The recovery and reconstruction of central nervous system function after spinal cord injury (SCI) is a worldwide problem. The difficulty lies in the feasibility issue of new axons passing through the injured area and the negative effect of scarring after injury. As a biological material, the human amniotic membrane (HAM) has the advantages of protecting nerve growth, inhibiting scar formation, and promoting neovascularization, but its weak physical properties are difficult to apply in treating SCI. In this study, HAMs were first decellularized and then chemically grafted with methacrylic anhydride. Next, the composite was photocrosslinked with gelatin methacrylate to prepare a cross-network biological complex. The final complexes prepared by appeal were used for in vitro and in vivo studies of SCI in rats, separately. In the in vitro experiment, the composite scaffold inherited abundant biological factors from the amniotic membrane and had the physical properties of a hydrogel, thus providing a favorable environment for the growth and development of neurons and blood vessels. In the in vivo experiment, the composite reduced scarring and promoted the growth of new nerves. Overall, the composite scaffolds can stably simulate the extracellular microenvironment in SCI defects, regulate pathological changes, and promote the generation of new neurons. Therefore, decellularized HAM hydrogels are promising biocomposite materials for central nerve repair after SCI. Graphic abstract

The human amniotic membrane (HAM) collagen matrix derived from human placenta can be decellularized (dHAM) to form a natural biocompatible material. dHAM has different bioactive substances and has been used widely in human tissue engineering research. However, dHAM has some disadvantages, e.g., poor mechanical properties , easy degradation and inconvenient operation and use, so it is not conducive to large-area or full-thickness skin defect healing. To overcome these limitations, for the first time, dHAM was grafted with methacrylic anhydride (MA) to form photocrosslinked dHAM methacrylate (dHAMMA); dHAMMA was then blended with methacrylated gelatin (GelMA), followed by the addition of a photosensitizer for photocrosslinking to obtain the fast-forming GelMA-dHAMMA composite hydrogel. Further, GelMA-dHAMMA was found to have the porous structure of a bicomponent polymer network and good physical and chemical properties. In vitro experiments, GelMA-dHAMMA was found to promote fibroblast proliferation and α-smooth muscle actin (α-SMA) expression. In vivo investigations also demonstrated that GelMA-dHAMMA promotes wound collagen deposition and angiogenesis , and accelerates tissue healing. GelMA-dHAMMA inherits the good mechanical properties of GelMA and maintains the biological activity of the amniotic membrane, promoting the reconstruction and regeneration of skin wounds. Thus, GelMA-dHAMMA can serve as a promising biomaterial in skin tissue engineering. Statement of significance Since the early 20th century, the human amniotic membrane (HAM) has been successfully used for trauma treatment and reconstruction purposes. dHAM has different bioactive substances and has been used widely in human tissue-engineering research. In this work, the dHAM and gelatin were grafted and modified by using methacrylic anhydride (MA) to form photocrosslinked dHAMMA and methacrylated gelatin (GelMA). Then, the dHAMMA and GelMA were blended with a photosensitizer to form the GelMA-dHAMMA composite hydrogel derived from gelatin-dHAM. GelMA-dHAMMA exhibits a bicomponent-network (BCN) interpenetrating structure. dHAM dydrogel has advantages, e.g., good mechanical properties, slow degradation and convenient operation, so it is conducive to large-area or full-thickness skin defect healing.

Spinal cord injury (SCI) results in devastating consequences for the motor and sensory function of patients due to neuronal loss and disrupted neural circuits, confronting poor prognosis and lack of effective therapies. A new therapeutic strategy is urgently required. Here, human amniotic epithelial cells (hAEC), featured with immunocompatibility, non-tumorgenicity and no ethical issues, were induced into neural-like cells by a compound cocktail, as evidenced with morphological change and the expression of neural cell markers. Interestingly, the hAEC-neural-like cells maintain the characteristic of low immunogenicity as hAEC. Aiming at SCI treatment in vivo, we constructed a 3D-printed GelMA hydrogel biomimetic spinal cord scaffold with micro-channels, in which hAEC-neural-like cells were well-induced and grown. In a rat full transection SCI model, hAEC-neural-like cell scaffolds that were implanted in the lesion demonstrated significant therapeutic effects; the neural circuit and hindlimb locomotion were partly recovered compared to little affection in the SCI rats receiving an empty scaffold or a sham implantation operation. Thus, the establishment of hAEC-neural-like cell biomimetic scaffolds may provide a safe and effective treatment strategy for SCI.Keywords:human amniotic epithelial cell;neural cell;biomimetic scaffold;spinal cord injury

