Carotenoid profiling of Mytilus coruscus with unialgal cultures: Insights into biosynthetic pathways in tissues

Shuang Li,Keyi Fang,Shubing Chen,Jilin Xu,Haimin Chen,Juanjuan Chen

The carotenoid profiling of Mytilus coruscus generally depends on the dietary microalgae, and the ingested carotenoids are modified and processed depending on the properties of different tissues. Although much research effort has focused on analyzing carotenoid profiling between microalgae and shellfish, the detailed metabolic pathways of dietary carotenoids in shellfish are poorly understood. In the present study, we carried out feeding experiments with different unialgal cultures ( Chaetoceros mulleri and Dicrateria inornata ) during the growth stages and examined the carotenoid profiles between the dietary microalgae and the corresponding tissues in order to study the resorption and metabolic transformations of dietary carotenoids in M. coruscus . The results showed that 1) carotenoids mainly occurred in the pre-spawning phase of M. coruscus; the contents of most carotenoids showed the same trends as in dietary microalgae. 2) An association analysis of the total content of carotenoids and relevant gene expression demonstrated that the main tissue for carotenoid synthesis in M. coruscus was the hepatopancreas . The synthetic reactions of the allenic to acetylenic carotenoids pathway and the 4-keto oxidative pathway in M. coruscus coexisted. 3) Quantitative analysis revealed that the carotenoids mytiloxanthin, alloxanthin, and pectenolone containing acetylene groups were selectively assimilated in gonad (69.3–74.1%), mantle (83.2–102%), and muscle (86.9–100%), forming a large percentage of the carotenoids present in those tissues. These results provide new insights into the biosynthetic and metabolic transformation pathways of carotenoids in M. coruscus .

Exploring the impact of nonylphenol exposure on Litopenaeus vannamei at the histological and molecular levels

Xianbin Su,Teng Li,Xiaowen Zhu,Peihua Zheng,Huakang Pan,Hui Guo

Nonylphenol (NP) is one of the common pollutants in the environment that have toxic effects on aquatic animals. Nevertheless, little is known about the possible toxicity mechanism of NP on the hepatopancreas of Litopenaeus vannamei . In the present study, the detrimental effects of NP on the hepatopancreas of the L. vannamei were explored at the histological and transcriptomic levels. The findings indicated that after NP exposed for 3, 12, and 48 h, the hepatopancreas histology was changed significantly. Transcriptomic analysis showed that a total of 4302, 3651, and 4830 differentially expressed genes (DEGs) were identified at 3, 12, and 48 h following NP exposure. All these DEGs were classified into 12 clusters according to the expression patterns at different time points. GO and KEGG enrichment analyses of DEGs were also performed, immunological, metabolic, and inflammatory related pathways, including arachidonic acid metabolism (ko00590), the PPAR signaling pathway (ko03320), and the regulation of TRP channels by inflammatory mediators (ko04750) were significantly enriched. Six DEGs were selected for validation by quantitative real-time PCR (qRT-PCR) and the results confirmed the reliability of transcriptome data. All results indicated that NP is toxic to L. vannamei by damaging the histopathological structure and disrupting the biological function. The findings would provide a theoretical framework for lowering or limiting the detrimental impacts of NP on aquaculture and help us to further study the molecular toxicity of NP in crustaceans.

Lipotoxic Effects of Gossypol Acetate Supplementation on Hepatopancreas Fat Accumulation and Fatty Acid Profile in Cyprinus carpio

Yuru Zhang,Junmei Zhang,Yanhua Ren,Mengdan Zhang,Junping Liang

Cottonseed meal is an excellent protein source with low price and abundant nutrients, but the existence of free gossypol restricts its wide application in animal feeds. To explore the toxic effects of free gossypol, the effects of gossypol acetate on the growth performance, hepatopancreatic fat accumulation, and muscle fatty acid composition of the common carp (Cyprinus carpio) were investigated in the present study. Carp were randomly divided into four groups, which were fed a diet supplemented with 0, 170, 340, and 680 mg/kg gossypol acetate, respectively. After 60 days of feeding, the gossypol residue in the tissue, growth performance, blood biochemical indices, hepatopancreas histology, fatty acid components, and expression of key genes involved in lipid metabolism pathways were measured. It was found that the concentrations of free gossypol were increased significantly in the serum, bile, and hepatopancreas of carp fed 340 mg/kg and 680 mg/kg gossypol acetate (). Dietary gossypol acetate had a limited effect on growth performance, and high concentrations of gossypol acetate induced excessive hepatopancreatic fat deposition and hepatocyte injury in C. carpio. In addition, gossypol acetate decreased total polyunsaturated fatty acids (PUFAs) and docosahexaenoic acid in muscle. In the hepatopancreas, decreased HSL, PPARα, and CD36 may result in fat accumulation. In the muscle, decreased FADS2 may be the key reason for the decrease in PUFAs. Taken together, these findings indicated that dietary gossypol acetate can cause lipid metabolism disorders, such as hepatopancreatic damage and significant changes in tissue fatty acid profile, which are regulated by the key lipid metabolic genes in common carp.

