsequent etching process, photosensitizer conjugation, and bacterial membrane encapsulation. The manganese dioxide and bacterial membrane endow the BCBM with bacteria-like characteristics, enabling a specific response to the tumor microenvironment (TME). The manganese dioxide can react with hydrogen peroxide to produce large amounts of oxygen and manganese ions under acidic conditions. Upon near-infrared irradiation, the BCBM exhibits an excellent ability to generate reactive oxygen species because of sufficient oxygen supply. Furthermore, the released manganese ions activate cGAS-STING pathway, leading to the efficient apoptosis of cancer cells. The process of apoptosis is accompanied by the release of tumor-associated signaling molecules, which enhances therapeutic efficacy. Therefore, this work offers insights into designing biomimetic multifunctional nanoagents against cancers.

AimsTo investigate the effects of the ginsenoside Rh2 on monospecies and multispecies cariogenic biofilms and explore the mechanism of the antibiofilm effect of Rh2 in vitro. Methods and ResultsStreptococcus mutans, Streptococcus sobrinus and Streptococcus sanguinis were chosen to form the monospecies or multispecies biofilms. Crystal violet staining and laser scanning confocal microscopy were used to observe the effect of Rh2 on biofilms in vitro. Cytotoxicity was examined by the Cell Counting Kit‐8. The effects of Rh2 on bacterial membranes were observed via transmission electron microscopy (TEM). The isobaric tags for relative and absolute quantification (iTRAQ) method were used to profile the common differentially expressed proteins. Gene expression was analysed by reverse transcription quantitative polymerase chain reaction. In general, the treatment of cariogenic biofilms with Rh2 significantly decreased biomass accumulation by inhibiting bacterial growth and extracellular polysaccharide synthesis without any cytotoxic effects. TEM imaging showed that Rh2 could disrupt the cell membranes of these bacteria. The iTRAQ results indicated that the levels of mannose‐specific IIC/D and acetaldehyde/alcohol dehydrogenase were substantially down‐regulated, while the mRNA expression of the corresponding genes were significantly changed.ConclusionsOur data revealed a potential application for Rh2 in the protection against dental caries via the inhibition of cariogenic biofilms.Significance and Impact of the StudyThis study describes the first application of a ginsenoside against multispecies cariogenic biofilms. Rh2 may serve as an alternative agent to prevent dental caries by effectively modulating the pathogenic potentials of oral biofilms.

s Ovotransferrin (OVT), displaying multiple bioactivities, is an important egg antimicrobial component. In this study, a novel antimicrobial peptide (OVTp12) was purified from the hydrolysate of OVT by ion-exchange chromatography, Sephadex G-15 gel filtration, and reversed-phase HPLC. The sequence of OVTp12 was identified as AGLAPYKLKPIA using LC-MS/MS with molecular mass of 1240.76 Da. Moreover, OVTp12 showed effective antimicrobial activity against both Gram-positive and Gram-negative bacteria (minimal inhibitory concentrations, MIC values = 32 − 128 μg/mL). The outer/inner cell membrane permeabilization induced by OVTp12 indicated that the isolated peptide exhibited significant membrane permeability of Escherichia coli . To further reveal the antibacterial mechanism, CLSM and flow cytometry analyses demonstrated that OVTp12 could destroy cell membrane integrity and increase the permeability of pathogen's membrane. SEM analyses expressed that OVTp12 caused morphological changes in both Escherichia coli and Staphylococcus aureus . These results suggested that peptide OVTp12 from ovotransferrin may be a promising antimicrobial agent for food preservation or pharmaceuticals.

Bioinspired materials based on self-assembling peptides are promising for tackling various challenges in biomedical engineering. While contemporary data-driven approaches have led to the discovery of self-assembling peptides with various structures and properties, predicting the functionalities of these materials is still challenging. Here we describe the deep learning-guided de novo design of antimicrobial materials based on self-assembling peptides targeting bacterial membranes to address the emerging problem of bacterial drug resistance. Our approach integrates non-natural amino acids for enhanced peptide self-assembly and effectively predicts the functional activity of the self-assembling peptide materials with minimal experimental annotation. The designed self-assembling peptide leader displays excellent in vivo therapeutic efficacy against intestinal bacterial infection in mice. Moreover, it exhibits an enhanced biofilm eradication capability and does not induce acquired drug resistance. Mechanistic studies reveal that the designed peptide can self-assemble on bacterial membranes to form nanofibrous structures for killing multidrug-resistant bacteria. This work thus provides a strategy to discover functional peptide materials by customized design.