Human amniotic membrane (hAM) is a promising material for tissue engineering due to several benefits, including desirable biocompatibility, stem cell source, antibacterial activity, etc. However, because of its low elasticity, the clinical application of hAM is severely restricted. To solve this issue, we employed polydopamine/polyacrylamide (PDA/PAM) hydrogels to toughen hAM. The test results indicated that the PDA/PAM hydrogel can enhance the toughness of hAM dramatically due to the formation of abundant chemical bonds and the strong mechanical properties of the hydrogel itself. Compared to pure hAM, the break elongation and tensile strength of PDA/PAM-toughened hAM rose by 154.15 and 492.31%, respectively. And most importantly, the fracture toughness was almost 15 times higher than untreated hAM. In addition, the cytotoxicity of the PDA/PAM-coated hAM was not detected due to the superior biocompatibility of the chemicals used in the study. Treating hAM with adhesive hydrogels to increase its mechanical characteristics will further promote the application of hAM as a tissue engineering material.

The diabetic wounds do not heal easily in part because they are susceptible to infection due to environmental influences. Wound dressing is crucial to wound healing, as it can basically protect the wound from external damages and provide a suitable microenvironment for tissue regeneration. In this study, a double-layer membrane that consists of chitosan sponge and decellularized bovine amniotic membrane (dBAM) has been developed by freeze-casting method. The results showed that the porous structure of the sponge layer improved the performances of blood coagulation and swelling. The dense dBAM can optimize the mechanical property of wound dressing. In vitro studies revealed that the bilayer membrane had favorable biocompatible, especially for human foreskin fibroblast cells (HFF-1) cell adhesion and proliferation. Moreover, the full-thickness skin defects of diabetic model mice that treated with bilayer membrane showed over 80% closure in 8 days. Our findings imply that the double-layer dressing has great potentials to be used in diabetic patients. Keywords:chitosan;decellularized bovine amniotic membrane;diabetic wound healing;double-layer dressing

Bone tissue engineering is a novel and efficient repair method for bone tissue defects, and the key step of the bone tissue engineering repair strategy is to prepare non-toxic, metabolizable, biocompatible, bone-induced tissue engineering scaffolds of suitable mechanical strength. Human acellular amniotic membrane (HAAM) is mainly composed of collagen and mucopolysaccharide; it has a natural three-dimensional structure and no immunogenicity. In this study, a polylactic acid (PLA)/Hydroxyapatite (nHAp)/Human acellular amniotic membrane (HAAM) composite scaffold was prepared and the porosity, water absorption and elastic modulus of the composite scaffold were characterized. After that, the cell–scaffold composite was constructed using newborn Sprague Dawley (SD) rat osteoblasts to characterize the biological properties of the composite. In conclusion, the scaffolds have a composite structure of large and small holes with a large pore diameter of 200 μm and a small pore diameter of 30 μm. After adding HAAM, the contact angle of the composite decreases to 38.7°, and the water absorption reaches 249.7%. The addition of nHAp can improve the scaffold’s mechanical strength. The degradation rate of the PLA+nHAp+HAAM group was the highest, reaching 39.48% after 12 weeks. Fluorescence staining showed that the cells were evenly distributed and had good activity on the composite scaffold; the PLA+nHAp+HAAM scaffold has the highest cell viability. The adhesion rate to HAAM was the highest, and the addition of nHAp and HAAM could promote the rapid adhesion of cells to scaffolds. The addition of HAAM and nHAp can significantly promote the secretion of ALP. Therefore, the PLA/nHAp/HAAM composite scaffold can support the adhesion, proliferation and differentiation of osteoblasts in vitro which provide sufficient space for cell proliferation, and is suitable for the formation and development of solid bone tissue. Keywords:human acellular amniotic membrane;acellular process;osteoblasts;polylactic acid;hydroxyapatite

