Comparison of gamithromycin residue depletion in yellow-feather and white-feather broilers after one single subcutaneous injection

Xing-Ping Li,Jun Liang,Ze-En Li,Ming-Hui Duan,Yan Dai,Yan-Ni Zhang,Yue Liu,Yang-Guang Jin,Fan Yang

ABSTRACT This study aimed to compare the residue depletion of gamithromycin in yellow-feather and white-feather broilers, using Sanhuang and Arbor Acres chickens as typical examples, respectively. Each breed (54 chickens) received a single subcutaneous dose of gamithromycin at 7.5 mg/kg bodyweight ( BW ). Tissues, including muscle, skin + fat, liver, kidney, and injection site, were collected at 6 h, 3, 5, 7, 10, 14, 21, 28, and 35 d postdrug administration. Gamithromycin concentrations in these tissues were determined using ultra‐performance liquid chromatography–tandem mass spectrometry ( UPLC‐MS/MS ). The kinetics of gamithromycin were analyzed in different tissues using a noncompartmental method in the Phoenix software. Differences were observed in gamithromycin concentrations and kinetic characteristics in both breeds of chickens, with higher residue concentrations and longer residue times found in yellow-feathered broilers. In Sanhuang broilers, the elimination rates of gamithromycin followed this order: injection site > muscle > liver > kidney > skin + fat. The corresponding elimination half-lives ( t 1/2λz s ) in these samples were 1.22, 1.30, 1.71, 2.04, and 2.52 d, respectively. In contrast, in Arbor Acres broilers, a different order was noted: muscle > injection site > kidney > liver > skin + fat, with corresponding t 1/2λz s of 1, 1.23, 1.88, 1.93, and 2.21 d, respectively. These differences may be related to variations in pigments in various tissues of chickens of the 2 breeds. However, further investigations are warranted to discern the underlying reasons.

Oregano Oil Combined with Macleaya Cordata Oral Solution Improves the Growth Performance and Immune Response of Broilers

Cheng Zhang,Weihao Li,Ligong Chen,Zhaoliang Chen,Xuejing Wang,Qianqian Xu,Hailong Zhang,Huan Chen,Juxiang Liu

Simple SummaryDeveloping safe and effective antibiotic growth promoters (AGPs) substitutes is particularly important to improve animal health and production performance. As an essential plant oil, the oregano oil's main bioactive substance is carvacrol, which has been proven to have antioxidant, anti-inflammatory, antibacterial, and antiviral properties. The sanguinarine from macleaya cordata is the primary bioactive substance. Sanguinarine has anti-tumor, immune-enhancing, antibacterial, and anti-inflammatory effects. However, it has not been reported whether the compatibility of oregano oil and macleaya cordata extract could produce better results. This study is the first to report the effect of oregano oil combined with macleaya cordata oral solution on the growth of broilers. The oregano oil combined with macleaya cordata oral solution significantly improved the growth performance of broilers. At the same time, serum biochemical indices, serum antioxidant indices, serum immune indices, serum cytokines, and intestinal morphology were significantly improved. In summary, our results demonstrated that the mixed solution of oregano oil and macleaya cordata has substantial potential to be an alternative to AGPs for broilers to reduce costs and improve benefits. This study provides basic data and technical support for further research.AbstractThe abuse of AGPs in animal husbandry has led to severe problems such as drug resistance and ecological, and environmental destruction, which seriously threaten human health and public health security. In recent years, extracts of oregano oil and macleaya cordata have become a hot spot in the research and application of AGP substitutes for their safety and high efficiency. This study is the first to report the effect of oregano oil combined with macleaya cordata oral solution on broiler growth performance. A total of 960 one-day-old broiler chickens were randomly divided into four treatment groups (240 chickens per group). Each treatment group was divided into six replicate groups (40 birds per replicate group). There were four groups in this study: the solvent control group, the oregano essential oil combined with macleaya cordata extract oral solution group (OS group), the oregano essential oil oral solution group (OEO group), and the macleaya cordata extract oral solution group (MCE group). Two chickens from each replicate group were collected and mixed into a composite sample. Six composite samples were obtained for each treatment group. The results showed that the oregano oil combined with macleaya cordata oral solution significantly improved the growth performance of broiler chickens. At the same time, serum biochemical indices, serum antioxidant indices, serum immune indices, serum cytokines, and intestinal morphology were significantly improved by the OS group. This study shows that oregano oil combined with macleaya cordata oral solution has substantial potential to be an alternative to AGPs for broilers. Keywords:oregano oil;macleaya cordata;oral solution;broilers;growth performance

