Application of a new packaging material in the postharvest quality and spoilage microorganisms bacteria of morels

Kai Jiang,Yiwei Huang,Rui Cui,Yuyue Qin,Thanapop Soteyome,Margaret Brennan,Charles Brennan

In this study, we screened out the dominant spoilage microorganisms during the cold storage of morels, prepared composite paper with a targeted selection of antimicrobial active substances, applied the composite paper to morels for cold storage and preservation, and investigated the changes of physicochemical indexes, flavor-presenting nucleotides and enzyme activities throughout the process of storage, and evaluated the freshness preservation effect. The pathogenic microorganisms during the natural spoilage of Morels were isolated and purified at 4 °C. The dominant spoilage microorganisms obtained were: bacterium B1 ( Lelliia amnigena ), fungi F2 ( Paecilomyces penicillatus ) and F3 ( Mucor hiemalis ). Inhibition experiments were carried out by selecting various active antimicrobial agents—lemon essential oil, clove essential oil and nisin (NI)—against the dominant spoilage microorganisms B1, F2 and F3. Comparing the inhibitory effects, NI was selected as an inhibitor for the dominant spoilage microorganisms following comprehensive consideration. The antimicrobial system (NI/C-CNC) was prepared by incorporating NI into carboxylated cellulose nanocrystals (C-CNC), and it exhibited a strong blocking effect on ultraviolet rays, which became more pronounced with the increase of NI addition. The composite paper NI-C was prepared using the NI/C-CNC antimicrobial system and absorbent paper (100 % virgin wood pulp), which showed better mechanical properties and reduced water vapor transmission rate (WVP). NI-C composite paper was applied to the preservation of morels, which effectively inhibited the dominant pathogenic bacteria. The flavor 5′-nucleotides were effectively protected, and it significantly slowed down the change in hardness during storage of morels. It also inhibited lipid peroxidation, as evidenced by the accumulation of malondialdehyde (MAD) content and reactive oxygen species (ROS), and had a positive effect on the control of enzyme activity. NI-C antimicrobial composite paper has good application prospects in the field of packaging of morels.

Metabolomic profiles and differential metabolites in the fruiting bodies of Morchella spp. infected with white mold at different degrees

Huijuan Liu,Lin Ma,Zhifei Cheng,Shuyu Yang,Jiao Xie

White mold is one of the most common and serious fungal diseases of morel affecting the entire growth cycle. Studies show that when infected, plants defend against pathogens by regulating metabolic responses within their systems. A total of 310 metabolites for infecting Paecilomyces penicillatus in morel mushrooms were identified by UPLC-MS/MS . Most metabolites showed an intuitive downward trend during infection and reached their lowest levels at the third stage. For PCA and HCA, infection period had a significant effect on metabolites, and the second stage was the key turning point for metabolite accumulation in response to disease. Correlation analysis of the top 50 differential metabolites with the highest VIP values screened by OPLS-DA suggested that lipids, nucleotides and their derivatives, sugars, organic acids, phenolic acids and alkaloids may respond synergistically to disease during infection. Taken together, this study provided an entry point for studying white mold pathogenesis in morel.

Antioxidative and Protective Effect of Morchella esculenta against Dextran Sulfate Sodium-Induced Alterations in Liver

Shutong Chen,Min Wang,Suresh Veeraperumal,Bo Teng,Rui Li,Zhengming Qian,Jianping Chen,Saiyi Zhong,Kit-Leong Cheong

Morchella esculentais an edible mushroom with special flavor and high nutritional value for humans, primarily owing to its polysaccharide constituents.M. esculentapolysaccharides (MEPs) possess remarkable pharmaceutical properties, including antioxidant, anti-inflammatory, immunomodulatory, and anti-atherogenic activities. The aim of this study was to evaluate the in vitro and in vivo antioxidant potential of MEPs. In vitro activity was determined using free radical scavenging assays, whereas in vivo activity was evaluated through dextran sodium sulfate (DSS)-induced liver injury in mice with acute colitis. MEPs effectively scavenged 1,1-diphenyl-2-picrylhydrazyl and 2,2-azinobis-6-(3-ethylbenzothiazoline sulfonic acid) free radicals in a dose-dependent manner. Additionally, DSS-induced mice showed severe liver damage, cellular infiltration, tissue necrosis, and decreased antioxidant capacity. In contrast, intragastric administration of MEPs showed hepatoprotective effects against DSS-induced liver injury. MEPs remarkably elevated the expression levels of superoxide dismutase, glutathione peroxidase, and catalase. Additionally, it decreased malondialdehyde and myeloperoxidase levels in the liver. These results indicate that the protective effects of MEP against DSS-induced hepatic injury could rely on its ability to reduce oxidative stress, suppress inflammatory responses, and improve antioxidant enzyme activity in the liver. Therefore, MEPs could be explored as potential natural antioxidant agents in medicine or as functional foods to prevent liver injury. Keywords:Morchella esculenta;polysaccharide;antioxidant;ulcerative colitis;oxidative stress

