Heparin sodium-selective ‘on–off’ and lysine-selective ‘off–on’ fluorescence switching of cadmium telluride quantum dots and their analytical applications

Hong Zhi Zhang,Rong Sheng Li,Ni Wang,Li Qi,Cheng Zhi Huang,Jian Wang

Sensing biomolecules such as heparin sodium and lysine are of great significance. In this work, a “turn-off–on” fluorescence switching of cadmium telluride quantum dots (CdTe QDs) was designed for both heparin sodium and lysine. Even though both heparin sodium and thioglycolic acid (TGA)-capped CdTe QDs are negatively charged, they are capable of forming a self-assembly and even aggregates through hydrogen bonding, depending on the concentration of heparin sodium, which leads to the sensitive fluorescence quenching of CdTe QDs. Thus, a fluorescence ‘turn-on’ analytical method for heparin sodium sensing could be established with a detection range of 0.200–5.000 μg mL−1 and the detection limit was 0.033 μg mL−1, which is applicable to determining heparin sodium in injection samples. What's more, a sensitive and selective “turn-off–on” nanosensor was developed for lysine analysis with the detection range of 2–200 μmol L−1 and the detection limit as low as 0.146 μmol L−1. This approach offers a new, simple, fast and selective method for determining heparin sodium and lysine.

A Novel Fluorescent Detection Strategy for Lysozyme in Tears Based on Glycoside Bond Hydrolysis

Wei Wei,Yiwei Zhang,Mengdi Yu,Di Shen,Xiyu Sun,Mingshi Fu,Chi Zhang,Zhenzhen Jia,Mingzhen Zhang

Dry eye disease (DED) is a prevalent condition characterized by a multifaceted etiology, with its incidence exhibiting an upward trajectory. Consequently, it is imperative to develop a sensitive, straightforward, and convenient method for the analysis of biomarkers associated with DED to facilitate its auxiliary diagnosis. Lysozyme (LYZ), produced by the lacrimal gland, is a class of antibacterial enzymes believed to play a crucial role in immunity and is associated with DED. In this study, a novel fluorescent sensing platform utilizing neutral red-heparin sodium (NR-HS) was developed with LYZ as the target. The platform operates on the principle of static quenching, where HS effectively quenches the fluorescence of NR. As a hydrolase, LYZ can catalyze the hydrolysis of the glucoside bond in HS, thereby modulating the transformation of NR-HS fluorescence signal. This provides a straightforward fluorescent method for monitoring LYZ levels. Under optimal conditions, the developed "on-off-on" NR-HS sensing platform demonstrated the capability to detect LYZ within a range of 0.5 to 10 µg/mL, with a detection limit of 0.42 µg/mL, and exhibited enhanced specificity for LYZ. In conclusion, a cost-effective, rapid, and efficient LYZ sensing platform was established, which facilitates the diagnosis of DED and shows potential as a diagnostic detection technique.

High strength and antibacterial polyelectrolyte complex CS/HS hydrogel films for wound healing

Mengmeng Shu,Shijun Long,Yiwan Huang,Dapeng Li,Haiyan Li,Xuefeng Li

We report a simple and facile self-assembly approach to fabricate polyelectrolyte complex (PEC) hydrogel films with positively charged chitosan (CS) and negatively charged heparin sodium (HS) by combining hydrogen bonding and electrostatic interactions. The CS/HS hydrogel films exhibited excellent tensile strength and toughness, good self-recovery ability, superior water absorbency, and pH-dependent surface charge characteristics. The gelation mechanism was investigated by zeta potential measurements. The CS/HS hydrogel films exhibited high antibacterial efficacy against E. coli at selected pHs or when coordinated with various metal ions and a significant effect on accelerating wound healing. The self-assembly approach presented in this work may serve as a generic strategy for the fabrication of novel multi-functional PEC hydrogels for broad biomedical applications.

