Dihydromyricetin as a Dietary Supplement Enhances Hypoglycemic Efficacy: Synergistic Modulation of the Gut-Liver with Sangzhi Alkaloids

Yuwen Guan,Qiuyue Lv,Xinyu Wang,Baoping Jiang,Peigen Xiao

Chronic hyperglycemia, a hallmark of Type 2 diabetes mellitus, presents significant challenges in long-term management due to drug resistance and adverse effects. Dihydromyricetin (DHM), a natural flavanol from vine tea ( Ampelopsis grossedentata ), widely recognized as a nutraceutical, shows potential in enhancing the efficacy of pharmacological therapies. This study investigates the role of DHM in improving the hypoglycemic effects of Sangzhi Alkaloids (SZ-A), a commercially used glucosidase inhibitor. Our results demonstrate that DHM supplementation allows for a reduction in SZ-A dosage while maintaining or surpassing its glucose-lowering efficacy, thereby mitigating gastrointestinal side effects. Mechanistic studies reveal that this combination modulates the gut-liver axis, reducing ER stress, and enhancing gut microbiota diversity, including increased beneficial bacteria and non-12OH bile acids. These findings highlight DHM’s potential as an effective adjunctive nutraceutical, offering a safer and more efficient therapeutic strategy for managing hyperglycemia and related metabolic disorders.

The interaction between polyethylene microplastics and ciprofloxacin on inducing hepatotoxicity in Carassius auratus via the gut-liver axis

Peng Zhang,Guanghua Lu,Leibo Zhang,Zhenhua Yan,Min Wang

Microplastics (MPs) can accumulate and enrich antibiotics in aquatic environments, with their interactions exacerbating threats to aquatic organisms and complicating hepatotoxicity. Therefore, this study aimed to elucidate the effects of single or combined exposure to polyethylene MPs and ciprofloxacin (CIP) on hepatic injury in crucian carp, and to reveal the potential gut-liver axis toxicity mechanisms. The results showed that MPs enhanced the bioaccumulation of CIP, which increased with increasing MPs concentration. Co-exposure induced extensive eosinophilic necrosis and localised hepatocyte clustering in hepatocytes, alongside severe vacuolization and lymphocytic infiltration in intestinal cells. Glutamate aminotransferase and alanine aminotransferase activities, and total antioxidant capacity levels in the liver were significantly elevated, whilst glutamyltransferase activities were suppressed by the co-exposure. High-concentration MPs in combination with CIP significantly enriched the pathways of amino acid metabolism, glycerophospholipid metabolism, and primary bile acid biosynthesis in the liver. Moreover, co-exposure markedly suppressed the abundance of Cetobacterium while promoting that of Gemmobacter and Bosea . The imbalances in Bacteroides and pathogens ( Aeromonas and Bose ) are closely associated with related products in primary bile acid biosynthesis and glycerophospholipid metabolism pathways. These findings underscore that exposure to MPs and CIP induces hepatitis and reveal the pivotal role of the indirect gut-liver axis in elucidating the potential mechanisms underlying hepatotoxicity induced by co-exposure, thereby furnishing critical evidence for the holistic assessment of health hazards posed by these pollutants.

Dual-Carriers of Tartary Buckwheat-Derived Exosome-Like Nanovesicles Synergistically Regulate Glucose Metabolism in the Intestine-Liver Axis

Dan Li,Gaoyang Yi,Guifang Cao,Adam C. Midgley,Yongli Yang,Dan Yang,Wenguang Liu,Yujuan He,Xiaolin Yao,Guoliang Li

The utilization of plant-derived exosome-like nanovesicles (ELNs) as nanocarriers for oral delivery of bioactives has garnered significant attention. However, their distinctive lipid membrane composition may result in elevated membrane permeability within the gastrointestinal environment, leading to the leakage of carried bioactives. Inspired by the concept of projectile design, Tartary buckwheat-derived ELNs (TB-ELNs) based dual-carriers are fabricated by loading chlorogenic acid (CGA) into the cores and bonding selenium nanoparticles (SeNPs) to the lipid membrane. The results indicate that SeNPs bond markedly augments the membrane rigidity, and therefore enhances the stability of TB-ELNs and the retention rate of the loaded CGA during gastrointestinal digestion. In vitro and in vivo studies indicates that the TB-ELNs based dual-carriers are internalized by epithelial cells and transcytosis via the endoplasmic reticulum, and show the synergistic regulatory effect on high-fat diet-induced hyperglycemia in the intestine-liver axis. These results may be attributed to the fact that SeNPs combination reduces the gastrointestinal degradation of the carried bioactives. Moreover, SeNPs with antioxidant property can protect ELNs and their carried bioactives from oxidative damage, thereby enhancing their biological activities. Collectively, this study offers a new strategy to develop highly efficient oral delivery systems for bioactives to alleviate hyperglycemia and diabetes.

