Objective Sargassum fusiforme (SF), a brown algae, is a marine traditional Chinese medicine with nutritional value. However, studies on the bioactive components in SF are limited. The aim of this study is to investigate phytochemical and biological properties of SF from different geographical origins and botanical parts, as well as to explore bioactive components within SF. Methods SF from different origins and botanical parts were collected for integrated phytochemical analyses using ultra-high performance liquid chromatography-tandem quadrupole time of flight mass spectrometry (UPLC-Q-TOF MS), mass spectral molecular networking and liquid chromatography-single quadrupole mass spectrometry (LC-MSD), etc., followed by the systematic chemical profile evaluation. Subsequently, the glycolipid metabolism regulation and anticoagulation of SF were examined. Furthermore, the overall information acquired was assembled using multivariate analysis and machine learning to discover bioactive components. Results A total of 123 compounds in SF were identified with the assistance of mass spectral molecular networking. The variations in the content of amino acids, fucoidan, alginate, phlorotannin, alkaloids, terpenoids, etc. were observed in SF from different origins and botanical parts. SF possessed the hypolipidemic, hypoglycemic and anticoagulant effects, and SF from Dongtou, Wenzhou, Zhejiang, was outstanding among origins in both phytochemical and biological evaluations. A heterogeneous information ensemble approach was adopted to discover the bioactive components in SF, namely 3-indoleacrylic acid, phlorotannin and fucoidan. Conclusion This study evaluated the phytochemical and biological properties of SF, revealed the bioactive components using heterogeneous information and provided a reference for the rational consumption and further development of SF.

Macroalgae Laminaria japonica (MLJ) has been reported to exhibit various biological activities including improving immunity, anti-aging, anti-tumor, anti-atherosclerosis and anti-diabetic, but the protective mechanisms of MLJ consumption against non-alcoholic fatty liver disease (NAFLD) associated with hyperlipidemia remain poorly understood. This study demonstrated that MLJ consumption prevented high-fat diet (HFD)-induced NAFLD associated with hyperlipidemia in a rat model, and improved hyperlipidemia-related parameters, e.g. serum and hepatic lipid profiles. Moreover, histological analysis showed that MLJ reduced lipid deposition in adipocytes and hepatocytes compared with the HFD group. Such beneficial effects may be associated with the modulation of the intestinal microbiota, especially some key microbial phylotypes involved in lipid metabolism homeostasis. The underlying protective mechanisms of MLJ consumption against HFD-induced NAFLD associated with hyperlipidemia were also studied by ultra-high performance liquid chromatography with quadruple-time of flight mass spectrometry (UPLC-QTOF/MS)-based liver metabolomics coupled with pathway analysis. The metabolic pathway enrichment analysis of the differentially abundant hepatic metabolites indicated that primary bile acid biosynthesis metabolism and cysteine and methionine metabolism were the two main metabolic pathways altered by MLJ consumption when compared with the model group. The analysis of the transcription levels of liver-related genes by RT-qPCR and the expressions of liver-related proteins by immunohistochemistry (IHC) showed that MLJ consumption could regulate the levels of mRNA transcription and protein expression related to hepatic lipid metabolism. In short, this study indicates that MLJ could be developed as functional food supplement for the prevention or treatment of NAFLD associated with hyperlipidemia.

Background Laminaria japonica polysaccharide (LJP), a fucose enriched sulfated polysaccharide has been demonstrated to have excellent anticoagulant and antithrombotic activities. However, the antithrombotic effect of low molecular weight polysaccharide from enzymatically modified of LJP (LMWEP) remains unknown. Material/Methods LMWEP was prepared by fucoidanase enzymatic hydrolysis, and the antithrombotic and anticoagulant activities, and the underlying mechanism were investigated thoroughly. Rats were randomly divided into 6 groups (8 rats in each group): the blank control group, the blank control group treated with LMWEP (20 mg/kg), the model group, the model group treated with heparin (2 mg/kg), the model group treated with LJP (20 mg/kg), and the model group treated with LMWEP (20 mg/kg). After 7 days of intravenous administration, blood was collected for biochemical parameters examinations. Results LMWEP increased the activated partial thromboplastin time (APTT), thrombin time (TT), prothrombin time (PT), 6-keto prostaglandin F1α (6-Keto-PGF1α), and endothelial nitric oxide synthase (eNOS). In addition, LMWEP decreased fibrinogen (FIB), endothelin-1 (ET-1), thromboxane B2 (TXB2), erythrocyte sedimentation rate (ESR), and hematocrit (HCT). Conclusions LMWEP, an enzymatically modified fragment with a molecular weight of 25.8 kDa, is a potential antithrombotic candidate for treatment of thrombosis related diseases.
