Myristic acid as a checkpoint to regulate STING-dependent autophagy and interferon responses by promoting N-myristoylation

Jia Mutian,Wang Yuanyuan,Wang Jie,Qin Danhui,Wang Mengge,Chai Li,Fu Yue,Zhao Chunyuan,Gao Chengjiang,Jia Jihui,Zhao Wei

Stimulator of interferon gene (STING)-triggered autophagy is crucial for the host to eliminate invading pathogens and serves as a self-limiting mechanism of STING-induced interferon (IFN) responses. Thus, the mechanisms that ensure the beneficial effects of STING activation are of particular importance. Herein, we show that myristic acid, a type of long-chain saturated fatty acid (SFA), specifically attenuates cGAS-STING-induced IFN responses in macrophages, while enhancing STING-dependent autophagy. Myristic acid inhibits HSV-1 infection-induced innate antiviral immune responses and promotes HSV-1 replication in mice in vivo. Mechanistically, myristic acid enhances N-myristoylation of ARF1, a master regulator that controls STING membrane trafficking. Consequently, myristic acid facilitates STING activation-triggered autophagy degradation of the STING complex. Thus, our work identifies myristic acid as a metabolic checkpoint that contributes to immune homeostasis by balancing STING-dependent autophagy and IFN responses. This suggests that myristic acid and N-myristoylation are promising targets for the treatment of diseases caused by aberrant STING activation.

Direct Acylation of Cyanidin-3-Glucoside with Lauric Acid in Blueberry and Its Stability Analysis

Li-yi Zhao,Jian Chen,Zhi-qiang Wang,Rui-meng Shen,Nan Cui,Ai-dong Sun

Cyanidin-3-glucoside is an anthocyanin that is abundant in blueberry. Lauric acid was used as the acyl donor in the acylation of cyanidin-3-glucoside. Preparative high performance liquid chromatography was used to separate and purify the acylated cyanidin-3-glucoside, the acylated rate was 30.78%. Fourier transform infrared spectroscopy and liquid chromatography mass spectrometry were used to confirm the basic structure of cyanidin-3-glucoside. Lauric acid was combined with cyanidin-3-glucoside. Lauric acid reacted with the glucoside’s primary hydroxyl group and removed a molecule of water, thereby resulting in the obtained structure. The compositions of the unacylated cyanidin-3-glucoside and acylated cyanidin-3-glucoside were compared under different temperatures and under illumination. The effects of different concentrations of additive compounds on the stability of acylated cyanidin-3-glucoside were compared. Stability of acylated cyanidin-3-glucoside was obviously higher than that of the unacylated cyanidin-3-glucoside because of the higher stability of the ester group than the hydroxyl group. The primary hydroxyl esterification of glucoside improved cyanidin-3-glucoside.

Speed of sound and thermal diffusivity of ethyl myristate

Ying Zhang,Junshuai Chen,Taotao Zhan,Xiangyang Liu,Maogang He

Ethyl myristate is one of the main components of biodiesels and its thermophysical properties are critical to be supplemented and perfected. In this work, the speed of sound and thermal diffusivity of ethyl myristate were measured by the light scattering method. The investigated temperature and pressure is ranged from 303.15 K to 560.15 K and up to 10 MPa. The relative expanded uncertainties of the speed of sound and thermal diffusivity were estimated to be less than 1.3% and 2.6% (k = 2), respectively. The correlations for the speed of sound and thermal diffusivity as function of temperature and pressure were presented based on the experimental data. The absolute average deviations (AADs) between the experimental data and the calculated results are 0.04% for the speed of sound at atmospheric pressure, 0.07% for the speed of sound at p  > 0.1 MPa and 0.28% for the thermal diffusivity , respectively.

Preparation and properties of decyl – myristyl alcohol/expanded graphite low temperature composite phase change material

Shihua Zhang,Xuelai Zhang,Xiaofeng Xu,Yi Zhao

In order to find a low temperature phase change material with a temperature range of 2–8°C, decyl alcohol (DA) and myristyl alcohol (MA) binary eutectic mixture was prepared based on theoretical calculation, with a mass ratio of DA: MA=87:13. DA-CA/EG with an optimal mass ratio of DA-MA: EG=14:1 was obtained after vacuum adsorption. Scanning electron microscope and Fourier transformation infrared spectroscopy results show that DA-MA was uniformly distributed in the porous network structure of EG due to physical action. Phase change temperature and latent heat of DA-MA/EG were measured at 3.55°C and 172.3 J·g−1, and the thermal conductivity was 0.942 W·(m·K)−1, which was 3.4 times higher than DA-MA. After 500 high and low temperature cycles, DA-MA/EG did not change much and still maintained good stability. Overall, the prepared DA-MA/EG composite PCM had great application prospects in cold chain logistics.

