Dietary sodium butyrate supplementation improves fish growth, intestinal microbiota composition, and liver health in largemouth bass (Micropterus salmoides) fed high-fat diets

Weijun Chen,Shiyang Gao,Kuo Chang,Xiaoyu Zhao,Ben Niu

The present study evaluated the effect of dietary sodium butyrate (NaBT) supplementation on the growth performance, intestinal microbiota composition, and liver health of largemouth bass Micropterus salmoides fed high-fat diets. Four isonitrogenous and isolipid diets were formulated: a high-fat diet and the high-fat diet supplemented by 0.05% NaBT, 0.1% NaBT, or 0.2% NaBT. Each diet was randomly distributed to triplicate tanks (200 L ) containing 30 fish (initial weight 4.0 ± 0.1 g) per tank. The experimental fish were fed to apparent satiation twice daily for 60 days. The orthogonal polynomial contrasts showed that the weight gain rate (WGR) and specific growth rate (SGR) increased linearly ( P  < 0.05) with increasing dietary NaBT levels (P  < 0.05). No significant differences were found for feed intake (FI), feed conversion ratio (FCR), condition factor (CF), viscerosomatic index (VSI), and hepatosomatic index (HSI). There were no differences in the alpha diversity indices (including Operational Taxonomic Units, Chao index, and Simpson index) of intestinal microbiota between the various dietary groups ( P  > 0.05). For the intestinal microbiota composition, the proportion of Proteobacteria were linearly and cubically increased, whereas the proportion of Tenericutes and Mesomycoplasma were linearly and cubically decreased with the increase of dietary NaBT levels ( P  < 0.05). The increasing dietary NaBT levels led to significantly quadratic decrease of alanine aminotransferase (ALT) ( P  < 0.05), linear and quadratic decrease of aspartate aminotransferase (AST) ( P  < 0.05), and linear, quadratic, and cubic decrease of diamine oxidase (DAO) ( P  < 0.05). Hepatic triglyceride (TG) and total cholesterol (TCHO) contents decreased linearly and quadratically with the increase of dietary NaBT levels ( P  < 0.05). Increasing dietary NaBT levels linearly, quadratically, and cubically decreased the malondialdehyde (MDA) contents ( P  < 0.05). Total superoxide dismutase (T-SOD) and glutathione peroxidase (GPx) activity levels increased linearly with the increasing dietary NaBT levels ( P  < 0.05). As the dietary NaBT levels gradually increased, the relative expression of carnitine palmitoyl transferase (CPT1) and peroxisome proliferator-activated receptor α (PPARα) increased linearly, quadratically, and cubically ( P  < 0.05), while the relative expression of sterol regulatory element binding protein 1 (SREBP 1) decreased linearly, quadratically, and cubically ( P  < 0.05). The relative expression of tumor necrosis factor α (TNF-α) and cysteinyl aspartate specific protease 3 (Caspase 3) decreased linearly and quadratically with the increasing NaBT levels ( P  < 0.05). Pearson correlation analysis showed that fish growth was closely correlated with microbiota composition, hepatic lipid accumulation, hepatic antioxidant-related parameters, and hepatic gene expression ( P  < 0.05). These results indicated that NaBT could be used to alleviate the negative influence of high-fat diets on fish growth, intestinal microbiota homeostasis , and liver health.

Dietary Sodium Butyrate Supplementation Attenuates the Detrimental Effects of High-Fat Diets on Growth Performance, Liver Health, and Disease Resistance in Grass Carp

