As a natural and healthy beverage, tea is widely enjoyed; however, the pesticide residues in tea leaves affect the quality and food safety. To develop a highly selective and efficient method for the facile removal of pesticide residues, the subcritical butane extraction (SBE) technique was employed, and three variables involving temperature, time and extraction cycles were studied. The optimum SBE conditions were found to be as follows: extraction temperature 45 °C, extraction time 30 min, number of extraction cycles 1, and in such a condition that the extraction efficiency reached as high as 92%. Further, the catechins, theanine, caffeine and aroma components, which determine the quality of the tea, fluctuated after SBE treatment. Compared with the uncrushed leaves, pesticide residues can more easily be removed from crushed leaves, and the practical extraction efficiency was 97%. These results indicate that SBE is a useful method to efficiently remove the bifenthrin, and as appearance is not relevant in the production process, tea leaves should first be crushed and then extracted in order that residual pesticides are thoroughly removed. Keywords:subcritical butane;pesticide residue;bifenthrin;black tea

Cytochrome P450 monooxygenases (CYPs or P450s) play paramount roles in detoxification of insecticides in a number of insect pests. However, little is known about the roles of P450s and their responses to insecticide exposure in the codling mothCydia pomonella(L.), an economically important fruit pest. Here we report the characterization and expression analysis of the first P450 gene, designated asCYP9A61, from this pest. The full-length cDNA sequence ofCYP9A61is 2071 bp long and its open reading frame (ORF) encodes 538 amino acids. Sequence analysis shows that CYP9A61 shares 51%–60% identity with other known CYP9s and contains the highly conserved substrate recognition site SRS1, SRS4 and SRS5. Quantitative real-time PCR showed thatCYP9A61were 67-fold higher in the fifth instar larvae than in the first instar, and more abundant in the silk gland and fat body than other tissues. Exposure of the 3rd instar larvae to 12.5 mg L−1of chlorpyrifos-ethyl for 60 h and 0.19 mg L−1of lambda-cyhalothrin for 36 h resulted in 2.20-and 3.47-fold induction ofCYP9A61, respectively. Exposure of the 3rd instar larvae to these two insecticides also significantly enhanced the total P450 activity. The results suggested thatCYP9A61is an insecticide-detoxifying P450.Keywords:Cydia pomonella;cytochrome P450;induction;response;insecticides

Based on the fluorescence quenching phenomenon, a smart fluorescent microsensor was synthesized. The bifenthrin (BI) microsensor inherited the high selectivity of molecular imprinted polymers (MIPs) and the excellent fluorescence properties of aqueous CdTe quantum dots (QDs). Aqueous CdTe QDs are functionalized by octadecyl-4-vinylbenzyl-dimethyl-ammonium chloride (OVDAC). A type of functional monomer, 4-vinylphenylboronic acid (VPBA), was used and its boronic acid groups could covalently combine with a cis-diol compound for direct imprinting polymerization. The OVDAC-functionalized aqueous CdTe QDs were used as solid supports and auxiliary monomers. Under optimal conditions, experimentation showed that BI had a linear detection range of 10 to 300 μmol L−1 with a correlation coefficient of 0.9968 and a high imprinting factor (IF) of 4.53. In addition, the prepared MIP-OVDAC/CdTe QDs were successfully used to detect BI in water samples. Therefore, this work provided a highly selective and sensitive fluorescence probe for the detection of BI. In addition, the fluorescence probe could be used to detect other targets by changing the functional monomers.

Polyaniline/silver (PANI/Ag) composites were prepared with a one-step synthesis method, and fabricated into solid-phase microextraction fiber coatings through three different means, i.e. in situ growth, gluing and chemical bonding. This work explored the utility of the as-fabricated PANI/Ag SPME fibers as dual-functional platforms for microextraction and matrix-free laser desorption/ionization (LDI) of the target analyte. The results showed that after incorporating Ag into the PANI polymer , the LDI efficiency was improved by over two times, which was 92.8% the LDI efficiency of the commercial LDI plate. In addition, the in situ grown fiber coatings were observed the most sensitive as dual platforms for the analysis of bifenthrin . The signal intensity was increased over twenty times by using in situ grown fiber, comparing to direct analysis with commercial LDI plate, which were attributed to the excellent extraction ability of PANI backbone, as well as the high LDI efficiency of the PANI/Ag composites. Low detection limit (lower than 10 ng L −1 ) and good fiber-to-fiber repeatability (relative standard deviation 11.0%, n=3) were obtained by using the in situ grown coating for the analysis of bifenthrin , and bifenthrin in real water samples was successfully analyzed. The work successfully coupled SPME with matrix-free LDI, which greatly simplified the analytical process and improved the analytical sensitivity.