Encapsulated crystalline lysine and DL-methionine have higher efficiency than the crystalline form in broilers

Mingfa Sun,Hongchao Jiao,Xiaojuan Wang,Victoria A. Uyanga,Jingpeng Zhao,Hai Lin

Crystalline amino acids ( AAs ) exhibit high nutritional values when supplemented AA-deficient diets. However, the AAs in crystalline form in the diet are absorbed quickly than protein-bound AAs, which may take an effect on AA utilization efficiency. In this study, 2 experiments were conducted to investigate the effect of encapsulated lysine-HCl ( Ly s) and DL-methionine ( DL-Met ) on the growth performance of broiler chickens. In experiment 1, a total of 432 one-day-old male Arbor Acres broilers were subjected to 3 dietary treatments (27 pens; 16 birds per pen) for 42 d. The control group was basal diets supplemented with crystalline Lys and DL-Met, and treatment groups had basal diets supplemented with encapsulated Lys and DL-Met at the levels of 80% and 60% of control diets ( 80CLM , 60CLM ), respectively. The growth performance, intestinal development, and transcription of AA transporters were determined. In experiment 2, 24 broiler chickens were subjected to the same treatments as in experiment 1. The plasma concentrations of free AAs were measured 0, 2, 4, and 6 h after feeding. The results showed that 80CLM treatment had no significant influence on production performance, carcass characteristics, and plasma free AAs content during the experiment compared with the control group ( P > 0.05). In addition, the 80CLM group moderately enhanced gut morphology development and increased AAs' absorption capacity. However, broilers fed the 60CLM diet had lower production performance and breast muscle weight than the control group ( P < 0.05), but increased villi height and B 0 AT mRNA expression level ( P < 0.05). At h 4 after feeding, the 60CLM broilers exhibited higher concentration of Ala, Cys, and total dispensable AAs than the control group ( P < 0.05). In conclusion, the result suggests that the supplemental levels of crystalline Lys and DL-Met can be effectively saved approximately for 20% by using the encapsulated form in broilers, with improvements to AAs utilization efficiency, while posing no detrimental effects on production performance. Encapsulated Lys and DL-Met would have greater potential for application when replacing crystalline AAs in broiler chickens.

Effects of different levels of rutin on growth performance, immunity, intestinal barrier and antioxidant capacity of broilers

Shun Chen,Huijuan Liu,Jiaqi Zhang,Binbin Zhou,Su Zhuang,Xiaofang He,Tian Wang,Chao Wang

This study investigated the effects of different doses of rutin on the growth performance, immunity, intestinal barrier, and antioxidant capacity of broilers. A total of two hundred and fifty-six 1-day-old male broilers were divided into 4 groups and fed basal diets supplemented with 0 (control group), 250, 500 and 1,000 mg rutin/kg, respectively, for 42 days. In the starter period (days 1–21) and whole trial period (days 1–42), broilers fed diets containing 500 mg rutin/kg had significantly (p < 0.05) higher average daily feed intake, body weight and average daily gain and better feed conversion ratio. Dietary 500 mg rutin/kg increased the villus height, villus height to crypt depth ratio and villus area, while reducing the B cell lymphoma 2 associated X mRNA expression (p < 0.05). Dietary 500 mg rutin/kg increased the level of IgA in serum and decreased serum tumour necrosis factor-α (TNF-α) content, and the nuclear factor kappa-B, interleukin-2 and TNF-α mRNA expression in jejunal mucosa (p < 0.05). Simultaneously, 500 and 1,000 mg rutin/kg significantly decreased serum diamine oxidase activity, D-lactic acid and lipopolysaccharide concentration, increased activities of total superoxide dismutase and total antioxidant capacity in jejunal mucosa (p < 0.05), and upregulated mRNA expressions of genes related to intestinal barrier and antioxidant capacity in jejunal mucosa (p < 0.05). These findings indicate that dietary rutin, especially at 500 mg/kg, improves broilers’ growth performance, intestinal barrier function, immunity, and antioxidant capability, which may be related to the nuclear factor erythroid-2-related factor 2 (Nrf2) pathway.HighlightsDietary rutin improved growth performance, jejunal morphology, and intestinal barrier function.Dietary rutin enhanced the immunity via inhibiting NF-κB, and improved antioxidant capacity via activating Nrf2/HO-1 pathway in broilers. The optimal dose of rutin was 500 mg/kg in broiler diet.

