Osteomyelitis poses substantial therapeutic challenges due to the prevalence of multidrug-resistant bacterial infections and associated inflammation. Current treatment regimens often rely on a combination of corticosteroids and antibiotics, which can lead to complications and impede effective bacterial clearance. In this study, we present CADP-10, a Cecropin A-derived peptide, designed to target methicillin-resistant Staphylococcus aureus (MRSA) and multidrug-resistant Escherichia coli (MRE), while simultaneously addressing inflammatory responses. CADP-10 self-assembles into nanobacterial net (NBacN) that selectively identify and bind to bacterial endotoxins (LPS and LTA), disrupting membrane integrity and depolarizing membrane potential, which culminates in bacterial death. Importantly, these NBacN are bound to LPS and LTA from dead bacteria, preventing their engagement with TLR receptors and effectively blocking downstream inflammatory pathways. Our assessments of CADP-10 demonstrate good biosafety in both in vitro and in vivo models. Notably, in a rabbit osteomyelitis model, CADP-10 eliminated MRSA-induced bone infections, mitigated inflammation, and promoted bone tissue regeneration. This research highlights the potential of CADP-10 as a multifunctional antimicrobial agent for the management of infectious inflammatory diseases.

The growing resistance of bacteria to antibiotics has posed challenges in treating associated bacterial infections, while the development of multi-model antibacterial strategies could efficient sterilization to prevent drug resistance. High-entropy MXene has emerged as a promising candidate for antibacterial synergy with inherent photothermal and photodynamic properties. Herein, a high-entropy nanomaterial of MXene/CDs was synthesized to amplify oxidative stress under near-infrared laser irradiation. Well-exfoliated MXene nanosheets have proven to show an excellent photothermal effect for sterilization. The incorporation of CDs could provide photo-generated electrons for MXene nanosheets to generate ROS, meanwhile reducing the recombination of electron-hole pairs to further accelerate the generation of photo-generated electrons. The MXene/CDs material demonstrates outstanding synergistic photothermal and photodynamic effects, possesses excellent biocompatibility and successfully eliminates drug-resistant bacteria as well as inhibits biofilm formation. While attaining a remarkable killing efficiency of up to 99.99% against drug-resistant Escherichia coli and Staphylococcus aureus , it also demonstrates outstanding antibacterial effects against four additional bacterial strains. This work not only establishes a synthesis precedent for preparing high-entropy MXene materials with CDs but also provides a potential approach for addressing the issue of drug-resistant bacterial infections.

Recently, increasing attention has been paid to antibiotic resistance in stormwater runoff. However, there is no available literature about the control of antibiotic resistant bacteria (ARB) through 365 nm ultraviolet light-emitting diode (UVA/LED). In this study, batch experiments were conducted to investigate ARB inactivation kinetics, effects of light intensity and water matrix (including suspended solid (SS) concentration, initial pH and bacteria concentration), and potential transmission risks after UVA/LED irradiation. Results showed that ARB inactivation efficiencies reached 6.31 log reduction at 8 mW/cm 2 (86 J/cm 2 ) of UVA/LED for 180 min. ARB inactivation efficiencies increased with the increase of light intensity, and showed a linear relationship. ARB inactivation decreased with increasing SS levels, and the largest inactivation efficiencies was 3.56 log reduction at 50 mg/L of SS. Initial pH had slight effect on ARB inactivation through UVA/LED irradiation. A low initial bacteria concentration (10 5 CFU/mL) was not necessarily associated with good ARB inactivation (3.59 log reduction). After UVA/LED irradiation, ARB was hardly detected during 12 hr of dark repair, and the transfer frequency of kanamycin resistance gene was increased to 5.43 × 10 −4 . These suggested that the application of UVA/LED to inactivate ARB in stormwater runoff was feasible and desirable in this study. ARB inactivation reached 6.31 log reduction at 8 mW/cm 2 of UVA/LED for 180 min. UVA/LED inactivation kinetics could be well described by the first-order model. Initial pH and bacteria levels had weak effects for ARB inactivation. Potential risks of ARB could be blocked after 60 min of UVA/LED disinfection.

