The present study attempted to synthesize carbon quantum dots (CQDs) through Aldol polymerization reaction , wherein acetone was used as the carbon source. A nano composite CQDs/Ag 2 S/CS was developed by loading as prepared CQDs and Ag 2 S nanoparticles on a chitosan substrate (CS). An in-situation growth of nanocomposites was adopted to study their antibacterial properties . Staphylococcus aureus (Gram-positive), Escherichia coli (Gram-negative) and methicillin-resistant Staphylococcus aureus were selected as the model bacteria. The CQDs/Ag 2 S/CS nanocomposites exhibited excellent inhibition not only against common pathogenic bacteria, but also those well-known drug-resistant bacteria. Moreover, compared to traditional antibiotics, the as prepared nanocomposites in the present work do not likely cause bacterial drug resistance , which make them a potential candidate for a new type of clinically applicable antibiotics.

Recently, there has been increasing information about the removal of chemical pollutants through biochar and intermittent aeration. However, little attention was paid to the application of biochar and intermittent aeration for the improvement of antibiotic resistant bacteria (ARB) removal in stormwater bioretention cells. Target ARB ( E. coli K-12 carrying bla TEM, tet R and aph A) was used in batch experiments for bioretention cells with different biochar contents, ratios of air to water (A/W) and aeration time (AT). ARB removal decreased with the increase of M/B in bioretention cells, where the highest ARB removal (3.73 Log) was found under 10 % of biochar, while intermittent aeration could contribute to the enhancement of ARB removal with the largest (3.94 Log) under A/W of 40:1 and AT of 4 hr/d. Both biochar chemisorption and H 2 O 2 oxidation were responsible for ARB removals with the maximal adsorption capacity of 33843 CFU/g, and the highest ARB removal less than 0.20 Log due to H 2 O 2 yields (0.021–0.042 mmol/L). ARGs conjugation frequencies tended to decrease with the increase of biochar levels and AT, and ARGs transformation decreased obviously under A/W of 40:1 and 20:1. There were around 1.13-fold changes for both intracellular reactive oxygen species (ROS) and cell membrane permeability (CMP) in bioretention cells with the combination of biochar and intermittent aeration, suggesting the slightly increasing risks for both conjugation and transformation when the application of the combined method on ARB removals.

The wide use of antibiotics in the animal husbandry and the relevant sustainable industries may promote the emergence of antibiotic-resistant bacteria (ARB), which constitutes a growing threat to human health. The objective of this study was to determine the abundance and diversity of sulfonamide- and tetracycline-resistant bacteria within an eco-agricultural system (EAS) in Hangzhou, China. We investigated samples at every link in the EAS, from livestock manure, to biogas residues and biogas slurry, to vegetable and ryegrass fields, to a fish pond. A combination of culture-based and 16S rRNA gene-based sequencing method was used in this study. Within the studied system, the average rate of bacterial resistance to sulfonamide (46.19 %) was much higher than that of tetracycline (8.51 %) ( p < 0.01). There were 224 isolates that were enumerated and sequenced, 108 of which were identified to species level. The genera comprising the sulfamethoxazole-resistant (SMX r ) bacteria were generally different from those of tetracycline-resistant (TC r ) bacteria. Staphylococcus and Acinetobacter were the most dominant genera of SMX r bacteria (19.30 % of the total resistant bacteria) and TC r bacteria (14.04 % of the total resistant bacteria), respectively. Several strains of resistant opportunistic pathogens (e.g., Pantoea agglomerans ) were detected in edible vegetable samples, which may exert a potential threat to both pig production and human health. In general, this study indicates that the EAS is an important reservoir of antibiotic-resistant bacteria, some of which may be pathogenic.

