Attenuation effects of iron on dissemination of antibiotic resistance genes in anaerobic bioreactor: Evolution of quorum sensing, quorum quenching and dynamics of community composition

Yi Xu,Shihong Yang,Guoxiang You,Jun Hou

Zero valent iron (ZVI) coupled with bioreactors is arising as a promising technology for antibiotic resistance genes (ARGs) mitigation, whereas the succession and behaviors of microbes caused by ZVI in relieving ARGs propagation remain unclear. Herein, the effects of ZVI on microbial quorum sensing (QS), quorum quenching (QQ) system and community dynamics were examined in anaerobic bioreactor fed with oxytetracycline ( tet ), to illustrate the roles of evolutive microbial communication and community composition in ARGs attenuation. With the addition of 5 g/L ZVI, the total absolute abundance of tet ARGs was retarded by approximate 95% and 72% in sludge and effluent after 25 days operation. The abundance of mobile genetic elements and the heredity of antibiotic resistant bacteria revealed the declined horizontal and vertical transfer of ARGs, which directly led to the reduced ARGs propagation. Potential mechanisms are that the positive effects of ZVI on QQ activity via the functional bacteria enrichment inhibited QS system and thus ARGs transfer. Partial least--squares path modeling further demonstrated that ARGs abundance was strongly limited by the dynamics of bacterial composition and thereby less frequent microbial communication. These results provide new insights into the mechanisms of antibiotic resistome remission in anaerobic bioreactor modified by ZVI.

Effects of metformin on Streptococcus suis LuxS/AI-2 quorum sensing system and biofilm formation

Jing Zuo,Yamin Shen,Haikun Wang,Shuji Gao,Shuo Yuan,Dong Song,Yuxin Wang,Yang Wang

Streptococcus suis ( S. suis ) regulates biofilm formation through LuxS/AI-2 quorum sensing system, increasing drug resistance and exacerbating infection. The anti-hyperglycaemic agent metformin has anti-bacterial and anti-biofilm activities. This study aimed to investigate the anti-biofilm and anti-quorum sensing activity of metformin in S. suis . We first determined the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of metformin on S. suis . The results indicated that metformin showed no obvious inhibitory or bactericidal effect . Crystal violet staining showed that metformin significantly inhibited the formation of S. suis biofilm at sub-MIC concentration, which was also confirmed by scanning electron microscopy. Then, we quantified the AI-2 signal molecules in S. suis , and the results showed that metformin had a significant inhibitory effect on the production of AI-2 signal in S. suis . Inhibition of enzyme activity and molecular docking experiments showed that metformin has a significant binding activity to LuxS protein. In addition, qRT-PCR results showed that metformin significantly down-regulated the expression of AI-2 synthesis-related genes luxS and pfs , and adhesion-related genes luxS , pfs , gapdh , sly , fbps, and ef . Western blotting also showed that metformin significantly reduced the expression of LuxS protein. Our study suggests that metformin seems to be a suitable candidate for the inhibition of S. suis LuxS/AI-2 QS system and prevention of biofilm formation, which provided a new idea for the prevention and control of S. suis .

Phytol inhibits quorum sensing and biogenic amine production in Pseudomonas fluorescens PF12

Hongwei Zhan,Xiang Zou,Jiao Li,Lixiu Zhang,Guanyuan Qu,Yanqing Li,Yongjun Sun,Wenming Ju,Mengqi Ye,Yang Deng,Ting Ding

Pseudomonas fluorescens is a prevalent spoilage bacterium in various chilled aquatic products, producing toxic biogenic amines (BAs) through the decarboxylation of amino acids via amino acid decarboxylases. Our previous studies revealed that P. fluorescens PF12 regulates BA production through its quorum sensing (QS) system, suggesting that disrupting QS could be a potential approach to control BA generation. In this study, we targeted the three-dimensional structure of the LuxI-type protein in P. fluorescens PF12 to screen for QS inhibitors (QSIs) through high-throughput screening from Seaweed Metabolite Database and Comprehensive Marine Natural Products Database. Nine compounds with high binding scores to the LuxI-type protein were identified. Considering cost-effectiveness and safety, phytol was selected for further investigation. Experimental results demonstrated that sub-inhibitory concentrations of phytol significantly suppressed BA production, siderophore production, motility, extracellular protease secretion, and biofilm formation in P. fluorescens PF12. Specifically, when the salmon fillets were stored for 288 h, the histamine content in the fillets treated with 25 mg/mL phytol decreased by 43.81 %. RT-qPCR revealed that the expression levels of key genes hisD and hisC related to histamine synthesis in the phytol treatment group were reduced by 7.68 ( p  < 0.05) and 8.23 % ( p  < 0.05), respectively, compared to the control group. Molecular docking analysis revealed that phytol interacts with key residues in the LuxI-type protein, thereby blocking the synthesis of acyl-homoserine lactones (AHLs) and downstream QS signaling. These findings indicate that phytol has great potential for controlling BA production in aquatic products and ensuring food safety.

