A rapid quantitative method by liquid chromatography–tandem mass spectrometry for the measurement of anthocyanins and their metabolic characteristics in Sprague–Dawley rats

Lian Yueru,Niu Liyun,Meng Zhiyun,Zhu Xiaoxia,Gu Ruolan,Wu Zhuona,Li Jian,Han Peng,Liu Taoyun,Gan Hui,Dou Guifang

As a major class of phenolic compounds, anthocyanins are the functional components of blueberry. Vaccinium corymbosum ‘Lanmei 1’ is a novel blueberry variety and detailed study of their anthocyanins remains scarce. Therefore, a simultaneously quantitative method for five anthocyanins using ultra-performance liquid chromatography – tandem mass spectrometry (UPLC – MS/MS) was established here. Good linear relationships were obtained ranging from 1 to 100 ng/mL ( R 2  > 0.99). In addition, the specificity, recovery, stability, and matrix effect met requirements of analytical methodology. The method successfully monitored the concentrations of five anthocyanins in plasma and studied their pharmacokinetics after oral administration of 270 mg of anthocyanin extract in Sprague – Dawley rats. Pharmacokinetic parameters for delphinidin-3- O -glucoside (D3G), malvidin-3- O -glucoside (M3G), malvidin-3- O -arabinoside (M3A), delphinidin-3- O -arabinoside (D3A), and petunidin-3- O -arabinoside (P3A) showed the similar short T max 0.25 – 0.28 h and medium speed T 1/2 3.24 – 3.52 h. It indicated anthocyanins were quickly absorbed and taken effectively, but eliminated in blood at an appropriate rate. Contents of D3G and M3G in anthocyanin extract are approximately equivalent, while the total amount and maximum concentration of M3G were twice those of D3G in plasma. This demonstrated that there were many differences between the substance contents of anthocyanin monomers in vitro and their absorption and distribution behaviors in vivo. Multiple peaks in both concentration – time profiles and bile excretions for five anthocyanins suggested the possibility of enterohepatic circulation in rats. The present study developed and validated a rapid and sensitive bioanalytical method based on UPLC – MS/MS for quantifying major anthocyanins from a novel blueberry extract in rat plasma to further understand their absorption, distribution, and metabolism.

The intestinal absorption properties of flavonoids in Hippophaë rhamnoides extracts by an in situ single-pass intestinal perfusion model

Lei Xin,Xiao-Hui Liu,Jun Yang,Hong-Yi Shen,Guang Ji,Xiu-Feng Shi,Yan Xie

The purpose of this study was to investigate the absorption properties of isorhamnetin (IS), quercetin (QU), and kaempferol (KA) in total flavones of Hippophaë rhamnoides L. (TFH) by an in situ single-pass intestinal perfusion model. The results indicated that IS, QU, and KA in TFH were absorbed site-dependently, and both enterohepatic circulation and intestinal flora could participate in their absorption processes. The absorption mechanisms of IS, QU, and KA in TFH were involved in both passive diffusion and active transport, and the mediation of efflux transporter multidrug resistance-associated proteins (MRPs) should not be neglected.

The Function of Multidrug Resistance-associated Protein 3 in the Transport of Bile Acids under Normal Physiological and Lithocholic Acid-induced Cholestasis Conditions

Wang Wenyu,Zeng Sijing,Hu Ming,Liu Zhongqiu,Gong Lingzhi

Background: The role of multidrug resistance-associated protein 3 (Mrp3) in the transport of bile acid (BA) in drug-induced cholestasis has not been well studied. Objective: In this study, wild type and Mrp3 knockout (Mrp3-/-) mice under normal physiological and lithocholic acid (LCA)-induced cholestatic conditions were employed to investigate the role of Mrp3 in BA transport. Methods: The levels of BA in serum, liver, gallbladder, intestine, kidney, feces and urine were quantified in both wild type and Mrp3-/- mice via ultra-high performance liquid chromatography triple quadrupole mass spectrometry (UHPLC-MS/MS). Quantitative real-time PCR (RT-PCR) analysis was used to measure the expression of genes related to the transport and synthesis of BA. Results: The results showed that the liver did not suffer more serious damage as a result of cholestasis when Mrp3 was depleted. The level of some individual bile acids changed apparently in the compartments of enterohepatic circulation (EHC) between the two control and model groups, respectively, but the level of serum total bile acid was only slightly reduced for Mrp3-/- groups. In addition, the level of BA-related efflux transporters and synthases increased significantly when Mrp3 was knocked out under normal physiological conditions, but a negligible alteration appeared under cholestatic conditions. Conclusion: Our results indicated that Mrp3 could be responsible for the transport of some specific bile acids, and part of the Mrp3 role could be compensated for by other transporters. Moreover, Mrp3 deficiency has a direct effect on the expression of BA-related synthases and efflux transporters under normal physiological conditions, but this effect could be less prominent under cholestatic conditions. This study could provide much valuable insight into the physiological function of Mrp3 in the transport of bile acids.