Synbiotic Intervention with Inulin and Lactiplantibacillus plantarum LPm77 Attenuates Type 2 Diabetes via Enhanced TUDCA Metabolism and Gut–Liver Axis Modulation

Minghan Li,Jialin Guo,Jinxin Meng,Xueting Wang,Tianxu Pan,Dongyu Zhao,Hongye Li,Ruyi Gao,Ya Wang,Hechun Li,Songyang Li,Jiagan Wang,Haibin Huang,Nan Wang,Shuyuan Yu,Jiayao Guan,Mingxiao Liu,Chunfeng Wang,Guilian Yang

Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by inflammation and immune dysregulation, with the gut–liver axis playing a crucial role in immunometabolic balance. While dietary fibers show therapeutic potential, their mechanisms in T2DM remain unclear. This study investigates the effects of synbiotic intervention combining inulin and Lactiplantibacillus plantarum LPm77 on T2DM, focusing on the gut–liver axis. After 7 weeks of synbiotic treatment, significant improvements in hyperglycemia, insulin resistance, and hepatic steatosis were observed in T2DM mice. Inulin promoted L. plantarum LPm77 colonization, enhancing the gut microbiota balance and restoring the intestinal barrier function. It also regulated bile acid levels in the enterohepatic circulation. The synbiotic promoted tauro-ursodeoxycholic acid (TUDCA) biosynthesis, improving hepatic lipid metabolism and activating TGR5. TGR5 activation suppressed M1 macrophage polarization via the TGR5-TLR4-NF-κB pathway, reducing pro-inflammatory cytokines and increasing IL-10, thus alleviating systemic inflammation. This study highlights a novel therapeutic approach targeting immune-metabolic dysregulation in T2DM.

The combined effects of polypropylene microplastics and sulfonamide antibiotics on the gut-liver axis of Gymnocypris przewalskii

Ping Zhu,Minxin Qin,Xinyu Wang,Qiang Gao,Dan Liu,Miaomiao Nie,Junmei Jia,Delin Qi,Cunfang Zhang

Microplastics (MPs) and antibiotics (ATs) accumulating in Qinghai Lake threaten aquatic health, yet their impact on Gymnocypris przewalskii ( G.przewalskii ) remains unclear. This multi-omics study investigated polypropylene MPs (PP-MPs) and sulfonamide antibiotics (SGD) on G.przewalskii. Groups included control group (C), single exposure groups (1 g/L SGD, 1 mg/L PP-MPs, 5 mg/L PP-MPs), and co-exposure groups (1 mg/L PP-MPs + 1 g/L SGD, 5 mg/L PP-MPs + 1 g/L SGD) exposed for 28 days. Using 16S rRNA sequencing, UHPLC, and MS/MS, we analyzed changes in the intestinal microbiota, metabolome, and transcriptome of G. przewalskii . Compared to control gruop, intestinal microbiota alpha diversity changed significantly in all treatments. The SGD group decreased notably, while co-exposure groups (SPL) showed significantly lower alpha diversity than single exposures. SPL groups exhibited higher beta diversity than controls and single exposures. In SPL, genera like Legionella, Fusobacterium, Phascolarctobacterium , and Bacteroides decreased, whereas Shinella, Lactobacillus , and Prevotella increased. Differential metabolites and genes decreased with rising concentrations across exposure groups. Most metabolites enriched in the ABC transporter pathway. Differentially expressed genes primarily enriched in carbon metabolism and cytochrome P450 pathways. Correlation analysis identified pathways like Neisseriaceae and Acinetobacter affecting Bilirubin in the gut, which in turn influenced liver genes LOC107556838, LOC109072261 and notum1a . This demonstrated G. przewalskii adaptation to PP-MPs and SGD through the gut-live. The study provides a scientific basis for protecting G. przewalskii and assessing health risks of MPs and ATs to aquatic ecosystems on the Qinghai-Tibet Plateau.

Low molecular fucoidan alleviated alcohol-induced liver injury in BALB/c mice by regulating the gut microbiota-bile acid-liver axis

Yiyun Sun,Qiuyue Men,Xiaomeng Ren,Chunhong Yan,Shuang Song,Chunqing Ai

Fucoidan has attracted significant attention owing to its remarkable bioactivities, but the effect of molecular weight (Mw) on its activities in the context of alcoholic liver diseases (ALD) is poorly understood. In this study, low Mw fucoidan (OSLF) was prepared, and its protective effect against alcohol-induced liver injury was assessed in a mouse model. OSLF increased weight gain and colon length, improved lipid disorders, and reduced oxidative stress in mice exposed to alcohol, alleviating liver injury. OSLF alleviated inflammation in the liver by inhibiting alcohol-activated NF-κB and MAPK pathways. The underlying mechanism can be attributed to the improvement of alcohol-induced dysbiosis of the gut microbiota, including a decrease in Proteobacteria and Bacteroidetes and an increase in microbiota diversity, as well as the abundances of Parabacteroides , Bacteroides , and Faecalibaculum . Metabolomics results showed that OSLF improved alcohol-induced abnormalities of microbiota metabolites, primarily involving amino acid metabolism and short chain fatty acids production. In addition, OSLF ameliorated bile acid metabolism in the gut and regulated the expression of bile acid-associated genes in the liver, affecting bile acid synthesis, regulation, and transport. It suggested that OSLF had the potential for the management of ALD by regulating the gut microbiota-bile acid-liver axis.