From byproduct to taste enhancer: Effective preparation of γ-glutamyl peptides from Morchella esculenta stipes and their mechanism of taste enhancement

Xiaoli Gong,Qiuyan Zhang,Na Zhang,Yuhao Zhang,Yu Fu

Long-term consumption of high-salt diets can lead to an increased risk of cardiovascular diseases. γ-Glutamyl peptides (GGP) display a taste-enhancing effect, which contributes to salt reduction. However, preparation of GGP in a sustainable fashion is challenging. Morchella esculenta stipe is a byproduct abundant in Glu/Gln, serving as a potential precursor of GGP. In this study, Morchella esculenta stipe was employed to prepare GGP by transpeptidation reaction. The reaction conditions were optimized, and their taste-enhancing effects were evaluated by sensory and electronic tongue analyses. Several novel GGP were identified by LC-MS/MS, and their binding modes with CaSR were analyzed by molecular docking. It was found that the optimal conditions were enzymatic hydrolysis for 5 h, transpeptidation time for 2 h, and enzyme of 12 U/g. GGP displayed a significant taste-enhancing effect based on sensory and electronic tongue analyses. Several novel di-, tri-, and oligo-GGPs were found. Molecular docking revealed that GGP can bind to CaSR through hydrogen bonds and hydrophobic interaction to enhance taste perception. Overall, this study highlights the potential of utilizing Morchella esculenta stipes as a sustainable source of γ-glutamyl peptides, which effectively enhance salt and umami taste perception and may contribute to salt reduction without compromising the taste.

Polysaccharide from wild morels alters the spatial structure of gut microbiota and the production of short-chain fatty acids in mice

Wenyan HUO,Peng QI,Langjun CUI,Liguang ZHANG,Lu DAI,Yu LIU,Suying HU,Zhengping FENG,Ting QIAO,Junzhi LI

Polysaccharides from morels possess many characteristics beneficial to health, such as anti-tumor and immunomodulatory activities. The gut microbiota plays a critical role in the modulation of immune function. However, the impact of morel polysaccharides on the gut microbiota has not yet been explored. In this study, a high-throughput pyrosequencing technique was used to investigate the effects of MP, a new heteropolysaccharide extracted from wild morels, on the diversity and composition of microbiota along the intestine in mice, as well as the production of short-chain fatty acids (SCFAs). The results showed that MP treatment increased the number of operational taxonomic unit (OTUs) and diversity along the intestine, especially in the small intestine. MP treatment induced a significant decrease in the number of Firmicutes and a significant increase in the number of Bacteroidetes in the small intestine microbiota. It was also observed that the relative abundance of SCFA-producing bacteria, especially Lachnospiraceae, was increased in both the cecum and colon of MP-treated mice. Moreover, MP promoted the production of SCFAs in mice. These results provide a foundation for further understanding the health benefits conferred by morel polysaccharides.

Structural characterization, in vitro and in vivo antioxidant activities of a heteropolysaccharide from the fruiting bodies of Morchella esculenta

Zheng-Nan Cai,Wan Li,Shomaila Mehmood,Wen-Juan Pan,Ya Wang,Fan-Ju Meng,Xiao-Fang Wang,Yong-Ming Lu,Yan Chen

This study aimed to investigate the structural features, in vitro and in vivo antioxidant activities of a heteropolysaccharide from the fruiting bodies of Morchella esculenta (FMP-1). FMP-1 had an average molecular weight of 4.7 × 10 3  Da and consisted of mannose, glucose and galactose. By methylation and NMR analysis, the backbone of FMP-1 was deduced to be made up of 1,4-linked Glc p and 1,6-linked Gal p . Hydroxyl, DPPH and superoxide radicals could be efficiently scavenged by FMP-1, with IC 50 values of 74.26, 119.32 and 161.49 μg/mL, respectively. Furthermore, FMP-1 could significantly protect zebrafish embryos against AAPH-induced oxidative damage. Decrease in malformations and mortalities was observed along with the reduction of ROS production, NO production and cell death. The protective effects were by decreasing MDA content and increasing SOD, CAT and GSH-Px levels. The current work provided a good suggestion of the potential utilization of FMP-1 as an attractive natural antioxidant.

Synthesis of Fe(III)-doping Morchella-like porous magnesium hydroxide for the enhanced heterogeneous Fenton degradation of amphoteric antibiotics and anionic dyes

Yonghui Lei,Wendan Chen,Qiwei Lin,Fangjie Lin,Xuesong Wang,Hu Zhu

A Morchella-like hierarchical porous Fe-doping magnesium hydroxide (Fe(III)-PMH) was synthesized via a one-step hydrothermal process using iron-bearing brucite with the modulation of recyclable L-threonate. The L-threonate facilitated formatting Morchella-like porous structure through controlling specific lattice plane of Mg(OH) 2 . The as-prepared Fe(III)-PMH features a high specific surface area porous structure and even distribution of steady Fe(III) active sites doped in Mg(OH) 2 lattice, contributing to its degradation of amphoteric antibiotics or anionic dyes in the presence of H 2 O 2 . The surface alkalinity of Fe(III)-PMH drives the anionic conversion of amphoteric antibiotics or enhances the ionization of anionic dyes. Then these anionic contaminants are rapidly absorbed and oxidized in situ on the positively charged surface of Fe(III)-PMH. For the stable presence of superoxide radicals in Fe(III)-PMH/H 2 O 2 system, a wide range of working pH was obtained (pH 3–11). Moreover, a negligible amount of leached iron ions, and excellent reusability make it durable and promising for industrial applications. This work provides a simple and low-cost preparation of heterogeneous catalysts with good performance, especially for the highly effective enhanced Fenton-like system in the wastewater treatment of amphoteric antibiotics and anionic dyes.