The emergence and prevalence of multidrug-resistant (MDR) bacteria have posed a serious threat to public health. Of particular concern are methicillin-resistantStaphylococcus aureus(MRSA) andblaNDM,mcr-1andtet(X)-positive Gram-negative pathogens. The fact that few new antibiotics have been approved in recent years exacerbates this global crisis, thus, new alternatives are urgently needed. Antimicrobial peptides (AMPs) originated from host defense peptides with a wide range of sources and multiple functions, are less prone to achieve resistance. All these characteristics laid the foundation for AMPs to become potential antibiotic candidates. In this study, we revealed that peptide WW307 displayed potent antibacterial and bactericidal activity against MDR bacteria, including MRSA and Gram-negative bacteria carryingblaNDM-5,mcr-1ortet(X4). In addition, WW307 exhibited great biofilm inhibition and eradication activity. Safety and stability experiments showed that WW307 had a strong resistance against various physiological conditions and displayed relatively low toxicity. Mechanistic experiments showed that WW307 resulted in membrane damage by selectively targeting bacterial membrane-specific components, including lipopolysaccharide (LPS), phosphatidylglycerol (PG), and cardiolipin (CL). Moreover, WW307 dissipated membrane potential and triggered the production of reactive oxygen species (ROS). Collectively, these results demonstrated that WW307 represents a promising candidate for combating MDR pathogens. Keywords:antimicrobial peptide;antibiotic resistance;multidrug-resistant bacteria;membrane damage

A series of novel pleuromutilin derivatives were designed and synthesized based on the twin drugs theory. Piperazinyl and thioether were used as the linkage to connect the pleuromutilin nuclear and sulfonamide to improve the biological activity and water solubility of derivatives. The in vitro antibacterial activities against drug-sensitive and drug-resistance bacteria were evaluated by agar dilution method. Most of the 25 compounds displayed excellent antibacterial activities against drug-sensitive bacteria, particularly, five compounds (9, 10, 11, 14a and 14b) exerted the excellent antibacterial activities against drug-resistance bacteria.

The rapid proliferation of multidrug-resistant (MDR) bacterial pathogens poses a serious threat to healthcare worldwide. Carbapenem-resistant (CR) Enterobacteriaceae, which have near-universal resistance to available antimicrobials, represent a particularly concerning issue. Herein, we report the identification of AMXT-1501, a polyamine transport system inhibitor with antibacterial activity against Gram-positive and -negative MDR bacteria. We observed minimum inhibitory concentration (MIC)50/MIC90 values for AMXT-1501 in the range of 3.13–12.5 μM (2.24–8.93 μg /mL), including for methicillin-resistant Staphylococcus aureus (MRSA), CR Escherichia coli, Klebsiella pneumoniae, and Pseudomonas aeruginosa. AMXT-1501 was more effective against MRSA and CR E. coli than vancomycin and tigecycline, respectively. Subinhibitory concentrations of AMXT-1501 reduced the biofilm formation of S. aureus and Enterococcus faecalis. Mechanistically, AMXT-1501 exposure damaged microbial membranes and increased membrane permeability and membrane potential by binding to cardiolipin (CL) and phosphatidylglycerol (PG). Importantly, AMXT-1501 pressure did not induce resistance readily in the tested pathogens.

Background: Currently, bacteria are getting resistant to antibiotics which pose a serious threat to human health. In this scenario, new nanoantibiotics have the potential to avoid the emerging bacterial resistance mechanisms. Hence, this study attempts to unveil the activity enhancement of Cephradine (Cfdn) by using experimental and theoretical investigations. Methods: Silver (Ag) and gold (Au) nanoparticles (NPs) stabilized with Cfdn (Cfdn-AgNPs and Cfdn-AuNPs) were synthesized using a one-pot reduction method, and their characteristics were analyzed using atomic force microscope (AFM), scanning electron microscope (SEM), Fouriertransform infrared (FT-IR) spectroscopy, UV-visible spectroscopy. The stability of the NPs was evaluated under different physiological conditions. The antimicrobial action of Cfdn, when conjugated with the Ag and Au NPs, was demonstrated by conducting experiments with multidrugresistant Staphylococcus aureus (S. aureus) cell cultures. AFM images were used to observe changes in S. aureus cell morphology before and after treatment with free and conjugated Cfdn. Moreover, theoretical analysis was carried out to further validate the enhanced activities of Cfdn-AgNPs and Cfdn-AuNPs.The spherical (20-55 nm) Cfdn-AgNPs and Cfdn-AuNPs demonstrated good stability under saline, pH, and thermal conditions. Interestingly, a significant improvement was achieved in the antibacterial activity, with Cfdn-AgNPs and Cfdn-AuNPs exhibiting activity enhancements of up to 2 and 6 times, respectively. AFM images confirmed improved kinetics of the antibiotic upon conjugation, where Cfdn alone did not cause sufficient damage to bacterial membranes even after a 4-hour treatment, whereas treatment with Cfdn-AgNPs and Cfdn-AuNPs resulted in effective cell membrane damage and complete disruption of cell morphology within the same timeframe. Moreover, theoretical simulation employing density functional theory (DFT) validated the enhanced antibacterial potential of Cfdn-AgNPs and Cfdn-AuNPs. Interestingly, cytotoxicity analysis revealed improved cytocompatibility of the NPs after conjugation with Cfdn.We established a systematic strategy for the development of innovative nanoantibiotics based on experimental and theoretical evidence, which can be exploited in the battle against bacterial resistance.