Chitosan is a promising biomaterial for tissue engineering, but its functionality is limited by a lack of bioactive sites. This study develops chitosan/amniotic membrane microcarriers to enhance vascularization and tissue regeneration for subcutaneous adipose tissue. The incorporation of decellularized amniotic membrane enhances the bioactivities of chitosan in promoting cell differentiation and angiogenesis. Optimized preparation yielded porous microcarriers with a particle size of 261.2 ± 28 μm and an average pore size of 19.0 ± 4 μm. In vitro degradation analysis showed accelerated degradation with higher amniotic membrane content. Cytocompatibility and adipogenic capacity assessments indicated that the microcarriers supported cell adhesion and proliferation over 7 days, with amniotic membrane facilitating adipogenic differentiation of adipose-derived stem cells. When injected subcutaneously into nude mice, these microcarriers formed neoplastic adipose tissues, which were harvested 8 weeks later. Fluorescence staining, oil-red O staining and CD31 labeling demonstrated that amniotic membrane incorporation significantly enhanced in vivo adipose tissue formation and angiogenesis.

Adhesions and poor healing are major complications after Achilles tendon injury, and there is no effective solution to this problem. The purpose of this study is to determine whether the biomimetic peritoneurosis can solve the above problems in the process of Achilles tendon healing; This study investigated the adhesion and proliferation of tendon-derived stem cells (TDSCs) on dAM in vitro, as well as their tenogenic differentiation. The effects of Achilles tendon rupture on tendon regeneration were assessed in vivo by using an Achilles tendon rupture model in rats; Finally, from in vitro mRNA transcriptome sequencing combined with in vivo Achilles tendon protein to omics analysis to explore the repair mechanism of Achilles tendon rupture. Student’s t-tests were used to assess the significance of observed differences between the two experimental groups. Multiple groups were compared using one-way analysis of variances (ANOVAs), followed by post hoc Bonferroni comparisons. The main findings of this study are that cell adhesion, proliferation, and differentiation of TDSCs were enhanced by dAM. Implanted dAM + TDSCs significantly accelerated tendon regeneration in vivo. In addition, extracellular matrix-related differential genes and proteins were screened by mRNA transcriptometry in vitro and proteomic analysis of Achilles tendon in vivo, and ERK signaling pathway was further explored to participate in the repair of Achilles tendon rupture. The dAM-TDSCs composite biomimetic peritendinous membrane material can effectively promote the healing of Achilles tendon. It provides a new direction for the development of biomimetic peritendinous membrane materials.

Background: Amniogenesis is a key event in biochemical pregnancy, and its failure may result in human embryonic death. However, whether and how environmental chemicals affect amniogenesis remain largely unknown. Objectives: The objective of the present study was to screen chemicals that may disrupt amniogenesis in an amniotic sac embryoid model and to investigate the potential mechanism of amniogenesis failure, with a focus on organophosphate flame retardants (OPFRs). Methods: This study developed a high-throughput toxicity screening assay based on transcriptional activity of octamer-binding transcription factor 4 ( Oct4 ). For the two positive OPFR hits with the strongest inhibitory activity, we used time-lapse and phase-contrast imaging to assess their effects on amniogenesis. Associated pathways were explored by RNA-sequencing and western blotting, and potential binding target protein was identified through a competitive binding experiment. Results: Eight positive hits exhibiting Oct4 expression were identified, with 2-ethylhexyl-diphenyl phosphate (EHDPP) and isodecyl diphenyl phosphate (IDDPP) showing the strongest inhibitory activity. EHDPP and IDDPP were found to disrupt the rosette-like structure of the amniotic sac or inhibit its development. Functional markers of squamous amniotic ectoderm and inner cell mass were also found disrupted in the EHDPP- and IDDPP-exposed embryoids. Mechanistically, embryoids exposed to each chemical exhibited abnormal accumulation of phosphorylated nonmuscle myosin (p-MLC-II) and were able to bind to integrin β 1 ( ITG β 1 ). Conclusion: The amniotic sac embryoid models suggested that OPFRs disrupted amniogenesis likely by inhibiting the ITG β 1 pathway, thus providing direct in vitro evidence associating OPFRs with biochemical miscarriage. https://doi.org/10.1289/EHP11958