Effects of nanopolystyrene and/or phoxim exposure on digestive function of Eriocheir sinensis

Mengting Huang,Yuan Ma,Qianru Fan,Shunli Che,Jun Zhang,Shuquan Ding,Shuren Zhu,Xilei Li

Nanopolystyrene (NP) and phoxim (PHO) are pervasive environmental contaminants that pose a significant threat to the health of aquatic organisms, prompting widespread concern among researchers and the public alike. The hepatopancreas play important roles in the Chinese mitten crab ( Eriocheir sinensis ), such as digestion, absorption and detoxification. This study assessed the hepatopancreatic toxicity caused by the exposure of Eriocheir sinensis to environmentally relevant concentrations of NP and/or PHO. After a 21-day exposure period, NP (1.0 × 10 10  particles/L) and PHO (24 μg/L) exposure resulted in reduced number of blister-like, resorptive, and fibrillar cells and an elevation in lipid droplets within the hepatopancreas compared to the control group. Furthermore, trypsin and lipase activity decreased, amylase activity increased, and a significantly decrease in the expression of digestion-related genes, including CHT , CarL , and CarB , suggested impairment in both digestive and metabolic functions. The marked upregulation of key genes, including PPARγ , GYK , PEPCK , and SCD , as well as key metabolites such as 4-methylzymosterol-carboxylate, zymosterone, lathosterol, 7-dehydro-desmosterol, vitamin D2, 24-methylene-cycloartanol, 5-dehydroepisterol, and sitosterol in the lipid metabolic pathway, showed that the peroxisome proliferator-activated receptor (PPAR) and steroid biosynthesis signaling pathways were highly affected by exposure to NP and/or PHO. These findings indicated that exposure to NP and/or PHO might adversely affect the hepatopancreatic physiological homeostasis in E. sinensis by causing tissue damage and interfering with digestive and metabolic functions. Our results provide ecotoxicological insights into the effects of nanopolystyrene and/or phoxim exposure on the digestive function of Eriocheir sinensis .

Identification of Isthmin-1 in common carp (Cyprinus carpio. L) and the effects on glucose metabolism in vivo and in vitro

Leya Qu,Liping Yang,Yiran Wang,Shaoyang Zhi,Mengjuan Zhao,Jinrui Xiong,Xiao Yan,Chaobin Qin,Guoxing Nie

Isthmin-1 (Ism1) plays roles in glucose uptake in mammals as an adipokine. To investigate its role in the glucose metabolism of common carp ( Cyprinus carpio. L), the Ism1 sequence was cloned, and its expression and distribution in tissues were detected. In addition, we prepared and purified the recombinant Ism1 protein using the E. coli expression system and assessed changes in the expression of key genes related to glucose metabolism through both in vivo injection experiments and primary hepatocyte experiments in vitro. The results revealed that the open reading frame of Ism1 was 1377 bp long, encoding 458 amino acids. Similarity analysis indicated that Ism1 exhibited a close evolutionary relationship with goldfish ( Carassius auratus ), sharing 98.35% amino acid similarity. Ism1 was expressed in all tissues of common carp, with the highest level observed in the heart, followed by the gill, head kidney, and hepatopancreas. Distinct patterns of Ism1 expression were identified during the oral glucose tolerance test and long-term high-carbohydrate and high-fat diet feeding experiments. In vivo studies demonstrated that the serum glucose concentration was reduced on treatment with Ism1, accompanied by a significant upregulation of mRNA levels for gk , hk , and pfk genes in hepatopancreas; conversely pepck and g6pase mRNA levels were significantly downregulated in the hepatopancreas under these conditions as well. Furthermore, our primary hepatocyte experiment confirmed that Ism1 could inhibit pepck and g6pase mRNA expression, while promoting gk , hk , and pfk mRNA expression levels. In conclusion, Ism1, in common carp, could participate in the glucose metabolism, which provides essential information for future studies on the function of Ism1.