Ultrasonically Prepared Sodium Heparin-Stabilized Indocyanine Green/Nano-Hydroxyapatite Suspension for Collaborative Photodynamic and Photothermal Tumor Therapy

Qifa Song,Zi-You Ding,Chengyu Li,Yiran Fan,Xinli Tang,Yingchao Han

Indocyanine green (ICG) shows promise for diagnosing and treating tumors due to its good photothermal and fluorescent properties. In this study, sodium heparin (SH)-stabilized ICG/nano-hydroxyapatite (SH-ICG/nHAP) suspension was ultrasonically prepared to achieve photothermal and photodynamic collaborative therapy (PTT/PDT) for treating tumors. The nHAP had a short rod-like shape and a drug-loading capacity of 13.3% for ICG, corresponding to a drug-loading efficiency of 88.6%. In addition, the SH-ICG/nHAP suspension showed a very low release of ICG in PBS (7.4) and a slightly improved release in acidic buffers (6.5, 5.5), indicating an excellent binding ability of nHAP for ICG. The resulting SH-ICG/nHAP showed good suspension stability. Under an 808 nm near-infrared (NIR) laser, SH-ICG/nHAP showed good photothermal properties and could produce reactive oxygen species (ROS). Under the irradiation of an 808 nm NIR laser at 0.8 W/cm2for 5 min, SH-ICG/nHAP was found to significantly inhibit HepG2 cells proliferation (78.58%), similar to free ICG.In vivo, SH-ICG/nHAP was found to exert an improved inhibitory effect on tumor growth compared to free ICG. Biocompatible and stable SH-ICG/nHAP suspension like this could be a promising system for the PTT/PDT of tumors.Keywords:ICG;nano-hydroxyapatite;suspension;PTT;PDT

Rapid and fast fluorescent sensing through heparin and chitosan encapsulated Eu3+ complexes for rapid detection of pesticides

Wei Ding,Xiaoyan Liu,Waquar Ahmed Khokhar,Chengbiao Wei,Faisal U.L. Rehman,Guoxu Wang,Zafar Mahmood,Abdur Raheem Aleem

Vegetables and fruits are essential sources of nutrients, minerals, vitamins, and dietary fiber for a well-balanced diet. However, excessive pesticide use, particularly 2,6-dichloro-4-nitroaniline (DCN), poses serious health and environmental risks. Traditional pesticide detection methods require expensive equipment, trained personnel, making them unsuitable for on-site sensing. In this study, we synthesized europium (Eu 3+ ) complexes incorporating heparin sodium (Hep) and chitosan (CS) to develop a rapid, cost-effective, and highly sensitive fluorescent sensor for DCN detection. The optimized Eu 3+ complex (ETP-HS3) demonstrated excellent water stability, strong red fluorescence, and a long fluorescence lifetime (888.09 μs). It exhibited a low limit of detection (LOD = 0.9049 μM), a rapid sensing response (20 s), and a high quenching constant (Ksv = 1.226 × 10 4  M −1 ), making it an effective and portable fluorescence sensor for real-time pesticide monitoring. The fluorescence quenching mechanism was thoroughly investigated, confirming the selectivity and efficiency of the sensor.

Composite nanofibrous dressing loaded with Prussian blue and heparin for anti-inflammation therapy and diabetic wound healing

Mengmeng Li,Xilan Li,Yujie Gao,Yang Yang,Chenggang Yi,Wei Huang,Bingbing Shen,Dongming Qi,Zhengwei Mao,Jindan Wu

Diabetic ulcer is a severe complication of diabetes that can lead to amputation due to the overproduction of pro-inflammatory factors and reactive oxygen species (ROS). In this study, a composite nanofibrous dressing was developed by combining Prussian blue nanocrystals (PBNCs) and heparin sodium (Hep) through electrospinning, electrospraying, and chemical deposition. The nanofibrous dressing (PPBDH) was designed to take advantage of the excellent pro-inflammatory factor-adsorbing capability of Hep and the ROS-scavenging capabilities of PBNCs, resulting in synergistic treatment. It is worth noting that the nanozymes were firmly anchored to the fiber surfaces through slight polymer swelling caused by the solvent during electrospinning, thereby guaranteeing the preservation of the enzyme-like activity levels of PBNCs. The PPBDH dressing was found to be effective in reducing intracellular ROS levels, protecting cells from ROS-induced apoptosis, and capturing excessive pro-inflammatory factors, including chemoattractant protein-1 (MCP-1) and interleukin-1β (IL-1β). Furthermore, a chronic wound healing evaluation conducted in vivo demonstrated that the PPBDH dressing was able to effectively alleviate the inflammatory response and accelerate wound healing. This research presents an innovative approach to fabricate nanozyme hybrid nanofibrous dressings, which have great potential in accelerating the healing of chronic and refractory wounds with uncontrolled inflammation.

