One-pot solvent-free green strategy to fabricate ultra-efficient polyphosphoester flame retardant for Poly(Lactic acid)

Zhicheng Bai,Feixiang Yin,Jiahao Shen,JinJia Xu,Fengqing Chen,Teng Huang,Hufeng Li,Dan Xie,Yi An,Wenbiao Zhang,Jinfeng Dai

Tremendous attention has been dedicated to crafting eco-friendly flame retardants with excellent comprehensive performance via green means. Herein, a novel ultra-efficient polyphosphoester (PPE) flame retardant was synthesized via a facile one-pot method of solid-phase polycondensation reaction. PPE was then employed as a multifunctional additive for high-performance poly (lactic acid) (PLA). Benefiting from the dual-phase flame-retardant effect, the resultant PLA composite with merely 0.5 wt% PPE exhibited a desired limiting oxygen index (LOI) value of 26.1 %, and a satisfactory V0 rating of UL-94 (1.6 mm) with well-preserved mechanical properties. Due to the nucleating effect of PPE on PLA, the mechanical properties and crystallization ability of the PLA composites were both gradually enhanced as PPE content increased from 0.3 to 1.5 wt%. The designed ultra-efficient approach, enhancing fire safety and mechanical properties simultaneously at a low content, lays a foundation of an affordable, eco-friendly, and versatile flame retardant for potential applications of PLA.

Effect of Hydrophobicity of Core on the Anticancer Efficiency of Micelles as Drug Delivery Carriers

Chun-Yang Sun,Yin-Chu Ma,Zi-Yang Cao,Dong-Dong Li,Feng Fan,Jun-Xia Wang,Wei Tao,Xian-Zhu Yang

Recently, micelles, which are self-assembled by amphiphilic copolymers, have attracted tremendous attention as promising drug delivery systems for cancer treatment. Thus, the hydrophobic core of the micelles, which could efficiently encapsulate small molecular drug, will play a significant role for the anticancer efficiency. Unfortunately, the effect of hydrophobicity of micellar core on its anticancer efficiency was rarely reported. Herein, the amphiphilic diblock polymers of poly(ethylene glycol) and polyphosphoester with different side groups (butyl, hexyl, octyl) were synthesized to tune the hydrophobicity of the micellar core. We found that the in vitro cytotoxicity of the DOX-loaded micelles decreased with the increasing hydrophobicity of micellar core due to the drug release rate. However, following systemic delivery, the DOX-loaded micelles with the most hydrophobic core exhibited the most significant inhibition of tumor growth in a MDA-MB-231 tumor model, indicating the importance of hydrophobicity of core on the antitumor efficacy of drug delivery systems.

Functionalized Polyphosphoester via Living Ring-opening Polymerization and Photochemical Thiol-ene Click Reaction

Wang Ying,Wang Qiliao,Hou Liman,Zhou Mingdong,Dong Dewen,Zhang Ning

Poly(ethylene phosphonate) was synthesized via the living ring-opening polymerization of cyclic phosphonate monomer catalyzed by organocatalyst. The pendant vinyl functionalities were employed to perform the photochemical click reactions with thiols. We demonstrated that both small thiol molecules and macromolecular thiols could be efficiently coupled into the PPE side chains, enabling the rapid and efficient functionalization of polyphosphoesters( PPE).

Synthesis of an Oxidation-Sensitive Polyphosphoester Bearing Thioether Group for Triggered Drug Release