Elemental sulfur is the oldest known antimicrobial agent. However, conventional sulfur in the clinic suffers from poor aqueous solubility and limited antibacterial activity, greatly hindering its practical use. Herein, we report a reform strategy coupling dimension engineering with chirality transfer to convert conventional 3D sulfur particles into chiral 2D sulfur nanosheets (S-NSs), which exhibit 50-fold improvement of antibacterial capability and dual-selective inhibition against Gram-positive bacteria. Benefiting from the inherent selectivity of S-NSs and chirality selectivity from decorated d-histidine, the obtained chiral S-NSs are proven to precisely kill Gram-positive drug-resistant bacteria, while no obvious bacterial inhibition is observed for Gram-negative bacteria. Mechanism studies reveal that S-NSs produce numerous reactive oxygen specipoes and hydrogen sulfide after incubation with bacteria, thus causing bacterial membrane destruction, respiratory chain damage, and ATP production inhibition. Upon spraying chiral S-NSs dispersions onto MRSA-infected wounds, the skin healing process was greatly accelerated in 8 days due to metabolism inhibition and oxidative damage of bacteria, indicating the excellent treatment efficiency of MRSA-infected wounds. This work converts the traditional well-known sulfur into modern antibacterial agents with a superior Gram-selectivity bactericidal capability.

Various antibiotic resistant bacteria (ARB) can thrive in soil and resist such environmental pressures as antibiotics through cooperative resistance, thereby promoting ARB retention and antibiotic resistance genes transmission. However, there has been finite knowledge in regard to the mechanisms and potential ecological risks of cooperative resistance in soil microbiome. In this study, soil minerals and organic matters were designed to treat a mixture of two Escherichia coli strains with different antibiotic resistance ( E. coli DH5α/pUC19 and E. coli XL2-Blue) to determine how soil components affected cooperative resistance, and Luria-Bertani plates containing two antibiotics were used to observe dual-drug resistant bacteria (DRB) developed via cooperative resistance. Results showed quartz, humic acid , and biochar promoted E. coli XL2-Blue with high fitness costs, whereas kaolin, montmorillonite , and soot inhibited both strains. Using fluorescence microscope and PCR, it was speculated DRB could resist the antibiotic pressure via E. coli XL2-Blue coating E. coli DH5α/pUC19. E. coli DH5α/pUC19 dominated cooperative resistance. Correlation analysis and scanning electron microscope images indicated soil components influenced cooperative resistance. Biochar promoted cooperative resistance by increasing intracellular reactive oxygen species , thereby reducing the dominant strain concentration required for DRB development. Kaolin inhibited cooperative resistance the most, followed by soot and montmorillonite.

Due to the abuse of antibiotics, bacteria produce serious drug resistance, which makes the existing clinical antibiotic treatment scheme fail and seriously endangers human health. The construction of antimicrobial nanometer systems including antimicrobial peptides and antimicrobial nanomaterials has become the focus of research on new antimicrobial agents, and may significantly improve the antimicrobial efficiency. In this study, we successfully explored a novel antimicrobial nano-system Dap@Au/Ag nanorods (Dap@Au/Ag NRs) based on gold nanorods, and investigated the antimicrobial properties of this novel antimicrobial agent with methicillin-resistant Staphylococcus aureus (MRSA) as the experimental strain. Under the action of H 2 O 2 , Dap@Au/Ag NRs can considerably release silver ions and antimicrobial peptides (Dap), destroying the integrity of the MRSA membrane and resulting in content leakage and bacterial death. Furthermore, in vivo and in vitro experiments showed that after a certain intensity of laser irradiation (808 nm, 0.8 W/cm 2 , 40–60 s), the antibacterial properties of Dap@Au/Ag NRs were significantly improved by using gold nanorods with good photothermal effect. At the same time, compared with traditional photothermal therapy (PTT), we found that laser irradiation with the appropriate intensity was given at the initial stage of infection and controlled at a mild temperature (below 47 °C), it not only significantly inhibited the growth of MRSA, prevented large area of wound ulceration and promoted wound healing, but also had no obvious thermal damage to the wound surface and surrounding skin. The results showed that novel antibacterial nano-system Dap@Au/Ag NRs have significant bactericidal properties, and can be used as a potential promising antimicrobial agent, which provides a new strategy for facilitating and enhancing the effective antimicrobial efficiency to inhibit drug-resistant bacterial infection.