Pest management is mostly accomplished by the use of insecticides. However, the overuse of insecticides has led to the development of resistance. Glutathione S-transferases (GSTs) are vital detoxification enzymes involved in insecticide resistance in insects. In this study, we report the involvement of GSTs in insecticide resistance to lambda-cyhalothrin in Cydia pomonella, a globally quarantined fruit pest. A total of 25 GST, including 22 cytosolic genes and 3 microsomal genes, are identified from the genome database of C. pomonella. These cytosolic genes are further classified into six classes, including four in delta, eight in epsilon, three in omega, three in sigma, one in theta, and one in zeta class, as well as two unclassified genes. The real-time quantitative polymerase chain reaction (RT-qPCR) shows that the majority of these genes are mainly expressed throughout the larval stage and in the midgut of the fourth-instar larvae. Exposure to an LD10 dose of lambda-cyhalothrin resulted in the upregulation of 17 GST genes. Moreover, mRNA levels of most GST genes, with the exception of CpGSTe6, CpGSTd2, CpGSTd4, and CpGSTz1, are considerably higher in a lambda-cyhalothrin-resistant population (ZW_R) than those of susceptible strains. Recombinant CpGSTd1, CpGSTd3, CpGSTe3, and CpGSTs2 can bind and metabolize lambda-cyhalothrin, with the highest metabolic rate observed for CpGSTd3 but no metabolite(s) was detected, supporting the role of GSTs in sequestration of lambda-cyhalothrin. Molecular dynamics simulation analysis indicates that key residues of hydrophobic pocket-derived lipophilic energy S(lipo) interactions with a hydrophobic pharmacophore of lambda-cyhalothrin are crucial for metabolism by CpGSTd3 and further lead to resistance. Our study is the first to experimentally confirm the involvement of GSTs in lambda-cyhalothrin resistance via sequestration and provides new insights into resistance management in C. pomonella.

Nitrite (NO 2 − ) and bifenthrin (BF), common environmental pollutant, have been proven to perform toxic effects on marine organisms. To have a better understanding of their effects on marine organism, Haliotis discus hannai was exposed to 0.1 mg/L NO 2 − and/or 10 μg/L BF for 28 days. The results showed that the gills in direct contact with the water environment exhibited the highest levels of BF bioaccumulation (89.37–142.36 μg/kg), especially after NO 2 − exposure ( P < 0.05). Transcriptome analysis of gills showed that significant enrichments were observed in metabolic, signaling and immune pathways after exposure, such as cGMP-PKG signaling pathway. Additionally, superoxide dismutase (SOD) activity was decreased in gills after exposure to NO 2 − and/or BF, parallel with the increase in catalase (CAT) activity, malondialdehyde (MDA) content, and total antioxidant capacity (AOC). Regarding genes related to immune, the expression of Defensin was significantly decreased while the expressions of B-cell lymphoma-2 ( Bcl-2 ), Tumor Necrosis Factor-α ( TNF-α ) and Caspase3 were increased. These results suggested that NO 2 − and BF exposure disrupted metabolic processes, impaired immune function, and dysregulated antioxidant defenses in the gills of abalone. Particularly, significant alterations were observed under the combined stress of NO 2 − and BF. In hemocytes, the ROS production could be induced by NO 2 − and/or BF stress, and the number of hemocytes and phagocytosis decreased significantly, especially in NO 2 − and BF groups. Integrated biomarker response (IBR) analysis proved that NO 2 − and/or BF exposure detrimentally affected the overall fitness of abalone, and combined NO 2 − and BF stress was the most stressful condition. This study emphasizes the impact of NO 2 − and/or BF on the abalone to better elucidate the risk of environmental pollution on abalone.

