Investigating the Influence of Different Umami Tastants on Brain Perception via Scalp Electroencephalogram

Ben Wu,Imre Blank,Yin Zhang,Yuan Liu

Three types of tastants are known as perceptually associated with umami taste: monosodium glutamate (MSG), disodium succinate (WSA), and disodium inosine monophosphate (IMP). While these tastants were confirmed to be perceptually similar in a sensory study, they could be discriminated (p < 0.05) by electroencephalogram (EEG) analysis on a time scale of 5–6 s. In comparison of the EEG responses of the participants, the brain could partly distinguish (p < 0.05) between different sensory intensities of MSG, WSA, or IMP. The EEG data indicated that the brain is partially sensitive to perceiving different sensory intensities (L, low; M, medium; and H, high) of the same umami stimuli; i.e., for MSG in μV2/Hz, L, 2.473 ± 0.181; M, 3.274 ± 0.181; and H, 3.202 ± 0.181. However, brain responses of perceptually equi-umami intensities could partially be discriminated, suggesting that the brain could partially discriminate (p < 0.05) MSG, WSA, and IMP, despite similar sensory intensities. Moreover, umami tastants were also found to significantly enhance (p < 0.05) the α wave activity, with the most responsive being at 10 Hz, particularly in the frontal and parietal and occipital regions of the brain (p < 0.001). This study shows the potential of EEG to investigate brain activity triggered by umami stimuli.

Nicotine plays a protective role in rats with induced viral pneumonia with polyinosinic-polycytidylic acid through α7nAChR

Wei Liu,Yi Zhu,Hong Yan,Lingyun Ren,Jingli Chen

Objective To study the effect of nicotine in rat model of pneumonia induced by polyinosinic-polycytidylic acid [Poly (I:C)] and explore the underlying mechanism. Methods Twenty-four healthy adult male Sprague-Dawley (SD) rats (200–250 g) were randomly divided into normal saline control group (NS group); Poly (I:C) group; nicotine group (NIC group); and α7 nicotinic acetylcholine receptor (α7nAChR) antagonist group (α-BGT group) (n = 6 each). Rats in the Poly (I: C), NIC, and α-BGT groups were administered 1.5 mg/mL 100 μL Poly (I:C) intranasally to establish pneumonia model. In α-BGT group, 1 μg/kg α-bungarotoxin (α-BGT) was intraperitoneally injected 45 min before intranasal Poly (I:C), and 400 μg/kg nicotine was intraperitoneally injected 15 min after α-BGT injection. The NIC group received an equal volume of NS in place of α-BGT while the other treatments were same. The Poly (I:C) group received equal volume of NS in place of nicotine while the other treatments were same as in NIC group. In the NS group, only NS was administered at all three time points. PaCO 2 , PaO 2 , and PaO 2 /FiO 2 levels were determined 24 h after administration of Poly (I:C). After euthanization, rat lung tissues were extracted for pathological examination, and wet weight/dry weight (W/D ratio) was determined. Expression of tumor necrosis factor-α (TNF-α), interleukin (IL)-6, IL-1β, and interferon (IFN)-γ in lung tissue was determined by ELISA. q-PCR was used to detect nuclear factor kappa-B P65 (NF-κBP65). Results Compared with NS group, Poly (I:C) and α-BGT groups showed significantly increased W/D ratio, PaCO 2 , TNF-α, IL-6, IL-1β, and IFN-γ content, NF-κB P65 expression, and reduced PaO 2 and PaO 2 /FiO 2 ( p  < 0.05), along with obvious signs of pathological injury. Nicotine pre-treatment reduced W/D ratio, PaCO 2 , proinflammatory cytokines, NF-κBP65 expression, and increased PaO 2 and PaO 2 /FiO 2 levels. The above effects were negated in α-BGT group. Conclusion Pre-administration of nicotine improved Poly (I:C)-induced pneumonia by activating the cholinergic anti-inflammatory pathway.

Biocompatible polypyrrole-block copolymer-gold nanoparticles platform for determination of inosine monophosphate with bi-enzyme biosensor

Chong Sun,Ling Gao,Daoying Wang,Muhan Zhang,Yuan Liu,Zhiming Geng,Weimin Xu,Fang Liu,Huan Bian

Inosine monophosphate (IMP) is considered to be an important constituent in the formation and development of meat flavor. In this paper, a bi-enzyme biosensor system for determination of IMP was developed to evaluate the freshness and quality of stored muscles. More details of preparing for polypyrrole-Pluronic F127-gold nanoparticles (PPy-F127-Au NPs) and immobilizing bi-enzyme (peroxidase and 5′-nucleotidase) on PPy-F127-Au NPs modified glassy carbon electrode (GCE) were presented. The electrochemical behaviors of the bi-enzyme biosensor were analyzed, and the concentration of IMP was determined based on the bi-enzyme biosensor response. The results of linear sweep voltammetric (LSV) and square wave voltammetry (SWV) indicated that the catalytic reaction was occurred favorably on the surface of the bi-enzyme biosensor, with a wide detection range (0.05–15   g/L) and a low detection limit of 3.85   ng/mL. The good anti-interference ability, stability and repeatability of this biosensor were also proved. The analytical results by the biosensor system did correlate well with those obtained by liquid chromatography.