Multidrug resistant bacterial infection has become one of the most serious threats to human health. Antimicrobial peptides (AMPs) have been identified as potential alternatives to antibiotics owing to their excellent bactericidal activity. However, the complicated bactericidal mechanism of AMPs is still poorly understood. Fluorescence imaging has many advantages in terms of dynamic monitoring, easy operation, and high sensitivity. In this study, we developed an aggregation-induced emission (AIE)-active probe AMP-2HBT by decorating the antimicrobial peptide HHC36 (KRWWKWWRR) with an AIEgen of 2-(2-hydroxyphenyl)benzothiazole (HBT). This AIE-active probe exhibited an excellent light-up fluorescence after binding with bacteria, enabling a real-time monitoring of the binding process. Moreover, a similar time-dependent bactericidal kinetics was observed for the AIE-active probe and HHC36 peptide, which indicated that the bactericidal activity of the peptide was not compromised by decorating with the AIEgen. The bactericidal mechanism of HHC36 peptide was further investigated by super-resolution fluorescence microscopy, transmission electron microscopy (TEM), and scanning electron microscopy (SEM), which suggested that the probe tended to accumulate on the bacterial membrane and efficiently disrupt the membrane structure to kill both Gram-positive and -negative bacteria. This AIE-active probe thus provided a convenient tool to investigate the bactericidal mechanism of AMPs.

Enterococcus faecalis infections pose a significant clinical challenge due to their multidrug resistance and propensity for biofilm formation. Exploring alternative treatment options, such as repurposing existing drugs, is crucial in addressing this issue. This study investigates the antibacterial activity of candesartan cilexetil against E. faecalis and elucidates its mechanism of action. Candesartan cilexetil exhibited notable antibacterial activity against both E. faecalis and Enterococcus faecium, with minimum inhibitory concentration (MIC) of ≤25 μM. Time-kill curves demonstrated concentration-dependent bactericidal effects. Candesartan cilexetil could significantly inhibited biofilm formation at the concentration of 1/4× MIC and induced alterations in biofilm structure. Permeability assays revealed compromised bacterial membranes, accompanied by the dissipation of membrane potential in E. faecalis cells after treatment with candesartan cilexetil. Checkerboard analysis showed that bacterial membrane phospholipids phosphatidylglycerol and cardiolipin could neutralize the antibacterial activity of candesartan cilexetil in a dose-dependent manner. Biolayer interferometry (BLI) assay indicated specific interactions between candesartan cilexetil and phosphatidylglycerol or cardiolipin. This study demonstrates the promising antibacterial and antibiofilm activities of candesartan cilexetil against multidrug-resistant E. faecalis. The mechanism of action involves disruption of bacterial membranes, possibly by interacting with membrane phospholipids. These findings underscore the potential utility of candesartan cilexetil as an effective therapeutic agent for combating E. faecalis infections, offering a valuable strategy in the battle against antibiotic-resistant pathogens.

Integrating kinds of bactericides as one entirety shows prospect for improving antibacterial efficiency, thus a new type of nanoclusters based on bactericide-directed herein emerged as a combination for achieving a higher antibacterial effect. Specifically, an easily operated approach for preparing bacitracin-directed silver, gold, and copper nanoclusters (AgNCs, AuNCs, and CuNCs, respectively) was proposed. Meanwhile, AgNCs@Bacitracin, AuNCs@Bacitracin, and CuNCs@Bacitracin emitted cyan, yellow, and red colors of fluorescence, respectively, and different sizes and nitrogen contents of these nanoclusters played a critical role for their fluorescence alterations. Significantly, AgNCs@Bacitracin exhibited robust bacteria-killing efficiency than other nanoclusters, owing to its marked damage on the bacterial membrane. Additionally, propidium iodide (PI) staining indicated the bacterial membrane damage of 72.3% to the bacteria population followed by the treatment with AgNCs@Bacitracin, and the introduction of AgNCs@Bacitracin lead to the higher reactive oxygen species (ROS) production, facilitating their attractive antibacterial activity. Moreover, this strategy of developing new nanoclusters broadened the avenues for designing the improved antibacterial materials.

Enveloped viruses such as SARS-CoV-2 frequently have a highly infectious nature and are considered effective natural delivery systems exhibiting high efficiency and specificity. Since simultaneously enhancing the activity and selectivity of lipopeptides is a seemingly unsolvable problem for conventional chemistry and pharmaceutical approaches, we present a biomimetic strategy to construct lipopeptide-based mimics of viral architectures and infections to enhance their antimicrobial efficacy while avoiding side effects. Herein, a surface-nanoengineered antimicrobial liposome (SNAL) is developed with the morphological features of enveloped viruses, including a moderate size range, lipid-based membrane structure, and highly lipopeptide-enriched bilayer surface. The SNAL possesses virus-like infection to bacterial cells, which can mediate high-efficiency and high-selectivity bacteria binding, rapidly attack and invade bacteria via plasma membrane fusion pathway, and induce a local “burst” release of lipopeptide to produce irreversible damage of cell membrane. Remarkably, viral mimics are effective against multiple pathogens with low minimum inhibitory concentrations (1.6–6.3 μg mL −1 ), high bactericidal efficiency of >99% within 2 h, >10-fold enhanced selectivity over free lipopeptide, 99.8% reduction in skin MRSA load after a single treatment, and negligible toxicity. This bioinspired design has significant potential to enhance the therapeutic efficacy of lipopeptides and may create new opportunities for designing next-generation antimicrobials.