Enrichment of Nutmeg Essential Oil from Oil-in-Water Emulsions with PAN-Based Membranes

Huilan Yin,Haoyu Zhang,Jiaoyang Cui,Qianlian Wu,Linlin Huang,Jiaoyue Qiu,Xin Zhang,Yanyu Xiang,Bo Li,Hongbo Liu,Zhishu Tang,Yue Zhang,Huaxu Zhu

This study used polyacrylonitrile (PAN) and heat-treated polyacrylonitrile (H-PAN) membranes to enrich nutmeg essential oils, which have more complex compositions compared with common oils. The oil rejection rate of the H-PAN membrane was higher than that of the PAN membrane for different oil concentrations of nutmeg essential oil-in-water emulsions. After heat treatment, the H-PAN membrane showed a smaller pore size, narrower pore size distribution, a rougher surface, higher hydrophilicity, and higher oleophobicity. According to the GC-MS results, the similarities of the essential oils enriched by the PAN and H-PAN membranes to those obtained by steam distillation (SD) were 0.988 and 0.990, respectively. In addition, these two membranes also exhibited higher essential oil rejection for Bupleuri Radix, Magnolia Officinalis Cortex, Caryophylli Flos, and Cinnamomi Cortex essential oil-in-water emulsions. This work could provide a reference for membrane technology for the non-destructive separation of oil with complex components from oil-in-water emulsions.

Measurements of the thermal conductivity of three pure alcohols and their binary mixtures with methyl myristate component of biodiesel

Jing Fan,Shuangshuo Liu,Fenhong Song,Xiaopo Wang

New experimental thermal conductivity values of 1-propanol, 1-butanol and 1-pentanol in the liquid phase have been reported, and the thermal conductivity of three binary mixtures (1-propanol + methyl myristate, 1-butanol + methyl myristate and 1-pentanol + methyl myristate) have been measured at atmospheric pressure and with the temperature range 292–362 K for the first time. The total standard uncertainty of the experimental results was estimated to be less than 2% (in the 0.68 confidence level) and the repeatability was better than ±0.5%. The liquid thermal conductivity results of three alcohols, 1-butanol/methyl myristate blends and 1-pentanol/methyl myristate blends were fitted as a polynomial function of temperature. In addition, three useful correlations have been optimized by the results of 1-propanol/methyl myristate blends.

Preparation, characterization and antimicrobial activity of inclusion complexes of Myristica fragrans Hott. essential oil in β-cyclodextrins

Wang Jin,Zhang Hai-Hong,Liu Fengxi,Zhang Yu-Ping,Zhao Yong-Ming

Inclusion complexes of essential oils with cyclodextrins are an effective way to improve stability and solubility, and turn liquid materials into easy to handle powders. In this work, an essential oil of Myristica fragrans Hott. (MFEO), already used in the food and cosmetics industries, was formulated with beta-cyclodextrins (β-CD) using a co-precipitation method. The orthogonal array scheme was adapted for the optimization of preparation process. DSC and FT-IR spectroscopy analysis indicated the successful formation of MFEO/β-CD inclusion complexes, which improved the thermal stability of MFEO. Furthermore, comparing the antimicrobial activity of MFEO/β-CD inclusion complexes and free essential oil against Staphyloccocus aureus, Staphyloccocus epidermidis, Escherichia coli, Klebsiella pneumoniae, yeast Saccharomyces cerevisiae and Bacillus subtilis, it was found that the antimicrobial effect was enhanced after the formation of inclusion complexes. This study demonstrates the potential for the use of MFEO/β-CD inclusion complexes in the treatment of bacterial infection.

Experimental investigations on the thermophysical properties of methyl myristate in alcoholic solutions

Fuxin Yang,Xiaopo Wang,Houzhang Tan,Siyuan He,Zhigang Liu

Due to the limitations of using biodiesel directly or blended with petroleum diesel, alcohols are always employed as the additives to improve the certain properties of biodiesel. In this work, experimental investigations are conducted on the thermophysical properties, especially the density and viscosity, of binary mixtures containing the component of the biodiesel, namely methyl myristate, with higher alcohols, including 1-propanol, 1-butanol and 1-pentanol at atmospheric pressure from 303.15 K to 333.15 K. The Vogel-Fulcher-Tammann equation is introduced to correlate the viscosity. The derived property of thermal expansion coefficient is calculated based on the experimental density. The excess properties are obtained to further understand the interactions between the ester and the alcohols. The positive value of the excess molar volume and negative value of viscosity deviation are observed, indicating that the increasing contribution of the repulsive forces between methyl ester and the alcohols.