Shiyang Gao,Xiaoyu Zhao,Ben Niu,Kuo Chang,Weijun Chen

An 8-week experiment was conducted to investigate the effects of sodium butyrate (NaBT) inclusion in high-fat (HF) diets on growth performance, liver health, and disease resistance in Grass Carp Ctenopharyngodon idella . Three diets (Control diet containing crude lipid at 58 g/kg, HF diet with 108-g/kg crude lipid, and NaBT diet with 108-g/kg crude lipid and 1-g/kg NaBT) were randomly assigned to nine tanks with 30 fish (9.50 ± 0.06 g) in each tank. After the feeding trial, disease resistance was assessed by injecting the fish with Aeromonas hydrophila . Compared to the Control diet group, the HF diet group showed lower specific growth rate, feed efficiency, and survival rate (15.7%) after the A. hydrophila challenge; significantly higher activity levels of alanine aminotransferase and aspartate aminotransferase in plasma; higher malondialdehyde content; higher messenger RNA (mRNA) expression of interleukin-8, cysteinyl aspartate specific protease (caspase) 9, and caspase 3; lower activity level of glutathione peroxidase; and lower mRNA expression of nuclear factor erythroid 2-related factor 2 in liver. However, the NaBT diet significantly increased fish growth performance and survival rate (39.7%) after the A. hydrophila challenge and reduced hepatic oxidative stress, inflammation, and apoptosis compared to the HF diet. In conclusion, NaBT can ameliorate the detrimental effects of HF diets on fish growth performance and fish health.

Astaxanthin, bile acid and chlorogenic acid attenuated the negative effects of high-fat diet on the growth, lipid deposition, and liver health of Oncorhynchus mykiss

Wei Zhao,Rong Yao,Han-Lin Wei,Yu-Cai Guo,An-Qi Chen,Bao-Yang Chen,Jin-Niu

This study aimed to examine the effects of high-fat diets supplemented with astaxanthin , chlorogenic acid, and bile acid on the growth, lipid accumulation, oxidation resistance, inflammatory response, and liver morphology in Oncorhynchus mykiss cultured in the convection-water cages. Fish were fed five isonitrogenous diets (D1: 20% lipid; D2: 30% lipid; D3: 30% lipid +0.1% BASF astaxanthin; D4: 30% + 0.06% chlorogenic acid; and D5: 30% lipid +0.1% bile acid) for 6 weeks. The results found that fish fed the D2 diet achieved a lower growth, while the optimal growth was identified in D4 diet. The higher crude lipid contents in the whole body were identified in fish fed the D3-D5 diets. Fish fed the high-fat diet showed liver vacuolization and lower superoxide dismutase (SOD) activity and transcriptional levels of NF-E2-related nuclear factor 2 ( Nrf2 ), catalase ( CAT ) , and SOD , as well as higher malondialdehyde content. Supplementing above three additives can reverse above results and enhance the total antioxidant capacity. Besides, the three additives alleviated the inflammatory responses caused by the high-fat diet via inhibiting proinflammatory cytokines. Moreover, high-fat diet reduced the endogenous lipid production and promoted the lipoprotein hydrolysis by increasing the lipoprotein lipase and inhibiting the fatty acid synthesis . The above results showed that three additives could mitigate the negative effects caused by the high-fat diet. Among them, astaxanthin showed the best antioxidant and anti-inflammatory properties and chlorogenic acid could elevate the growth greatest.

Sodium alginate ameliorates health in freshwater fish through gut-liver axis modulation under high carbohydrate diets

Wenlu Zhu,Huilin Zhang,Haitao Pan,Hao Zeng,Wenjun Wang,Yunfeng Liu,Zirui Wang,Qiubai Zhou,Chuanqi Yu

