Hydrogel-derived implants have proven value in bone tissue regeneration, and current efforts have concentrated on devising strategies for producing functional implants with desired structures and functions to improve therapeutic outcomes. Herein, a novel black phosphorus (BP) tagged responsive strontium (Sr) hydrogel particles are presented for bone defect repair. By applying microfluidic technology, Sr and carboxymethyl chitosan, and BP are integrated into poly(N-isopropyl acrylamide) (pNIPAM) hydrogel matrix to generate such microparticles called pNBCSMs. Upon exposure to near-infrared irradiation, the pNBCSMs experience volume shrinkage and provoke the extrusion of the incorporated Sr, ascribed to the photothermal conversion ability of BP and the thermosensitivity of pNIPAM. In vitro and in vivo experimental results reveal that pNBCSMs subjected to near-infrared light display superior anti-inflammatory, anti-apoptotic, bacterial inhibitory, as well as osteogenesis-promoting effects, thereby effectively improving defective cranial bone repair. These features suggest that the proposed pNBCSMs can be promising candidates for bone repair.

(1) Background: Osteoarthritis (OA) is a crippling condition characterized by chondrocyte dedifferentiation, cartilage degradation, and subsequent cartilage defects. Unfortunately, there is a lack of effective medicines to facilitate the repair of cartilage defects in OA patients. In this study, we investigated the role of lncRNA NEAT1_2 in maintaining the chondrocyte phenotype and identified tanshinone IIA(TAN) as a natural medicine that enhances NEAT1_2 levels, resulting in efficient cartilage regeneration under inflammatory cytokines. (2) Methods: The transcriptional levels of NEAT1_2 and cartilage phenotype-related genes were identified by RT-qPCR. The siRNA interference approach was utilized to silence NEAT1_2; the Alamar Blue assay was performed to determine chondrocyte viability under inflammatory conditions. To evaluate the concentrations of collagen type II and glycosaminoglycans distributed by chondrocytes in vitro and in vivo, immunohistochemical staining and Safranin O staining were used. (3) Results: IL-1β suppresses NEAT1_2 and genes related to the chondrocytic phenotype, whereas TAN effectively upregulates them in a NEAT1_2-dependent manner. Consistently, TAN alleviated chondrocyte oxidative stress inhibited cartilage degradation by modulating the relevant genes and promoted efficient cartilage regeneration in vitro and in vivo when chondrocytes are exposed to inflammatory cytokines. (4) Conclusions: TAN enhances the expression of NEAT1_2 inhibited by IL-1β and affects the transcription of chondrocytic phenotype-related genes, which promotes cartilage regeneration in an inflammatory environment. Keywords:osteoarthritis;tanshinone IIA;NEAT1_2;cartilage defects repair;inflammatory

Bone tumors may lead to osteoporosis. It is of great significance to fabricate functional scaffolds for treating bone tumor, osteoporosis and repairing bone defects. In this study, LaB 6 micro-nanoparticles/poly( d , l -lactide)-modified β-tricalcium phosphate scaffolds (TCP-PDLLA-LB) were successfully prepared. The LaB 6 surface chemistry-reinforced TCP-PDLLA-LB scaffolds were endowed with the following excellent properties: (1) the enhanced mechanical strength. The TCP-PDLLA-5LB scaffolds possessed higher compression resistant capacity than TCP scaffolds owing to the strong adhesive property of PDLLA that bound the LaB 6 micro-nanoparticles and scaffolds tightly; (2) the excellent photothermal performance. TCP-PDLLA-LB scaffolds presented distinct photothermal property because of localized surface plasmon resonance effect of LaB 6 micro-nanoparticles when exposed to the near-infrared (NIR) light (wavelength: 808 nm) and it was flexibly controllable through changing the quantity of LaB 6 micro-nanoparticles, power density of NIR light and environmental humidity; (3) the excellent biological properties in vitro . When culturing with saos-2 bone tumor cells, the TCP-PDLLA-5LB scaffolds effectively killed the saos-2 cells through adjusting power density of NIR light, irradiation duration time of NIR light and irradiation times of NIR light. Additionally, the TCP-PDLLA-5LB scaffolds were able to release bioactive ions which were beneficial to the in vitro osteogenesis of rabbit bone marrow stromal cells (rBMSCs) by supporting the attachment and proliferation of rBMSCs as well as promoting the expressions of osteogenic genes BMP2, RUNX2 and COL 1; and (4) the abilities of effectively ablating bone tumors and supporting bone tissue regeneration in vivo . The bone tumor tissues were significantly ablated and suppressed through hyperthermia due to the existence of LaB 6 micro-nanoparticles on the scaffolds. Simultaneously, TCP-PDLLA-5LB scaffolds implanted in bone defects effectively helped new bone formation regardless of NIR light irradiation. In conclusion, LaB 6 surface chemistry-reinforced scaffolds possess the dual functions of ablating bone tumor and repairing bone defects, which offers a promising therapy strategy for tumor/disease-related bone tissue engineering.

