For preparation of resveratrol and arctigenin from peanut hulls and arctium lappa fruits, respectively, a recombinant β-glucosidase (TmBglA) from hyperthermophile Thermotoga maritima was purified and characterized. The hydrolytic activity was the highest at 90 °C and pH 6.2 for arctiin with Km of 1.61 mM and kcat of 197.4 s−1, and 90 °C and 5.8 for polydatin with Km of 0.38 mM and kcat of 47.6 s−1. The enzyme produced 215.4 mg L−1 resveratrol and 355.7 mg L−1 arctigenin from 400 mg L−1 polydatin and 540 mg L−1 arctiin after 60 min of incubation at 80 °C, with capable of hydrolyzing up to 92.1 and 94.9% of polydatin and arctiin, respectively. The enzymatic hydrolysis of peanut hulls and fructus arctii displayed a conversion yield of 3.8 and 0.33 mg resveratrol and arctigenin per gram of substrate material flour, respectively. Of the reported β-glucosidase, TmBglA exhibited the highest thermostability, kcat, kcat/Km, and conversion productivity for hydrolyzing polydatin and arctiin, and has great potential applications in functional food and medicine production.
Gout is a disease that affects quality of life and has economic implications. However, most patients with gout are not adequately managed due to the adverse effects of current medical treatments. This study was performed to investigate the transdermal delivery of polydatin via the combination of inclusion complexes and dissolving microneedles and the therapeutic effect of polydatin on the treatment of acute gout arthritis . In vitro cytotoxicity of polydatin on HaCaT cells was measured by a CCK-8 assay. The results showed that polydatin exerted less cytotoxicity compared to indomethacin and colchicine . The aqueous solubility of polydatin increased considerably to 124.47 mg/mL via the formation of polydatin/Hydroxypropyl-β-cyclodextrin inclusion complexes. The complexes loaded dissolving microneedles showed significant improvement for in vitro polydatin permeation compared to the absence of complexes. Pharmacodynamic studies indicated that the polydatin complexes-loaded dissolving microneedles had significant inhibitory effects on swelling of the ankle in the arthritic mice. Furthermore, compared to the oral colchicine treatment, the body weights of the polydatin-treated mice increased significantly during the treatment, which implied a lower toxicity of polydatin microneedles delivery. The overall results indicate that the combination of inclusion complexes with dissolving microneedles provides a potential route for treatment of acute gout arthritis by enhanced transdermal delivery of polydatin.

Based on similar chemical structures and reactive sites with dopamine, polyphenols and some amines can react and generate adhesive polymers through oxidation and Michael addition/Schiff base reactions. In this study, four kinds of polyphenol-amine codeposition solutions were applied in surface modification of anion exchange membranes used in electrodialysis (ED). Catechol (CA)/tannic acid (TA) and m-phenylenediamine (MPD)/polyethylenimine (PEI) were respectively selected as polyphenol monomer and amine monomer, with chemical structural differences in the length of the carbon chain, the number of benzene rings, and functional groups. The effects of monomer type on copolymerization and codeposition process were investigated. Results showed that polyphenol types had important influences on chemical reactions and deposition. The codeposition solutions containing CA, especially the CA-MPD solution with two small molecular monomers, exhibited active and fast reaction rates, reflected in the changes in solution color and UV–visible absorption. By contrast, the flocculent between macromolecular monomers of TA and PEI changed the presence of solution, intermediates, and polymeric products, promoting deposition rates. In ED experiments, all modified AEMs maintained the desalination efficiency and demonstrated a better antifouling ability. However, the modified AEMs could not totally resist foulants and protect from the negative effects of fouling; regulating and improving codeposition modification need more investigation.
