Volumetric muscle loss (VML) results in the impediment of skeletal muscle function. Tissue engineering scaffolds have been widely developed and used in skeletal muscle regeneration. However, scaffold implantation causes an immune response that endogenously regulates implant integration and tissue regeneration. Moreover, vascularization is thought to be a principal obstacle in the reconstruction of skeletal muscle defects. Thus, creating a pro-regenerative microenvironment that facilitates muscle regeneration and supports angiogenesis represents a promising strategy for tissue repair following volumetric muscle loss (VML) injury. Previously, the electrospun silk fibroin–silk sericin (SF-SS) film could regulate macrophage polarization and promote neovessel formation. This study aimed to investigate if the electrospun SF-SS scaffold was capable of supporting functional muscle regeneration. The results indicate that the conditioned medium collected from macrophages co-cultured with the 7:3 SF-SS scaffold significantly enhanced the proliferation and migration of myoblast C2C12 cells and improved the tube formation of HUVECs. Data from animal studies showed that the 7:3 SF-SS scaffold significantly enhanced M2 macrophage polarization, vascularization, and muscle fiber regeneration, reduced fibrosis, and improved muscle function after VML injury, thereby promoting the repair of muscle tissue. Therefore, the 7:3 SF-SS scaffold might represent a potential candidate for skeletal muscle regeneration following VML injury.

Background To explore the role of skeletal muscle specific TGF-β signaling on macrophages efferocytosis in inflamed muscle caused by Cardiotoxin (CTX) injection. Methods CTX myoinjury was manipulated in TGF-βr2 flox/flox (control) mice or transgenic mice with TGF-β receptor 2 (TGF-βr2) being specifically deleted in skeletal muscle (SM TGF-βr2 −/− ). Gene levels of TGF-β signal molecules, special inflammatory mediators in damaged muscle or in cultured and differentiated myogenic precursor cells (MPC-myotubes) were monitored by transcriptome microarray or qRT-PCR. TGF-β pathway molecules, myokines and embryonic myosin heavy chain in regenerating myofibers, the phenotype and efferocytosis of macrophages were evaluated by immunofluorescence, immunoblotting, Luminex, or FACS analysis. In vitro apoptotic cells were prepared by UV-irradiation. Results In control mice, TGF-β-Smad2/3 signaling were significantly up-regulated in regenerating centronuclear myofibers after CTX-myoinjury. More severe muscle inflammation was caused by the deficiency of muscle TGF-β signaling, with the increased number of M1, but the decreased number of M2 macrophages. Notably, the deficiency of TGF-β signaling in myofibers dramatically affected on the ability of macrophages to conduct efferocytosis, marked by the decreased number of Annexin-V − F4/80 + Tunel + macrophages in inflamed muscle, and the impaired uptake of macrophages to PKH67 + apoptotic cells transferred into damaged muscle. Further, our study suggested that, the intrinsic TGF-β signaling directed IL-10-Vav1-Rac1 efferocytosis signaling in muscle macrophages. Conclusions Our data demonstrate that muscle inflammation can be suppressed potentially by activating the intrinsic TGF-β signaling in myofibers to promote IL-10 dependent-macrophages efferocytosis.

2-ethylhexyl diphenyl phosphate (EHDPHP), a ubiquitous organic phosphorus flame retardant, is extensively employed in food packaging, flame retardancy applications, and other industrial processes as a crucial chemical. However, recent discoveries have underscored its diverse toxic effects, posing a severe potential threat to biological health. Skeletal muscle, vital for locomotion, relies heavily on proper differentiation during development and maintenance for strength and functionality. This study investigated the impact of EHDPHP on skeletal muscle differentiation by culturing C2C12 cells with varying concentrations of EHDPHP during induction of differentiation, as well as constructing a mouse muscle injury repair model through oral administration of 30 μg/kg EHDPHP for 7 weeks. Exposure to EHDPHP was found to impede skeletal muscle differentiation in mice. Utilizing techniques such as western blot analysis, RNA-seq, and qRT-PCR, our preliminary analysis revealed that low concentrations of EHDPHP inhibited the expression of ACTC1, subsequently suppressing MyoG and MYH2 expression levels, thereby hindering C2C12 myoblast differentiation. Furthermore, EHDPHP exposure disrupted the repair process of muscle injury in mice and compromised the integrity of the repaired tissue. In conclusion, these findings elucidate the detrimental effects of EHDPHP exposure on muscle differentiation, uncovering its muscle toxicity.