Jihong Wang,Dongdong Li,Wei Tao,Yang Lu,Xianzhu Yang,Jun Wang

In this work, novel amphiphilic diblock copolymers of polyethylene glycol and polyphosphoester with pendant thioether groups, denoted as mPEG-b-PMSPEP, were synthesized through the ring-opening polymerization of functionalized cyclic phosphoester monomer using methoxy poly(ethylene glycol) and Sn(Oct)2 as the macroinitiator and catalyst, respectively. The successful synthesis was confirmed by 1H, 13C, 31P nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC). These amphiphilic block copolymers self-assembled spontaneously in the aqueous solution, and the formed nanoparticles were sensitive to the oxidation that induced the hydrophobic to hydrophilic transition for its PMSPEP core under triggering of H2O2 and the subsequent dissociation of the nanoparticles. In addition, the reactive oxygen species (ROS) generated by light and the photosensitizer were also capable of carrying out the oxidation of these nanoparticles. Their oxidation profiles were systemically evaluated by 1H NMR. Finally, the mPEG-b-PMSPEP nanoparticles were used to coencapsulate the photosensitizer chlorin e6 (Ce6) and anticancer drug paclitaxel (PTX), achieving the photoaccelerated PTX release via oxidation of the nanoparticles by the generated ROS under light irradiation. Meanwhile, the in vitro cytotoxicity assays indicated that these nanoparticles coencapsulated with PTX and Ce6 showed a combined cell-killing effect toward MDA-MB-231 tumor cells, exhibiting great potential for drug delivery systems that realize the synergistic chemo-photodynamic therapy for cancer treatment.

ROS-responsive selenium-containing polyphosphoester nanogels for activated anticancer drug release

Yan Zhang,Chuan Ma,Sihang Zhang,Chao Wei,Ying Xu,Wei Lu

Nowadays, the stability and on-demand release of drug carriers are still to be solved. To meet the demand of these issues, we developed a reactive oxygen species (ROS) responsive selenium-containing polyphosphoesters nanogel (PSeP) by a facile one-step ring-opening polymerization of the novel monomer 4-selenoctane-1,8-diyl bis(propylphosphatelane) (Se-COP) with polyethylene glycol (mPEG) employed as the macroinitiator. Their structure was confirmed by NMR, FT-IR and GPC. The crosslinked core-shell structure imparted the nanogels with excellent dimensional stability according to the dynamic light scattering (DLS) and transmission electron microscopy (TEM). Moreover, the selenide groups endowed the nanogels with rich ROS responsiveness when subjected to the stimuli of H 2 O 2 , thus the drug-loaded PSeP nanogels displayed swollen behaviors leading to an activated doxorubicin hydrochloride (DOX · HCl) release. The release mechanisms fitted by the Ritger-Peppas power-law model also proved the swollen release process. MTT assays exhibited that the PSeP nanogels were nontoxic up to a tested concentration of 50 μg mL −1  by A549 and HEK293, and the DOX-loaded PSeP had a high anti-cancer behaviour against A549 cancer cells. Additionally, these nanogels possessed enhanced intracellular drug release by CLSM . Therefore, these results highlighted that the selenium-containing polyphosphoesters nanogels could be a potential platform for the ROS-sensitive drug delivery.

Polyphosphoester-Camptothecin Prodrug with Reduction-Response Prepared via Michael Addition Polymerization and Click Reaction

Xueqiong Du,Yue Sun,Mingzu Zhang,Jinlin He,Peihong Ni

Polyphosphoesters (PPEs), as potential candidates for biocompatible and biodegradable polymers, play an important role in material science. Various synthetic methods have been employed in the preparation of PPEs such as polycondensation, polyaddition, ring-opening polymerization, and olefin metathesis polymerization. In this study, a series of linear PPEs has been prepared via one-step Michael addition polymerization. Subsequently, camptothecin (CPT) derivatives containing disulfide bonds and azido groups were linked onto the side chain of the PPE through Cu(I)-catalyzed azidealkyne cyclo-addition “click” chemistry to yield a reduction-responsive polymeric prodrug P(EAEP-PPA)-g-ss-CPT. The chemical structures were characterized by nuclear magnetic resonance spectroscopy, gel permeation chromatography, Fourier transform infrared, ultraviolet–visible spectrophotometer, and high performance liquid chromatograph analyses, respectively. The amphiphilic prodrug could self-assemble into micelles in aqueous solution. The average particle size and morphology of the prodrug micelles were measured by dynamic light scattering and transmission electron microscopy, respectively. The results of size change under different conditions indicate that the micelles possess a favorable stability in physiological conditions and can be degraded in reductive medium. Moreover, the studies of in vitro drug release behavior confirm the reduction-responsive degradation of the prodrug micelles. A methyl thiazolyl tetrazolium assay verifies the good biocompatibility of P(EAEP-PPA) not only for normal cells, but also for tumor cells. The results of cytotoxicity and the intracellular uptake about prodrug micelles further demonstrate that the prodrug micelles can efficiently release CPT into 4T1 or HepG2 cells to inhibit the cell proliferation. All these results show that the polyphosphoester-based prodrug can be used for triggered drug delivery system in cancer treatment.

