Effects of glucose and biphenyl on aerobic cometabolism of polybrominated diphenyl ethers by Pseudomonas putida: Kinetics and degradation mechanism

Yuancai Lv,Lianghao Li,Yuancai Chen,Zhenghua Tang,Yongyou Hu

Polybrominated diphenyl ethers (PBDEs) are emerging persistent organic pollutants and the degradation of PBDEs is still a tremendous challenge owing to their extreme persistence and recalcitrance. In this study, Pseudomonas putida was employed for bioremediation of four PBDE congeners (BDE47, 15, 3 and diphenyl ether (DE)) with an initial concentration of 2 mg/L and the cometabolism of PBDEs was illustrated via kinetics analysis and metabolic mechanism. The biodegradation kinetics analysis suggested that PBDEs was inversely proportional to the number of bromine substituted on diphenyl ether. Both glucose (24 mg/L) and biphenyl (10 mg/L) played a positive role in promoting the biodegradation of PBDEs. The growth kinetics analysis indicated that glucose had uncompetitive inhibition on PBDEs consumption while both competitive and uncompetitive inhibition existed in biphenyl-PBDEs binary substrate system. Furthermore, the intermediates were identified by GC–MS and the main tentative degradation pathways were proposed. The angular dioxygenation was the main pathway for PBDEs degradation, while lateral dioxygenation could be induced when biphenyl was added.

Polybrominated diphenyl ethers and polychlorinated biphenyls induced rice "diabetes" by disturbing the transport and decomposition of soluble sugars

Qian Liu,Shuyuan Wang,Wei Wang,Jie Chen,Lizhong Zhu

Halogenated flame retardants in farmlands were observed to inhibit the growth of exposed crops. This study aimed to elucidate the mechanism of inhibition on rice by employing four representative polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs). The exposure to these contaminants at 200 nM led to a decrease of 0.63–0.95 fold in rice below-ground biomass and 0.49–0.66 fold in yield, and a corresponding 4%–10% increase in soluble sugars in leaves. PBDEs and PCBs were found to significantly disrupt the synthesis, decomposition, and transport of sugars in leaves, the three pivotal determinants of crop growth. Notably, these compounds promoted a 1.41- to 7.60-fold upregulation of the triose phosphate translocator, significantly enhancing soluble sugar synthesis. Conversely, a 0.45–0.97 fold downregulation was observed for sucrose transporters, thus impeding the leaf-to-shoot efflux of soluble sugars. Furthermore, PBDEs and PCBs were favorably bound to fructose-1,6-bisphosphate aldolase (FBA), inducing its substrate-specific dysfunction in fructose-1,6-diphosphate decomposition (3%–14%). Overall, PBDE and PCB exposure promoted a notable intracellular accumulation of soluble sugars in rice leaves, a typical symptom of plant diabetes, since the intensified synthesis of soluble sugars in leaves and the repressed decomposition and transportation of soluble sugars to other storage organs, thus impeding crop growth. This study provided an insightful understanding of the toxic effects and molecular mechanisms of halogenated flame retardants, highlighting their role in abnormal sugar accumulation and growth inhibition in crops and offering vital information for the risk assessment and administration of these compounds to guarantee the safety of agricultural products.

Sphingobium fuliginis HC3: A Novel and Robust Isolated Biphenyl- and Polychlorinated Biphenyls-Degrading Bacterium without Dead-End Intermediates Accumulation

Jinxing Hu,Mingrong Qian,Qian Zhang,Jinglan Cui,Chunna Yu,Xiaomei Su,Chaofeng Shen,Muhammad Z. Hashmi,Jiyan Shi