Effects of different selenium sources on duodenum and jejunum tight junction network and growth performance of broilers in a model of fluorine-induced chronic oxidative stress

Siyuan Chen,Yajie Xue,Yutian Shen,Hao Ju,Xiaodong Zhang,Jinsong Liu,Yongxia Wang

ABSTRACT The protective effects and underlying molecular mechanisms of sodium selenite ( SS ) and selenomethionine ( SM ) against chronic oxidative stress-induced duodenum and jejunum tight junction ( TJ ) network disturbance and growth inhibition of broilers were investigated in the current experiment. At the age of 1 d, 720 Lingnan Yellow broiler chicks were allocated to 4 experimental diets (with 6 replicates per diet and 30 birds per replicate) and offered either a control diet (fluorine [ F ] 23 mg/kg, control [ CoN ] group) or test diets (800 mg/kg F, high F [ HF ] group; 800 mg/kg F+0.15 mg selenium [ Se ]/kg as SS [ SS group ] or SM [ SM group ]) for 56 d. The results showed that HF group could induce chronic oxidative stress and subsequently increased ( P < 0.05) proinflammatory cytokines levels of duodenum and jejunum in comparison with the CoN group. Increased proinflammatory cytokines levels of HF group promoted myosin light chain kinase ( MLCK ) transcription, thus leading to a decrease ( P < 0.05) in TJ proteins expression of duodenum and jejunum when compared with the CoN group. A reduction of TJ proteins expression destroyed the TJ structures in the HF group, which in turn increased intestinal mucosal permeability of duodenum and jejunum and ultimately induced growth inhibition of broilers. Dietary Se supplementation could ameliorate HF-induced duodenum and jejunum TJ network impairment and growth retardation of broilers, potentially by increasing ( P < 0.05) the glutathione peroxidase and thioredoxin reductase activities, reducing ( P < 0.05) the reactive oxygen species and malondialdehyde levels, regulating the secretion of proinflammatory cytokines, and mediating the transcription level of MLCK in the duodenum and jejunum. Additionally, our data also suggested that the protective effects of SM were superior to those of SS. This study will provide a theoretical basis for developing SM into an efficient protective agent for intestinal mucosal barrier in poultry.

Dietary bile acids alleviate corticosterone-induced fatty liver and hepatic glucocorticoid receptor suppression in broiler chickens

Liu Jie,Zhang Ke,Zhao Mindie,Chen Liang,Chen Huimin,Zhao Yulan,Zhao Ruqian

The aim of this study was to investigate the alleviating effects and mechanisms of bile acids (BA) on corticosterone-induced fatty liver in broiler chickens. Male Arbor Acres chickens were randomly divided into three groups: control group (CON), stress model group (CORT), and BA-treated group (CORT-BA). The CORT-BA group received a diet with 250 mg/kg BA from 21 days of age. From day 36 to 43, both the CORT and CORT-BA groups received subcutaneous injections of corticosterone to simulate chronic stress. The results indicated that BA significantly mitigated the body weight loss, liver enlargement, and hepatic lipid deposition caused by corticosterone (P < 0.05). Liver RNA-seq analysis showed that BA alleviated corticosterone-induced fatty liver by inhibiting lipid metabolism pathways, including fatty acid biosynthesis, triglyceride biosynthesis, and fatty acid transport. Additionally, BA improved corticosterone-induced downregulation of glucocorticoid receptor (GR) expression (P < 0.05). Molecular docking and cellular thermal shift assays revealed that hyodeoxycholic acid (HDCA), a major component of compound bile acids, could bind to GR and enhance its stability. In conclusion, BA alleviated corticosterone-induced fatty liver in broilers by inhibiting lipid synthesis pathways and mitigating the suppression of hepatic GR expression.

Zhenglie Dai,Huixian Wang,Jinsong Liu,Haoran Zhang,Qing Li,Xiaorong Yu,Ruiqiang Zhang,Caimei Yang

