BACKGROUND The objectives of this study were to investigate the effects of taurine on rumen fermentation, rumen bacterial community and metabolomics, nitrogen metabolism and plasma biochemical parameters in beef steers. Six castrated Simmental steers (liveweight 402 ± 34 kg) and three levels of taurine (0, 20, 40 g d −1 ) were assigned in a replicated 3 × 3 Latin square design. Each experimental period included 15 days for adaptation and 5 days for sampling. RESULTS Supplementing taurine did not affect the ruminal pH or concentrations of ammonia nitrogen and volatile fatty acids ( P > 0.10), but linearly increased the ruminal concentrations of taurine ( P < 0.001) and microbial crude protein ( P = 0.041). Supplementing taurine linearly increased the neutral detergent fiber digestibility ( P = 0.018), and tended to linearly increase dry matter digestibility ( P = 0.095), tended to increase the fecal nitrogen excretion ( P = 0.065) and increased the urinary taurine excretion ( P < 0.001). Supplementing taurine quadratically increased the plasma concentration of triglycerides ( P = 0.017), tended to linearly decrease growth hormone ( P = 0.074), but did not affect other plasma parameters ( P > 0.10). Supplementing taurine modified the rumen bacterial community and increased the ruminal concentration of taurine metabolite 2-hydroxyethoxysulfonic acid ( P < 0.001). CONCLUSION It was concluded that taurine improved ruminal microbial crude protein synthesis and increased fiber digestibility through modifying rumen bacterial community. It is necessary to clarify the ruminal hydrolysis of taurine in steers. © 2023 Society of Chemical Industry.

Two consecutive experiments were conducted to investigate the effects of dietary supplementation with red cabbage extract (RCE) rich in anthocyanins on the nitrogen (N) metabolism in beef bulls and the nitrous oxide (N 2 O) emissions from the soil applied with the urine of beef bulls. In Experiment 1, 8 Simmental beef bulls (body weight 387.9 ± 40.7 kg) were used as experimental animals . Two levels of RCE, i.e. 0 and 114 g dry matter per day, were supplemented to a basal ration as experimental treatments. The animals and the treatments were allocated in a 2 × 2 crossover design. In Experiment 2, the static incubation technique was used to determine the N 2 O-N emissions from the soil applied with the urine samples collected from Experiment 1. The results of Experiment 1 indicated that RCE supplementation did not affect the N excretion, N retention, and N retention rate in beef bulls ( P > 0.10). However, RCE supplementation increased the ratio of hippuric acid-N/urinary N ( P < 0.05) and tended to increase the ratio of creatinine-N/urinary N ( P = 0.081) without changing the excretion of other urinary nitrogenous components ( P > 0.10). Plasma metabolome analysis indicated that RCE supplementation upregulated 12 metabolites while downregulated 18 metabolites ( P < 0.05). The results of Experiment 2 indicated that RCE supplementation reduced the estimated urine N 2 O-N emissions by 33.1% through decreasing the ratio of N 2 O-N/urine-N ( P < 0.05). In conclusion, RCE supplementation did not affect the N retention and N excretion of beef bulls, whereas it attenuated the urine N 2 O-N emissions of beef bulls through increasing urinary hippuric acid excretions.

The aims of the present study were to investigate the effects of dietary supplementation with rumen-protected taurine (RPT) on whole-body protein turnover, plasma metabolomics, and whole blood cell transcriptomics in steers. Eight steers, averaging 220 ± 3.26 (mean ± SD) kg of liveweight, were allocated in a replicate 4 × 4 Latin square design. The experimental treatments consisted of four levels of RPT supplementation: 0, 25, 50, and 75 g RPT per day, added to a basal diet. The results showed that supplementation with RPT linearly decreased the fecal nitrogen (N) excretion ( P = 0.001) and the 15 N fractional recovery rate ( P = 0.047), while it linearly increased the urinary excretion of taurine ( P = 0.045) as well as the average daily weight gain ( P = 0.003), protein synthesis ( P < 0.001), protein degradation ( P < 0.001) and the whole-body protein turnover ( P < 0.001). Supplementation with RPT linearly increased the plasma concentrations of growth hormone ( P = 0.005) and quadratically affected the plasma concentration of insulin-like growth factor-1 ( P = 0.013), and it linearly decreased the plasma concentration of albumin ( P = 0.022). Supplementation with RPT altered the whole blood cell mRNA expression and upregulated the expressions of the marker genes, including RPS6KB1 , PRSS42 , COL1A2 , ENSBTAG00000013055 and ENSBTAG00000038159 which are related to protein metabolism. The plasma metabolome profiling indicated that supplementation with RPT upregulated the plasma concentrations of taurine, lysine and methionine. The experiment revealed the impact and the mechanisms of taurine on driving whole-body protein turnover and protein accretion in steers. Two novel marker genes which could be related to body protein degradation in steers were identified.