Biphenyl and polychlorinated biphenyls (PCBs) are typical environmental pollutants. However, these pollutants are hard to be totally mineralized by environmental microorganisms. One reason for this is the accumulation of dead-end intermediates during biphenyl and PCBs biodegradation, especially benzoate and chlorobenzoates (CBAs). Until now, only a few microorganisms have been reported to have the ability to completely mineralize biphenyl and PCBs. In this research, a novel bacterium HC3, which could degrade biphenyl and PCBs without dead-end intermediates accumulation, was isolated from PCBs-contaminated soil and identified as Sphingobium fuliginis . Benzoate and 3-chlorobenzoate (3-CBA) transformed from biphenyl and 3-chlorobiphenyl (3-CB) could be rapidly degraded by HC3. This strain has strong degradation ability of biphenyl, lower chlorinated (mono-, di- and tri-) PCBs as well as mono-CBAs, and the biphenyl/PCBs catabolic genes of HC3 are cloned on its plasmid. It could degrade 80.7% of 100 mg L −1 biphenyl within 24 h and its biphenyl degradation ability could be enhanced by adding readily available carbon sources such as tryptone and yeast extract. As far as we know, HC3 is the first reported that can degrade biphenyl and 3-CB without accumulation of benzoate and 3-CBA in the genus Sphingobium , which indicates the bacterium has the potential to totally mineralize biphenyl/PCBs and might be a good candidate for restoring biphenyl/PCBs-polluted environments.

Biodegradation of polychlorinated biphenyls (PCBs) by the novel identified cyanobacterium Anabaena PD-1

Hangjun Zhang,Xiaojun Jiang,Liping Lu,Wenfeng Xiao

Polychlorinated biphenyls (PCBs), a class of hazardous pollutants, are difficult to dissipate in the natural environment. In this study, a cyanobacterial strain Anabaena PD-1 showed good resistance against PCB congeners. Compared to a control group, chlorophyll a content decreased 3.7% and 11.7% when Anabaena PD-1 was exposed to 2 and 5 mg/L PCBs for 7 d. This cyanobacterial strain was capable of decomposing PCB congeners which was conclusively proved by determination of chloride ion concentrations in chlorine-free medium. After 7 d, the chloride ion concentrations in PCB-treated groups (1, 2, 5 mg/L) were 3.55, 3.05, and 2.25 mg/L, respectively. The genetic information of strain PD-1 was obtained through 16S rRNA sequencing analysis. The GenBank accession number of 16S rRNA of Anabaena PD-1 was KF201693.1. Phylogenetic tree analysis clearly indicated that Anabaena PD-1 belonged to the genus Anabaena . The degradation half-life of Aroclor 1254 by Anabaena PD-1 was 11.36 d; the total degradation rate for Aroclor 1254 was 84.4% after 25 d. Less chlorinated PCB congeners were more likely to be degraded by Anabaena PD-1 in comparison with highly chlorinated congeners. Meta - and para -chlorines in trichlorodiphenyls and tetrachlorobiphenyls were more susceptible to dechlorination than ortho -chlorines during the PCB-degradation process by Anabaena PD-1. Furthermore, Anabaena PD-1 can decompose dioxin-like PCBs. The percent biodegradation of 12 dioxin-like PCBs by strain PD-1 ranged from 37.4% to 68.4% after 25 days. Results above demonstrate that Anabaena PD-1 is a PCB-degrader with great potential for the in situ bioremediation of PCB-contaminated paddy soils.

Colorless polyimide copolymers derived from isomeric biphenyltetracarboxylic dianhydrides and 2,2′-bis(trifluoromethyl)benzidine

Yuanzhen Xu,Mengru Zhang,Yuanyuan Pang,Tianyue Zheng,Chun Tian,Zhen Wang,Jingling Yan

Colorless polyimide (CPI) copolymers were synthesized by one-step or two-step methods, using 2,2′-bis(trifluoromethyl)benzidine (TFMB), 2,2′,3,3′-biphenyltetracarboxylic dianhydride (3,3′-BPDA), and 3,3′,4,4′-biphenyltetracarboxylic dianhydride (4,4′-BPDA) as the monomers . These polymers exhibited a d -spacing value of 5.7–6.7 Å, and their d -spacing values increased while the intensity of π-π stacking interactions decreased with the increase of 3,3′-BPDA/TFMB contents. The solubility of the copolymers were gradually enhanced as the increase of 3,3′-BPDA/TFMB contents. Their coefficients of thermal expansion and glass transition temperatures increased with 3,3′-BPDA/TFMB contents, ranging from 15 to 68 ppm K −1 and 300 to 359 °C, respectively. These polymers exhibited excellent optical transparency ( λ 0  < 390 nm, T 450  > 83.2%, and b* < 4). CPI-4 and CPI-5 with 40 mol% and 20 mol% of 3,3′-BPDA/TFMB contents exhibited well-balanced solution processability and physical properties. The CPI copolymers can be potentially used in optical and optoelectronic fields owing to their desirable overall properties.