Maternal procymidone exposure has lasting effects on murine gut-liver axis and glucolipid metabolism in offspring

Xiaofang Wang,You Weng,Shinan Geng,Caiyun Wang,Cuiyuan Jin,Liyun Shi,Yuanxiang Jin

There is increasing evidence that maternal exposure to environmental pollutants can cause intestinal and metabolic diseases , and these disease risks still exist in offspring . Here, female C57BL/6 mice were orally treated with procymidone (PRO) (10 and 100 mg/kg body weight/day) by dietary supplementation during the gestation and lactation periods. Then, we discovered PRO changed the physiology, intestinal barrier and metabolism both in the generations of F 0 and different developmental stages of F 1 (7 weeks and 30 weeks old, respectively). Maternal PRO exposure affected the growth phenotypes and the glucolipid metabolism related indicators and genes of mice, especially the male mice of F 1 generations. The changes in bile acids (BAs) metabolism demonstrated that PRO disordered glucolipid metabolism through enterohepatic circulation. Furthermore, PRO reduced mucus secretion in the gut and altered the composition of gut microbiota , leading more bacteria to disseminate in the gut and inflammatory responses both in F 0 and F 1 regenerations. And PRO-induced gut microbiota dysbiosis was tightly related to BAs metabolites. Together, the results indicated that PRO destructed the functional integrity of intestinal barrier and the inflammatory reaction was triggered. And then, the disorder of glucolipid metabolism was induced through the BAs enterohepatic circulation. This study indicated that the cross-generation effects of PRO could not be ignored.

Identification and Spatial Visualization of Dysregulated Bile Acid Metabolism in High-Fat Diet-Fed Mice by Mass Spectral Imaging

Qi Zhang,Zhen-Hua Wu,Shan-Shan Zhao,Jing Yang,Lei Chen,Xiao-Yu Wang,Zhan-You Wang,Hui-Xin Liu

Changes in overall bile acid (BA) levels and specific BA metabolites are involved in metabolic diseases, gastrointestinal, and liver cancer. BAs have become established as important signaling molecules that enable fine-tuned inter-tissue communication within the enterohepatic circulation. The liver, BAs site of production, displayed physiological and functional zonal differences in the periportal zone versus the centrilobular zone. In addition, BA metabolism shows regional differences in the intestinal tract. However, there is no available method to detect the spatial distribution and molecular profiling of BAs within the enterohepatic circulation. Herein, we demonstrated the application in mass spectrometry imaging (MSI) with a high spatial resolution (3 μm) plus mass accuracy matrix-assisted laser desorption ionization (MALDI) to imaging BAs and N -1-naphthylphthalamic acid (NPA). Our results could clearly determine the zonation patterns and regional difference characteristics of BAs on mouse liver, ileum, and colon tissue sections, and the relative content of BAs based on NPA could also be ascertained. In conclusion, our method promoted the accessibility of spatial localization and quantitative study of BAs on gastrointestinal tissue sections and demonstrated that MALDI-MSI was a valuable tool to investigate and locate several BA molecules in different tissue types leading to a better understanding of the role of BAs behind the gastrointestinal diseases.

Succinate communicates pro-inflammatory signals to the host and regulates bile acid enterohepatic metabolism in a pig model

Xuan Li,Yuting Ren,Guowen Huang,Ruofan Zhang,Yanan Zhang,Weiyun Zhu,Kaifan Yu

Succinate is produced by both the host and microbiota with pleiotropic functions in the modulation of intestinal inflammation and metabolic homeostasis, but the mechanisms remain elusive. This study aimed to determine whether dietary succinate influences the intestinal inflammatory response and to analyze the possible mechanisms by which succinate regulates enterohepatic metabolism. Sixteen growing barrows were randomly assigned to two groups, fed with a basal diet that consisted of a typical commercial diet or fed with a basal diet supplemented with 1% sodium succinate. Our data showed that dietary succinate activated the expression of succinate receptor 1 (SUCNR1) and increased the concentrations of pro-inflammatory cytokines in the intestine. Dietary succinate inhibited the expression levels of the ileal Farnesol X receptor (FXR) and its target genes, promoted hepatic bile acid secretion, and altered the bile acid metabolic profile. Then, we demonstrated that the pro-inflammatory cytokines triggered by succinate disrupted the ability of bile acids to activate FXR and fibroblast growth factor 19. Furthermore, dietary succinate reduced the abundance of bile-salt hydrolase enriched bacteria in the ileum. Taken together, dietary succinate activated the pro-inflammatory response via SUCNR1 in the intestine, and the pro-inflammatory cytokines induced by succinate blocked the activation of FXR and its target genes and disturbed bile acid enterohepatic circulation.