Microbial infections have always been serious challenges to human health considering that antibiotics almost inevitably induce microbial resistance. Therefore, it is urgent to develop a new antibacterial agent that is active against drug-resistant bacteria and is less susceptible to microbial resistance. In this work, a series of host defense peptide (HDP) mimicking antibacterial poly-β-peptides were synthesized, characterized and evaluated for their biological activities. The best poly-β-peptide within this study (20 : 80 Bu : DM) displays potent and broad spectrum antibacterial activity against antibiotic-resistant super bugs and low toxicity toward mammalian cells. Moreover, these poly-β-peptides are bactericidal and kill bacteria very fast within 5 min. An antimicrobial resistance test demonstrated that bacteria develop no resistance toward the selected poly-β-peptides even over 1000 generations. Our studies demonstrate that random copolymers of heterochiral poly-β-peptides, without the need for defined secondary structures, can mimic the antimicrobial HDP. These results imply the potential application of these poly-β-peptides as new antimicrobial agents to tackle drug resistant antimicrobial infections.

Antibiotic-resistant bacterial infections have led to an increased demand for antibacterial agents that do not contribute to antimicrobial resistance. Antimicrobial peptides (AMPs) with the facially amphiphilic structures have demonstrated remarkable effectiveness, including the ability to suppress antibiotic resistance during bacterial treatment. Herein, inspired by the facially amphiphilic structure of AMPs, the facially amphiphilic skeletons of bile acids (BAs) are utilized as building blocks to create a main-chain cationic bile acid polymer (MCBAP) with macromolecular facial amphiphilicity via polycondensation and a subsequent quaternization. The optimal MCBAP displays an effective activity against Gram-positive methicillin-resistant Staphylococcus aureus (MRSA) and Gram-negative Escherichia coli, fast killing efficacy, superior bactericidal stability in vitro, and potent anti-infectious performance in vivo using the MRSA-infected wound model. MCBAP shows the low possibility to develop drug-resistant bacteria after repeated exposure, which may ascribe to the macromolecular facial amphiphilicity promoting bacterial membrane disruption and the generation of reactive oxygen species. The easy synthesis and low cost of MCBAP, the superior antimicrobial performance, and the therapeutic potential in treating MRSA infection altogether demonstrate that BAs are a promising group of building blocks to mimic the facially amphiphilic structure of AMPs in treating MRSA infection and alleviating antibiotic resistance.

Drug-resistant bacterial pneumonia presents a substantial challenge for targeted drug delivery to the infected lung site due to the existence of mucus and gas-blood barriers. Allicin, renowned for its extensive bioactivity, holds promise as an antimicrobial agent. However, its unregulated bioavailability restricts both its antibacterial and anti-inflammatory efficacy. As a result, precise targeting of the infection site for controlled drug release is crucial in the treatment of drug-resistant bacterial pneumonia. In this study, we developed a macrophage cell membrane-modified nanoallicin composite material (Allicin@MSN@CM) by utilizing cell membrane modification technology. By harnessing the homing ability of macrophage cell membranes to inflammatory sites, we enabled the active aggregation of nanomaterials during circulation within the body. Furthermore, α-hemolysin secreted by MRSA bacteria was employed to perforate the cell membrane, facilitating on-demand drug release at the infection site. In vitro antibacterial experiments demonstrated that the material possesses strong antibacterial properties, alongside good stability and biocompatibility. Additionally, in a mouse model of lung infection, it not only effectively eradicated bacteria but also reduced lung inflammation. This study provides valuable insights into the application of nanocarriers in delivering traditional Chinese medicine to address the challenges posed by bacterial drug-resistant pneumonia.