Carbapenems are considered a last resort for the treatment of multi-drug-resistant bacterial infections in humans. In this study, we investigated the occurrence of carbapenem-resistant bacteria in feedlots in Alberta, Canada. The presumptive carbapenem-resistant isolates (n= 116) recovered after ertapenem enrichment were subjected to antimicrobial susceptibility testing against 12 different antibiotics, including four carbapenems. Of these, 72% of the isolates (n= 84) showed resistance to ertapenem, while 27% of the isolates (n= 31) were resistant to at least one other carbapenem, with all except one isolate being resistant to at least two other drug classes. Of these 31 isolates, 90% were carbapenemase positive, while a subset of 36 ertapenem-only resistant isolates were carbapenemase negative. The positive isolates belonged to three genera;Pseudomonas,Acinetobacter, andStenotrophomonas, with the majority beingPseudomonas aeruginosa(n= 20) as identified by 16S rRNA gene sequencing. Whole genome sequencing identified intrinsic carbapenem resistance genes, includingblaOXA-50and its variants (P. aeruginosa),blaOXA-265(A. haemolyticus),blaOXA-648(A. lwoffii),blaOXA-278(A. junii), andblaL1andblaL2(S. maltophilia). The acquired carbapenem resistance gene (blaPST-2) was identified inP. saudiphocaensisandP. stutzeri. In a comparative genomic analysis, clinicalP. aeruginosaclustered separately from those recovered from bovine feces. In conclusion, despite the use of selective enrichment methods, finding carbapenem-resistant bacteria within a feedlot environment was a rarity.Keywords:carbapenem-resistant bacteria;antimicrobial resistance;beef production system;whole genome sequencing

This study investigated the effects of β-alanine (β-Ala) on ruminal fermentation, nutrient digestibility, nitrogen (N) metabolism, plasma biochemical parameters, and rumen bacterial communities in beef steers. Six steers with initial liveweight of 252.8 ± 5.2 kg and 3 treatments of supplementing with 0, 30, or 60 g β-Ala per day to the basal diet were allocated in a replicated 3 × 3 Latin square design. Each experimental period was 20 d, of which the first 15 d were for adaptation and the subsequent 5 d were for sampling. The results showed that β-Ala linearly increased the ruminal concentration of microbial crude protein (MCP) ( P = 0.002), but it did not affect the ruminal concentrations of ammonia N and total volatile fatty acids ( P > 0.10). β-Ala also linearly increased the dry matter (DM) ( P = 0.009), organic matter (OM) ( P = 0.017) and crude protein (CP) ( P = 0.043) digestibility, tended to decrease the acid detergent fiber digestibility ( P = 0.077), but it did not affect the neutral detergent fiber digestibility ( P = 0.641). β-Ala quadratically increased the relative abundance of ruminal Bacteroidota ( P = 0.021) at the phylum level, and increased Prevotella ( P = 0.028), and Prevotellaceae_UCG-003 ( P = 0.014) and decreased the relative abundance of NK4A214_group ( P = 0.009) at the genus level. Feeding steers with β-Ala linearly increased the urinary N excretion ( P = 0.006) and urea excretion ( P = 0.002) and the N retention ( P = 0.004), but it did not affect the N utilization efficiency ( P = 0.120). β-Ala quadratically increased the plasma concentrations of albumin ( P = 0.048) and linearly increased the plasma concentration of insulin-like growth factor-1 ( P < 0.001) and the total antioxidant capacity ( P = 0.011). In summary, dietary supplementation with β-Ala improved the rumen MCP supply and increased the digestibility of DM, OM, CP and the N retention. Further research is necessary to verify the ruminal degradability of β-Ala and to investigate the mechanism of the impact of absorbed β-Ala on the anti-oxidative ability of the steers.