Background The repair of bone defects has always been a significant challenge in clinical medicine. To address this challenge, doctors often utilize autologous bone grafts, allogeneic bone grafts and artificial bone substitutes. However, the former two methods may result in additional trauma and complications, while allogeneic bone grafts carry the risks of immune rejection and disease transmission. Magnesium phosphate cement (MPC), as a artificial bone substitutes, has been a potential biomaterial for repairing bone defects, but its clinical application is limited by insufficient mechanical strength and poor osteoinductive activity. Methods In this study, the cement liquid phase base on rhBMP-2 and chitosan solution into MPC were obtained and investigated. After mixing with a cement liquid, the structural and phase composition, morphology, chemical structure, setting time, compressive strength, degradation behavior, solubility, and cellular responses and bone regeneration in response to CHI-rhBMP2 MPC were investigated in vitro and in vivo. Results After the chemical component modification, CHI-rhBMP2 MPC possessed controllable degradation rate, moderate setting time, appropriate cuing temperature, good injectability, and improved initial strength. In vitro tests showed that the CHIrhBMP2 MPC could promote cell proliferation and adhesion, as well as that contribute to osteoblast differentiation and mineralization. In addition, cement materials were implanted into the rabbit femoral condyles for in vivo osseointegration evaluation. The results displayed that more new bone grew around CHI-rhBMP2 MPC, verifying improved osseointegration capacity. Transcriptome analysis revealed that focal adhesion, Forkhead box O(FoxO) signaling pathway and P13K/AKT signaling pathway were may involved in CHI-rhBMP2 MPC induced new bone formation. Conclusion This work provides a new strategy for the rational design of potential bone repair candidate materials.

The management of bone defects, particularly those with irregular geometries resulting from osteoporotic fractures, remains fraught with challenges. Microspheres have emerged as a promising vehicle for tissue engineering, distinguished by their controlled release, safety, and ease of application. Various bioactive components are integrated into microspheres to improve their performance. Metal-organic frameworks, formed from metal ions interconnected by organic ligands, are increasingly utilized in tissue engineering. Specifically, magnesium-based MOFs are notable for their broad applicability; Mg 2+ ions are instrumental in bone reconstruction and repair, facilitating osteogenesis, angiogenesis, antibacterial effects, and anti-inflammatory properties. Mg-MOF was synthesized using magnesium chloride and gallic acid, and it was incorporated into gelatin microspheres to create Gel@Mg-MOF composite microspheres. Leveraging gelatin's biocompatibility, controlled release, and biodegradability, the composites' biocompatibility was evaluated through toxicity and adhesion assays. Moreover, the osteogenic and angiogenic potentials of the Gel@Mg-MOF microspheres were assessed, alongside their capacity for ROS scavenging. Results suggest that controlled Mg 2+ release from Gel@Mg-MOF microspheres promotes osteogenic activity in RBMSCs and enhances angiogenic potential in HUVECs. Additionally, the gallic acid-containing composite microspheres exhibited antioxidative properties. Collectively, the findings suggest that Gel@Mg-MOF microspheres could provide effective support for bone defect repair, with potential for clinical deployment.

