Immobilization is an effective method to promote the application of enzyme industry for improving the stability and realizing recovery of enzyme. To some extent, the performance of immobilized enzyme depends on the choice of carrier material. Therefore, the development of new carrier materials has been one of the key issues concerned by enzyme immobilization researchers. In this work, a novel organic–inorganic hybrid material, nickel-carnosine complex (NiCar), was synthesized for the first time by solvothermal method. The obtained NiCar exhibits spherical morphology, hierarchical porosity and abundant unsaturated coordination nickel ions , which provide excellent anchoring sites for the immobilization of proteins. His-tagged organophosphate-degrading enzyme (OpdA) and ω-transaminase (ω-TA) were used as model enzymes to evaluate the performance of NiCar as a carrier. By a simple adsorption process , the enzyme molecules can be fixed on the particles of NiCar, and the stability and reusability are significantly improved. The analysis of protein adsorption on NiCar verified that the affinity adsorption between the imidazole functional group on the protein and the unsaturated coordination nickel ions on NiCar was the main force in the immobilization process, which provided an idea way for the development of new enzyme immobilization carriers.

Quercetin and carnosine, natural antioxidants with anti-aging and photoprotective potential, face therapeutic limitations due to poor skin permeability. We engineered EV-QuerCar, a liposomal-based nanoparticles (LPNs) system co-encapsulating both compounds, to enhance transdermal delivery and evaluate its structural and functional efficacy. EV-QuerCar was prepared biomimetically using thermophilic bacterial lipids via ultracentrifugation, thin-film hydration, and high-pressure homogenization. Physicochemical properties, transdermal penetration, cellular uptake, and cytotoxicity were tested in vitro. Molecular docking and qPCR assessed pathway targeting. EV-QuerCar exhibited uniform size (94.70 ± 0.71 nm), high encapsulation efficiency (90.07%), sustained release, and 3.21-fold enhanced transdermal absorption compared to free compounds. Cellular uptake in fibroblasts increased time-dependently (up to 5.48-fold at 4 h). Molecular docking and qPCR confirmed interactions with NRF2 and HES1 targets, significantly upregulating their mRNA levels (2.29- and 3.35-fold, respectively) in UV-exposed human fibroblasts. EV-QuerCar improves skin permeability and antioxidant efficacy, highlighting its cosmeceutical promise.

Hypothesis: Taking advantage of photoinduced electron transfer , one dimensional organic nanomaterials with tunable donor-acceptor (D-A) interface provide a promising avenue to get high optoelectric properties. However, strong charge transfer interaction between D and A segments impedes the formation of long-range ordered structure, which limits the charge transport through efficient π electronic delocalization. Incorporation of chiral peptide offering various hydrogen bonding (H-bonding) along with asymmetric molecular structure enables substantially controllable D-A interface and tunable organization of the π -conjugates. Experiments: A new amphiphilic perylene diimide (CUPDI) with PDI as an acceptor is designed and synthesized. A polar chiral dipeptide composed of β -alanine and l -histidine with the imidazole ring as the donor i.e. , l -carnosine, is incorporated at one of imides . Transition of various supramolecular assemblies of CUPDI is realized by changing CUPDI concentration and solvents. The photoelectronic properties of the assemblies are investigated as well as their association with the microstructure of the nanomaterials. Findings: Delicately tuned hydrogen bonds between the peptides and π - π interaction between PDI cores in different solvents enable the formation of assemblies with multifarious microstructures such as small spherical aggregates, nanowires with uniform diameter, nanobelts , and irregular aggregates. The maximum amount of photocurrent enhancement is up to 1.08 µA observed for the nanobelt, four times higher than that of irregular aggregates. However, the nanowires show the best performance of 7.1-fold in response to ammonia. Thus, the photoelectric performances are strongly dependent on the the molecular arrangement within the nanomaterials.

Carnosine is a natural bioactive dipeptide with important physiological functions widely used in food and medicine. Dipeptidase (PepD) from Serratia marcescens can catalyze the reverse hydrolytic reaction of β-alanine with l-histidine to synthesize carnosine in the presence of Mn2+. However, it remains challenging to practice carnosine biosynthesis due to the low activity and high cost of the enzyme. Therefore, the development of biocatalysts with high activity and stability is of significance for carnosine synthesis. Here, we proposed to chelate Mn2+ to polyethylenimine (PEI) that induced rapid formation of calcium phosphate nanocrystals (CaP), and Mn-PEI@CaP was used for PepD immobilization via electrostatic interaction. Mn-PEI@CaP as the carrier enhanced the stability of the immobilized enzyme. Moreover, Mn2+ loaded in the carrier acted as an in situ activator of the immobilized PepD for facilitating the biocatalytic process of carnosine synthesis. The as-prepared immobilized enzyme (PepD-Mn-PEI@CaP) kept similar activity with free PepD plus Mn2+ (activity recovery, 102.5%), while exhibiting elevated thermal stability and pH tolerance. Moreover, it exhibited about two times faster carnosine synthesis than the free PepD system. PepD-Mn-PEI@CaP retained 86.8% of the original activity after eight cycles of batch catalysis without the addition of free Mn2+ ions during multiple cycles. This work provides a new strategy for the co-immobilization of PepD and Mn2+, which greatly improves the operability of the biocatalysis and demonstrates the potential of the immobilized PepD system for efficient carnosine synthesis.