A FITC-doped silica coated gold nanocomposite for both in vivo X-ray CT and fluorescence dual modal imaging

Jie Feng,Di Chang,Zhifei Wang,Bin Shen,Jinjin Yang,Yanyun Jiang,Shenghong Ju,Nongyue He

Dual-modal imaging contrast agents with unique X-ray computed tomography (CT) and optical imaging capabilities have attracted much attention in recent years. Herein, the new silica hybrid nanocomposites containing Au nanoparticles and FITC dyes (FITC–Au@SiO2) were investigated as this kind of contrast agent. TEM characterization showed that the as-synthesized FITC–Au@SiO2 nanoprobes were uniform in morphology with the average size of 90 nm. After the PEGylation, the obtained FITC–Au@SiO2@PEG exhibited good dispersion stability under different conditions. The MTT assay suggested that FITC–Au@SiO2@PEG didn't show appreciable toxicity toward KB cells even at high concentration of up to 1000 μg mL−1. In addition, it was found that FITC–Au@SiO2@PEG could cross the cell membrane and enter into the cytoplasm during the incubation with cells. In vivo dual modal optical/CT imaging of C57BL/6J mice uncovered that within 2 h post-injection FITC–Au@SiO2@PEG nanoprobes preferred to accumulate in the liver and were gradually cleared through the enterohepatic circulation system, which could bring novel opportunities to the next generation of dual-modality contrast agents.

Caffeoylquinic acids from Silphium perfoliatum L. show hepatoprotective effects on cholestatic mice by regulating enterohepatic circulation of bile acids

Guoying Zhang,Wenjing Jia,Liying Liu,Luya Wang,Jiyu Xu,Jihong Tao,Mingting Xu,Min Yue,Huiqin Luo,Ping Hai,Huilan Yue,Dejun Zhang,Xiaohui Zhao

Ethnopharmacological relevance The incidence of cholestatic liver disease (CLD), which is primarily marked by abnormal bile acids (BAs) metabolism and can result in significant hepatic injury, is rising. Nevertheless, there remains a lack of effective treatments and drugs in clinical practice. Silphium perfoliatum L. (SP) is rich in various structural types of caffeoylquinic acid (CQA) compounds, and it is a traditional herb of North American Indians with hepatobiliary therapy effects. However, its therapeutic effect and mechanism of action on CLD have never been studied. Aim of the study To determine if SP-8, an extract rich in CQAs from SP, protects against cholestatic liver injury induced by alpha-naphthylisothiocyanate (ANIT) and to clarify its mechanism based on the farnesoid x receptor (FXR) signaling pathway and enterohepatic circulation of BAs. Materials and methods The therapeutic efficacy of SP-8 was evaluated by assessing the serum biochemical indices, inflammatory factors, and liver histopathology. Targeted metabolomics of the BAs was studied in the feces, liver, serum, and bile using UPLC-MS/MS. Additionally, a Western blot analysis was used to examine the expression levels of the peroxisome proliferator-activated receptor γ (PPARγ), the FXR, and proteins related to the synthesis and transport of BAs. 16S rRNA gene sequencing was performed to evaluate the gut microbiota (GM). Finally, molecular docking simulations were conducted to assess the interaction between seven types of CQAs from SP-8 with FXR and PPARγ. Results SP-8 significantly enhanced the health status of cholestatic mice induced by ANIT as evidenced by a notable reduction in the liver function indices and pro-inflammatory factors, restoration of liver pathological damage, and acceleration of BAs excretion through the feces. In addition, the levels of harmful secondary BAs in the liver and blood were significantly reduced by SP-8. Furthermore, the results of the study on the mechanism of action confirmed that SP-8 not only regulated FXR and PPARγ but also significantly ameliorated the GM structure, thereby promoting the enterohepatic circulation of BAs and achieving the homeostasis of the BAs in the blood and liver. In addition, SP-8 successfully reduced the inflammatory response by strongly suppressing the nuclear translocation of NF-κBp65. According to the molecular docking results, the extract's primary active ingredients could be the seven CQAs in SP-8, as they exhibited a strong affinity for both FXR and PPARγ. Finally, the Mantel test analysis revealed a significant correlation among cholestatic-associated parameters, the GM, and BAs. Conclusion It was confirmed for the first time that the SP-8 extract of Silphium perfoliatum L. that is rich in seven CQAs had a strong therapeutic effect on ANIT-induced CLD. Its mechanism may involve the regulation of the FXR signaling pathway and the amelioration of the GM structure to promote the homeostasis of BAs enterohepatic circulation. This study provides a potential candidate medicinal herb and its components for the development of CLD therapeutic drugs.