Dual-Responsive PPy-AIPH@LA Nanoplatform for Synergistic Photothermal and Thermodynamic Therapy of Colorectal Cancer

Liang Zhang,Jiahao Qian,Xinyuan Zhang,Yanwei Lv,Jiulong Zhao,Shige Wang,Hao Xu

Graphical The dual-responsive polypyrrole-2,2'-Azobis(2-methylpropionamidine) dihydrochloride (AIPH)@Lauric acid nanoplatform integrates polypyrrole for photothermal therapy and AIPH for thermodynamic therapy, achieving synergistic colorectal cancer treatment. Under near-infrared irradiation, the system enables precise, oxygen-independent radical generation, enhancing tumor inhibition with minimal toxicity. This study highlights the platform's potential for minimally invasive cancer therapy. In this study, we introduce a novel nanoplatform, polypyrrole (PPy)-2,2′-Azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH)@lauric acid (LA) (PPy-AIPH@LA) nanoparticles (NPs), designed to overcome these limitations through synergistic photothermal therapy (PTT) and photodynamic therapy (PDT). This dual-responsive system incorporates PPy for efficient photothermal conversion, AIPH for thermos-responsive and oxygen-independent free radical generation, and LA as a thermally responsive encapsulation layer. The LA coating melts upon 808 nm near-infrared laser irradiation, releasing AIPH and free radicals to enable precise spatiotemporal activation of therapeutic effects. PPy-AIPH@LA demonstrates exceptional photothermal conversion efficiency (55.74%) and generates sufficient radicals to enhance PDT efficacy, even in hypoxic tumor microenvironments. In vitro studies revealed concentration-dependent tumor cell ablation and inhibition of migration, while in vivo experiments showed that the combined PTT-PDT treatment achieved an impressive 90.7% tumor growth inhibition rate in a mouse colon cancer cells CT-26 murine model, with no significant systemic toxicity. Molecular analyses further revealed modulations in pathways associated with tumor metabolism, apoptosis, and immune escape, highlighting the comprehensive therapeutic potential of this nanoplatform. These findings underscore the potential of PPy-AIPH@LA as a safe, effective, and minimally invasive nanotherapeutic platform for combating CRC and other solid tumors.

Eco-friendly biochar/CNTs-enhanced myristic acid composite phase change materials for efficient solar-thermal energy conversion

Wenwen Guo,Han Chen,Guoneng Li,Youqu Zheng,Lingcheng Qi

In the paper, we crafted a unique eco-friendly composite phase change material (PCM) with superior leak-proof performance by integrating modified biochar, carbon nanotubes (CNTs) of specific diameters, and myristic acid (MA) through a vacuum impregnation process. The biochar was modified with KOH, resulting in a specific surface area of 813.56 m 2 /g for modified biochar (KBC3) and an increased MA loading capacity of 63 %. Various characterizations confirmed the successful incorporation of MA into the porous materials without destroying their structure. To further enhance the PCM’s loading capacity and leakage resistance, CNTs of varying diameters were added to the KBC/MA PCM. With 75 % MA loading, KBC1.5/MA (75) showed significant shape leakage (8.5 % mass loss). However, KBC1.5/CNT1/MA (75) exhibited minimal leakage during testing, indicating improved leakage resistance. Notably, KBC1.5/CNT2/MA (75) exhibited high thermal conductivity enhancement (40.1 %), a high enthalpy of phase change at 143.5 J/g, outstanding enduring durability, and consistent thermal cycling curve after 50 cycles compared to single DSC curve. Biochar modification and CNTs significantly boosted the photothermal conversion efficiency of PCMs, from 0 % in pure MA and BC/MA (63) to 58.88 % in KBC1.5/MA, and further to 76.37 % in KBC1.5/CNT2/MA (63). The addition of CNTs with appropriate diameters had a profound impact on enhancing the PCMs’ performance, primarily due to carbon nanotubes’ high thermal conductivity and strong visible light absorption capabilities.

Insights into ultrasound-induced starch-lipid complexes to understand physicochemical and nutritional interventions

Jialin Tang,Qiufang Liang,Xiaofeng Ren,Husnain Raza,Haile Ma

Novel lotus root starch (LRS)-myristic acid (MA) complexes were prepared using an ultrasound-assisted hydrothermal method (UHM) to investigate its nutritional intervention in type 2 diabetes mellitus (T2DM). Ultrasonic treatment promoted the formation of the V-type crystal structure of the complex and improved the intermolecular interaction force, the order of the short-range starch molecules, and crystallinity. The volume of the ultrasound-assisted LRS-MA composite (U-LRS-MA) particles was enlarged, the particle distribution showed non-uniformity, and the surface grooves were deepened. The resistant starch content of U-LRS-MA was greatly increased from 34.58 % of native starch to 68.20 %. Dietary Supplements of 5 % and 15 % U-LRS-MA significantly reduced the body weight, the organ index and fasting blood glucose of T2DM mice, effectively adjusted its blood lipid level, alleviated its liver damage and increased the levels of colonic short-chain fatty acids. The addition of 5 % U-LRS-MA was more effective in T2DM than 15 % U-LRS-MA. Ultrasound could be effectively employed to prepare lipid-starch complexes, namely type 5 resistant starch, which was proved for the first time to have an excellent intervention effect on T2DM.