Renewable isosorbide-derived poly(phosphoester) for simultaneously enhanced flame-retardancy and mechanical property of polylactide

Xin Wang,Haoxin Niu,Wenwen Guo,Lei Song,Yuan Hu

Incorporating flame retardant fillers is the most efficient way to improve the fire resistance of polylactide (PLA). However, most flame retardant fillers result in deterioration of the mechanical property of PLA so far. Therefore, to create high-performance PLA materials combining excellent flame retardancy with superior mechanical properties remains highly attractive and challenging. In this study, a novel isosorbide-derived poly(phosphoester) (PIDI) was synthesized and used as a bio-based flame-retardant for PLA. The addition of PIDI showed a marginal effect on the change of the glass transition temperature of the PLA composites, but the crystallinity degree of the PLA composites was improved by the inclusion of PIDI. Benefitting from the improved crystallinity, the tensile strength, flexural strength, and impact strength of PLA composite containing 15 wt% PIDI was enhanced by 19.4%, 14.1%, and 36.9%, respectively, compared to the virgin PLA. With the addition of 20 wt% PIDI, the PLA composite showed an LOI value of 25.5% and a UL-94 V-0 rating in the vertical burning test. The flame retardant effect of PIDI was also demonstrated by a notable reduction (−46%) in the peak heat release rate (PHRR) of PLA composite containing 20 wt% PIDI. Such a renewable isosorbide-derived poly(phosphoester) is one of the promising candidates for PLA bio-composites with an outstanding balance between flame retardancy and intrinsic physical properties.

Polyphosphoester-modified Cellulose Nanocrystals for Stabilizing Pickering Emulsion Polymerization of Styrene

Che Kun-Ming,Zhang Ming-Zu,He Jin-Lin,Ni Pei-Hong

The structure and properties of functional nanoparticles are important for stabilizing Pickering emulsion polymerization. Recently, cellulose nanocrystals (CNCs) are increasingly favored as a bio-based stabilizer for Pickering emulsions. In this study, we reported a novel functionalized polyphosphoester-grafted CNCs for the stabilization of oil-in-water Pickering emulsions and the emulsion polymerization of styrene. First, polyphosphoester containing an amino group at one end of the chain, abbreviated as PBYP-NH 2 , was prepared by ring-opening polymerization (ROP) and hydrolysis reaction, wherein PBYP represents poly[2-(but-3-yn-1-yloxy)-2-oxo-1,3,2-dioxaphospholane]. Subsequently, CNC-COOH was obtained via 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) oxidation of CNCs. The functionalized nanocrystals CNC-PBYP-COOH with carboxyl groups and polyphosphoester on the surface were obtained by the amidation reaction of PBYP-NH 2 with CNC-COOH. Finally, we used CNC-PBYP-COOH as sole particle emulsifiers to stabilize styrene-in-water Pickering emulsions and studied its effects on the emulsions in details by using dynamic light scattering (DLS). The results indicated that the properties of these emulsions depended on the concentration of hydrophobically modified CNCs, volume ratios of oil to water, and pH values. The modified CNCs had higher ability to stabilize the styrene-in-water emulsions relative to the unmodified CNCs, and a stable oil-in-water (o/w) Pickering emulsion with diameter of hundreds of nanometers could be obtained. The resulting emulsions could be polymerized to yield nanosized latexes. The polyphosphoester-modified CNCs as green particle emulsifiers can efficiently stabilize nanoemulsions and latexes, which would promote the development of novel environmentally friendly materials.