Simple SummaryAntibiotics are banned in animal feed, and substances used to replace antibiotics are being found continuously.Yucca schidigera,Quillaja saponaria, andYucca saponinare natural plants and their components, which have many beneficial health functions for animals, are suitable as animal feed additives to replace antibiotics. By comparing the impacts of three substances on the growth, immunity, and intestinal health of broilers, this study found thatYucca schidigera,Quillaja saponaria, andYucca saponinall promote the healthy growth of broilers, andYucca saponinhas better effects than the other two substances.AbstractThe purpose of this study is to investigate the effects ofYucca saponin(YSa),Yucca schidigera(YS), andQuillaja Saponaria(QS) on growth performance, nitrogen metabolism, immune ability, antioxidant capability, and intestinal flora of yellow-feather broilers. This study randomly divided a total of 480 1-day yellow-feather broilers into 4 treatment groups. Factors in the 4 groups included CON group (basic diet), YSa group (basic diet mixed with 500 mg/kg YSa), YS group (basic diet mixed with 500 mg/kg YS), and QS group (basic diet mixed with 500 mg/kg QS). Throughout the 56-day study period, YSa, YS, and QS groups had higher average daily gain in broilers than the CON group (p< 0.01). The YS group had a lower feed gain ratio (F: G) in broilers than the CON group (p< 0.05). YSa, YS, and QS showed increased serum immunoglobin A (IgA), immunoglobin Y (IgY), immunoglobin M (IgM), and total antioxidant capacity (T-AOC) levels; enhanced acetic acid, butyric acid, and valeric acid levels of cecal content; and reduced contents of ammonia nitrogen, urea nitrogen, interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and malondialdehyde (MDA) in serum in broilers (p< 0.05). The relative abundance of Lachnoclostridium in the QS group was decreased compared with that in the CON group (p< 0.05). Higher IgA and IgY sera contents were observed in the YS group compared to the YSa and QS groups (p< 0.05). In contrast with the QS group, the serum IL-6 concentration of the YS group was reduced (p< 0.05). In conclusion, YSa, YS, and QS promoted growth performance, nitrogen metabolism, immunity, antioxidant capability, and intestinal flora in broilers. Through the comparison of YSa, YS, and QS, it was found that YS is more suitable as a feed additive to ameliorate the healthy growth of broilers. Keywords:Yucca saponin;Quillaja saponaria;yellow-feather broilers;growth performance;intestinal flora

Effects of the pesticide carbendazim on broiler chicken health and carbendazim residue levels in broiler tissues

Zhang Wanjun,Tang Yutong,Fan Ruiqi,Zhang Fude,Li Yongchen,Zang Jianjun,Yang Wenjun,Wang Zongyi,Yuan Xiaoyan,Yang Yuan,Chen Yiqiang

Pesticide residues may enter the human body through the food chain when livestock and poultry consume pesticide-contaminated feed. Therefore, monitoring and limiting pesticide residues in animal feed and animal-origin foods is necessary. Carbendazim is one of the most frequently detected pesticides in food and feed and has various toxic effects on non-target animals. This study investigated the effects of varying concentrations of carbendazim contamination in feed on broiler chicken growth performance, serum biochemical indicators, histopathology, and carbendazim residues in broiler muscles and livers. The results demonstrated that contamination of 5–100 mg/kg carbendazim in feed did not affect broiler growth performance or health. Carbendazim contamination in feed at 200–800 mg/kg slightly reduced growth performance. Broiler kidneys showed minor histopathological alterations after 400 mg/kg carbendazim exposure. Furthermore, when the carbendazim content in feed was less than 25 mg/kg, the residual carbendazim in the muscles and livers of broilers did not exceed the maximum residue level set by the European Union and China. Based on the above findings, carbendazim residues in the feed of less than 25 mg/kg can be considered safe for chicken products.

Toxicity induced by F. poae-contaminated feed and the protective effect of Montmorillonite supplementation in broilers

Lingchen Yang,Zhiyong Zhao,Yifeng Deng,Zhenlei Zhou,Jiafa Hou

The T-2 and HT-2 toxins, the main metabolites of Fusarium poae , induce toxicity in broilers and accumulate in tissues. Consequently, during the breeding process of broilers, diets are frequently supplemented with physical adsorbents to protect birds against the toxicity induced by mycotoxins. In the present research, T-2 and HT-2 were produced in maize inoculated with F. poae . Mont, the strongest adsorbent based on in vitro adsorption ratios, was added to the contaminated diet. One-day-old chickens were randomly and equally divided into the following four groups: control diet group, Mont supplemented diet group, contaminated diet group and detoxification diet group. The experiment lasted for 42 days. Compared to the control group, the contaminated group showed significant decrease in body weight, feed intake and TP ( P  < 0.05), and marked increase in FCR, ALP, AST and ALT activity, T-2/HT-2 residues in the tissues and the relative expressions of apoptosis-related mRNAs ( P  < 0.05). Mont supplementation provided protection for the treated broilers in terms of performance, blood biochemistry, hepatic function, T-2/HT-2 residue of tissues and apoptosis. Therefore, Mont may be suitable as a detoxification agent for T-2/HT-2 in feed for broilers.