Multiscale Cross-Linking via Polysaccharide-Assisted Robust Zwitterionic Hydrogel Interface Enabling Electrochemical Sensing in Complex Serum

Peng Sun,Jie Jin,Meiling Wang,Jiao Ma,Hua Wang

The serious fouling layer from the nonspecific adsorption in complex media would greatly impair the analytical performance and even lead to the complete failure of the electrochemical sensor. Zwitterionic hydrogel has been demonstrated to possess excellent antifouling capability, while it suffers from a weak interface and spontaneous detachment from the electrode surface due to the double-edged hydrophilicity. Here, we found that the anionic polysaccharide coating on the liquid metal (LM) nanoparticle surface, in combination with the cationic chitosan penetrated in the hydrogel network, could synergistically enhance the interface adhesion while retaining antifouling ability, providing an ingenious solution to this issue. Additionally, only ultrasonic treatment was involved for the preparation of this LM-based zwitterionic hydrogel because ultrasonication could easily induce the formation of LM nanoparticles, the self-assembly of anionic polysaccharide on its surface, and simultaneous free radical polymerization of zwitterionic monomer. Finally, an electrochemical immunosensor was successfully fabricated based on this polysaccharide cross-linked LM-based zwitterionic hydrogel, and an ultralow detection limit of 7.17 pg·mL–1, while a wide linear range from 10 pg·mL–1 to 10 μg·mL–1 was obtained in 100% human serum with negligible difference from that in PBS solution. We believe the polysaccharide-stabilized antifouling hydrogel interface presented in this work could inspire more innovative research for biosensing and bioelectronics in the future.

Rational Design of Cationic Nanogels with Serum Tolerance for Efficient siRNA Delivery and Antitumor Therapy

Yifan Gao,Yuening Qiu,Hongyang Zhao,Yilei Yu,Mingwei Wang,Xuhong Guo,Martien A. Cohen Stuart,Junyou Wang

Cationic polymers have been applied widely for siRNA delivery, yet most of the conventional linear or branched polymers make RNA polyplexes with poor serum stability and low delivery efficacy. We here propose cationic nanogels as a better alternative and demonstrate their advantages for siRNA delivery. Using a recently developed electrostatic-templated polymerization method, we synthesized a series of cationic nanogels with precisely controlled chemical composition, size, and cross-linking degree, enabling the systematic exploration of structure–performance relationships for nanogel-mediated siRNA delivery. We show that the nanogels, with their stable 3D structure, offer both surface and interior sites for siRNA loading depending on the N/P ratio. At high N/P ratios, siRNA binds primarily to the interior sites of the nanogels, which provide enhanced protection against competing serum proteins and facilitate efficient cellular uptake. Furthermore, the chemical composition, size, and degree of cross-linking of the nanogels cooperatively influence siRNA loading and cellular uptake. The optimal formulation, poly(vinylbenzyl trimethylammonium chloride) (PVBTMA) nanogel, incorporates a strong charge group and a hydrophobic phenyl spacer that facilitate siRNA loading and cellular uptake, respectively. With an optimal size of 107 nm and 5% cross-linking, PVBTMA nanogel achieves superior siRNA internalization compared to widely used transfection agents like Lipofectamine 3000 and branched PEI25k. Finally, when loaded with tumor-specific gene silencer siPLK1, the PVBTMA nanogel facilitates superior gene silencing in vitro and effectively inhibits tumor growth in vivo. Our findings clarify the distinct advantages of cationic nanogels as carriers for siRNA delivery and highlight their promising potential for therapeutic applications.