The Effect of Bacterial AHL on the Cyclic Adenosine Monophosphate Content in Plants According to High-Performance Liquid Chromatography

Xuemeng Zhao,Wen Li,Xiliu Li,Zhenhua Jia,Shuishan Song,Qian Zhao

Cyclic adenosine monophosphate (cAMP) is an important second messenger in cells, mediating various stimulation signals such as the growth and development of organisms and stress and participating in regulating various biological processes of cells. This article explores the quantitative determination of cAMP in plants using High-Performance Liquid Chromatography (HPLC) and applies this method to analyzing the changes in cAMP content during the process of plant response to the bacterial quorum sensing signalN-acyl homoserine lactone (AHL). Research has shown that the optimal detection conditions for HPLC are as follows: the chromatographic column is Venusil MP C18 (2), the mobile phase is methanol–water (0.1% trifluoroacetic acid) (v:v, 10:90), the detection wavelength is 259 nm, the column temperature is 35 °C, and the flow rate is 0.8 mL/min. The precision of the standard sample of this method is 98.21%, the precision of the sample is 98.87%, and the recovery rate is 101.067%. The optimal extraction conditions for cAMP in Arabidopsis are to use 15% methanol ultrasonic extraction for 10 min, followed by a 40 °C water bath for 4 h. Bacterial AHL signal processing can significantly stimulate an increase in cAMP levels in Arabidopsis leaves and roots. The establishment of HPLC detection methods for the cAMP content in plants is of great significance for in-depth research on the signal transduction mechanisms of plant–bacterial interactions.

Regulatory mechanism by which exogenous AHLs enhance the degradation of bisphenol A by Pseudomonas asiatica P1

Fenglin Zhang,Jinming Gu,Yibing Wang,Qing Qiu,Yue Yu,Xuejian Sun,Menghan Chang,Dandan Zhou,Hongliang Huo,Kejian Tian

Bisphenol A (BPA) poses multiple risks to human health, and it has become a critical environmental pollution issue. The advantage of bacterial degradation of BPA has become more prominent, and enhancement methods have become increasingly diverse. Using quorum sensing to regulate functional bacteria and genes to enhance the bacterial degradation efficiency of BPA serves as a highly promising new approach. This study investigated the effect of adding exogenous N-hexanoyl-L-homoserine lactone (C6-HSL) on the degradation of BPA by Pseudomonas asiatica P1 (strain P1) and analyzed the regulatory mechanism of the signaling molecules. The results showed that exogenous homoserine lactone signaling molecules (AHLs) could enhance the BPA degradation efficiency of strain P1, with 10 μM C6-HSL demonstrating the most significant enhancement, increasing the degradation rate by 12.4 % compared with the control group and producing products such as benzoic acid and succinic acid. In terms of the regulatory mechanism, C6-HSL preferentially induced a 5.18-fold upregulation in the expression of the AHL receptor-encoding gene LasR. Subsequently, LasR regulates the upregulation of hub genes such as bvgS and aruS , ultimately achieving an upregulation of BPA degradation genes through hierarchical regulation. This led to an enhancement in the initial degradation step of BPA and the activation of the benzoic acid degradation pathway. This study analyzed the potential of C6-HSL to enhance microbial BPA degradation and, for the first time, clarified the mechanism by which C6-HSL enhanced bacterial BPA degradation efficiency through hierarchical regulation.

Cinnamaldehyde Decreases the Pathogenesis of Aeromonas hydrophila by Inhibiting Quorum Sensing and Biofilm Formation

Shengping Li,Shun Zhou,Qiuhong Yang,Yongtao Liu,Yibin Yang,Ning Xu,Xiaohui Ai,Jing Dong

Antibiotics were the main fishery drugs for treatingAeromonas hydrophila(A. hydrophila) infection, which would generate selective pressure and result in the appearance of antibiotic-resistant bacteria. The bacterial quorum sensing (QS) system provides a new alternative strategy againstA. hydrophilainfection. QS inhibitors can reduce bacterial virulence behaviors by disrupting QS, which has no effect on bacterial growth. Therefore, we studied the effect of cinnamaldehyde from a natural plant extract on the QS ofA. hydrophilaaiming to reduce its pathogenicity. The efficacy of cinnamaldehyde againstA. hydrophilawas evaluated from various aspects, including the effects on aerolysin, lipase, protease, swarming motility, biofilm formation, acyl-homoserine lactones (AHLs), and QS-related genes. Moreover, the therapeutic effect of cinnamaldehyde in vitro and in vivo was studied. The results showed that cinnamaldehyde could decrease the virulence phenotypes ofA. hydrophilaregulated by QS. Moreover, the transcriptions of related genes (aerA,ahyR, andahyI) were downregulated following the addition of cinnamaldehyde. The in vitro and in vivo therapeutic assays show that cinnamaldehyde could reduce the aerolysin-mediated A549 cell injury and increase the survival rate of crucian carp infected withA. hydrophila. These results indicate that cinnamaldehyde would be a candidate QS inhibitor againstA. hydrophilainfection. Keywords:Aeromonas hydrophila;quorum sensing;cinnamaldehyde;virulence;biofilm

Effects of hydroxyl group in cyclo(Pro-Tyr)-like cyclic dipeptides on their anti-QS activity and self-assembly.

