Multi-Targeted Peptide-Modified Gold Nanoclusters for Treating Solid Tumors in the Liver

Feng Xiao,Yao Chen,Jie Qi,Qiaofeng Yao,Jianping Xie,Xingyu Jiang

Apoptosis and autophagy determine the fate of cancer cells. However, simply promoting apoptosis of tumor cells is limited in the treatment of unresectable solid liver tumors. Generally, autophagy is considered the anti-apoptotic “guardian”. But the pro-apoptotic effects of autophagy can be activated by excessive endoplasmic reticulum (ER) stress. Here, amphiphilic peptide-modified glutathione (GSH)-gold nanocluster aggregates (AP 1 P 2 -PEG NCs) were designed with the enrichment of solid liver tumors and the prolonged stress in the ER, which can achieve the mutual promotion of autophagy and apoptosis in liver tumor cells. In this study, orthotopic and subcutaneous liver tumor models show the anti-tumor effectiveness of AP 1 P 2 -PEG NCs, with a better antitumor effect than sorafenib, biosafety (Lethal Dose, 50% (LD 50 ) of 827.3 mg kg −1 ), wide therapeutic window (non-toxic in 20 times of therapeutic concentration) and high stability (blood half-life of 4 h). These findings identify an effective strategy to develop peptide-modified gold nanocluster aggregates with low toxicity, high potency, and selectivity for solid liver tumors treatment.

Intratumoral self-assembly of renal-clearable gold nanoparticles as precise photothermal nanomedicine for liver tumor therapy

Gangqiang Yuan,Xiaoxi Luo,Kui He,Yue Tan,Caiming Luo,Ben Liu,Yidan Sun,Jinbin Liu

Noninvasive photothermal therapy (PTT) for cancer with photothermal agents (PTAs) has recently achieved success in both preclinical and clinical trials. However, traditional PTAs tend to nonspecifically accumulate in normal liver tissue, hampering their use in PTT of liver tumors. By taking advantage of extremely low liver accumulation from ultrasmall renal-clearable gold nanoparticles (AuNPs), we report a biosafe therapeutic PTT strategy to treat liver tumors precisely through the intratumoral self-assembly of renal-clearable AuNPs at the tumor site via host-guest interactions. After active tumor targeting from the host AuNPs functionalized with both cyclo (Arg-Gly-Asp-d-Phe-Cys) and cyclodextrin, the guest AuNPs functionalized with both pH-responsive doxorubicin and adamantane are designed to precisely trigger intratumoral self-assembly, enhancing both PTT and chemotherapy toward the liver tumor microenvironment. This smart design principle generates a precise therapeutic action toward liver tumors without causing any noticeable heating effect or damage to the surrounding normal liver tissue.

Active-target T1-weighted MR Imaging of Tiny Hepatic Tumor via RGD Modified Ultra-small Fe3O4 Nanoprobes

Zhengyang Jia,Lina Song,Fengchao Zang,Jiacheng Song,Wei Zhang,Changzhi Yan,Jun Xie,Zhanlong Ma,Ming Ma,Gaojun Teng,Ning Gu,Yu Zhang

Developing ultrasensitive contrast agents for the early detection of malignant tumors in liver is highly demanded. Constructing hepatic tumors specific targeting probes could provide more sensitive imaging information but still faces great challenges. Here we report a novel approach for the synthesis of ultra-small Fe 3 O 4 nanoparticles conjugated with c(RGDyK) and their applications as active-target T 1 -weighted magnetic resonance imaging (MRI) contrast agent (T 1 -Fe 3 O 4 ) for imaging tiny hepatic tumors in vivo . RGD-modified T 1 -Fe 3 O 4 nanoprobes exhibited high r 1 of 7.74 mM -1 s -1 and ultralow r 2 /r 1 of 2.8 at 3 T, reflecting their excellent T 1 contrast effect at clinically relevant magnetic field. High targeting specificity together with favorable biocompatibility and strong ability to resist against non-specific uptake were evaluated through in vitro studies. Owing to the outstanding properties of tumor angiogenesis targeting with little phagocytosis in liver parenchyma, hepatic tumor as small as 2.2 mm was successfully detected via the T 1 contrast enhancement of RGD-modified T 1 -Fe 3 O 4 . It is emphasized that this is the first report on active-target T 1 imaging of hepatic tumors, which could not only significantly improve diagnostic sensitivity, but also provide post therapeutic assessments for patients with liver cancer.

