Huganning, a Chinese patent medicine, has been used in clinical practice for the treatment of hepatitis in China for more than 20 years, and its main physiologically active components are polydatin and emodin. In this study, a rapid and accurate proton nuclear magnetic resonance method for the quantification of these constituents was developed. 4-Nitrochlorobenzene was selected as an internal standard, and the signals for polydatin at δ 6.44 ppm and emodin at δ 7.54 ppm were chosen for the quantification. Validation of the method was performed in terms of specificity, precision, and stability. The results indicated that the method was precise and in line with the China Food and Drug Administration guidance. The method has been successfully applied to characterize tablets and capsules obtained from six pharmaceutical companies. This study indicated that the rapid 1 H NMR method is more suitable for the quality control of Huganning than the high-performance liquid chromatography method.

The discovery of enzyme inhibitors from natural products is a crucial aspect in the development of therapeutic drugs. However, the complexity of natural products presents a challenge in developing simple and efficient methods for inhibitor screening. Herein, we have developed an integrated analytical model for screening xanthine oxidase (XOD) inhibitors that combines simplicity, accuracy, and efficiency. This model utilizes a colorimetric sensor and affinity chromatography technology with immobilized XOD . The colorimetric sensor procedure can quickly identify whether there are active components in complex samples. Subsequently, the active components in the samples identified by the colorimetric sensor procedure were further captured, separated, and identified through affinity chromatography . The integrated analytical model can significantly enhance the efficiency and accuracy of inhibitor screening. The proposed method was applied to screen for an activity inhibitor of XOD in five natural medicines. As a result, a potential active ingredient for XOD , polydatin , was successfully identified from Polygoni Cuspidati Rhizoma et Radix. This work is anticipated to offer new insights for the screening of enzyme inhibitors from natural medicines.

Continuous fluid catalysis technology is widely used in industry as traditional fluid reactors have unstable reaction efficiency and poor production conversion. Herein, a continuous fluid reactor with porous directional channel was prepared from natural wood (xylem reactor). When the reactor was used for continuous fluid catalysis, fluid from adjacent channels collided through the pores in the inner walls of the directional channels, creating fluid disturbances that caused full contact between the fluid and enzymes in the reactor. Thus, the porous directional channel structure improved mass transfer and catalytic efficiency, which led to a high biological conversion rate. The xylem reactor exhibited satisfactory catalytic performance and catalyzed the hydrolysis of polydatin at 60 °C for 4 h with an optimum conversion of 99.44%. Additionally, the xylem reactor prevented pressure buildup (0.34 kPa) and displayed cycling stability. The xylem reactor with porous directional channel structure exhibited excellent application potential suitable for sustainable development and green chemistry owing to its low preparation cost, minimal environmental impact and high efficiency.

Cisplatin induced acute kidney injury (AKI) is clinically prevalent, with a complex pathogenesis and a lack of effective therapeutic drugs. Polydatin (Po) has excellent biological activity, but its low solubility and bioavailability limit its application. In this study, fucoidan (Fu) and carboxymethyl chitosan (Cs) self-assembled into nanoparticles through electrostatic interactions/hydrogen bonding and loaded Po (Fu/Cs Po NPs). In vitro studies found that Fu/Cs Po NPs protected human renal tubular epithelial (HK-2) cells from cisplatin induced damage and accumulation of reactive oxygen species (ROS). Mechanistic studies showed that Fu/Cs Po NPs inhibited cisplatin induced DNA damage and activation of cyclic guanosine monophosphate synthase (cGAS) and intron gene stimulator (STING) pathways. In vivo studies showed that Fu/Cs Po NPs treatment alleviated cisplatin induced AKI symptoms, including elevated blood urea nitrogen (BUN) and serum creatinine (SCr), as well as pathological damage to kidney tissues. In vivo mechanism studies also showed that Fu/Cs Po NPs treatment inhibited cisplatin induced DNA damage and activation of the cGAS-STING pathway. The pharmacokinetic and tissue distribution results demonstrated that the Fu/Cs delivery system enhanced the bioavailability and kidney accumulation of Po in vivo . In summary, our study provided potential drugs for the treatment of cisplatin induced AKI.