High carbohydrate (HC) diets have become a conventional approach in modern aquaculture. However, this feeding strategy frequently compromises the health of high-value economic fish species, which might ultimately impair the farmers’ final profits. Our prior research indicated that Saccharina japonica efficiently reduces harm from HC diets, likely due to its sodium alginate (SA) content. In this study, Monopterus albus were randomly grouped into treatments: a normal diet (20 % carbohydrate, NC), a high carbohydrate diet (32 % carbohydrate, HC), and high carbohydrate diets supplemented with 0.5 % SA (LA). Then our outcomes evinced that LA improves various health-related parameters, including reduction of hepatosomatic index (HSI), serum glucose, triglycerides, and aminotransferase levels, as well as the decrease in hepatic lipid droplets, glycogen, and collagen fiber content. Our transcriptomic analysis on liver tissue showed that detoxification-related differentially expressed genes (DEGs) were less active in HC group than in NC group. In contrast, DEGs linked to fatty acid synthesis increased in LA group versus HC group. A combined analysis of metagenomics and the short-chain fatty acids (SCFAs) profile in the posterior intestinal digesta indicated that LA diets greatly increased concentration of acetic acid. This effect was accompanied by a concurrent reduction in the abundance of the species Marinobacter guineae relative to HC diets. Overall, sodium alginate has demonstrated a substantial enhancement in Monopterus albus’ health by mitigating liver injury and ameliorating intestinal microbiota dysbiosis.

Integrative analysis of microbiome and metabolome reveals the effect of deoxynivalenol on growth performance, liver and intestinal health of largemouth bass (Micropterus salmoides)

Yi lian Yin,You xing Xu,Meng ting Wang,Dao zhi Yang,Xiao yan Wang,Xian ke Zhou,Meng meng Huang,Shun Yang,Hui Fei

This study explored the toxic effects of deoxynivalenol (DON) intake at low (100 µg·Kg −1 , LD group) and high doses (300 µg·Kg −1 , HD group) on largemouth bass ( Micropterus salmoides ). After a 56-day feeding trial, the HD group exhibited significantly reduced growth performance and weakened antioxidant capacity, along with elevated activities of enzymes related to metabolic dysregulation. Histopathological analysis showed an increase in hepatic cell vacuoles, as well as a shortened intestinal villi in both LD and HD groups. Notably, high-dose DON intake markedly down-regulated the expression of intestinal epithelial tight junction-related genes and proteins. Microbiome analysis indicated a significant increase in the Delftia and Acinetobacteria relative abundance, accompanied by diminished in Mycoplasma abundance in the HD group. Metabolomic profiling demonstrated that the dysregulated metabolites were mainly associated with the lysine biosynthesis pathway. Integrative multi-omics analysis revealed that pyridoxamine and diferuloylputrescine served as key biomarkers associated with Acinetobacteria . Collectively, we confirmed that high-dose DON intake induces hepatointestinal damage, thereby impairing the growth performance of largemouth bass. Our findings also further highlighted the link between DON-induced gut microbiota alterations and metabolic disorders, offering new intervention targets for alleviating DON toxicity.

The multi-protective role of dietary betaine in largemouth bass (Micropterus salmoides): coordinating antioxidant, inflammatory, and metabolic homeostasis under high-fat diet stress

Yi-Jia Liu,Jian Guo,Chang-Da Li,Si-Yin Han,Lu Cai

Introduction Intensive aquaculture frequently utilizes high-fat diets (HF) as a cost-effective strategy, yet this practice often induces hepatic steatosis, oxidative stress, and chronic inflammation in carnivorous fish. Betaine, a natural methyl donor, has shown potential as a functional feed additive, but its comprehensive protective mechanisms under HF stress remain to be fully elucidated. Methods Juvenile largemouth bass (Micropterus salmoides) were fed one of four isonitrogenous diets for 8 weeks: a normal-fat control (Control), a high-fat diet (HF), and two high-fat diets supplemented with 0.5% (HFB0.5) or 1.0% (HFB1) betaine. Growth performance, digestive enzyme activities, serum biochemical parameters, hepatic antioxidant capacity, and the expression of genes related to antioxidant defense, lipid metabolism, and inflammation were analyzed. Results The HF group exhibited significantly impaired growth, digestive function, and antioxidant capacity, along with elevated lipid peroxidation, dyslipidemia, and pro-inflammatory cytokine expression. Betaine supplementation restored growth performance and feed efficiency to control levels, ameliorated digestive enzyme activities (particularly enhancing lipase), and activated the hepatic Nrf2-Keap1 pathway, upregulating antioxidant genes (nrf2, sod1, cat, gpx, ho-1, gr) and enhancing enzyme activities. Betaine also improved serum lipid profiles, upregulated genes related to fatty acid oxidation (pparα, cpt-1) and lipolysis (lpl, hsl), suppressed lipogenic genes (srebp-1, fas), and rebalanced inflammatory cytokines by reducing tnf-α and il-1β while increasing tgf-β1 and il-10. Discussion Dietary betaine effectively counteracts HF-induced metabolic stress in M. salmoides through coordinated multi-pathway regulation. It enhances antioxidant defense, reprograms hepatic lipid metabolism toward catabolism, and restores inflammatory homeostasis. These findings underscore betaine’s role as a multi-functional feed additive capable of mitigating HF-related metabolic disorders and promoting overall health in carnivorous fish aquaculture.