Li Li,Zuxian Xu,Ruipin Cao,Jiaxin Li,Chang-Jer Wu,Yinglu Wang,Hu Zhu

We investigated the influence of hydroxyl groups on the anti-quorum-sensing (anti-QS) and anti-biofilm activity of structurally similar cyclic dipeptides, namely cyclo( L -Pro- L -Tyr), cyclo( L -Hyp- L -Tyr), and cyclo( L -Pro- L -Phe), against Pseudomonas aeruginosa PAO1. Cyclo( L -Pro- L -Phe), lacking hydroxyl groups, displayed higher virulence factor inhibition and cytotoxicity, but showed less inhibitory ability in biofilm formation. Cyclo( L -Pro- L -Tyr) and cyclo( L -Hyp- L -Tyr) suppressed genes in both the las and rh l systems, whereas cyclo( L -Pro- L -Phe) mainly downregulated rhl I and pqs R expression. These cyclic dipeptides interacted with the QS-related protein LasR, with similar binding efficiency to the autoinducer 3OC12-HSL, except for cyclo( L -Pro- L -Phe) which had lower affinity. In addition, the introduction of hydroxyl groups significantly improved the self-assembly ability of these peptides. Both cyclo( L -Pro- L -Tyr) and cyclo( L -Hyp- L -Tyr) formed assembly particles at the highest tested concentration. The findings revealed the structure-function relationship of this kind of cyclic dipeptides and provided basis for our follow-up research in the design and modification of anti-QS compounds.

Quorum Sensing System-Regulated Proteins Affect the Spoilage Potential of Co-cultured Acinetobacter johnsonii and Pseudomonas fluorescens From Spoiled Bigeye Tuna (Thunnus obesus) as Determined by Proteomic Analysis

Wang Xin-Yun,Xie Jing

Food spoilage by certain species of bacteria is reported to be regulated by quorum sensing (QS). Acinetobacter johnsonii and Pseudomonas fluorescens, the major specific spoilage organisms, are found to be limited in their QS and co-culture interactions. The aim of this study was to determine how QS-regulated proteins affect the spoilage potential of co-cultured A. johnsonii and P. fluorescens obtained from spoiled bigeye tuna (Thunnus obesus) using a proteomics approach. The A. johnsonii, P. fluorescens, and their co-culture tested the N-acyl-homoserine lactone (AHL) activities using reporter Chromobacterium violaceum CV026 and LC-MS/MS in qualitative and quantitative approaches, respectively. These latter showed that, of the 470 proteins and 444 proteins in A. johnsonii (A) and P. fluorescens (P), respectively, 80 were significantly up-regulated and 97 were significantly down-regulated in A vs. AP, whereas 90 were up-regulated and 65 were down-regulated in P vs. AP. The differentially expressed proteins included the AI-2E family transporter OS, 50S ribosomal protein L3, thioredoxin reductase OS, cysteine synthase CysM OS, DNA-binding response regulator, and amino acid ABC transporter ATPase OS. The cellular process (GO:0009987), metabolic process (GO:0008152), and single-organism process (GO:0044699) were classified into the gene ontology (GO) term. In addition, energy production and conversion, amino acid transport and metabolism, translation, ribosomal structure and biogenesis, post-translational modification, protein turnover, and chaperones were distributed into the clusters of orthologous groups of proteins (COG) terms. The KEGG pathways revealed that 84 and 77 differentially expressed proteins were divided into 20 KEGG pathways in A vs. AP and P vs. AP, respectively, and amino acid metabolism, carbohydrate metabolism, energy metabolism, and translation were significantly enriched. Proteins that correlated with the spoilage-related metabolic pathways, including thioredoxin reductase OS, cysteine synthase OS, and pyridoxal phosphate-dependent enzyme family protein OS, were identified. AI-2E family transporter OS and LuxR family transcriptional regulator OS were identified that related to the QS system. These findings provide a differential proteomic profile of co-culture in A. johnsonii and P. fluorescens, and have potential applications in QS and the regulation of spoilage potential.