Phenylboronic acid-functionalized unimolecular micelles based on a star polyphosphoester random copolymer for tumor-targeted drug delivery

Li Zhang,Dongjian Shi,Yunyun Gao,Tianyang Zhou,Mingqing Chen

To overcome the thermodynamic instability of polymeric micelles in an extremely high dilution bio-environment, unimolecular micelles with covalently-bound molecular architectures have been widely studied for drug delivery. In this work, an amphiphilic multi-arm star random copolymer, polyamidoamine-poly(butenyl phospholane-co-methoxy phospholane) with phenylboronic acid end groups (abbreviated as PAMAM-P(BEP-co-MP)-PBA), was synthesized by a simple one-pot ring-opening polymerization with the fourth-generation PAMAM dendrimer as a macro-initiator. Then, unimolecular micelles (UMs) were formed by the obtained copolymers in a dilute aqueous solution. Dynamic light scattering (DLS) and transmission electron microscopy (TEM) results showed that the UMs possess uniform spherical structures and stable hydrodynamic sizes (ca. 20 nm). Besides, further information on polymer repeat unit numbers could be theoretically calculated by comparison of hydrodynamic sizes and contour lengths. Doxorubicin (DOX) could be encapsulated into the inner hydrophobic regions of the UMs. The obtained DOX-loaded UMs exhibited a slow and sustainable release behavior in an intracellular simulated environment. Furthermore, in vitro cytotoxicity and cellular uptake studies demonstrated that the DOX-loaded UMs could specifically recognize and accumulate in HepG2 cells, and then inhibit tumor cell growth effectively. Therefore, this PBA-functionalized unimolecular micelle could be a promising candidate for targeted drug delivery.

Dual-Responsive Polyphosphoester-Doxorubicin Prodrug Containing a Diselenide Bond: Synthesis, Characterization, and Drug Delivery

Guoqing Ma,Jie Liu,Jinlin He,Mingzu Zhang,Peihong Ni

The development of novel stimuli-responsive and biodegradable polyphosphoester-anticancer prodrugs is of importance in designing water-soluble prodrugs utilized in the field of drug delivery. In this study, the focus is on the synthesis of biocompatible and biodegradable diselenide-containing polyphosphoester [PEEP-b-PBYP-Se]2 using reduction-responsive di(1-hydroxylundecyl) diselenide as an initiator to polymerize 2-(but-3-yn-1-yloxy)-2-oxo-1,3,2-dioxaphospholane (BYP) and 2-ethoxy-2-oxo-1,3,2-dioxaphospholane (EOP). After that, a doxorubicin (DOX) derivative containing an azide group was linked onto the side chain of [PEEP-b-PBYP-Se]2 via the Cu(I)-catalyzed azide–alkyne cycloaddition (CuAAC) “click” reaction to yield a pH/reduction-responsive polymeric prodrug, namely [PEEP-b-(PBYP-hyd-DOX)-Se]2. The chemical structures of various polymers were characterized by nuclear magnetic resonance spectroscopy, ultraviolet–visible spectrophotometer, Fourier transform infrared spectroscopy, and high-performance liquid chromatography. The self-assembly behavior measured by dynamic light scattering and transmission electron microscopy clearly supported the formation of the prodrug nanoparticles (NPs). The results indicated that the polymeric prodrug NPs were relatively uniform spheres that could maintain stability in a physiological condition but be cleaved in acidic or reductive medium. Furthermore, the pH- and reduction-responsive properties of the prodrug NPs were investigated via drug release in vitro in different media. It turned out that the drug was efficiently released in acidic or reductive medium compared with that under physiological conditions. The results of methyl thiazolyl tetrazolium assays confirmed the favorable biocompatibility of [PEEP-b-PBYP-Se]2. Moreover, the cell cytotoxicity and intracellular uptake experiments were carried out to verify the efficient cellular proliferation inhibition. This finding contributes to the design of a novel diselenide-containing polyphosphoester-doxorubicin prodrug.