Effects of dietary vitamin E supplementation on growth performance and liver health of Jinhu grouper (Epinephelus fuscoguttatus ♀ × Epinephelus tukula ♂) fed oxidized fish oil diets

Guoyong Huang,Weibin Huang,Guangcai Fu,Yansheng Liu,Hengyang Song,Wenshan Cai,Yubin Liu,Yuxuan Han,Yilin Yao,Beiping Tan,Xiaohui Dong

This study assessed the efficacy of dietary vitamin E in ameliorating growth and liver function in Jinhu grouper compromised by the inclusion of oxidized fish oil (OFO; peroxide value: 121.68 mmol/kg) in the diet, compared with a fresh fish oil control (FFO; peroxide value: 1.25 mmol/kg). The eight-week feeding trial utilized five experimental diets that were isonitrogenous and isolipidic: FFO, OFO (0 mg/kg VE), or OFO supplemented with 200 (VE1), 400 (VE2), or 600 (VE3) mg/kg vitamin E. Fish receiving oxidized fish oil showed slower growth, indicated by reduced weight gain rate (WGR) and increased feed conversion ratio (FCR). Intake of oxidized lipids altered lipid metabolism, lowering serum total cholesterol (TC), triglycerides (TG), and high-density lipoprotein cholesterol (HDL-C) but elevating low-density lipoprotein cholesterol (LDL-C) and malondialdehyde (MDA) levels. Hepatic antioxidant capacity was compromised, as revealed by the concurrent suppression in both the activities and gene expression levels of key antioxidant enzymes (SOD, CAT, and GPx). Concurrently, oxidized fish oil induced liver inflammation, significantly upregulating pro-inflammatory cytokine genes (notably il-1β , il-6 , il-8 , tnf-α ), and caused histopathological damage including hepatocyte nuclear displacement and lipid droplet accumulation. Vitamin E supplementation ameliorated these effects. The VE2 group (400 mg/kg) maximally restored growth performanc, reduced serum MDA and LDL-C levels, enhanced hepatic antioxidant enzyme activities and gene expression, suppressed inflammatory cytokine expression, and attenuated histopathological lesions. In conclusion, dietary vitamin E supplementation at 400 mg/kg effectively counteracts growth impairment and hepatotoxicity induced by oxidized fish oil in Jinhu grouper.

Dietary high lipid and high plant-protein affected growth performance, liver health, bile acid metabolism and gut microbiota in groupers