Quorum Quenching-Mediated Biofilm Mitigation on Functionalized Ultrafiltration Membranes via Atom Transfer Radical Polymerization

Shaojie Ren,Guohe Huang,Yunkun Wang

Quorum sensing (QS) critically affects biofilm formation during membrane filtration, which can be manipulated for membrane biofouling mitigation. This study demonstrates a quorum quenching strategy for combating membrane biofouling by functionalizing poly(vinylidene fluoride) (PVDF) membranes via atom transfer radical polymerization of a QS inhibitor, vanillin. Poly(vanillin) brushes are successfully grafted on the membrane surface, which is verified by extensive membrane characterization. The effective biofilm inhibition ability and mitigation mechanism of poly(vanillin) brushes are evaluated by analyzing the QS-related gene expression and acyl-homoserine lactone (AHL) signal molecule concentrations. We show that poly(vanillin) brushes can inhibit the bacterial QS system and decrease the AHL concentration to mitigate membrane biofouling. In dynamic filtration experiments, the vanillin-grafted PVDF membrane exhibits a 50% increase in stable flux compared to that of the pristine PVDF membrane. Our results provide an alternate strategy for mitigating membrane biofouling by regulating the bacterial QS system via inhibitor-functionalized membranes.

Regulation of Burkholderia cenocepacia virulence by the fatty acyl-CoA ligase DsfR as a response regulator of quorum sensing signal

Xia Li,Shihao Song,Xiaohan Kong,Xiayu Chen,Zhuoxian Zhao,Zizi Lin,Yantao Jia,Yong Zhang,Hai-Bin Luo,Qiao-Ping Wang,Lian-Hui Zhang,Wei Qian,Yinyue Deng

Quorum sensing (QS) is a cell-to-cell communication mechanism mediated by small diffusible signaling molecules. Previous studies showed that RpfR controls Burkholderia cenocepacia virulence as a cis -2-dodecenoic acid (BDSF) QS signal receptor. Here, we report that the fatty acyl-CoA ligase DsfR (BCAM2136), which efficiently catalyzes in vitro synthesis of lauryl-CoA and oleoyl-CoA from lauric acid and oleic acid, respectively, acts as a global transcriptional regulator to control B . cenocepacia virulence by sensing BDSF. We show that BDSF binds to DsfR with high affinity and enhances the binding of DsfR to the promoter DNA regions of target genes. Furthermore, we demonstrate that the homolog of DsfR in B. lata , RS02960, binds to the target gene promoter, and perception of BDSF enhances the binding activity of RS02960. Together, these results provide insights into the evolved unusual functions of DsfR that control bacterial virulence as a response regulator of QS signal.

Virtual Screening and Biological Evaluation of Anti-Biofilm Agents Targeting LuxS in the Quorum Sensing System

Zheng Liu,Lihua Zhang,Jincai Wang,Yanping Li,Yiqun Chang,Xiaoling Huang,Jun Duan,Yilong Ai,Xuxin Zeng,Jialiang Guo

Biofilm formation is considered as a crucial factor in various oral diseases, such as dental caries. The quorum sensing (QS) signaling system was proved to have a crucial role in the microbial dental plaque biofilm formation of Streptococcus mutans (S. mutans). LuxS was critical to regulating the QS system and survival of the bacterium, and, therefore, compounds which target LuxS may be a potential therapy for dental caries. The binding activities of 37,170 natural compounds to LuxS were virtually screened in this study. Baicalein and paeonol were chosen for further research of the binding mode and ΔG values with LuxS. Both baicalein and paeonol inhibited the biofilm formation without influence on the growth of S. mutans. Baicalein also distinctly reduced the production of both rhamnolipids and acids. The results provide us with a new approach to combat dental caries instead of the traditional use of antibacterial chemicals.

Dual-action gallium-flavonoid compounds for combating Pseudomonas aeruginosa infection

Xiaojun He,Bingjie Han,Runming Wang,Yu Guo,Richard Y T Kao,Hongyan Li,Hongzhe Sun,Wei Xia

The opportunistic pathogen Pseudomonas aeruginosa (P. aeruginosa) causes infections that are difficult to treat, which is due to the bacterial natural resistance to antibiotics. The bacterium is also able to form a biofilm that protects the bacterium from clearance by the human immune system and leads to chronic infection. Herein, we synthesized and characterized a novel gallium compound that interferes with both the iron metabolism and quorum sensing system of P. aeruginosa to achieve a significant bactericidal activity. The compound could substantially reduce the secretion of bacterial virulence factors as well as eliminate biofilm formation. Integrative omics analysis indicates that this compound can significantly disturb the gene transcription and metabolism of P. aeruginosa. The effectiveness of the gallium compound was further validated in mammalian cell and murine skin infection models. Our study offers a new strategy to design new gallium-based antimicrobials to combat P. aeruginosa infection.