Effects of dietary sweeteners supplementation on growth performance, serum biochemicals, and jejunal physiological functions of broiler chickens

Jingle Jiang,Siyi Liu,Tuniyaz Jamal,Tengxin Ding,Lina Qi,Zengpeng Lv,Debing Yu,Fangxiong Shi

The objective of this study was to investigate the effects of dietary 3 kinds of sweeteners supplementation on growth performance, serum biochemicals, and jejunal physiological functions of broiler chickens for 21 D. A total of one hundred ninety-two 1-day-old male Ross 308 broiler chicks were randomly divided into 4 treatments with 6 replicates for each treatment. The treatments were basal diet ( CON ), a basal diet supplemented with 250 mg/kg stevioside ( STE ), a basal diet supplemented with 100 mg/kg sucralose ( SUC ), and a basal diet supplemented with 600 mg/kg saccharin sodium ( SAC ). All birds were housed in 3-level battery cages. The results showed that dietary STE supplementation increased ( P  < 0.05) growth performance, serum total protein, serum albumin, and jejunal antioxidant capacity of broiler chickens. Both SUC and SAC supplementation decreased ( P  < 0.05) serum total protein and albumin. Dietary SAC supplementation impaired the intestinal integrity, permeability, and mucus layer of the jejunum in broiler chickens. In addition, SAC supplementation elevated ( P  < 0.05) the transcription expression level of jejunal bitter taste receptors and induced excessive jejunal apoptosis. Our data suggest that STE could be potentially applied as a growth-promoting and antioxidant feed additive in broiler chickens. Whereas, dietary supplementation with high level SAC has side-effects on the jejunal physiological functions of broiler chickens.

Dietary supplementation of silver-silica nanoparticles promotes histological, immunological, ultrastructural, and performance parameters of broiler chickens

Dosoky Waleed M.,Fouda Moustafa M. G.,Alwan Ali B.,Abdelsalam Nader R.,Taha Ayman E.,Ghareeb Rehab Y.,El-Aassar M. R.,Khafaga Asmaa F.

Silver nanoparticles (AgNPs) have been used as a promising alternative to antibiotics in poultry feed. In this study, silver-doped silica nanoparticles (SiO 2 @AgNPs) were prepared in powder form, using starch, via the chemical reduction method and sol–gel technique followed by full characterization. SiO 2 @AgNPs were added to the poultry diet at three doses (2, 4, and 8 mg/kg diet). The safety of the oral dietary supplementation was estimated through the evaluation of the growth performance and hematological, biochemical, and oxidative parameters of birds. Moreover, the immunohistochemical examination of all body organs was also performed. Results of this study showed that SiO 2 @AgNPs have no negative effects on the growth performance and hematological, biochemical, and oxidative parameters of birds. Moreover, the immunohistochemical examination revealed the minimum inflammatory reactions and lymphoid depletion under a dose level of 8 mg/kg. In conclusion, SiO 2 @AgNPs could be considered as a promising and safe nano-growth promoter in broilers when added to poultry diet under a dose level of 4 mg/kg diet.

Effects of phenylpyruvate on the growth performance and intestinal microbiota in broiler chicken

M. Z. Talpur,W. Peng,Y. Zeng,P. Xie,J. Li,H. Zhang,G. Shu,Q. Jiang

1. The purpose of this study was to see how dietary supplementation with phenylpyruvate affected broiler growth, slaughter performance, gut health microbiota and immunity. This information can be used to develop alternative approaches to antibiotic replacement in modern poultry production and health.2. A total of 288, one-day-old broiler chickens were randomly assigned to one of four groups (six replicates each replicate has 12 chickens). A control basal diet (NC), basal diet plus antibiotic virginiamycin 15ppm (PC), basal diet plus phenylpyruvate 1 kg/t or 2 kg/t, respectively (LCP and HCP).3. Results showed that the birds in the PC group had higher ADFI during the first 21 d, and better FCR than the NC group. The HCP-fed group had a higher all-eviscerated ratio than the NC group and less abdominal fat than the birds fed LCP. The birds fed HCP had increased villus length and crypt depth in the ileum compared to the NC group.4. The bursa index was lower in the HCP group whereas the thymus index was lower in LCP and PC groups. In contrast, birds fed HCP has lower pro-inflammatory cytokine IL-1, as well as lower TLR4. Phenylpyruvate improved number in the Selenomonadaceae, genus Megamonas bacteroides spp., which are known for their beneficial effects on the maintenance of the cell surface structure, regulating aromatic amino acids and Clostridia jejuni-suppressive treatment respectively.5. It was concluded that phenylpyruvate can be utilised in feed to improve growth performance and positively modulate gut microbiota. However, this was less efficient than antibiotics in improving growth performance, although more efficient in improving productive performance and gut morphology. Moreover, a high dose of phenylpyruvate is more effective than a low dose.
