Dietary resveratrol supplementation on growth performance, immune function and intestinal barrier function in broilers challenged with lipopolysaccharide

Lei-Zheng Zhang,Jian-Gang Gong,Jia-Hui Li,Yan-Shuang Hao,Hong-Jian Xu,Yan-Ci Liu,Zhi-Hua Feng

ABSTRACT This study discusses the effects of resveratrol ( RES ) on the productive performance, immune function and intestinal barrier function of broiler chickens challenged with lipopolysaccharide ( LPS ). Two hundred and forty 1-day-old male Arbor Acres broilers were randomly divided into 4 groups of 6 replicates each, with 10 broilers per replicate. This experiment used a 2 × 2 factorial design with dietary factors (basal diets or basal diets supplemented with 400 mg/kg RES were administered from d 1 to 21) and stress factors (intraperitoneal injection of 0.5 mg/kg BW of saline or LPS at 16, 18 and 20 d of age). The results showed that LPS challenge had a significant adverse effect on average daily gain ( ADG ) in broilers at 16 to 21 d of age ( P < 0.05), whereas the addition of RES to the diet inhibited the LPS-induced decrease in ADG ( P < 0.05). RES also alleviated LPS-induced immune function damage in broilers, which was manifested by the decrease of spleen index ( P < 0.05) and the recovery of serum immunoglobulin M and ileal secretory immunoglobulin A content ( P < 0.05). The LPS challenge also disrupts intestinal barrier function and inflammation, and RES mitigates these adverse effects in different ways. RES attenuated LPS-induced reduction of villus height in the jejunum and ileum of broilers ( P < 0.05). LPS also caused an abnormal increase in plasma D-lactic acid levels in broilers ( P < 0.05), which was effectively mitigated by RES ( P < 0.05). LPS challenge resulted in a significant decrease in mRNA expression of occludin in the intestinal mucosa ( P < 0.05), which was mitigated by the addition of RES ( P < 0.05). RES significantly decreased the mRNA expression of toll-like receptor 4, nuclear factor kappa-B and tumor necrosis factor alpha in the ileum tissue stimulated by LPS ( P < 0.05). Taken together, this study shows that RES exerts its beneficial effect on broilers challenged with LPS by alleviating immune function damage, relieving intestinal inflammation and barrier damage, and thus improving growth performance.

Effects of Dietary L-Theanine on Growth Performance, Antioxidation, Meat Quality, and Intestinal Microflora in White Feather Broilers With Acute Oxidative Stress

Wang Zixi,Tang Yanfang,Long Lina,Zhang Huihua

In order to reduce the negative effects caused by oxidative stress on broilers, it is particularly important to find ways to alleviate oxidative stress. As a natural plant extract, L-theanine has a variety of biological effects, such as improving antioxidant capacity, promoting growth, and enhancing immunity and antitumor. This trial evaluated the effects of dietary supplementation of L-theanine on growth performance, antioxidation, meat quality, and intestinal microflora in 817 White Feather Broilers. A total of 108 21-day-old 817 broilers with similar body weight (BW) were randomly divided into three groups with six replicates per group and six chickens within each replicate. The three groups were corn-soybean-based diet (NC group); basal diet plus drinking water with 30 mg hydrocortisone/kg (PC group); and basal diet supplemented with 400 mg L-theanine/kg plus drinking water with 30 mg hydrocortisone/kg (LT group). Compared with the NC group, from 21 to 24 days of age, the PC and LT groups had decreased BW, average daily gain (ADG), and average daily feed intake (ADFI), and increased feed to gain ratio (F/G; p < 0.05). At 24 days of age, the LT group had improved superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in serum as compared to the NC group (p < 0.05). The LT group broilers also had significantly higher concentrations of malondialdehyde (MDA) in serum and liver (p < 0.05). On the 42nd days, the PC group had lower PH45min (p < 0.05) than the NC and LT groups and higher cooking loss and shear force (p < 0.05). Moreover, the villi height of the PC group was significantly lower in jejunum than the NC group (p < 0.05). The LT group had a higher ZO-1 content in duodenum than the NC and PC groups (p < 0.05). The activity of GSH-Px in the liver of the LT group was increased than in the PC group (p < 0.05). The relative abundance of Firmicutes in the LT group was significantly higher than in the NC and PC groups (p < 0.05). These results suggested that the effects of acute oxidative stress on growth performance and meat quality of broilers are continuous, and dietary supplementation of L-theanine could improve the growth performance and meat quality, enhance the intestinal mucosal barrier and antioxidant capacity, and improve the composition of the intestinal flora of broilers caused by acute oxidative stress.