The overuse or abuse of antibacterial drugs has led to serious health problems. At present, among various antimicrobial materials (natural, organic, inorganic, etc.), inorganic antimicrobial materials, especially ZnO, have received widespread attention. However, an important factor that influences the function of nanoparticles is agglomeration. The agglomeration-sensitive nature of ZnO compromises its antibacterial properties . Here, talc is used as a substrate to control the size and dispersion of ZnO, forming composite minerals with excellent antibacterial properties against both Escherichia coli and Staphylococcus aureus . According to transmission electron microscopy , ZnO/talc is easily adhered to bacterial cells, whereas atomic force microscopy reveals that antibacterial effects are caused by non-covalent interactions between ZnO/talc and bacterial membranes. The comprehensive investigation of antibacterial performance and interfacial interaction is conducive to our understanding of the antibacterial mechanism of inorganic compounds , and also provides new perspectives on the biological effects of inorganic nanomaterials .

The biological effects of magnetic fields are pervasive in microorganisms, with significant attention given to alternating magnetic fields (AMFs). However, AMFs induce electrical and magnetothermal effects, which complicate the interpretation of magnetic field-induced biological effects and introduce uncertainties regarding cytotoxicity in practical applications. The static magnetic field (SMF) with few variables and high biocompatibility presents a promising alternative for both understanding biological mechanisms and ensuring safe applications, but has a remaining problem on weak interactions with microorganisms. Here we show that the combination of SMF with paramagnetic calcium-polypyrrole nanoparticles (Ca-PPy) remarkably enhances bactericidal activity. Our experiments indicate that the synergistic action of SMF and Ca-PPy significantly promotes the generation of reactive oxygen species (ROS), i.e., singlet oxygen and superoxide anion radicals, in Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), coupled with the physical disruption of bacterial membrane, exhibiting the extraordinary bactericidal performance (the bactericidal rate is over 94%). The mechanism disclosed by computations is that the singlet-to-triplet transition of radical pairs can be increased by the introduction of magnetic fields. These findings offer new insights into the biological effects of magnetic fields and pave the way for their safe, highly effective use in bactericidal applications.

Photodynamic antimicrobial therapy (PDAT) for efficient bacterial infection eradication critically relies on photosensitizers (PSs) that can specifically target and disrupt bacterial membranes. However, the complex membrane architecture of Gram-negative bacteria poses a significant challenge to the efficacy of most aggregation-induced emission (AIE) PSs. Herein, we introduce TPQ, an AIE PS meticulously designed to overcome this challenge by incorporating an outer membrane disruption ability, thereby boosting PDAT efficacy against Gram-negative bacteria. TPQ demonstrated excellent microbial imaging and potent PDAT activity against both Gram-positive and Gram-negative bacteria, attributed to its inherent fluorescence, high singlet oxygen generation, and balanced electrostatic and hydrophobic interactions with bacterial membranes. Notably, TPQ achieved exceptional PDAT activity (>97% efficacy) against Gram-negative bacteria while exhibiting minimal cytotoxicity to mammalian cells. Furthermore, TPQ-mediated PDAT effectively healed Escherichia coli-infected wounds on mice models with assured biosafety. This work provides valuable insights into the rational design of AIE PSs and highlights the synergistic effect of membrane disruption for advancing PDAT applications, particularly against recalcitrant Gram-negative bacterial infections.

The emergency of antibiotic-resistant bacteria and their wide spread has posed a worldwide threat to public health, and traditional antibiotics are gradually overwhelmed by infectious bacteria. Herein, we report an efficient and economical strategy to construct antimicrobial polymers with net cationic component and hyperbranched architecture, which exhibit highly selective toxicity toward bacteria over human cells. To this aim, cationic poly(aminoethyl acrylate) (PAEA) without hydrophobicity is chosen for low hemolysis activity and targeting the negative bacterial membranes, and hyperbranched architecture is introduced to solve the dilemma of low antimicrobial activity. Long-subchain hyperbranched PAEA ( lhb -PAEA) samples kills >99.99% gram-negative Escherichia coli and >98% gram-positive Staphylococcu aureus at the dose of ≤4 μg/mL. Moreover, lhb -PAEA samples exhibit great biocompatibility, for the hemolysis percentage was ≤35% even at the high dose of 1024 μg/mL. Thus, enhanced antimicrobial activity, reduced hemolytic toxicity, the feasible and low-cost production are achieved for lhb -PAEA as antimicrobial agents. © 2016 Wiley Periodicals, Inc. J. Polym.