Targeted delivery of Bi2Se3 Nanoflowers to orthotopic liver tumor via transarterial infusion for enhanced microwave ablation sensibilization

Long Gao,Haili Yan,Shuang Zhu,Xiaochun Wang,Yan Tan,Jiangfeng Du,Duiping Feng,Hui Zhang,Zhanjun Gu

Microwave ablation (MWA) is acknowledged as one of the optimal therapeutic strategies for unresectable hepatocellular carcinoma (HCC). Unfortunately, inadequate MWA at high-risk sites is very common in clinic owing to the scruple of probable damage to adjacent vital organs, which eventually leads to recurrence of residual tumors. Herein, we synthesize novel Bi 2 Se 3 nanoflowers (NFs) which possess the peculiarity of microwave sensibilization and explore their sensitization capability both in vitro and in vivo . The as-prepared NFs exhibits remarkable heating capacity under microwave irradiation compared to deionized water , mainly due to the multilayered petal structures that could strongly increase the reflectivity of microwave and stimulate the motion of water molecules. More importantly, we recruit clinically available transarterial infusion (TAI) method for precisely targeted delivery of Bi 2 Se 3 NFs to tumor site and perform sequential MWA on orthotopic liver tumor. In addition, the high Z element containing Bi 2 Se 3 NFs could realize efficient computed tomography to monitor the therapeutic process. What’s more, no obvious cell and organic toxicity are observed during the experimental period, which further verifies the good biocompatibility of Bi 2 Se 3 NFs. In this work, we utilize minimally invasive TAI for targeted delivery of microwave sensitive nanoflowers to improve MWA efficacy at tumor site and reduce side effect in normal tissues, which provides a neoteric theoretical and experimental strategy for clinical interventional therapy of HCC.

Preparation of microspheres encapsulating sorafenib and catalase and their application in rabbit VX2 liver tumor

Xihan Li,Hui Yu,Ying Huang,Yu Chen,Jialun Wang,Lei Xu,Feng Zhang,Yuzheng Zhuge,Xiaoping Zou

Background Transcatheter arterial chemoembolization (TACE) is extensively used in the treatment of advanced hepatocellular carcinoma (HCC). However, the efficacy of TACE is usually limited to secondary tumor hypoxia and hypoxia-related tumor angiogenesis. Methods In this study, poly(lactic-co-glycolic acid) (PLGA) microspheres (SOR-CAT-PLGA MSs) encapsulating sorafenib (SOR) and catalase (CAT) were prepared by double-emulsion solvent diffusion method. Sorafenib inhibits tumor angiogenesis, and catalase decomposes hydrogen peroxide (H 2 O 2 ) to generate oxygen in the tumor. Results In vitro and in vivo , SOR -CAT-PLGA MSs could significantly improve the efficacy of hepatic artery embolization in the treatment of rabbit VX2 liver tumors, regulate tumor hypoxia and immunosuppressive microenvironment, then achieved near-complete and rapid necrosis of liver tumors. Conclusions The application of new SOR -CAT-PLGA MSs in hepatic artery chemoembolization of rabbit VX2 liver tumor is a promising approach to improve the therapeutic effect of liver tumors and has a broad clinical application prospect.

Multimodal imaging of nano-assembled microspheres loaded with doxorubicin and Cisplatin for liver tumor therapy

He Yiwei,Zhang Yuqing,Gong Yuanchuan,Zhang Zhewei,Xu Tiancheng,Tian Liqiang,Pan Ting,Yang Hong,Pan Hao,Kou Quanming,Wang Hao,Shao Guoliang

Currently, clinically available drug-loaded embolic microspheres have some shortcomings, such as being invisible with standard medical imaging modalities and only being able to carry positively charged drugs. The visualization of drug-loaded microspheres is very important for real-time monitoring of embolic position to improve the therapeutic effect. Meanwhile, the visualization of microspheres can enable postoperative reexamination, which is helpful for evaluating the embolization area and guiding the subsequent treatment. In addition, microspheres capable of loading different charged drugs can increase the choice of chemotherapeutic drugs and provide more possibilities for treatment. Therefore, it is of great importance to explore drug-loaded microspheres capable of multimodal imaging and loading drugs with different charges for transarterial chemoembolization (TACE) treatment of liver tumors. In our study, we designed a kind of nano-assembled microspheres (NAMs) that can realize computer X-ray tomography (CT)/magnetic resonance imaging (MRI)/Raman multimodal imaging, be loaded with positively and negatively charged drugs and test their imaging ability, drug loading and biological safety. The microspheres have strong attenuation performance for CT, high T2 relaxation for MRI and good sensitivity for surface enhanced Raman spectroscopy (SERS). At the same time, our microspheres can also load the positively charged drug, doxorubicin (DOX), and negatively charged drug Cisplatin. One gram of NAMs can hold 168 mg DOX or 126 mg Cisplatin, which has good drug loading and sustained-release capacity. Cell experiments also showed that the nano-assembled microspheres had good biocompatibility. Therefore, as multimodal developed drug loaded microspheres, nano assembled microspheres have great potential in TACE treatment of liver cancer.