Reshaping fish intestinal microbiota and facilitating barrier function by ZnO nanoparticles

Shuoli Ma,Wen-Xiong Wang

Nano-Zn (nZnO) has been increasingly applied in different fields, but the mechanisms of their uses as nutritional sources are less understood. Our study presents a regulatory mechanism of intestinal microbiota and structure mediated by nano-Zn, which subsequently improves the health and growth of fish. Fish diets were supplemented with 60 mg Zn kg−1 ZnSO4 (metal salt as the control group), 60 mg Zn kg−1 nZnO (powder), or 170 mg Zn kg−1 nZnO (powder) and then fed to golden pompanos for 5 weeks. We employed a new analytical amplicon sequence variant (ASV) method to identify the bacterial species and achieved the first in situ spatial quantification of fish tissue samples. Our results indicated that 60 mg Zn kg−1 of dietary nZnO promoted the probiotic colonization (Lactic-acid bacilli spp.) in the intestine. Upon exposure to a higher nZnO dose (170 mg Zn kg−1), Brevinema and Mycoplasma colonized in the fish intestine as pro-inflammatory microorganisms, which activated the C5 isoprenoid biosynthesis pathway and NAD biosynthesis. Furthermore, dietary nZnO (60 mg Zn kg−1) up-regulated the expression of genes related to intestinal mucus secretion and accelerated the goblet cell renewal in the intestinal villi. At a dose of 170 mg Zn kg−1 nZnO, the fish intestine significantly down-regulated occludin and mucin gene expression while displaying lower villus height, villus area, and goblet cell numbers, which ultimately led to abnormal digestion and absorption and immune functions. The Mantel test confirmed that the intestinal microbial abundance was significantly correlated with the changes in the intestinal structure, and the value of villi-to-crypt ratio decreased with the increase in Vibrio abundance, which was validated by the key genes and serum biochemical indexes. Finally, nZnO could regulate the glucose and lipid metabolism of fish by regulating microorganisms. Our results provided important insights into the interaction between nZnO and fish intestinal microbial community and host.

Comparative chronic toxicities of PFOS and its novel alternatives on the immune system associated with intestinal microbiota dysbiosis in adult zebrafish

Jing Huang,Qiyu Wang,Shuai Liu,Hong Lai,Wenqing Tu

6:2 Chlorinated polyfluorinated ether sulfonate (F-53B) and sodium p -perfluorous nonenoxybenzene sulfonate (OBS) are widely used as perfluorooctane sulfonate (PFOS) alternatives in the Chinese market. Here, adult zebrafish were chronically exposed to 1 μM PFOS, F-53B, and OBS for 21 days to investigate the comparative immunotoxicity of these three per- and polyfluoroalkyl substances (PFAS). PFOS induced more severe oxidative stress in the liver than F-53B and OBS, and these three PFAS induced similar anti-inflammatory effects by repressing the expression of pro-inflammatory cytokines. The intestinal microbiota analysis showed that the relative abundance of Plesiomonas , Aeromonas , Cetobacterium , Shewanella , and Vibrio changed with the same trend in the three PFAS treatment groups. Furthermore, the PFAS increased the expression of hepcidin , muc , the immune-related genes mpo and saa , and decreased the expression of the tight junction-related gene occ in the intestine; moreover, villus height of the intestine was reduced after PFAS exposure, which indicated the functional disruption of the intestine. In particular, the significant correlation between the changed intestinal microbiota and liver and intestinal indicators also suggested the interaction between the immune system and intestinal microbiota. Taken together, our results indicate that exposure to PFOS and its alternatives F-53B and OBS can induce hepatic immunotoxicity associated with intestinal microbiota dysbiosis in adult zebrafish.

Effects of High-Dose Genistein on the Hypothalamic RNA Profile and Intestinal Health of Female Chicks

Zengpeng Lv,Chenhui Hu,Jingle Jiang,Song Jin,Quanwei Wei,Xihui Wei,Debing Yu,Fangxiong Shi

Genistein is abundant in animal feed. In this study, the side effects of high-dose genistein on intestinal health and hypothalamic RNA profile were evaluated. Chicks exposed to high-dose genistein by intraperitoneal injection (416 ± 21, 34.5 ± 2.5) and feed supplementation (308 ± 19, 27.2 ± 2.1) both showed a reduced body weight gain and feed intake in comparison with the control group (261 ± 16, 22.7 ± 1.6, P < 0.01). In comparison with the control (22.4 ± 0.5, 33.3 ± 2.4), serum levels of albumin and total protein were decreased after high-dose genistein injection (21.6 ± 0.5, 31.8 ± 1.6) and diet supplementation (20.6 ± 0.9, 29.9 ± 2.5, P < 0.001). Interestingly, the genistein diet presented the chick hypothalamus with downregulated expression of bitter receptors (TAS1R3, P < 0.05). Meanwhile, it upregulated the expressions of TAS2R1 (P < 0.05) and downstream genes (PLCB2 and IP3R3) in the ileum (P < 0.05). Accordingly, high-dose dietary genistein reduced villus height and the abundance of Lactobacillus, along with the increased abundance of pathogenic bacteria in the ileum (P < 0.05). Furthermore, transcriptomic analysis identified 348 differently expressed genes (168 upregulated and 224 downregulated) in the high-dose dietary genistein treated group in comparison with the control (P < 0.05, |log2FoldChange| > 0.585). Therefore, high-dose dietary genistein altered the hypothalamic RNA profile and signal processing. Cluster analysis further revealed that high-dose dietary genistein significantly influenced apoptosis, the immune process, and the whole synthesis of steroid hormones in the hypothalamus (P < 0.05). In conclusion, high-dose dietary genistein altered the hypothalamic RNA profile and intestinal health of female chicks.