Insight into the application of magnetic molecularly imprinted polymers in soil-washing effluent: Selective removal of 4,4′-dibromodiphenyl ether, high adaptivity of material and efficient recovery of eluent

Yao Ma,Changyu Mao,Xiaodong Du,Chunsheng Xie,Jiangmin Zhou,Xueqin Tao,Zhi Dang,Guining Lu

Soil washing techniques can effectively remove soil polybrominated diphenyl ethers (PBDEs), but further removal of PBDEs from washing effluent is disrupted by environmental factors and coexisting organic matter. Hence, this work prepared novel magnetic molecularly imprinted polymers (MMIPs) to selectively remove PBDEs in soil washing effluent and recycling surfactants, with Fe 3 O 4 nanoparticles as the magnetic core, methacrylic acid (MAA) as the functional monomer , and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. Later, the prepared MMIPs were applied to adsorb 4,4′-dibromodiphenyl ether (BDE-15) in Triton X-100 soil-washing effluent and characterized by scanning electron microscopy (SEM), infrared spectrometry (FT-IR), nitrogen adsorption and desorption experiments. According to our observations, BDE-15 equilibrium adsorptions on dummy-template magnetic molecularly imprinted adsorbent (D-MMIP, 4-bromo-4′-hydroxyl biphenyl as template) and part-template magnetic molecularly imprinted adsorbent (P-MMIP, toluene as template) were reached within 40 min, and their equilibrium adsorption capacities were 164.54 μmol/g and 145.55 μmol/g, respectively, with imprinted factor α  > 2.03, selectivity factor β  > 2.14, and selectivity S > 18.05. MMIPs exhibited good adaptability to pH, temperature, and cosolvent. Our Triton X-100 recovery rate reached as high as 99.9%, and MMIPs maintained a more than 95% adsorption capacity after being recycled five times. Our results offer a novel approach to selectively remove PBDEs in soil-washing effluent, with efficient recovery of surfactants and adsorbents in soil-washing effluent.

Phytotoxicity and metabolic responses induced by tetrachlorobiphenyl and its hydroxylated and methoxylated derivatives in rice (Oryza sative L.)

Fangjing Lin,Jianteng Sun,Na Liu,Lizhong Zhu

Polychlorinated biphenyls (PCBs) and their biotransformation products, hydroxylated (OH-PCBs) and methoxylated derivatives (MeO-PCBs), have been detected in the environment and biota, especially crops. However, to date, little information is available on the phytotoxicity and metabolic responses induced by these chemicals in crops. In this study, we exposed rice ( Oryza sative L.) seedlings to 2,3,4,5-tetrachlorobiphenyl (CB-61) and its hydroxylated (4′-OH-CB-61) and methoxylated derivatives (4′-MeO-CB-61) at 0, 10, 50, 100 and 500 μg/L, respectively. After exposure for 14 days, significantly growth inhibition and oxidative damage were observed, among which the toxicities of 4′-OH-CB-61 and 4′-MeO-CB-61 were greater than that of the parent PCBs. Metabolomics analysis indicated that exposure to the three chemicals induced different metabolic responses. 4′-MeO-CB-61 mainly affected the saccharide catabolism, including pyruvate metabolism, the TCA cycle, the transfer of acetyl groups into mitochondria and the Warburg effect, resulting in a greater energy consumption. Moreover, both CB-61 and 4′-OH-CB-61 promoted several amino acid metabolism and fatty acid biosynthesis, thereby alleviating the potential ROS damage. This study for the first time evaluates and reveals the phytotoxicity of OH-PCBs and MeO-PCBs at the metabolic level, which attempts to provide important information for accurately evaluating the environmental risks of PCBs from the perspective of metabolism.