Characterization of Magnetite/Iodized Oil Magnetic Fluid for Embolization and Hyperthermia Application

Sun Xiaoning,Song Kun,Wang Song,Sun Kangning

Fe 3 O 4 nanoparticles/iodized oil magnetic fluids (FIOMFs) have showed potential in targeted hyperthermia for liver tumors; in this paper, FIOMFs were simply and conveniently prepared by dispersing the synthesized hydrophobic Fe 3 O 4 nanoparticles (FNPs) in iodized oil; the stability, magnetothermal effects, and the biotoxicity of FIOMFs were evaluated. The results showed that the stability of FIOMFs was mainly influenced by the morphology and the size of FNPs. FNPs prepared with FeCl 3 ·6H 2 O, oleylamine (OAm), ethylene glycol (EG), and stearic acid exhibited the best suspension stability, which could suspend in iodized oil stably for a long time. Depending on the FNP content and the parameters of the alternating magnetic field (AMF), the temperature of FIOMF in AMF can reach up to more than 70 °C within 60 s, exhibited excellent magnetothermal effects. The in vitro cytotoxicity tests indicated the cytotoxicity of FNPs is mainly influenced by its concentration in cells.

Tumor vessel-adaptable adhesive and absorbable microspheres for sustainable transarterial chemoembolization therapy

Guo Jiakun,Huang Jintao,Huang Zuliang,Hu Di,Tan Hujing,Wang Yan,Deng Chao,Zhu Xiaoli,Zhong Zhiyuan

Transarterial chemoembolization (TACE) is a common clinical intervention used for unresectable liver tumors, but conventional embolic microspheres generally exhibit slack vascular stacking and suboptimal drug release. Here, we report tumor vessel-adaptable, adhesive, and absorbable microspheres (3Asphere) based on hyaluronic acid developed using microfluidic and radical polymerization techniques for sustained TACE therapy of liver tumors. 3Asphere presents uniform sizes, gradual degradation over two months, and fast encapsulation and sustained release of chemotherapeutics such as epirubicin, irinotecan, and cisplatin. Interestingly, 3Asphere with high elasticity enables robust vascular embolization, effectively blocking rabbit renal blood supply for over one month. In rabbit VX2 orthotopic liver tumors, 3Asphere actively binds to endothelial cells via CD44 and inhibits tumor growth, affording significant survival benefits compared to commercial Embosphere ® . TACE with epirubicin-loaded 3Asphere further enhances tumor inhibition and prevents lung metastasis. 3Asphere provides a versatile and advanced TACE therapy that might alleviate liver tumors.

Radioiodine-131-Labeled Theranostic Nanoparticles for Transarterial Radioembolization and Chemoembolization Combination Therapy of VX2 Liver Tumor

Guorong Jia,Tao Wang,Rou Li,Xiao Li,Gaofeng Sun,Wei Chen,Ye Peng,Chao Cheng,Jijin Yang,Changjing Zuo

In interventional treatment, materials are administered into the blood supply artery and directly delivered to tumors, offering proper scenarios for nanomedicine potential clinical applications. Transarterial chemoembolization (TACE) and transarterial radioembolization (TARE) are effective treatment methods for hepatocellular carcinoma (HCC), but postoperative residual tumor may result in intrahepatic recurrence and distant metastasis. The combination therapy of TACE and TARE based on multifunctional nanoparticles (NPs) is expected to overcome the drug resistance in hypoxic tumors and improve the therapeutic effect. Herein, BaGdF 5 NPs are synthesized and then coated with polydopamine (PDA), conjugated with the chemotherapeutic drug cis-diamminedichloride platinum (CDDP), radio-labeled with therapeutic radionuclide 131 I, yielding 131 I-BaGdF 5 @PDA-CDDP NPs. The in vitro anti-cancer effects of 131 I-BaGdF 5 @PDA-CDDP NPs are confirmed using CCK-8 and γ-H2AX assays in Huh7 cells. Mixed with Lipiodol, 131 I-BaGdF 5 @PDA-CDDP NPs are injected into the hepatic artery via a microcatheter to realize the TACE and TARE combination therapy in a rabbit VX2 liver tumor model. The results indicate that glucose metabolism is clearly decreased based on 18 F-FDG PET imaging and the apoptosis of tumor cells is increased. Furthermore, 131 I and BaGdF 5 NPs can be used for SPECT imaging and CT/MR imaging respectively, facilitating real-time monitoring of the in vivo biodistribution of 131 I-BaGdF 5 @PDA-CDDP NPs.