Drug-resistant bacterial infections cause significant harm to public life, health, and property. Biofilm is characterized by overexpression of glutathione (GSH), hypoxia , and slight acidity, which is one of the main factors for the formation of bacterial resistance . Traditional antibiotic therapy gradually loses its efficacy against multi-drug-resistant (MDR) bacteria. Therefore, synergistic therapy , which regulates the biofilm microenvironment, is a promising strategy. A multifunctional nanoplatform, SnFe 2 O 4 -PBA/Ce6@ZIF-8 (SBC@ZIF-8), in which tin ferrite (SnFe 2 O 4 , denoted as SFO) as the core, loaded with 3-aminobenzeneboronic acid (PBA) and dihydroporphyrin e6 (Ce6), and finally coated with zeolite imidazole salt skeleton 8 (ZIF-8). The platform has a synergistic photothermal therapy (PTT)/photodynamic therapy (PDT) effect, which can effectively remove overexpressed GSH by glutathione peroxidase-like activity, reduce the antioxidant capacity of biofilm, and enhance PDT. The platform had excellent photothermal performance (photothermal conversion efficiency was 55.7 %) and photothermal stability. The inhibition rate of two MDR bacteria was more than 96 %, and the biofilm clearance rate was more than 90 % (150 μg/mL). In the animal model of MDR S. aureus infected wound , after 100 μL SBC@ZIF-8+NIR (150 μg/mL) treatment, the wound area of mice was reduced by 95 % and nearly healed. The serum biochemical indexes and H&E staining results were within the normal range, indicating that the platform could promote wound healing and had good biosafety. In this study, we designed and synthesized multifunctional nanoplatforms with good anti-drug-resistant bacteria effect and elucidated the molecular mechanism of its anti-drug-resistant bacteria. It lays a foundation for clinical application in treating wound infection and promoting wound healing .

The growing problem of bacterial resistance caused by the abuse of antibiotics is a serious challenge for the world. In order to make the clinically available antibiotics regain their bactericidal effect, our study introduced photothermal therapy (PTT) to assist antibiotics to annihilate drug-resistant bacteria. To achieve the synergistic effect, nanoparticles (FeTGNPs) with an antibiotic core (gatifloxacin complexing with tannins) and a photothermal shell (ferric iron coordinating with tannins) were prepared directly in aqueous solution by a convenient yet efficient one-pot synthesis. The excellent photothermal properties of the shell of FeTGNPs were used to break the mechanism of bacterial resistance, and the sustained-release of gatifloxacin from the core regained the killing effect against drug-resistant bacteria. From the results of antibacterial experiments, with the synergistic effect of APTT and antibiotics, FeTGNPs (400 μg/mL) could effectively kill methicillin-resistant Staphylococcus aureus (sterilizing rate up to 96.5 %) and gatifloxacin-resistant Staphylococcus aureus (sterilizing rate up to 98.7 %) than equivalent antibiotics. Moreover, under slightly acidic microenvironment, such as infection area, gatifloxacin could accelerate its release from the core of FeTGNPs. Therefore, FeTGNPs would be a highly effective antibacterial agent against drug-resistant bacterial infections in the future.