Oxidative dehalogenation and mineralization of polychlorinated biphenyls by a resuscitated strain Streptococcus sp. SPC0

Qihua Lin,Xinru Zhou,Shusheng Zhang,Junliang Gao,Mengqi Xie,Linqin Tao,Faqian Sun,Chaofeng Shen,Muhammad Zaffar Hashmi,Xiaomei Su

Most functional microorganisms cannot be cultivated due to entering a viable but non-culturable (VBNC) state, which limits the characterization and application of polychlorinated biphenyl (PCB)-degrading strains. Resuscitating VBNC bacteria could provide huge candidates for obtaining high-efficient PCB degraders. However, limited studies have focused on the ability of resuscitated strains for PCBs degradation. In the present study, whole-genome analysis of a resuscitated strain SPC0, and its performances in degradation of three prevalent PCB congeners (PCBs 18, 52 and 77) were investigated. The results indicate that the strain SPC0 belonged to the genus Streptococcus , possessed the degradation potential for aromatic xenobiotics . The SPC0 could effectively degrade PCBs 18 and 52, but exhibited lower degradation efficiency of PCB 77. Degradation of PCBs 18 and 52 could be fitted well by zero-order model, whereas the fittest model for PCB 77 degradation was pseudo second-order kinetics. The bph genes expression, chloride ions release and degradation metabolites identification, suggest that SPC0 possessed the capability of oxidative dehalogenation and mineralization of PCBs. Interestingly, SPC0 can degrade PCBs via the bph -encoded biphenyl pathway, and further mineralize metabolite dichlorobenzoate via protocatechuate pathway. This study is the first to show that a strain belonging to genus Streptococcus possessed PCB-degrading capability, which uncovered the powerful potential of resuscitated strains for bioremediation of PCB-contaminated sites.

High chemical stability poly(oxindole biphenylene)/ZrO2 porous separator for alkaline water electrolysis

Hongjing Li,Min Liu,Bin Hu,Xu Hu,Meizi He,Junhao Xin,Chengyuan Niu,Yingda Huang,Nanwen Li,Zushun Xu,Quanyuan Zhang

Porous separators are essential for alkaline electrochemical energy technologies, such as alkaline water electrolyzers (AWEs). These separators are promising alternatives to dense membranes because they offer the potential for low cost and high ionic conductivity. However, two significant challenges in developing efficient and durable alkaline energy systems are the suppression of hydroxide ions-induced degradation and the reduction of gas permeation in porous separators. Compared to hydrophobic polysulfone (PSF), KOH-doped poly (oxindole biphenylene) (POBP) is a highly stable ion-solvating polymer that can conduct both potassium and hydroxide ions. Building upon this, a series of porous separators based on POBP polymer with different zirconia loads were designed using NIPS to investigate the chemical stability and water electrolysis performance where the Z60 separator exhibits remarkable characteristics, including low area resistance (0.227 Ω cm 2 ), high bubble point pressure (0.813 MPa), low H 2 permeability (0.5 L min −1 ·cm −2 ) and exceptional 5040 h of ex-situ durability in 6 M KOH at 80 °C. Furthermore, the Z60 diaphragm showed a higher breaking time and lower weight loss after 261 h at 80 °C in the Fenton reagent, with the remaining mass being Z60 was 38 %. Notably, the Z60 diaphragm, when equipped with Ni–Al catalysts, achieved a current density of 0.5 A/cm 2 at 1.7 V, which surpasses that of Zirfon (0.5 A/cm 2 at 1.8 V). More importantly, when using Ni foam catalysts, the Z60 diaphragm demonstrated stable operation under 0.125 and 0.5 A/cm 2 current densities at 80 °C for more than 900 h and 1200 h, significantly improving the durability of the porous separators. The results of this study showed that the blending of POBP can lead to the development of highly chemically stable porous diaphragms, thereby opening up a new avenue for advanced AWEs.