Effects of waterborne cadmium exposure on Spinibarbus sinensis hepatopancreas and kidney: Mitochondrial cadmium accumulation and respiratory metabolism

Hanxun Qiu,Wenming Liu,Yulian Yan,Jing Long,Xiaojun Xie

To examine the relationship between heavy metal accumulation in mitochondria and their respiration function in fish during in vivo exposure, juvenile Spinibarbus sinensis were exposed to different waterborne cadmium (Cd) concentrations for up to 28 days. We measured the state III respiration rate and cytochrome c oxidase (CCO) activity of mitochondria in hepatopancreas and kidney and the accumulated Cd concentrations in mitochondria and heat-stable protein (HSP) fractions. Dose- and time-dependent Cd accumulation occurred at different levels in both organs, but was lower in hepatopancreas. When hepatopancreas mitochondrial Cd concentrations in Cd-exposed groups were > 5.5 μg/g dwt, their state III respiration rates were significantly lower than the control. CCO activity of hepatopancreas mitochondria exhibited decreasing dose- and time-dependent trends. However, kidney mitochondria respiratory activities were not affected significantly by Cd exposure. Cd concentrations in kidney HSP fraction were 2–5 times higher than in hepatopancreas under all exposure conditions, and were mainly present as non-deleterious metallothionein (MT)-Cd complexes. These results suggest that Cd accumulation occurred in hepatopancreas and kidney mitochondria of S. sinensis following waterborne Cd exposure, which significantly inhibited the respiration function of hepatopancreas mitochondria but did not have a deleterious effect on kidney mitochondria. The inhibitory pattern of hepatopancreas mitochondrial Cd concentrations related to function exhibited threshold and saturation effects, suggesting the capacity of S. sinensis to manage Cd toxicity. The difference in the relative proportion of Cd occurring as MT-Cd complexes in organs likely causes the organ-specific effects of Cd on hepatopancreas and kidney mitochondrial function.

Cadmium-induced oxidative stress, histopathology, and transcriptomic changes in the hepatopancreas of Fenneropenaeus merguiensis

Zhimin Ju,Charles Brighton Ndandala,Yiguo Lei,Vicent Michael Shija,Junliang Luo,Panmei Wang,Chongqing Wen,Huafang Liang

Cadmium (Cd) is a threatening heavy metal pollutant to aquatic organisms. The hepatopancreas plays a crucial role in combating environmental stress and enhancing the immune defense of aquatic species. The exact mechanism behind the damage induced by Cadmium in the hepatopancreas of Fenneropenaeus merguiensis is unclear. Thus, this study aimed to elucidate the potential mechanisms of Cadmium's impact on the hepatopancreas of F. merguiensis through physiological, histological, and transcriptomic analyses. After 72 h of F. merguiensis exposure to the 0.5 mg/L concentration of cadmium results in the reduction of superoxide dismutase and catalase activities, coupled with an increase in hydrogen peroxide and malondialdehyde levels, which leads to damage in both the gills and hepatopancreas. This causes severe histological changes in the gills and hepatopancreas of F. merguiensis , characterized by irregularities in the gill filaments and ruptures in the basal membrane of the hepatopancreas. Comparative transcriptomic analysis of F. merguiensis revealed the upregulation of several genes associated with pathways related to metabolism (ko01100), antigen processing and presentation (ko04612), and cell apoptosis (ko04215), indicating adaptive response mechanisms to Cadmium exposure. Transcriptomic analysis also showed that Cadmium exposure induced the expression of genes related to detoxification and apoptosis, such as heat shock proteins, metallothionein, and Caspase-3 , suggesting adverse effects on the normal physiological functions of F. merguiensis . Our study provides comprehensive insights into the toxicological mechanisms and impacts of Cadmium on F. merguiensis.

New insights into the regulation mechanism of Pacific white shrimp (Litopenaeus vannamei) hepatopancreas under 4-nonylphenol exposure using transcriptome analysis

Pei-Hua Zheng,Yao-Peng Lu,Xiu-Xia Zhang,Ke-Er Luan,Ze-Long Zhang,Jia-Jun Li,Tong Xu,Jun-Tao Li,Jian-An Xian,Hui Guo,An-Li Wang