In insects, glutathione S -transferases (GSTs) play a pivotal role in the detoxification of a wide range of pesticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest insect of stored products. Recently, pyrethroid insecticides have been used to control this pest. However, little is known concerning the responses and functions of GSTs in L. serricorne under pyrethroid exposure. In this study, transcriptome sequencing was performed on L. serricorne , and a total of 14 GSTs were identified by retrieving the unigene dataset. Of these, 13 predicted GSTs fell into six cytosolic classes, namely, delta, epsilon, omega, sigma, theta, and zeta, and one was assigned to an “unclassified” group. The GST genes were differentially expressed in various larval tissues and at different developmental stages. Exposure to the pyrethroid insecticide lambda-cyhalothrin (LCT) caused oxidative stress in L. serricorne larvae and led to significantly elevated expression levels of six genes, among which LsGSTe1 was the most upregulated. Recombinant LsGSTE1 protein displayed LCT-metabolizing activity. Furthermore, LsGSTE1 protects cells against oxidative stress. Moreover, knockdown of LsGSTe1 by RNA interference dramatically increased the susceptibility of L. serricorne larvae to LCT treatment. The results from this study provide sequence resources and expression data for GST genes in L. serricorne . Our findings indicate that LsGSTE1 plays a dual role in LCT detoxification by metabolizing the pesticide and by preventing LCT-induced oxidative stress. Thus, the LsGSTe1 gene could be used as a potential target for sustainable management of the cigarette beetle.

Although several approaches have been reported on the development of nanoemulsions over the last few years, studies on the formation of biodiesel nanoemulsions for bifenthrin formulation by the low-energy phase inversion composition (PIC) method are still scarce. Herein, the preparation of oil-in-water (O/W) nanoemulsions suitable for pesticide application has been achieved in biodiesel by dissolving a bifenthrin/mixture of a non-ionic surfactant (NP-6) and an anionic surfactant (ABSCa)/water system by the PIC method. The mechanism of the formation of bifenthrin nanoemulsions by dripping the water phase into the oil–surfactant phase was exemplified via the pseudo-ternary phase diagram. The effects of the mass ratio of NP-6 and ABSCa, mROS, stirring rate, the addition rate of water and the emulsification temperature on the mean droplet size of the nanoemulsion were investigated by dynamic light scattering (DLS). In addition, the interfacial tension and the contact angle of bifenthrin nanoemulsions for the spraying application were investigated. The insecticidal activity of bifenthrin nanoemulsions against cabbage maggots was further studied. Moreover, the emulsion stability of the bifenthrin nanoemulsions against Ostwald ripening behavior was evaluated, and the long-term stability of the bifenthrin formulation was studied by the HPLC method to assess the shelf life of the pesticide formulation. Experimental results showed that the optimum emulsification conditions for the mass ratio of NP-6 and ABSCa, mROS, stirring rate, the addition rate of water and the emulsification temperature were respectively 5/5, 1.4, 8000 rpm, 0.7 mL min−1 and 25 °C. The bifenthrin nanoemulsion with low interfacial tension and contact angle, easy adsorption on plant leaf surfaces and good shelf life has great potential for use as a pesticide formulation.

A sensitive monoclonal antibody-based enzyme monoclonal antibody-based enzyme-linked immunosorbent assay (ELISA) was developed to detect bifenthrin in a chemical soil barrier for termite control. LBc ((2-methyl[1,1-biphenyl]-3-methoxy) carbonyl propionic acid) was conjugated to bovine serum albumin (BSA) for producing monoclonal antibody. After optimization, the IC 50 and limit of detection (LOD, IC 10 ) were 0.05 mg L − 1 and 0.004 mg L − 1 , respectively. The sensitivity was improved 40-fold compared to polyclonal antibody-based ELISA reported earlier. No cross-reactivity was measured for the other analogues such as cyhalothrin, cyhalothric acid, cypermethrin, deltamethrin, fenpropathrin, fenvalerate, ethofenprox and tetramethrin except 2-methyl-3-biphenylmethyanol. Spiked recoveries were between 83.5% and 104.7% for the detection of bifenthrin in loess, red soil and black soil. All the relative standard deviations (RSDs) were less than or equal to 15.0%. Moreover, the ELISA for authentic samples showed reliability and high correlation with gas chromatography. The developed ELISA is an alternative tool for simple, sensitive and accurate monitoring of bifenthrin in chemical soil barriers.