Bleeding in outdoor environments is often accompanied by bacterial infection. Due to poor outdoor conditions, it is essential to use the same materials to achieve one-stop treatment of fast hemostasis and simultaneously sterilizing bacteria, especially multidrug-resistant bacteria. Photodynamic therapy (PDT) can kill superbacteria, and local PDT through a nanofiber platform can effectively reduce damage to normal tissue. However, current photosensitizers whether in the interior or on the surface of fibers would leak into the wound and inhibit collagen regeneration. Herein, we use a battery-powered handheld electrospinning device that can work outdoors. It directly spins fibers onto the wound, which facilitates fast hemostasis due to its excellent adhesion to the wound. Eluting holes in the hydrophobic fibers by wound tissue fluid are also proposed to accelerate the escape of reactive oxygen species (ROS) from the interior of the fibers to the wound. After photosensitizers were coated on upconverting nanoparticles (UCNPs), they formed clusters whose size (∼55 nm) was much larger than the uniform elution hole (∼4 nm), which prevented photosensitizers from leaking out into the wound tissue. This cluster structure can also tailor the photosensitizers to be triggered by near infrared (NIR) light, whose deeper penetration depth in tissue can facilitate treating deep infections. Because of the combination of the in situ fiber deposition method with the designed elution mode, ROS is effectively poured out onto the fiber surface and is quickly delivered to the wound. Thus, after rapid hemostasis (<7 s), this one-stop treatment followed by photodynamic sterilizing of superbacteria can promote collagen regeneration and reduce wound healing time from 24 to 16 days.

Introduction In recent years, cationic amphipathic antimicrobial peptides (AMPs) have shown great promise in combating antibiotic resistance on account of their distinctive membrane-disruptive mechanism. However, the clinical application of AMPs is restricted by their unsatisfactory stability and safety. Although attempts have been made to improve the stability and safety of AMPs, many of them are accompanied by a decline in their antimicrobial activity and bacterial selectivity. Objectives To develop AMPs with excellent and balanced antimicrobial activity, stability, and safety using a combination strategy. Methods A series of sC18 4b -derived peptide analogues were designed by a combination strategy of subtly adjusting the charges, hydrophobic properties, and introducing specific unnatural amino acids in a well-balanced manner. The antimicrobial activity, cytotoxicity, hemolytic activity, stability, anti-biofilm activity, mechanism of action, synergistic effects, in vivo efficacy, and pharmacokinetics of the analogues were evaluated. Results Among these analogues, P-α-02-B stood out for its broad-spectrum and potent antimicrobial activity, anti-biofilm activity, desirable bacterial selectivity, high plasma stability, and synergistic effect with antibiotic levofloxacin. P-α-02-B exhibited strong membrane disturbance effect, which could be explained by its rigid α-helical structure revealed by molecular dynamics simulations. More importantly, P-α-02-B showed favorable therapeutic efficacy in vivo , whether used alone or in combination with levofloxacin. Conclusion P-α-02-B is a promising antimicrobial agent for MDR bacterial infections, demonstrating the effectiveness of the combination strategy for AMP development.

The overexpression of virulence factors and the induced inflammatory cytokine storm, resulting in tissue damage, represent significant challenges in treating antibiotic-resistant bacterial infections with conventional antibiotics. Herein, we have developed a bifunctional nanoparticle loaded with antibiotics (PSA@PLGA-TD/SMT) designed for precise response to the virulence proteins of drug-resistant bacteria while protecting the host from excessive inflammatory damage. This is achieved by modifying polylactic- co -glycolic acid (PLGA) nanoparticles with polysialic acid (PSA). Tildipirosin (TD), through hydrophobic interactions, is encapsulated within the core, while S-Methylisothiourea (SMT) is electrostatically adsorbed onto the shell layer. In vitro, the polysialic acid in the nanoparticles interacts with the neuraminidase overexpressed on the surface of Glaesserella parasuis (Gps), triggering disintegration and subsequent release of TD, which effectively kills the bacteria. Additionally, SMT functions to suppress the secretion of inducible nitric oxide synthase (NOS2), promoting the phenotypic transformation of macrophages and reducing the expression of pro-inflammatory factors. Ultimately, PSA@PLGA-TD/SMT achieves an extended drug circulation time through its prolonged retention effect, effectively alleviating discomfort caused by resistant bacteria. Therefore, the construction of nanocarriers for precise targeting of drug-resistant pathogens, delivering antibiotics, and preventing host-induced inflammatory damage presents an effective treatment strategy. This approach not only addresses the direct issue of bacterial eradication but also mitigates the collateral damage typically associated with severe infections, offering a more comprehensive and nuanced approach to managing drug-resistant bacterial infections.