Attainment and characterization of a microbial consortium that efficiently degrades biphenyl and related substances

Tao Xu,Teng Liu,Dawei Jiang,Ziyue Yuan,Xiaoqiang Jia

Persistent organic pollutants (POPs) mainly including polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), and organochlorine pesticides (OPs), have long-term harmful effects on human health and the environment. In this study, a natural consortium BP-W was attained from oil-contaminated soil using biphenyl as the sole carbon source, which is a relatively simple PAH and can form PCBs under certain catalytic conditions. PCR-DGGE techniques were used to analyze the structure of the consortium BP-W. The results showed that this consortium was composed of Chryseobacterium BP-1, Pseudomonas putida BP-2, Stenotrophomonas BP-3, Achromobacter BP-4, and an uncultured bacterium. The consortium BP-W degraded 99.88 % of 1 g L −1 biphenyl in 72 h. The degradation ratios of 100 mg L −1 naphthalene, phenanthrene, pyrene, and PCB77 in 5 days were 100 %, 97.54 %, 42.26 %, and 26.73 %, respectively. Notably, the consortium BP-W exhibited a high tolerance for these POPs. In the binary-substrate and ternary-substrate systems, the consortium BP-W was still able to rapidly degrade organic substrates. The results of gas chromatography-mass spectrometry showed that biphenyl was completely degraded by BP-W and entered the TCA cycle through the protocatechuic acid pathway, and no other highly toxic metabolites were produced during the degradation process. Thus, this consortium BP-W may have potential utility for the environmental-friendly and cost-effective bioremediation of POPs.

Biodegradation and chemotaxis of polychlorinated biphenyls, biphenyls, and their metabolites by Rhodococcus spp.

Wang Hui,Hu Jinxing,Xu Kai,Tang Xianjin,Xu Xinhua,Shen Chaofeng

Two biphenyl-degrading bacterial strains, SS1 and SS2, were isolated from polychlorinated biphenyl (PCB)-contaminated soil. They were identified as Rhodococcus ruber and Rhodococcus pyridinivorans based on the 16S rRNA gene sequence, as well as morphological, physiological and biochemical characteristics. SS1 and SS2 exhibited tolerance to 2000 and 3000 mg/L of biphenyl. And they could degrade 83.2 and 71.5% of 1300 mg/L biphenyl within 84 h, respectively. In the case of low-chlorinated PCB congeners, benzoate and 3-chlorobenzoate, the degradation activities of SS1 and SS2 were also significant. In addition, these two strains exhibited chemotactic response toward TCA-cycle intermediates, benzoate, biphenyl and 2-chlorobenzoate. This study indicated that, like the flagellated bacteria, non-flagellated Rhodococcus spp. might actively seek substrates through the process of chemotaxis once the substrates are depleted in their surroundings. Together, these data provide supporting evidence that SS1 and SS2 might be good candidates for restoring biphenyl/PCB-polluted environments.

Degradation and mineralization of 2-chlorobiphenyl by gamma radiation-activated persulfate system

Qin Wen,Shizong Wang,Jianlong Wang,Hai Chen,Yuedong Zhou

In this study, the degradation and mineralization of 2-chlorobiphenyl (2-PCB) by gamma irradiation were investigated in the presence of persulfate (PS), to examine the synergistic effect of radiation and persulfate oxidation. The results showed that the absorbed dose required for complete removal of 2-PCB decreased in the presence of PS. In addition, compared to a single irradiation system, the removal efficiency and mineralization of 2-PCB in deionized water , groundwater, and secondary effluent increased when irradiation combined with PS. 2-PCB at concentration of 4 mg L −1 to 8 mg L −1 could be efficiently degraded at a wide pH range (3.0–9.5) in irradiation-activated PS (irradiation/PS) system. The presence of 10 mM or 100 mM of Cl − , NO 3 − , CO 3 2− , and SO 4 2− in irradiation/PS system had no obvious influence on 2-PCB removal. 2-PCB was degraded under the combined action of sulfate radicals (SO 4 •− ) and hydroxyl radicals (·OH), in which ·OH contributed more. The irradiation/PS system is promising to treat PCB-containing wastewater.