Severe trauma not only damages the skin, but also affects the bones, causing osteoarthritis or bone defects. Traditional treatment strategies (such as sutures, metal stents, etc. ) have problems with the need for a second surgery and difficulty in degradation, which cannot meet the treatment expectations of clinical doctors well. As the novel biomedical material, hydrogels have great potential in the treatment of bone defects due to the advantages of seamless filling, degradability and easy operation. Here, we designed a composite hydrogel (Gel-Gym/HS-Dopa) composed of glycidyl methacrylate-modified gelatin (Gel-Gym) and dopamine-functionalized chondroitin sulfate (HS-Dopa). The hydrogel exhibited good tissue adhesion, self-healing, biodegradability, anti-inflammatory and biocompatibility. In animal experiments, Gel-Gym/HS-Dopa hydrogel could significantly promote the rat skin wound healing and bone defect reconstruction in vivo . Therefore, the hydrogel was promising as a potential deep trauma dressing.

Cranial defect repair remains a significant challenge in neurosurgery, and designing material complexes that can support bone regeneration while minimizing complications such as infection and inflammation could help alleviate this clinical challenge. This study presents a photothermal hydrogel complex with a controlled rapid gelation process, PDA-G-A-H, which integrates photothermal polydopamine nanoparticles (PDA NPs) with gentamycin (G) and alendronate acid (A). Furthermore, the incorporation of the injectable hydrogel Pluronic F127 and collagen (H) made this composite hydrogel (PDA-G-A-H) suitable for the multifaceted needs of cranial defects. The PDA-G-A-H hydrogel exhibited superior biocompatibility, as evidenced by high cell viability and minimal hemolysis, making it a safe candidate for biomedical applications. In vitro assessments with MC3T3-E1 cells demonstrated that this hydrogel enhanced mineralization and osteogenic differentiation, and significant upregulation of key osteogenic markers was subsequently detected. The antibacterial activity of the hydrogel against Staphylococcus aureus and Staphylococcus epidermidis was also investigated. The results of the RT‒PCR analysis revealed the potential for inhibiting biofilm formation. The hydrogel composite combines biocompatibility, osteoinductive, and antibacterial potential. It has translational potential for cranial defect repair and other bone regeneration therapies.

Bone defects caused by trauma, tumor resection, and surgery present significant clinical challenges, often resulting in complications such as delayed union, nonunion, and even long-term functional impairment. Current treatments, including autografts and allografts, are limited by donor site morbidity, immune rejection, and pathogen transmission, highlighting the need for developing reliable synthetic alternatives. To address these challenges, we report a binary composite hydrogel combining gelatin methacryloyl (GelMA) and κ-Carrageenan, reinforced with calcium phosphate cements (CPC). GelMA ensures rapid gelation and biocompatibility, κ-carrageenan improves injectability, and CPC enhances mechanical strength and osteogenic activity, collectively creating a robust and versatile hydrogel system. Furthermore, the hydrogel’s injectable, adaptive, and self-healing characteristics enable it to conform to irregular bone defect sites, providing mechanical support and osteogenic stimulation. It also releases bioactive components to accelerate bone regeneration. With exceptional toughness and resilience, this hydrogel recovers its shape after deformation, positioning it as a promising candidate for clinical bone defect repair applications.