Bifenthrin (BF) is a broad-spectrum insecticide that has gained widespread use due to its high effectiveness. However, there is limited research on the potential toxic effects of bifenthrin pollution on amphibians. This study aimed to investigate the 50 % lethal concentration (LC 50 ) and safety concentration of Chinese giant salamanders (CGS) exposed to BF (at 0, 6.25,12.5,25 and 50 μg/L BF) for 96 h. Subsequently, CGS were exposed to BF (at 0, 0.04, and 4 μg/L BF) for one week to investigate its toxic effects. Clinical poisoning symptoms, liver pathology, oxidative stress factors, DNA damage, and transcriptome differences were observed and analyzed. The results indicate that exposure to BF at 4 μg/L significantly decreased the adenosine-triphosphate (ATP), superoxide dismutase (SOD), glutathione (GSH), and catalase (CAT) contents in the brain, liver, and kidney of CGS. Additionally, the study found that the malondialdehyde (MDA), reactive oxygen species (ROS), and 8-hydroxydeoxyguanosine (8-OHdG) contents were increased. The liver tissue exhibited significant inflammatory reactions and structural malformations. RNA-seq analysis of the liver showed that BF caused abnormal antioxidant indices of CGS. This affected molecular function genes such as catalytic activity, ATP-dependent activity, metabolic processes, signaling and immune system processes, behavior, and detoxification, which were significantly upregulated, resulting in the differential genes significantly enriched in the calcium signaling pathway, PPARα signaling pathway and NF-kB signaling pathway. The results suggest that BF induces the abnormal production of free radicals, which overwhelms the body's self-defense system, leading to varying degrees of oxidative stress. This can result in oxidative damage, DNA damage, abnormal lipid metabolism, autoimmune diseases, clinical poisoning symptoms, and tissue inflammation. This work provides a theoretical basis for the rational application of bifenthrin and environmental risk assessment, as well as scientific guidance for the conservation of amphibian populations.

Numerous application of pyrethroid insecticides has led to their accumulation in the environment, threatening ecological environment and human health. Its fate in the presence of iron-bearing minerals and natural organic matter under light irradiation is still unknown. We found that goethite (Gt) and humic acid (HA) could improve the photodegradation of bifenthrin (BF) in proper concentration under light irradiation. The interaction between Gt and HA may improve BF degradation. On one hand, the adsorption of HA on Gt may decrease the photocatalytic activity of HA through decreasing HA content in solution and sequestering the functional groups realated with the production of reactive species. On the other hand, HA could improve the photocatalytic activity of Gt through extending light absorption, lowing of bandgap energy, hindering the recombination of photo-generated charges, and promoting the oxidation and reduction reaction on Gt surface. The increased oxygen vacancies on Gt surface along with the reduction of trivalent iron and the nucleophilic attack of hole to surface hydroxyl group contributed to the increasing photocatalytic activity of Gt. Electron paramagnetic resonance and quenching studies demonstrated that both oxidation species, such as hydroxyl radical (•OH) and singlet oxygen ( 1 O 2 ), and reducing species, such as hydrogen atoms (H•) and superoxide anion radical (O 2 •− ), contributed to BF degradation in UV-Gt-HA system. Mass spectrometry, ion chromatography, and toxicity assessment indicated that less toxic OH–BF (C 23 H 22 ClF 3 O 3 ), TFP (C 9 H 10 ClF 3 O), OH-MBP (C 14 H 14 O 2 ), MBP acid (C 14 H 12 O 2 ), and OH-MBP acid (C 14 H 12 O 3 ), and chloride ions were the main degradation products. The production of OH–BF, MPB, and TFP acid through oxidation and the production of MPB and TFP via reduction were the two primary pathways of BF degradation.

In this work, covalent organic framework (COF)-derived magnetic nanocomposites (Fe 3 O 4 @PDA@COF TABP ), comprising Fe 3 O 4 as the magnetic core, polydopamine (PDA) as the hydrophilic interlayer, and COF TABP as the outer shell, were first constructed and utilized as the powerful adsorbents for the magnetic solid-phase extraction (MSPE) of pyrethroids (PYRs). The introduction of the PDA coating facilitated the growth of the COF TABP shell and simultaneously provided additional adsorption sites, thereby enhancing the extraction efficiency. Molecular docking simulations revealed that Fe 3 O 4 @PDA@COF TABP exhibited excellent affinity toward PYRs through synergistic π-π stacking, hydrogen bonding, and hydrophobic interactions. The developed MSPE method coupled with GC-μECD exhibited excellent linearity (R 2 ≥ 0.9988), low limits of detection (0.025–0.064 μg kg −1 ), satisfactory recoveries (82.4 %–106.8 %), and low relative standard deviations (RSDs <7.1 %). Finally, the established method was successfully applied to the quantification of trace PYRs in vegetable samples, demonstrating its strong potential for routine pesticide residue monitoring in vegetables.