Jia Xu,Fan Wang,Chaoqun Hu,Junxiang Lai,Shiwei Xie,Kefu Yu,Fajun Jiang

High lipid diets (HLD) and high plant-protein diets (HPD) exhibit potential fishmeal-saving effects but negatively impact liver health and growth performance in fish. We hypothesized that HLD and HPD impair liver health in pearl gentian groupers ( Epinephelus fuscoguttatus ♂ × Epinephelus lanceolatus ♀) through the modulation of intestinal microbiota and bile acid (BA) metabolism. Four diet groups were tested: control diet (CD, 46.21% crude protein, 9.48% crude lipid), HLD (46.37% crude protein, 16.70% crude lipid), HPD (46.50% crude protein, 9.38% crude lipid), and high lipid-high plant-protein diet (HLPD, 46.54% crude protein, 16.67% crude lipid). A total of 300 fish (average body weight = 15.22 ± 0.03 g) were randomly divided into 4 diet treatments (ensuring 3 tanks replicates of each diet treatment, each tank containing 25 fish). After an eight-week feeding period, the HLD and HPD significantly decreased the final body weight (FBW), weight gain rate (WGR), specific growth rate (SGR) and feed intake (FI) in comparison to CD group, with HLPD exacerbating these indicators ( P < 0.05). Compared to CD group, the content of total cholesterol (T-CHO) and triglyceride (TG) in liver and serum were significantly increased in HLD group ( P < 0.05). Compared to HPD group, the content of T-CHO in liver was significantly decreased, the content of TG in liver and serum were significantly increased in HLPD group ( P < 0.05). HLD, HPD, and HLPD impaired liver health by inducing histological damage, inflammation, and oxidative stress. Compared to CD group, the mRNA relative expression of bile salt export pump ( bsep ) and multidrug resistance protein 3 ( mdr3 ) were significantly increased in HLD group, whereas the mRNA relative expression of sterol-27-hydroxylase ( cyp27a1 ), microsomal epoxide hydrolase ( meh ), apical sodium-dependent bile acid transporter ( asbt ), multidrug resistance-associated protein 3 ( mrp3 ), farnesoid X receptor ( fxr ) and G protein-coupled bile acid receptor 5 ( tgr5 ) were significantly decreased ( P < 0.05). Compared to CD group, the mRNA relative expression of mdr3 , asbt , mrp3 , organic anion transporters 1 ( oatp1 ), meh , fxr and tgr5 were significantly decreased in HPD group ( P < 0.05). In summary, HLD affects intestinal microbiota, BA metabolism, and lipid metabolism, leading to lipid deposition and liver damage. HPD regulates gut microbiota, BA metabolism, inflammatory responses, and BA receptor expression, impairing grouper liver health. HLPD synergistically combines the adverse effects of HLD and HPD on grouper liver health.

Effects of Dietary Fish Oil Levels on Growth Performance, Lipid Metabolism, Hepatic Health, Nonspecific Immune Response, and Intestinal Microbial Community of Juvenile Amur Grayling (Thymallus grubii)

Shaoxia Lu,Chang’an Wang,Yang Liu,Bing Liu,Ying Zhang,Honghe Shi,Gefeng Xu,Shicheng Han,Hongbai Liu

This trial was conducted to assess the effects of different levels of dietary fish oil on growth performance, hepatic health, nonspecific immune responses, and intestinal microbial community of Amur grayling ( Thymallus grubii ). Five isonitrogenous diets containing 60 (6FO), 90 (9FO), 120 (12FO), 150 (15FO), and 180 g/kg (18FO) fish oil were fed to triplicate groups of 60 fish per tank for 8 weeks, respectively. The results revealed that specific growth rate (SGR) and weight gain (WG) of fish in the 15FO group were significantly greater than those in the 6FO group ( p < 0.05). Somatic indices and whole-body lipid levels were positively correlated with increases in dietary fish oil levels. Trypsin and lipase activities in 15FO and 18FO groups were significantly higher than those in the 6FO and 9FO groups ( p < 0.05). The activities of intestinal catalase (CAT) and liver superoxide dismutase (SOD), CAT, lysozyme (LZM), alkaline phosphatase (AKP), and acid phosphatase (ACP) improved significantly as the dietary lipid content increased to 185.3 g/kg and decreased thereafter ( p < 0.05). The lipid metabolism-related genes peroxisome proliferator-activated receptor gamma (PPAR γ ) and carnitine palmitoyltransferase 1A (CPT1A) were significantly downregulated and upregulated ( p < 0.05), respectively, in the 15FO group. Immune-related genes in the liver and intestine, such as interleukin (IL-8), were significantly upregulated in the 15FO group ( p < 0.05). The liver sections from 18FO group presented more numerous and larger lipid vacuoles. Both low- (6FO) and high-lipid (18FO) diets reduced the relative abundance of intestinal Lactococcus . The relative abundances of intestinal Staphylococcus and Bacillus (mainly Bacillus anthracis ) increased in the low-lipid diet group and that of Pedobacter increased in the high-lipid diet group. Second-order polynomial analysis of WG and the feed conversion ratio (FCR) for varying levels of dietary lipid revealed that a range of 194.76–198.90 g/kg dietary lipid was optimal for the growth and health of Amur grayling.