Despite the rapid development of the application of tissue engineering techniques in regenerative bone repair, the repair of severe pathologic bone defects, represented by osteoporotic bone defects, is still a huge challenge. Recent studies have identified local delivery of parathyroid hormone relative peptides (PTHrPs) as a novel pathway for bone repair as well, but how to load and locally release PTHrPs for good bone repair is a major obstacle. Here, a biopolymer of PLGA-PLL loaded with PTHrP1 is electrospunned into nanofiber membranes for guided bone regeneration. In vitro experiments show that PLGA-PLL synergized with PTHrP1 significantly promote osteogenic differentiation and angiogenesis, providing a favorable bone regeneration microenvironment. In vivo experiments show that the membrane recruits endogenous stem cells and promotes their ectopic osteogenic differentiation in an osteoporotic environment. Furthermore, the membrane can promote osteogenic differentiation and angiogenesis in situ and improve the local osteoporotic microenvironment, ultimately promoting the repair of bone defects. Therefore, present study provides a promising strategy for local delivery of PTH, which opens a new pathway for the treatment of osteoporotic bone defects.

Managing large, critical-sized bone defects poses a complex challenge, especially when autografts are impractical due to their size and limited availability. In such situations, the development of synthetic bone implants becomes crucial. These implants can be carefully designed and manufactured as potential bone substitutes, offering controlled parameters such as porosity, hardness, and osteogenic cues. In this study, we employed digital light processing (DLP) technology to construct an alumina ceramic scaffold featuring a triply periodic minimal surface (TPMS) structure for bone transplantation. The scaffold was filled with type I collagen to enhance cell infiltration [1], thereby increasing the total surface area. In addition, type I collagen is a carrier for both bone morphogenetic protein-2 (BMP-2) and zoledronic acid (ZA). Using a clinically relevant rabbit cranium defect model, the scaffold underwent in vivo assessment for its functionality in repairing critical-sized bone defect (approximately 8 mm). Four groups of animal experiments were carried out including the control group, the gyroid scaffold group, the type I collagen-loaded scaffold group, and the bioactive factor-functionalized scaffold group. Our animal-based study results revealed that the gyroid scaffold, functionalized with bioactive molecules, provided a conductive surface for promoting increased bone formation and enhancing the healing process in critical-sized long bone and cranium defects. These findings offer preclinical evidence, supporting the use of a TPMS structure composite scaffold and present compelling support for its application as an advanced synthetic bone substitute in the future.

The light-curable composite of siloxane/hydroxyapatite (HA) had been successfully achieved by photopolymerization of a kind of gel combined tetraethoxyorthosilicate (TEOS) and 3-methacryloxypropyltrimethoxysilane (MAPS) with siloxane-modified nano-HA (HA- g -Si). HA- g -Si was prepared by grafting reaction of 3-isocyanatopropyltrimethoxysilane (IPS) onto the surface of HA. Then, TEOS and MAPS were mixed as precursor, and photoinitiator 2-hydroxy-4′-(2-hydroxyethoxy)-2-methylpropiophenone (Irgacure 2959) and as-prepared HA- g -Si were added into the precursor to form a homogenous and sticky gel. The movable gel that could be cured with UV light resulted in various sharp immovable composites of siloxane/HA in different mold. The HA, HA- g -Si nanoparticles, and composites were analyzed by Fourier transformed infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, thermogravimetric analysis, transmission electron microscopy, scanning electron microscopy, mechanical testing, and water contact angles. It could be a promising bone repairing material for orthopedic application. POLYM. COMPOS., © 2011 Society of Plastics Engineers.

Point-wise suturing is the standard method for ensuring that patches effectively perform mechanically supportive functions in tissue repair. However, stress concentrations around suture holes can compromise the mechanical stability of patches. In this study, we develop a suturable hydrogel patch with flaw-tolerance capabilities by leveraging multiscale stress deconcentration, inspired by natural silk. This design mitigates stress concentration across two scales through the synergistic integration of nanoscale high-energy crystalline domains and intermolecular interactions. The resulting integral hydrogel patch exhibits superior flaw resistance compared to conventional patches and effectively addresses tissue adhesion issues. To validate the efficacy of the patch, we demonstrate successful in vivo repair of abdominal wall defects in rats, comparing the performance of the proposed patch to commercial mesh patches (Prolene). The integral patch design strategy present here offers a valuable approach for developing patches that can be tailored to meet the mechanical support needs of various tissue repair applications.