Acute blunt skeletal muscle injury occurs frequently in sports and traffic accidents, and even leads to muscle necrosis and impaired functionality. Current treatment options for muscle injuries remain suboptimal and often result in delayed/incomplete recovery of damaged muscles. Tanshinone IIA is extracted from Salvia Miltiorrhizae, which is effective in the treatment of injury repair. But the clinical application of tanshinone IIA is limited due to its low water solubility, low permeability to biofilm and low bioavailability. In this study, tanshinone IIA liposomes were prepared to improve the bioavailability and sustained release of tanshinone IIA. The particle size, dispersion coefficient, zeta potential, encapsulation efficiency (EE) and drug loading (DL) of tanshinone IIA liposomes were 150.67 ± 27.23 nm, 0.20 ± 0.015, -8.73 ± 2.28 mV, 70.32 ± 4.04% and 15.63%, respectively. The results of quantitative real-time polymerase chain reaction (QRT-PCR) showed that tanshinone IIA liposome significantly promoted the expression of vimentin and reduce MHCIIB expression compared with other groups (P < 0.05). Western blotting showed that tanshinone IIA liposome could effectively promote the expression of autophagy-related proteins (VPS34, Beclin 1 and CTSD) and decrease p62 expression levels to treat injured muscle. Through HE, immunohistochemistry, ELISA and serological tests, we found that tanshinone IIA liposome not only effectively promoted the expression of desmin, but also reduced the expression of collagen-I and inhibited the production of pro-inflammatory factors, such as tumor necrosis factor α (TNF-α) and interleukin-6 (IL-6) (P < 0.05). In addition, tanshinone IIA liposome therapy significantly reduced the level of malondialdehyde (MDA) and increased the activity of superoxide dismutase (SOD) after muscle injury compared with other groups (P < 0.05). In conclusion, tanshinone IIA liposome possesses an effective therapeutic effect on acute blunt muscle injury in rats by augmenting autophagy and alleviating oxidative stress. The continuous release of tanshinone IIA encapsulated by liposomes for disease treatment provide a new idea for the efficient and safe use of drugs with low lipid solubility and bioavailability for the treatment of acute blunt muscle injury and repair of other injuries.

Surface electromyography (sEMG) can provide multiplexed information about muscle performance. If current sEMG electrodes are stretchable, arrayed, and able to be used multiple times, they would offer adequate high-quality data for continuous monitoring. The lack of these properties delays the widespread use of sEMG in clinics and in everyday life. Here, we address these constraints by design of an adhesive dry electrode using tannic acid, polyvinyl alcohol, and PEDOT:PSS (TPP). The TPP electrode offers superior stretchability (~200%) and adhesiveness (0.58 N/cm) compared to current electrodes, ensuring stable and long-term contact with the skin for recording (>20 dB; >5 days). In addition, we developed a metal-polymer electrode array patch (MEAP) comprising liquid metal (LM) circuits and TPP electrodes. The MEAP demonstrated better conformability than commercial arrays, resulting in higher signal-to-noise ratio and more stable recordings during muscle movements. Manufactured using scalable screen-printing, these MEAPs feature a completely stretchable material and array architecture, enabling real-time monitoring of muscle stress, fatigue, and tendon displacement. Their potential to reduce muscle and tendon injuries and enhance performance in daily exercise and professional sports holds great promise.