Sustained photodynamic H2O2 generation via disodium 5′-inosinate enhances cooked meat preservation by membrane-targeted antibacterial action

Lijun Tan,Shasha Zheng,Yong Xie,Wendi Zhang,Baocai Xu

Photodynamic inactivation (PDI) is a promising non-thermal antimicrobial technology known for broad-spectrum efficacy and low resistance potential. However, conventional PDI face limitations in bactericidal efficacy due to the short-lived and poorly diffusing reactive oxygen species (ROS). This study proposes a novel food-grade PDI strategy that incorporates disodium 5′-inosinate (IMP) into a vitamin B2 (VB2)-mediated system, significantly enhancing H 2 O 2 production. Acting as an electron donor, IMP promotes type I photochemical reactions, enabling sustained H 2 O 2 generation and achieving >6 log CFU/mL inactivation of Bacillus safensis , a spoilage bacterium often overlooked in cooked meat products. Mechanistic analyses showed that H 2 O 2 oxidized key phospholipids in the cell membrane, increased permeability, caused membrane potential hyperpolarization and ion leakage, and ultimately disrupted multiple membrane domains. Moreover, this strategy also exhibited persistent antimicrobial activity, effectively delaying spoilage in cooked meat products stored at 4 °C and 25 °C. Overall, this study introduces a safe and effective non-thermal sterilization method with promising industrial potential and offers a novel pathway to enhance PDI efficacy via intensified type I ROS mechanisms.

Light-controllable charge-reversal nanoparticles with polyinosinic-polycytidylic acid for enhancing immunotherapy of triple negative breast cancer

Lei Fang,Zitong Zhao,Jue Wang,Ping Xiao,Xiangshi Sun,Yaping Ding,Pengcheng Zhang,Dangge Wang,Yaping Li

Nucleic acid drugs are highly applicable for cancer immunotherapy with promising therapeutic effects, while targeting delivery of these drugs to disease lesions remains challenging. Cationic polymeric nanoparticles have paved the way for efficient delivery of nucleic acid drugs, and achieved stimuli-responsive disassembly in tumor microenvironment (TME). However, TME is highly heterogeneous between individuals, and most nanocarriers lack active-control over the release of loaded nucleic acid drugs, which will definitely reduce the therapeutic efficacy. Herein, we have developed a light-controllable charge-reversal nanoparticle (LCCN) with controlled release of polyinosinic-polycytidylic acid [Poly(I:C)] to treat triple negative breast cancer (TNBC) by enhanced photodynamic immunotherapy. The nanoparticles keep suitably positive charge for stable loading of Poly(I:C), while rapidly reverse to negative charge after near-infrared light irradiation to release Poly(I:C). LCCN-Poly(I:C) nanoparticles trigger effective phototoxicity and immunogenic cell death on 4T1 tumor cells, elevate antitumor immune responses and inhibit the growth of primary and abscopal 4T1 tumors in mice. The approach provides a promising strategy for controlled release of various nucleic acid-based immune modulators, which may enhance the efficacy of photodynamic immunotherapy against TNBC

Dynamic sensations of fresh and roasted salmon (Salmo salar) during chewing

Meng Dong,Yu-Ying Zhang,Xu-Hui Huang,Ran Xin,Xiu-Ping Dong,Kunihiko Konno,Bei-Wei Zhu,Ian Fisk,Lei Qin

The sensory perception of food is a dynamic procedure, which is closely related to the released flavor stimuli. Thus, we evaluated the dynamic sensations of fresh and roasted salmon during the chewing process and investigated the tastants released in saliva. For fresh salmon, the fishy, umami, salty, and sweet attributes were perceived successively. Meanwhile, the smoky and fried flavors were the most dominant attributes of roasted salmon at the beginning, then various attributes were perceived. During the chewing process, free amino acids and 5′-nucleotides released in saliva were quantified. Compared to the sensory data, the results demonstrated that glutamic acid and inosine 5′-monophosphate released in saliva might induce the umami perception. The sweet-tasting amino acids alanine and glycine may contribute to sweetness. Therefore, we suggested that the time dimension of tastants dissolved in saliva would affect the dynamic sensation of food, even for complex food materials.