Beneficial effects of soluble dietary fiber from Dendrocalamus brandisii Munro shoots on high-fat diet-induced metabolic disorders in mice: impact on liver, adipose tissue, and intestinal health

Mingtian Tian,Jiahong Dong,Yuhong Guo,Yihe Zhao,Jianxin Cao

Bamboo shoot soluble dietary fiber (DBS-SDF) has numerous health-promoting benefits, but its protective effects and potential mechanism against obesity-related metabolic disorders remain unclear. The effects of DBS-SDF on metabolic disorders induced by a high-fat diet (HFD) in mice was studied. Supplementation with DBS-SDF alleviated the development of HFD-induced obesity and related symptoms by lowering serum lipid levels, preventing lipid degeneration, and modulating the expression levels of genes related to lipid synthesis and decomposition in the liver. At the same time, DBS-SDF reversed the enlargement of white adipose cells, prevented the whitening of brown adipose tissue, and activated fat browning to improve energy balance. In addition, DBS-SDF reshaped the structure of the gut microbiota, significantly reducing the ratio of Firmicutes/Bacteroidetes, promoting the proliferation of SCFA producing bacteria Ligilactobacillus , unclassified_ Muribaculaceae , and Lachnospiraceae _NK4A136_group, while inhibiting harmful bacterial populations such as Faecalibaculum and Desulfovibrio . These effects are related to the enhancement of the intestinal barrier, such as upregulation of Intectin expression, increased secretion of mucin MUC2, and increased expression of tight junction proteins. Spearman correlation analysis revealed significant associations between obesity related indicators, intestinal barrier, and gut microbiota, emphasizing the improvement effect of DBS-SDF on metabolic disorders through a healthy gut microenvironment. These findings indicate that DBS-SDF has enormous potential in the development of prebiotics or functional foods.

Redox-Responsive AIEgen Diselenide-Covalent Organic Framework Composites Targeting Hepatic Macrophages for Treatment of Drug-induced Liver Injury

Jialu Zhuang,Hao Zhang,Jin Wu,Danyou Hu,Tao Meng,Jing Xue,Hanyang Xu,Gang Wang,Hua Wang,Guiyang Zhang

The escalating misuse of antipyretic and analgesic drugs, alongside the rising incidents of acute drug-induced liver injury, underscores the need for a precisely targeted drug delivery system. Herein, two isoreticular covalent organic frameworks (Se-COF and Se-BCOF) are developed by Schiff-base condensation of emissive tetraphenylethylene and diselenide-bridged monomers. Leveraging the specific affinity of macrophages for mannose, the first precise targeting of these COFs to liver macrophages is achieved. The correlation is also explored between the therapeutic effects of COFs and the NLRP3/ASC/Caspase-1 signaling pathway. Utilizing this innovative delivery vehicle, the synergistic delivery of matrine and berberine are accomplished, compounds extracted from traditional Chinese medicine. This approach not only demonstrated the synergistic effects of the drugs but also mitigated their toxicity. Notably, berberine, through phosphorylation of JNK and up-regulation of nuclear Nrf-2 and its downstream gene Mn-SOD expression, simultaneously countered excessive ROS and suppressed the activation of the NLRP3/ASC/Caspase-1 signaling pathway in injured liver tissues. This multifaceted approach proved highly effective in safeguarding against acute drug-induced liver injury, ultimately restoring liver health to normalcy. These findings present a novel and promising strategy for the treatment of acute drug-induced liver injury.