Effects of Dietary Astragalus Polysaccharide Supplementation on the Th17/Treg Balance and the Gut Microbiota of Broiler Chickens Challenged With Necrotic Enteritis

Bochen Song,Peng Li,Shaojia Yan,Yan Liu,Mingkun Gao,Huiyuan Lv,Zengpeng Lv,Yuming Guo

This study aimed to investigate the effects of dietary astragalus polysaccharide (APS) supplementation on the immune function, gut microbiota and metabolism of broiler chickens challenged with necrotic enteritis (NE). Two hundred forty Arbor Acres broiler chicks (one day old) were randomly assigned using a 2 × 2 factorial arrangement into two groups fed different levels of dietary APS (0 or 200 ppm of diet) and two disease challenge groups (control or NE challenged). The results showed that NE infection significantly increased FCR, mortality rate, Th17/Treg (Th17 cells% in blood and ileum, Th17/Treg, IL-17 and IL-17/IL-10 in blood), NO, lysozyme activity and IL-1β in blood, intestinal immune cell proportion and activity (Tc%, Treg% and monocyte phagocytic activity in ileum), intestinal inflammatory cytokines (TLR2, NF-κB, TNF-α and IL- 6) gene expression levels, and the number of Clostridium perfringens in cecum. NE infection significantly reduced body weight gain, thymus index, lymphocyte proliferation activity in blood and ileum, villus height and V/C in jejunum, Th cells% and Mucin2 gene expression in ileum. Dietary APS supplementation significantly increased body weight, feed intake, proportion of immune cells (T cells in blood and Tc, Treg in ileum), lymphocyte proliferation activity, V/C in jejunum, and ZO-1 gene expression in ileum. Dietary APS supplementation significantly reduced FCR and mortality rate, Th17/Treg, Th17%, intestinal pathology scores, intestinal inflammatory cytokine gene expression levels, and the number of Clostridium perfringens in cecum. In addition, broilers challenged with NE significantly increased Staphylococcus and Turicibacte r and reduced α diversity of microbiota in ileum. Dietary APS supplementation significantly increased α diversity, Romboutsia , Halomonas , propionic acid, butyric acid, formononetin, taurine, cholic acid and equol and downregulated uric acid, L-arginine and serotonin in ileum. Spearman’s correlation analysis revealed that Romboutsia, Turicibacter, Staphylocpccus, Halomonas, Streptococcus, Escherichia-Shigella, Prevotella , uric acid, L-arginine, jerivne, sodium cholate and cholic acid were related to inflammation and Th17/Treg balance. In conclusion, APS alleviated intestinal inflammation in broilers challenged with NE probably by regulating intestinal immune, Th17/Treg balance, as well as intestinal microbiota and metabolites.

Microbiomic and metabonomic analysis provide new insights into the enhanced intestinal health in large-size largemouth bass (Micropterus salmoides) when fed novel proteins: Novel proteins are promising foods for future aquaculture

Lukuan Li,Yu Wang,Zhen Zhang,Chunfang Wang

The novel proteins, i.e., Clostridium autoethanogenum (CAP), Tenebrio molitor (TM), Chlorella vulgaris (ChM), and cottonseed protein concentrate (CSM), are promising food for the future aquaculture due to the limited fishmeal (FM) resources. However, in-depth investigations on the intestinal barriers of fish when fed novel proteins are sparse, and the association between intestinal flora and physiological biomarkers is still unknown. Five isonitrogenous and isolipidic diets comprising various proteins were developed, with fishmeal serving as the control and novel proteins as the experimental diets. Largemouth bass ( Micropterus salmoides ) with an initial body weight of 211.9 ± 1.2 g were used as experimental model and fed with these five diets for eight weeks. The findings from the current study indicated that the intestinal histological structure as well as immunochemistry barrier performance of the ChM, CAP, and TM groups were superior to those of the CSM and FM groups, despite the fact that the villi height of FM was the greatest of all groups. Similarly, the intestinal inflammatory response and the permeability were elevated in CSM while decreased in ChM, CAP, and TM group. Concerning the intestinal short fatty acids (SCFAs), the levels in the CAP as well as ChM group were greater than the other proteins. From the data analysis of 16S rRNA gene sequencing, the abundance of opportunistic pathogens including (Ralstonia , Acinetobacter , Aeromonas , and Protebacteria) were higher in CSM group while attenuated in ChM, CAP and TM group. In addition, the abundance of nine types of intestinal-barriers-repairing probiotics were elevated in all novel proteins compared with the fishmeal group. These altered microbiota were highly correlated with intestinal inflammatory response, antioxidant status, immunochemistry barrier, physical barrier and SCFAs indexes by Mantel test and Pearson's correlation analysis. Collectively, the ranking of the beneficial functions among these five proteins was ChM > CAP>TM > FM > CSM according to Z -score and this study provides in-depth data manifesting the effects of novel proteins on the gut health status of large-size largemouth bass.