Highly-controllable drug release from core cross-linked singlet oxygen-responsive nanoparticles for cancer therapy

Jiayan Zhou,Chunyang Sun,Chunshui Yu

Highly-controllable release consisting of preventing unnecessary drug leakage at physiologically normal tissues and triggering sufficient drug release at tumor sites is the main aim of nanoparticle-based tumor therapy. Developing drug-conjugation strategies with covalent bonds in response to a characteristic stimulus, such as reactive oxygen species (ROS) generated by photodynamic therapy (PDT) has attracted much attention. ROS can not only cause cytotoxicity, but also trigger the cleavage of ROS-responsive linkers. Therefore, it is feasible to design a new model of controlled drug release via the breakage of ROS-responsive linkers and degradation of nanoparticles. The self-supply of the stimulus and highly-controllable drug release can be achieved by encapsulation of photosensitizer (PS) and chemotherapeutic drugs simultaneously without any support of tumor endogenous stimuli. Therefore, we used thioketal (TK) linkers as the responsive linkers due to their reaction with singlet oxygen (1O2, SO), a type of ROS. They were conjugated to the side groups of polyphosphoesters (PPE) via click chemistry to acquire the core cross-linked SO-responsive PPE nanoparticles poly(thioketal phosphoesters) (TK-PPE). TK-PPE coated with the photosensitizer chlorin e6 (Ce6) and chemotherapeutic drug doxorubicin (DOX) simultaneously were prepared and named as TK-PPECe6&DOX. TK-PPECe6&DOX kept stable due to the high stability of the TK-linkers in the normal physiological environment. With self-production of SO as the stimulating factor from the encapsulated Ce6, highly-controlled drug release was achieved. After incubation of tumor cells, 660 nm laser irradiation induced SO generation, resulting in the cleavage of TK-linkers and boosted-release of DOX. Highly-controllable drug release of TK-PPECe6&DOX through self-production of stimulus increased antitumor efficacy, offering a promising avenue for clinical on-demand chemotherapy.

Synthesis and characterization of UPPE-PLGA-rhBMP2 scaffolds for bone regeneration

Tian Zhichao,Zhu Yuanli,Qiu Jinjun,Guan Hanfeng,Li Liangyu,Zheng Shouchao,Dong Xuehai,Xiao Jun

A novel unsaturated polyphosphoester (UPPE) was devised in our previous research, which is a kind of promising scaffold for improving bone regeneration. However, the polymerization process of UPPE scaffolds was unfavorable, which may adversely affect the bioactivity of osteoinductive molecules added if necessary, such as recombinant human bone morphogenetic protein-2 (rhBMP2). The purpose of this study was to build a kind of optimal scaffold named UPPE-PLGA-rhBMP2 (UPB) and to investigate the bioactivity of rhBMP2 in this scaffold. Furthermore, the cytotoxicity and biocompatibility of UPB scaffold was assessed in vitro . A W1/O/W2 method was used to fabricate PLGA-rhBMP2 microspheres, and then the microspheres were added to UPPE for synthesizing UPB scaffold. The morphological characters of PLGA-rhBMP2 microspheres and UPB scaffolds were observed under the scanning electron microscopy and laser scanning confocal microscopy. The cumulative release of UPB scaffolds was detected by using ELISA. The cytotoxicity and biocompatibility of UPB scaffolds were evaluated through examining the adsorption and apoptosis of bone marrow stromal cells (bMSCs) seeded on the surface of UPB scaffolds. The bioactivity of rhBMP2 in UPB scaffolds was assessed through measuring the alkaline phosphates (ALP) activity in bMSCs seeded. The results showed that UPB scaffolds sequentially exhibited burst and sustained release of rhBMP2. The cytotoxicity was greatly reduced when the scaffolds were immersed in buffer solution for 2 h. bMSCs attached and grew on the surface of soaked UPB scaffolds, exerting well biocompatibility. The ALP activity of bMSCs seeded was significantly enhanced, indicating that the bioactivity of rhBMP2 remained and still took effect after the unfavorable polymerization process of scaffolds. It was concluded that UPB scaffolds have low cytotoxicity, good biocompatibility and preserve bioactivity of rhBMP2. UPB scaffolds are promising in improving bone regeneration.