Molecular cloning, characterization and expression analysis of CXCR3a and CXCR3b from Nile tilapia (Oreochromis niloticus)

Along Gao,Liting Wu,Lan Li,Biao Han,Jianmin Ye

The CXC chemokine receptors (CXCRs) are members of the seven transmembrane (7-TM) G-protein-coupled receptor superfamily that involves innate and adaptive immune systems. In this study, CXCR3a and CXCR3b from Nile tilapia ( Oreochromis niloticus ) were cloned and identified, designated as OnCXCR3a and OnCXCR3b . The open reading frames of OnCXCR3a and OnCXCR3b were 1074 and 1080 bp, encoding the predicted proteins of 357 and 359 amino acids, respectively. Multiple alignment analysis of OnCXCR3a- and OnCXCR3b-deduced protein sequences with the mammalian and bird sequences indicated the presence of typical structural features of chemokine receptors, including a 7-TM domain and conserved motifs. Quantitative real-time PCR analysis revealed that OnCXCR3a and OnCXCR3b were constitutively expressed in a wide range of tissues. When stimulated with Streptococcus agalactiae , Aeromonas hydrophila , polyinosinic:polycytidylic acid and lipopolysaccharide in vivo or in vitro on leukocytes, the mRNA levels of OnCXCR3a and OnCXCR3b were significantly upregulated. Overall, these results indicated that OnCXCR3s might be involved in host immune responses in Nile tilapia.

Spoilage microbes’ effect on freshness and IMP degradation in sturgeon fillets during chilled storage

Jiatong Li,Genghui Zhou,Peng Xue,Xiuping Dong,Yongtao Xia,Joe Regenstein,Ming Du,Liming Sun

Bacterial spoilage is an important reason causing freshness deterioration of fish during postmortem storage. The aim of this study is mainly to investigate the role of bacteria in freshness and the degradation of inosine 5′-monophosphate (IMP) as well as nucleotide derived values in chilled sturgeon (hybrid, Acipenser schrenckiid  ×  Huso dauricus ) fillets using a microbial reduction pretreatment. During storage at 4 or -1 °C, lower total bacterial count and total volatile base nitrogen in the treated samples was observed when compared to the control samples. But very close K, Ki value and IMP was observed in both groups, although higher IMP was recorded at day 6 and 9 at 4 °C. In later storage period, accelerated decrease of inosine (Ino) and increment of hypoxanthine (Hx) occurred in two control groups, resulting in a rapid increase of G and H values, significant higher than the treated groups. Six predominant spoilage bacteria ( Acinetobacter johnsonii , Acinetobacter lwoffii , Shewanella baltica , Pseudomonas Fragi , Pseudomonas koreensis and Pseudomonas antarctica ) were isolated from the sturgeon fillets during storage. Based on the results of respective co-cultivation of IMP with the six species, only Acinetobacter johnsonii had ability to degrade IMP when reached a high density of 2.84 × 10 9  CFU/mL. Results suggested that microbial reduction pretreatment could better the freshness of chilled sturgeon fillet. Comparing to endogenous IMPase, spoilage bacteria probably played a small role in IMP degradation, but affected considerably the decomposition of Ino to Hx.

A peptide-AIEgen nanocomposite mediated whole cancer immunity cycle-cascade amplification for improved immunotherapy of tumor

Jun Dai,Xiaoqi Dong,Rui Liu,Biao Chen,Xiyuan Dong,Quan Wang,Jing-Jing Hu,Fan Xia,Xiaoding Lou

Immunotherapy maintains the cancer-immunity cycle via re-activating the immune system, so as to achieve the purpose of anti-tumor. However, the response rate of current tumor immunotherapy strategies is still low. Even the most reported immune checkpoint block (ICB), the objective response rate (ORR) is only about 10–30%. Here, aiming at obtaining a higher response rate, we designed a cascade amplification nanocomposite consisting of the immune adjuvant polyinosinic:polycytidylic acid [Poly (I:C)] and aggregation-induced emission luminogen (AIEgen)-modified modular peptide (named PMRA). The PMRA includes: D PPA-1 peptide (P), an immune checkpoint inhibitor; PLGLAG peptide (M), a matrix metalloproteinase 2 (MMP-2) responsive sequence to promote the release of D PPA-1; RRRRRRRR peptide (R), for loading the Poly (I:C); PyTPA (A), a photosensitizer with AIE property. In cancer-immunity cycle, photodynamic therapy (PDT) mediated by PyTPA promotes the release of tumor-associated antigens (TAAs), and primes T lymphocytes . The cytokines coming from the stimulation of PDT and Poly (I:C) promote the activation of T lymphocytes. The high level of chemokines in tumor microenvironment promotes immune cells migration and infiltration in tumor with the assistance of PDT. Finally, through ICB with D PPA-1 peptide, T lymphocytes enhance the recognition of tumor cells and killing tumor cells. Immunogenic cell death induces the release of more TAAs, which will enter the next cycle and complete the full-loop again. Taking advantages of whole cancer-immunity cycle, the cascade amplification nanocomposite achieved almost 100% ORR in vivo . This concept of whole cancer-immunity cycle enhanced immunotherapy provides a novel perspective for tumor treatment.