4-Nonylphenol (4-NP) is one of the common endocrine-disrupting chemicals (EDCs) in estuaries and coastal zones, which can exert detrimental effects on the physiological function of aquatic organisms . However, the molecular response triggered by 4-NP remains largely unknown in Pacific white shrimp ( Litopenaeus vannamei ). In this study, transcriptomic analysis was performed to investigate the underlying mechanisms of 4-NP toxicity in the hepatopancreas of L. vannamei . Nine RNA-Seq libraries were generated from L. vannamei at 0 h, 24 h, and 48 h following exposure to 4-NP. Compared with 0 h vs 24 h, 962 up- and 463 down-regulated differentially expressed genes (DEGs) were identified, indicating that many genes in L. vannamei were induced to resist adverse circumstances by 4-NP exposure. In contrast, 902 up- and 1027 down-regulated DEGs were revealed in the comparison of 0 h vs 48 h, demonstrating that prolonged exposure to the stress from 4-NP resulted in more inhibited genes. To validate the accuracy of the transcriptome data, eight DEGs were selected for quantitative real-time polymerase chain reaction (qRT-PCR), which were consistent with the RNA-Seq results. Through KEGG pathway enrichment analysis, three specific pathways related to hormonal effects and endocrine function of L. vannamei were enriched significantly, including tyrosine metabolism, insect hormone biosynthesis , and melanogenesis . After 4-NP stress, genes involved in tyrosine metabolism (Tyr) and melanogenesis pathway (AC, CBP , Wnt, Frizzled, Tcf, and Ras) were induced to promote melanin pigment to help shrimp resist adverse environments. In the insect hormone biosynthesis, ALDH , CYP15A1, CYP15A1/C1, and JHE genes were activated to synthesize juvenile hormone (JH), while Spook, Phm, Sad, and CYP18A1 were induced to generate molting hormone. There is an enhanced interaction between the molting hormone and JH, with JH playing a dominant role and maintaining its “classic status quo action”. Our study demonstrated that 4-NP exposure led to impairments of biological functions in L. vannamei hepatopancreas. The genes and pathways identified provide novel insights into the molecular mechanisms underlying 4-NP toxicity effects in prawns and enrich the information on the toxicity mechanism of crustaceans in response to EDCs exposure.

In vitro disinfection efficacy and clinical protective effects of common disinfectants against acute hepatopancreatic necrosis disease (AHPND)-causing Vibrio isolates in Pacific white shrimp Penaeus vannamei

Zou Peizhuo,Yang Qian,Wang Hailiang,Xie Guosi,Cao Zhi,Chen Xing,Gao Wen,Huang Jie

Acute hepatopancreatic necrosis disease (AHPND) is one of the most significant bacterial diseases in global shrimp culture, causing severe economic losses. In the present study, we carried out in vitro antimicrobial tests to investigate the disinfection efficacy of 14 common disinfectants toward different AHPND-causing Vibrio spp., including eight isolates of V. parahaemolyticus , four isolates of V. campbellii , and one isolate of V. owensii . Polyhexamethylene biguanidine hydrochloride (PHMB) was revealed to possess the strongest inhibitory activity. Through analyzing and evaluating the results of antimicrobial tests and acute toxicity test, we selected PHMB and hydrogen peroxide (H 2 O 2 ) for further clinical protection test. Clinical manifestations indicated that both PHMB (2 mg/L and 4 mg/L) and H 2 O 2 (12 mg/L) could effectively protect juvenile Penaeus vannamei from the infection of V. parahaemolyticus isolate Vp 362 at 10 6 CFU/ml, and the survival rate was over 80%. When the bacterial concentration was reduced to 10 5 CFU/ml, 10 4 CFU/ml, and 10 3 CFU/ml, the survival rate after treated by 1 mg/L PHMB was 64.44%, 93.33%, and 100%, respectively. According to the results, PHMB and H 2 O 2 showed a lower toxicity while a better protection activity, particularly against a lower concentration of the pathogens. Therefore, these two disinfectants are proved to be promising disinfectants that can be applied to prevent and control AHPND in shrimp culture. Moreover, the methods of this study also provided valuable information for the prevention of other important bacterial diseases and suggested a reliable means for screening potential drugs in aquaculture.

Toxicological effects of sulfamethoxazole (SMX) and tire micro-particles (TMPs), alone or in combination, on the hepatopancreas of Litopenaeus vannamei

