Climate change significantly threatens crops, mainly through drought stress, affecting barley, which is essential for food and feed globally. Ten barley cultivars were evaluated under normal and drought stress conditions during the 2019/20 and 2020/21 seasons, focusing on traits such as days to heading and maturity, plant height, number of spikes m−2, spike length, 1000-kernel weight, and biological and grain yield. Drought stress significantly reduced most of these traits. The genotypes showed significant differences in their responses to irrigation treatments, with the interaction between seasons and cultivars also being significant for most traits. The grain yield and 1000-kernel weight were among the least affected traits under drought stress, respectively. Notably, Giza138 and Giza126 showed strong drought tolerance, suitable for drought-resilient breeding. In season one, Giza126, Giza134, and Giza138 yielded 13%, 9%, and 11%, respectively, while Giza135 and Giza129 showed higher reductions at 31% and 39%. In season two, Giza126, Giza134, and Giza138 had reductions of 14%, 10%, and 13%, respectively, while Giza135 and Giza129 again exhibited higher reductions at 31% and 38%. These cultivars also showed strong performance across various stress tolerance indices, including the MP, YSI, STI, GMP, and YI. Giza 134 demonstrated the lowest values for the SDI and TOL, indicating superior drought stress tolerance. On the other hand, Giza 129 and Giza 135 were the most sensitive to drought stress, experiencing significant reductions across critical traits, including 6.1% in days to heading, 18.37% in plant height, 28.21% in number of kernel spikes−1, 38.45% in grain yield, and 34.91% in biological yield. In contrast, Giza 138 and Giza 2000 showed better resilience, with lower reductions in the 1000-kernel weight (6.41%) and grain yield (10.61%), making them more suitable for drought-prone conditions. Giza 126 and Giza 132 also exhibited lower sensitivity, with minimal reductions in days to heading (2%) and maturity (2.4%), suggesting potential adaptability to water-limited environments. Giza 126 maintained the highest root lengths and had the highest stomatal conductance. Giza 138 consistently had the highest chlorophyll content, with SPAD values decreasing to 79% under drought. Despite leading in shoot length, Giza 135 decreased to 42.59% under drought stress. In conclusion, Giza 126 and Giza 138 showed adaptability to water-limited conditions with minimal impact on phenological traits. Giza 126 had the longest roots and highest stomatal conductance, while Giza 138 consistently maintained a high chlorophyll content. Together, they and Giza 134 are valuable for breeding programs to improve barley drought tolerance.

SUMMARY Calcineurin B-like (CBL)-interacting protein kinases (CIPKs) play a central role in Ca 2+ signalling and promote drought tolerance in plants. The CIPK gene family in pigeon pea ( Cajanus cajan L.), a major food crop affected by drought, has not previously been characterised. Here, we identified 28 CIPK genes in the pigeon pea genome. Five CcCIPK genes were strongly upregulated in roots upon drought treatment and were selected for further characterisation. Overexpression of CcCIPK13 and CcCIPK14 increased survival rates by two- to three-fold relative to controls after 14 days of drought. Furthermore, the three major flavonoids, genistin, genistein and apigenin, were significantly upregulated in the same transgenic plants. Using CcCIPK14 as bait, we performed a yeast two-hybrid screen and identified six interactors, including CcCBL1. CcCIPK14 exhibited autophosphorylation and phosphorylation of CcCBL1 in vitro . CcCBL1 -overexpressed plants displayed higher survival rates upon drought stress as well as higher expression of flavonoid biosynthetic genes and flavonoid content. CcCIPK14 -overexpressed plants in which CcCBL1 transcript levels were reduced by RNA interference had lower survival rates, which indicated CcCBL1 in the same pathway as CcCIPK14. Together, our results demonstrate a role for the CcCIPK14-CcCBL1 complex in drought stress tolerance through the regulation of flavonoid biosynthesis in pigeon pea.

Virus-induced drought tolerance presents a fascinating facet of biotic-abiotic interaction in plants, yet its molecular intricacies remain unclear. Our study shows that cowpea mild mottle virus (CPMMV) infection enhances drought tolerance in common bean (Phaseolus vulgaris) plants through a virus-derived small interfering RNA (vsiRNA)-activated autophagy pathway. Specifically, a 21-bp vsiRNA originating from the CPMMV Triple Gene Block1 (TGB1) gene targeted the 5' untranslated region (UTR) of the host Teosinte branched 1, Cycloidea, Proliferating Cell Factor (TCP) transcription factor gene PvTCP2, independent of the known role of TGB1 as an RNA silencing suppressor. This targeting attenuated the expression of PvTCP2, which encodes a transcriptional repressor, and in turn upregulated the core autophagy-related gene (ATG) PvATG8c, leading to activated autophagy activity surpassing the level induced by drought or CPMMV infection alone. The downstream EARLY RESPONSIVE TO DEHYDRATION (ERD) effector PvERD15 is a homologue of Arabidopsis thaliana AtERD15, which positively regulates stomatal aperture. PvERD15 was degraded in PvATG8c-mediated autophagy. Therefore, we establish a TGB1-PvTCP2-PvATG8c-PvERD15 module as a trans-kingdom fine-tuning mechanism that contributes to virus-induced drought tolerance in plant-drought-virus interactions.

Plant yield is severely hampered by drought, affirming the urgent need to develop strategies to improve plant growth and drought tolerance. The synergistic application of ABA and calcium nanoparticles (Ca-NPs) has emerged as a promising approach for sustainable crop production and improved resistance to abiotic stress, although the underlying mechanism for foliar application of growth regulator ABA and Ca-NPs in regulating plant growth and drought tolerance in rapeseed ( Brassica napus L.) remains poorly understood. To address this knowledge gap, the current study was conducted to investigate the protective role of 100-µM ABA and 100 ppm Ca-NPs in alleviating the drought stress (15% PEG-6000) in B. napus following further application with 100 µM solution of photosynthesis inhibitor 3-(3,4-dichlorophyllorophenyl)-1,1-dimethylurea (DCMU). Foliar application of 100 ppm Ca-NPs and 100-µM ABA significantly reduced the drought stress by increasing chl a, chl b, total chlorophyll, carotenoids, xanthophyll by 86, 63, 78, 56, 68%; nutritional profile Ca, K, P, Mg, Mn, B by 131, 108, 98, 111, 82, 67% of B. napus plants, respectively. Moreover, under drought condition the combined treatment of Ca-NPs + ABA along with the DCMU increased the Pn, Gs, Ci, Tr by 30, 24, 37, 22% and transcript abundance of transcription factors and ROS homeostasis genes ( CDPK5, MYC2, TGA1, CAS , and POD ), and photosynthetic proteins expressions ( NCED3, PsbB, CP29, CP24, PCaA, CP26 and cytochrome f) . Overall, this study provides the evidence of effectiveness of combined application of Ca-NPs and ABA in alleviating drought stress in rapeseed.