A nitric oxide-releasing organic selenium-grafted phosphorylcholine coating for thrombosis and inflammation prevention in extracorporeal membrane oxygenation

Lietao Wang,Fanjun Zhang,Rifang Luo,Xin Fu,Wei Lai,Hui Yan,Yao Xiong,Wei Zhang,Zhongwei Zhang,Min He,Yubing Wang

Thrombosis and inflammation represent major challenges limiting the functionality of asymmetric poly(4-methyl-1-pentene) (PMP) hollow fiber membranes in extracorporeal membrane oxygenation (ECMO) systems. Moreover, the interplay between these two pathological processes can further exacerbate both coagulation and inflammatory responses. In this study, a dual-functional surface modification strategy was developed by first pre-functionalizing PMP hollow fiber membranes with selenocystamine, followed by grafting a phosphorylcholine copolymer MA(PCLA) to form a coating with combined anti-thrombotic and anti-inflammatory properties. Selenocystamine mimics endothelial nitric oxide release to suppress inflammatory responses, while MA(PCLA) mimics the structure of cell membranes to reduce protein adsorption and cellular adhesion. This approach is designed to provide a safer and more effective strategy for mitigating thrombotic and inflammatory complications. In vitro blood circulation assays demonstrated that the modified membranes exhibited significantly reduced protein adsorption, platelet adhesion, and thrombosis compared to both unmodified and commercially modified PMP membranes. Furthermore, both in vitro and in vivo experiments confirmed that the coating effectively modulates inflammatory cell differentiation and attenuates inflammatory responses. The dual-functional coating, exhibiting synergistic anti-thrombotic and anti-inflammatory effects, holds considerable promise for application in blood-contacting medical devices, particularly ECMO systems.

Targeted delivery of PTX by lactoferrin-modified nanoemulsions for the treatment of glioblastoma

Haiming Huang,Jiyong Peng,Zhiting Gao,Yongtong Huang,Wenyang Song,Wenhao Wu,Song Gao,Songsen Chen,Qingchun Xie,Shu Zhang,Jiu Wang

Glioblastoma treatment is hindered by the blood-brain barrier (BBB), which limits the penetration and accumulation of chemotherapeutic agents. Paclitaxel (PTX), an effective chemotherapeutic drug, faces clinical challenges due toits poor solubility and restricted ability to traverse the BBB. Consequently, there is an urgent need for advanced drug delivery systems to facilitate the efficient and safe translocation of PTX across the BBB. In this study, PTX was encapsulated within nanoemulsions (NEs) conjugated to lactoferrin (Lf) via electrostatic interactions, followed by the optimization of its formulation. To investigate cellular uptake and BBB penetration, fluorescent dye coumarin 6 (C6) was incorporated into NEs. Uptake was evaluated in GL261 cells and BBB penetration in hCMEC/D3 cells. Further studies were conducted on the biodistribution in mice and the therapeutic efficacy in murine intracranial glioblastoma model. Characterization of PTX@Lf-NE demonstrated stability, biological safety, and favorable release properties. Notably, the fluorescence intensity of C6@Lf-NE was twice of C6@NE in one hour post-administration, and the drug uptake rate decreased with the addition of free Lf, suggesting that Lf promotes the ability of NEs to traverse the BBB. In vivo distribution further revealed that Lf-NE increased brain distribution while reduced accumulation in other organs. In the glioblastoma model, it was found that the bioluminescent intensity of PTX@Lf-NE was significantly decrease than that of PTX@NE on the 15th day of administration, indicating that the modification with Lf facilitated the targeted delivery of PTX and enhanced its therapeutic efficacy. This study successfully designed and developed an effective drug delivery system for glioblastoma treatment, which improves the translocation of drugs across the BBB.

Single-channel electrochemical biosensor for simultaneous monitoring of glucose and uric acid

Changyun Quan,Liping Wen,Minghui Yang,Binjie Xu

Given the metabolic interplay between glucose and uric acid, individuals should proactively monitor and be alert of elevated levels of both substances. For the first time, this paper describes a novel design of electrochemical biosensor based on screen-printed electrode of a single channel but capable of simultaneously measuring blood glucose and uric acid concentrations. Experimental results demonstrated that this single-channel biosensor exhibits good performance towards glucose and uric acid, such as broad linear range, good reproducibility, good stability, and strong resistance against interference. For spiked recovery experiments on human venous blood samples, the glucose recovery rate ranged from 95.9 % to 103.2 %, while the uric acid recovery rate ranged from 99.7 % to 104.9 %. And the relative standard deviation for both parameters was less than 5 %, indicating that this biosensor holds promising prospects for commercial application. This is truly the first time that enable simultaneous detection of glucose and uric acid concentrations using just one drop of blood and a single test strip. It reduces the number of blood draws required, lowers analysis costs, and significantly improves screening efficiency for individuals with diabetes and hyperuricemia.