Patients with clinical bone defects often experience varying degrees of muscle injury. Due to the high heterogeneity of hard and soft tissue structures, the integrated treatment of muscle injuries associated with bone defects remains a challenge. To address this challenge, this study proposes the construction of a heterogeneous bilayered electrospun nanofibrous scaffold based on the structural heterogeneity of bone and muscle to guide bone-muscle regeneration. The oriented fiber layer of the asymmetric bilayered scaffold promoted myofibroblast orientation and myogenic differentiation, whereas the random fiber layer promoted bone marrow mesenchymal stem cells (BMSCs) differentiation. Furthermore, immobilizing BMSCs-derived exosomes (BMSCs-Exo) on a heterogeneous bilayered scaffold via surface chemical modification of PEI enhances angiogenesis and immunoregulation. The foreign body response and muscle repair effect of the exosome-laden scaffolds were evaluated by subcutaneous implantation and a volumetric muscle loss model in mice. The performance of guided bone regeneration was assessed using a critical-sized cranial bone defect model in rats. BMSCs-Exo-functionalized heterogeneous bilayered electrospun nanofibrous scaffolds have great application potential in repairing bone-muscle injury.

The extracellular matrix (ECM) plays a crucial role in regulating cellular behaviors and functions. However, the impact of ECM topography on muscle cell adhesion and differentiation remains poorly understood from a mechanosensing perspective. In this study, we fabricated aligned and random electrospun polycaprolactone (PCL) nanofibers to mimic the structural characteristics of ECM. Mechanism investigations revealed that the orientation of nanofibers promoted C2C12 polarization and myogenesis through Rac-related signaling pathways. Conversely, cells cultured on random fibers exhibited spreading behavior mediated by RhoA/ROCK pathways, resulting in enhanced stress fiber formation but reduced capacity for myogenic differentiation. Our findings highlight the critical role of an ECM structure in muscle regeneration and damage repair, providing novel insights into mechanosensing mechanisms underlying muscle injury diseases.

Exhaustive exercise is known to induce muscle damage characterized by inflammation and oxidative stress. Although “regular” and “weekend warrior” exercise regimens have been shown to confer comparable health benefits in human studies, such as reduced risks of all-cause, cardiovascular disease (CVD), and cancer mortality, their differential impacts on muscle damage post-exhaustive exercise remain unclear. This study aimed to compare the effects of long-term, moderate-intensity (LTMI) and short-term, high-intensity (STHI) training modalities, matched for total exercise volume, on gut microbiota, short-chain fatty acids (SCFAs), and exhaustive exercise-induced muscle damage in mice, as well as to evaluate the correlation between these factors. LTMI is considered a regular exercise regimen, while STHI shares some similarities with the “weekend warrior” pattern, such as promoting exercise intensity and condensing training sessions into a short period. Our findings indicate that LTMI training significantly enhanced the abundance of SCFA-producing bacteria, including Akkermansia , Prevotellaceae_NK3B31_group , Odoribacter , Alistipes , and Lactobacillus , thereby increasing SCFA levels and attenuating muscle damage following exhaustive swimming. In contrast, STHI training increased the abundance of opportunistic pathogens such as Staphylococcus and Bilophila , without altering SCFA levels, and was associated with exacerbated muscle damage. Moreover, we observed a significant negative correlation between the abundance of SCFA-producing bacteria and SCFA levels with the expression of inflammatory cytokines in the muscle of mice post-exhaustive exercise. Conversely, the abundance of Staphylococcus and Bilophila showed a notable positive correlation with these cytokines. Additionally, the effects of LTMI and STHI on exhaustive exercise-induced muscle damage were transmissible to untrained mice via fecal microbiota transplantation, suggesting that gut microbiota changes induced by these training modalities may contribute to their contrasting impacts on muscle damage. These results underscore the significance of selecting an appropriate training modality prior to engaging in exhaustive exercise, with implications for athletic training and injury prevention.