Folate-functionalized SMMC-7721 liver cancer cell membrane-cloaked paclitaxel nanocrystals for targeted chemotherapy of hepatoma

Wenwen Shen,Shuke Ge,Xiaoyao Liu,Qi Yu,Xue Jiang,Qian Wu,YuChen Tian,Yu Gao,Ying Liu,Chao Wu

In this study, we prepared a folic acid-functionalized SMMC-7721 liver cancer cell membrane (CM)-encapsulated paclitaxel nanocrystals system (FCPN) for hepatoma treatment. Transmission electron microscopy (TEM) characterization showed that FCPN was irregular spherical shapes with a particle size larger than 200 nm and a coated thickness of approximately 20 nm. In an in vitro release experiment, FCPN indicated a slowly release effect of paclitaxel (PTX). Cell experiments demonstrated that FCPN was taken up by SMMC-7721 cells and significantly inhibited the proliferation of SMMC-7721 cells, which illustrated that FCPN had good targeting ability compared with PN and CPN. According to the results of in vivo animal experiments, FCPN significantly inhibited tumor growth. Tissue distribution experiments proved that FCPN could accumulate significantly in tumor tissues, which further explained why FCPN had good targeting ability. These results clearly suggested that folate-functionalized homotypic CM bionic nanosystems might represent a very valuable method for liver cancer treatment in the future.

Colloidally Stabilized Magnetic Carbon Nanotubes Providing MRI Contrast in Mouse Liver Tumors

Yue Liu,Benjamin W. Muir,Lynne J. Waddington,Tracey M. Hinton,Bradford A. Moffat,Xiaojuan Hao,Jieshan Qiu,Timothy C. Hughes

The use of medical imaging contrast agents may lead to improved patient prognosis by potentially enabling an earlier detection of diseases and therefore an earlier initiation of treatments. In this study, we fabricated superparamagnetic iron oxide (SPIO) nanoparticles within the inner cavity of multiwalled carbon nanotubes (MWCNTs) for the first time; thereby ensuring high mechanical stability of the nanoparticles. A simple, but effective, self-assembled coating with RAFT diblock copolymers ensured the SPIO-MWCNTs have a high dispersion stability under physiological conditions. In vivo acute tolerance testing in mice showed a high tolerance dose up to 100 mg kg–1. Most importantly, after administration of the material a 55% increase in tumor to liver contrast ratio was observed with in vivo MRI measurements compared to the preinjection image enhancing the detection of the tumor.

Iron oxide nanoparticles for targeted imaging of liver tumors with ultralow hepatotoxicity

Juanjuan Li,Ruitao Cha,Yulong Zhang,Hongbo Guo,Keying Long,Pangye Gao,Xiaohui Wang,Fengshan Zhou,Xingyu Jiang

Even though iron oxide (Fe3O4) nanoparticles are promising materials for magnetic resonance imaging (MRI) contrast agents, their biocompatibility and targeting efficacy still need to be improved. Herein, we modified glycyrrhetinic acid (GA) groups on Fe3O4 nanoparticles (Fe3O4@cGlu-GA) for liver tumor-targeted imaging. To evaluate the biocompatibility of these nanoparticles, we studied their cytotoxicity, hemolysis, and hepatotoxicity. We measured the uptake of Fe3O4@cGlu-GA nanoparticles in normal and liver tumor cells, then we investigated the specificity of Fe3O4@cGlu-GA nanoparticles in mouse models bearing subcutaneous and orthotopic liver tumors. With good biocompatibility and targeting efficacy both in vitro and in vivo, the Fe3O4@cGlu-GA nanoparticles are promising MRI contrast agents with ultralow hepatotoxicity and show great improvement on existing Fe3O4-based nanoparticles.