Morchella-inspired solar evaporator for anti-salt accumulation and highly efficient solar-driven interfacial evaporation

Rong Zhang,Renzhi Li,Zhiqiang Song,Pengjin Ye,Bo Du,Yongsheng Zhang,Bo Xiang

Solar-driven interfacial evaporation utilizing aerogels holds promising potential for applications in seawater desalination and wastewater treatment. Nevertheless, developing aerogels that simultaneously achieve high energy conversion efficiency, superior salt resistance, and cost-effectiveness continues to pose a significant challenge. Herein, we develop a morchella -inspired solar evaporator composed of carbonized straw serving as the solar absorber to achieve high solar absorption (95.2 %) and effectively reduce the material cost, as well as a polyvinyl alcohol (PVA) aerogel matrix ensuring excellent thermal insulation, rapid water transport, and efficient vapor escape. Specially, the surface of the solar evaporator is hydrophobically functionalized to enhance salt resistance, which in turn improves the solar steam generation performance. Benefiting from its ingenious design, the solar evaporator achieves a high water evaporation rate of 1.82 kg m −2 h −1 and a high solar energy conversion efficiency of 97.7 % under 1-sun illumination (1 kW m −2 ). Overall, the developed solar evaporator achieves high efficiency, excellent salt resistance, cost-effectiveness, and practicability, holding significant potential for applications in seawater desalination and wastewater purification.

Polysaccharide FMP-1 from Morchella esculenta attenuates cellular oxidative damage in human alveolar epithelial A549 cells through PI3K/AKT/Nrf2/HO-1 pathway

Wan Li,Zheng-Nan Cai,Shomaila Mehmood,Ya Wang,Wen-Juan Pan,Wen-Na Zhang,Yong-Ming Lu,Yan Chen

Oxidative stress is considered to involve cell death in severe pulmonary diseases like idiopathic pulmonary fibrosis (IPF). Polysaccharide FMP-1 from Morchella esculenta can exert significant antioxidant activity. However, its effects on alveolar epithelial cells remain unperceived. Herein, the effects of FMP-1 against H 2 O 2 -induced oxidative damage in human alveolar epithelial A549 cells were investigated. FMP-1 could inhibit H 2 O 2 -induced cytochrome C and Caspase-3 release to prevent cell apoptosis via attenuation of MDA and ROS levels, and enhancement the enzymatic activities of SOD and T-AOC. Furthermore, the underlying molecular mechanisms were clarified. The phosphorylation of AKT and the nuclear translocation of Nrf2 were observed to be promoted by FMP-1 as well as the level of HO-1. These findings suggested that FMP-1 attenuate cellular oxidative stress through PI3K/AKT pathway, and FMP-1 could be explored as natural potential antioxidants to lower oxidative stress relevant to the progression of IPF.

De novo assembly and characterization of the transcriptome of Morchella esculenta growth with selenium supplementation

Mengxiang Du,Shengwei Huang,Zihan Huang,Lijuan Qian,Yang Gui,Jing Hu,Yujun Sun

Although Morchella esculenta (L.) Pers. is an edible and nutritious mushroom with significant selenium (Se)-enriched potential, its biological response to selenium stimuli remains unclear. This study explored the effect of selenium on mushroom growth and the global gene expression profiles of M. esculenta . While 5 µg mL −1 selenite treatment slightly promoted mycelia growth and mushroom yield, 10 µg mL −1 significantly inhibited growth. Based on comparative transcriptome analysis, samples treated with 5 µg mL −1 and 10 µg mL −1 of Se contained 16,061 (452 upregulated and 15,609 downregulated) and 14,155 differentially expressed genes (DEGs; 800 upregulated and 13,355 downregulated), respectively. Moreover, DEGs were mainly enriched in the cell cycle, meiosis, aminoacyl-tRNA biosynthesis, spliceosome, protein processing in endoplasmic reticulum pathway, and mRNA surveillance pathway in both selenium-treated groups. Among these, MFS substrate transporter and aspartate aminotransferase genes potentially involved in Se metabolism and those linked to redox homeostasis were significantly upregulated, while genes involved in isoflavone biosynthesis and flavonoid metabolism were significantly downregulated. Gene expression levels increased alongside selenite treatment concentration, suggesting that high Se concentrations promoted M. esculenta detoxification. These results can be used to thoroughly explain the potential detoxification and Se enrichment processes in M. esculenta and edible fungi.