Overuse of antibiotics has led to the emergency of vancomycin-resistant Enterococcus (VRE), considered one of the leading causes of severe diseases and even death. The phototherapy strategy of combined photothermal therapy (PTT) and photodynamic therapy (PDT) has achieved considerable attention since it could effectively enhance synergistic efficacy and avoid new drug resistance. In this paper, a kind of hybrid nanocomposite was fabricated and applied in the phototherapy-based combination strategy against VRE pathogens, in which graphene oxide (GO) not only covalent conjugated with vancomycin (Van) but also loaded photosensitizer phthalocyanine (Pc) via π–π stacking interaction. Outstanding PTT and PDT effects were observed in the as-prepared nanocomposite (GO@Van@Pc), giving almost 2–3 logs of bacterial reduction in vitro. Similar data were also obtained when GO@Van@Pc was used to treat VRE-infected BALB/C mice, exhibiting obvious infection regression and accelerated wound healing with negligible damage to normal cells and major organs. Therefore, the as-prepared GO@Van@Pc integrating PTT and PDT has great potential in the phototherapy of drug-resistant pathogens. Graphical Abstract

The misuse of antibiotics leads to the emergence of a variety of drug-resistant bacteria, which poses a serious threat to human life. Novel antimicrobial strategies are urgently needed. Herein, peroxidase-like enzyme material of Cu 2 O/NPC/Pal was prepared from Cu 2 O/nanoporous carbon (Cu 2 O/NPC) derived from Cu-MOF in the presence of palygorskite (Pal) by the pyrolysis-etching-pyrolysis strategy. Cu 2 O/NPC/Pal could catalyze the production of ·O 2 − from H 2 O 2 to cause the oxidative stress and cell membrane damage to bacteria. The antibacterial rates against Staphylococcus aureus and Pseudomonas aeruginosa reached 98.99 % and 92.70 %, respectively. In addition, Cu 2 O/NPC/Pal can promote the oxidation of glutathione and the consumption of intracellular effectively. Introduction of Pal into Cu 2 O/NPC significantly enhances the binding affinity and forms a highly concentrated substrate microenvironment. The study provided the pyrolysis strategy for the synthesis of carbon-based metal oxide nanomaterials in combination with mineral-based materials for the enhancement of peroxide-like activity in antimicrobial application.

The antimicrobial effect of antimicrobial peptides is typically slow; they can be rapidly biodegraded and often have non-selective toxicity and elaborate sequences. Here we report a short peptide that is activated by ultrasound, that shows high broad-spectrum antibacterial efficiency (>99%) against clinically isolated methicillin-resistant bacteria (specifically, Staphylococcus aureus , Escherichia coli , Staphylococcus epidermidis , Enterobacter cancerogenus and Pseudomonas aeruginosa ) with 15 min of ultrasound irradiation, and that has negligible toxicity and low self-antibacterial activity. We selected the peptide, FFRKSKEK (a segment from the human host-defence LL-37 peptide), from a library of peptides with piezoelectric diphenylalanine (FF) sequences, low toxicity, hydrophobicity and net positive charge. We show via all-atom molecular dynamics simulations that ultrasound amplifies the membrane-penetrating ability of peptides with FF sequences and that its piezoelectric polarization generates reactive-oxygen species and disturbs bacterial electron-transport chains. In a goat model of hard-to-treat intervertebral infection, the sonosensitive peptide led to better outcomes than vancomycin. Antimicrobial peptides activated by ultrasound may offer a clinically relevant strategy for combating antibiotic-resistant infections.