Yu Han,Xingzhou Yu,Yanan Liu,Min Zhang,Xianke Huang,Yinan Zhang,Hangjun Zhang

As the metabolic center of crustacean invertebrates, hepatopancreas plays a crucial role in metabolic detoxification and nutrient storage. Although sulfamethoxazole (SMX) and tire micro-particles (TMPs) are emerging marine pollutants, their combined hepatotoxic effects on lipid metabolism in crustaceans remain poorly understood. In this study, Litopenaeus vannamei were exposed to SMX and TMPs for 14 days. Hepatopancreatic lesions, lipid metabolism parameters, SMX bioaccumulation, and transcriptomic analysis were evaluated. Results revealed that SMX and TMPs co-exposure induced: (1) significant histological damage to hepatopancreas, accompanied by elevated levels of hepatopancreatic lesion markers; (2) a marked increase in lipid accumulation and altered fatty acid composition in hepatopancreas; (3) enhanced activities of lipid synthesis enzymes, increased triglyceride and total cholesterol levels, and reduced lipase activity; (4) dysregulation of metabolic pathways, characterized by upregulation of fatty acid synthesis-related pathways and downregulation of fatty acid decomposition-related pathways, alongside disruption of oxidative stress and detoxification pathways. Comparative analysis illustrated that SMX and TMPs co-exposure exhibited more toxicity than individual treatments. These findings demonstrated that SMX and TMPs synergistically impair hepatopancreatic function via lipid metabolism disruption and bioaccumulation, posing a potential threat to crustacean health and significant ecological risks in marine environments.

Effects of dietary carbohydrate levels on growth, body composition, and gene expression of key enzymes involved in hepatopancreas metabolism in mud crab Scylla paramamosain

Qiuyu Zhan,Tao Han,Xinyu Li,Jiteng Wang,Yunxia Yang,Xiaojun Yu,Puqiang Zheng,Teng Liu,Hanying Xu,Chunlin Wang

This study estimated the effects of different dietary carbohydrate levels on growth performance, feed utilization, body composition, and the gene expression of key enzymes involved in hepatopancreas lipogenesis and glucose metabolism in mud crab Scylla paramamosain . Crabs with an initial mean weight of 1.84 ± 0.06 g were used in an 8-week experiment to test five diets (C6, C12, C18, C24, C30) formulated to contain increasing levels of carbohydrate, measured as 8.55%, 13.36%, 17.76%, 25.64% and 32.50%, respectively. The results showed that the levels 25.64% and 32.50% improved growth performance, with the best gains at 25.64%. The highest dietary carbohydrate level led to higher hemolymph glucose concentrations and more fat deposition in the body. Furthermore, real-time quantitative PCR analysis revealed that the mRNA expression of key enzymes in glycolytic and lipogenic metabolism (i.e. glucokinase , fructose-2,6-diphosphatase, and fatty acid synthetase) were significantly affected by the dietary carbohydrate level. The mRNA expression of glucokinase was lowest in the group fed the carbohydrate-rich C30 diet, and the mRNA of fructose-2 and 6-diphosphatase were remarkably expressed in the C24 group ( P  < .05). The mRNA expression of fatty acid synthase at the dietary carbohydrate levels 32.50% and 25.64% were respectively 10-fold and 5-fold greater than with 8.55% dietary carbohydrate. The overall data suggest that S. paramamosain can be fed 25.64%–32.50% carbohydrate to achieve satisfactory growth performance. A two-slope broken-line regression analysis of weight gain showed that 24.08% dietary carbohydrate would be optimal for this species reared under similar conditions.

Comparative assessment of silver nanoparticle and silver nitrate toxicities in Mytilus galloprovincialis

Zimin Cai,Zihan Xing,Mingzhe Xu,Yuting Zhao,Lin Ye,Wei Sun,Ruijia Tao,Liuya Mi,Bowen Yang,Lei Wang,Yancui Zhao,Xiaoli Liu,Liping You

Silver nanoparticles (AgNPs) and Ag + ions are both detected in aquatic environments, posing potential risks to marine ecosystems. Mytilus galloprovincialis is an effective model for monitoring marine environments. In this study, AgNPs were synthesized using a chemical approach, and to distinguish the toxicological effects of AgNPs and AgNO 3 in M. galloprovincialis , experiments were conducted using various treatments (control, AgNO 3 , AgNPs, and AgNPs + cysteine). Our findings revealed that the uptake rate of AgNPs and AgNO 3 was different, they predominantly accumulated in the hepatopancreas and gills. qRT-PCR analysis showed varying degrees of alterations in immune genes of HSPA12A, TCTP, sHSP22, sHSP24.1 , P63, Bcl-2, and Ras . Histopathological analysis demonstrated disrupted epithelial cell arrangements and connective tissue damage in the hepatopancreas, with the AgNPs exhibiting the most severe damage compared to AgNO 3 . In addition, AgNPs significantly induced oxidative stress in hemocytes compared to AgNO 3 , resulting in elevated apoptosis rates. These findings contribute to a deeper understanding of the AgNPs and AgNO 3 interactions in marine environments and provide a theoretical basis for the evaluation of marine pollution and biomonitoring strategies.