Background: This study aimed to explore the regulatory effect of anserine on HUVEC cell injury and thrombosis in deep venous thrombosis (DVT) rats, and to elucidate the underlying molecular mechanisms. Methods: Non-targeted metabolomics data analyses were conducted using an ultra-performance liquid chromatography system Vanquish UHPLC and mass spectrometer to detect plasma metabolism profiles. The transcriptome sequencing and gene intervention experiments were performed to verify the regulatory effect. Further in vivo and in vitro experiments were performed. Enzyme-linked immunosorbent assay was used to detect the levels of P-selectin, E-selectin, and vWF, hematoxylin-eosin (HE) staining was performed to observe thrombotic and inflammatory cell infiltration, flow cytometry and TUNEL assays were performed to detect apoptosis, and qPCR and WB assays were conducted to determine the gene and protein expression. Results: Anserine alleviated HUVECs injury, reduced adhesion molecule expression, and inflammation. It decreased P-selectin, E-selectin, vWF, THBD, TFPI levels, and apoptosis while promoting NOS3, ET-1, and NO release in HUVECs. In DVT rats, anserine reduced P-selectin, E-selectin, vWF, thrombosis, cell infiltration, apoptosis, and promoted NO release. Transcriptome sequencing and gene intervention confirmed anserine’s regulation of the PI3K-Akt pathway and coagulation via MYB. CARNMT1, a regulatory enzyme for anserine metabolism, increased anserine content, inhibiting coagulation, thrombosis, cell infiltration, and promoting NO release in rats. Conclusion: This study confirmed anserine could alleviate DVT by improving the inflammatory response, inhibiting blood agglutination, and promoting vasodilation, providing new potential therapeutic targets, important scientific evidence for the development of DVT management, and new clues for an in-depth understanding of its molecular mechanisms.

Natural bioactive peptides (NBPs) possess diverse physiological and pharmacological activities but face challenges in separation and detection due to weak ultraviolet absorption, similar structures, and coexisting chiral and achiral components. A new chiral stationary phase (CSP), Sil-Star-multi-CD CSP, was constructed by synthesizing a multi-terminal star cyclodextrin polymer (Star-multi-CD) with a full 2,3,6-derived CD as the parent nucleus. Sil-Star-multi-CD CSP has multiple interaction sites and altered spectral properties of guests, enabling the simultaneous separation and quantification of chiral and achiral physiological indicators, including endogenous peptides and common metabolites in serum. A simplified strategy was established for analyzing structurally similar NBPs in chicken breast. The limits of detection and quantitation of carnosine analogues ranged from 1.50 to 6.88 μg mL −1 and 5.00 to 22.9 μg mL −1 , respectively. Achieving highly selective separations and sensitive detection in a single column for complex biological matrices provides a convenient strategy in the analysis of functional NBPs.

Aqueous zinc-ion batteries (AZIBs) are promising for grid-scale energy storage but suffer from Zn anode instability caused by dendrite growth, hydrogen evolution, and corrosion. In this work, L-carnosine (L-C), a natural dipeptide, is introduced as an electrolyte additive to address these challenges, due to its dual ion-chelating and Zn-adsorbing capabilities. It is demonstrated that L-C can regulate Zn 2+ solvation structures by displacing water molecules via strong coordination bonds. Simultaneously, L-C can preferentially adsorb on Zn surfaces, guiding uniform Zn deposition along the (101) plane and forming a dynamic protective layer that dynamically repairs interfacial defects. This self-healing capability enables short-circuited Zn anodes to recover stable cycling for over 2000 h, while pre-scratched surfaces are fully restored during cycling. These synergistic mechanisms enable Zn||Zn symmetric cells to achieve exceptional cycling stability (>5400 h at 1 mA cm −2 , 1 mAh cm −2 ) and high-rate durability (>1100 h at 5 mA cm −2 , 5 mAh cm −2 ). Full cells, paired with NH 4 V 4 O 10 (NHVO) cathodes, retain 82.26% capacity after 2000 cycles at 5 A g −1 , highlighting practical viability. The bioinspired solvation-interfacial dual-regulation strategy provides a universal approach for designing eco-friendly, high-performance AZIBs.

Understanding the interrelationships between the differentially abundant microorganisms and metabolites of traditional “Fuet” fermented sausages (FSs) and inoculated fermented sausages (IFSs) can help us identify key species that are missing from commercial starter cultures to reproduce the flavor compounds and nutrients of traditional Fuet FSs. IFSs, inoculated with P. pentosaceus , P. acidilactici , S. xylosus , S. carnosus (SBM-52) or P. pentosaceus , and S. xylosus (THM-17), were deficient in reproducing the volatilome profile (in particular esters, methyl aldehydes, and methyl ketones) of traditional Fuet FSs because of the lack of diverse Staphylococci ( S. carnosus , S. xylosus , S. equorum , and S. saprophyticus ). Moreover, the combination of Pediococcus and Staphylococcus were positively associated with amino acid, fatty acid, l -anserine, and l -carnosine levels. Pyridoxal and indolelactic acid levels were significantly increased in IFSs with the addition of P. acidilactici and S. carnosus , which were positively associated with vitamin B 6 and tryptophan metabolic pathways.