Defects within the oral mucosa can result from a variety of causes, including tumors and trauma. Adults have limited ability to regenerate after tissue loss or injury, meaning autografts to reconstruct defects are often required. Due to shortcomings associated to tissue transplantation,
the use of tissue engineering technology to reconstruct and regenerate oral tissues has gained attention. In this regard, HAM (human amniotic membrane) was utilized as a biological material for tissue engineering applications. However, its unfavorable properties have limited its use, and hybrid
scaffolds based on HAM have been investigated. Thus, in this study, a biomimetic hybrid scaffold composed of gelatin methacryloyl and decellularized human amniotic particles (GelMA-dHAP) was prepared and used as a tissue substitute to promote wound healing of the oral mucosa in rabbits. First,
the degradation and swelling characteristics of the scaffold were evaluated and found to be more desirable than those of GelMA alone. Then, the effects of the GelMA-dHAP stent on the proliferation and differentiation of human foreskin fibroblasts were studied in vitro using a cell counting
kit-8 (CCK8) and immunofluorescence, respectively. This showed that the hybrid scaffold could significantly promote fibroblast proliferation and differentiation in comparison to controls. Following these initial assessments and to better understand the underlying mechanism of wound healing,
in vivo histological and immunohistochemical studies were undertaken using a rabbit oral mucosa defect model. The results showed that GelMA-dHAP can promote angiogenesis and collagen expression within the oral mucosa. Importantly, the GelMA-dHAP scaffold was shown to assist wound healing
better than GelMA alone, demonstrating the potential of the hybrid scaffold in tissue engineering.

Extrusion-based 3D bioprinting of cell-laden hydrogels is a potential technology for regenerative medicine, which enables the fabrication of constructs with spatially defined cell distribution. However, the limited assessment of rheological behaviors of hydrogel before printing is still a major issue for the advancement of 3D bioprinting. In this work, we systematically investigated the rheological behaviors (i.e. viscosity, storage modulus (G’), and loss modulus (G”)) of alginate/gelatin composite hydrogels first for 3D printing complex constructs. The rheological studies revealed that viscosity of alginate/gelatin hydrogels is temperature-dependent and shear thinning. Sol-gel transition (intersection of G’ and G”) study provided indication for printing temperature, which are in the range of 18.8 °C ( H2/7.5 ) to 24.5 °C ( H2/24.5 ). The alginate (2 wt%) /gelatin (15 wt%) composite hydrogel sample was chosen to print the constructs and subsequent bioprinting. Complex constructs (i.e. nose and ear) were obtained with high printing resolution (151 ± 13.04 μm) in a low temperature (4 °C) chamber and crosslinking with 2 wt% CaCl 2 subsequently without extra supports. Human amniotic epithelial cells (AECs) showed superior potential to differentiate into epithelial cells, while Wharton’s jelly derived mesenchymal stem cells (WJMSCs) showed a superior angiogenic potential and fibroblastic phenotype. For the in vitro study, AECs and WJMSCs as seed cells, encapsulated in alginate/gelatin composite hydrogels, were bioprinted to form biomimetic bilayered membranous construct. High cell viability (> 95%) were observed up to 6 days after printing. The presented 3D bioprinting of human AECs and WJMSCs-laden alginate/gelatin composite hydrogels provides promising potentials for future skin tissue engineering.

Human amniotic transplantation has been proposed to improve the therapeutic efficacy of intrauterine adhesions (IUAs). Human amniotic mesenchymal stem stromal cells (hAMSCs) can differentiate into multiple tissue types. This study aimed to investigate the mechanism by which hAMSCs transplantation promotes endometrial regeneration. The rat models with IUA were established through mechanical and infective methods, and PKH26-labeled hAMSCs were transplanted through the tail vein (combined with/without estrogen). Under three different conditions, hAMSCs differentiated into endometrium-like cells. HE and Mason staining assays, and immunohistochemistry were used to compare the changes in rat models treated with hAMSCs and/or estrogen transplantation. To define the induction of hAMSCs to endometrium-like cells in vitro, an induction medium (cytokines, estrogen) was used to investigate the differentiation of hAMSCs into endometrium-like cells. qRT-polymerase chain reaction (PCR) and western blotting were performed to detect the differentiation of hAMSCs into endometrium-like cells. A greater number of glands, fewer endometrial fibrotic areas, and stronger expression of vascular endothelial growth factor and cytokeratin in the combined group (hAMSCs transplantation combined with estrogen) than in the other treatment groups were observed. hAMSCs could be induced into endometrium-like cells by cytokine treatment (TGF-β1, EGF, and PDGF-BB). Transplantation of hAMSCs is an effective alternative for endometrial regeneration after injury in rats. The differentiation protocol for hAMSCs will be useful for further studies on human endometrial regeneration.
