The nutritional and functional properties of 1-oleoyl-2-palmitoyl-3-linoleoylglycerol-rich oil: promoting early-life growth and intestinal health with alterations in the intestinal microbiota of Micropterus salmoides

Meng Kang,Konglong Feng,Weijie Dai,Jianyin Miao,Guo Liu,Huaiyi Fang,Yong Cao

1-Oleoyl-2-palmitoyl-3-linoleoylglycerol (OPL), a key structural lipid in the breast milk fat, plays a critical role in providing nutrients and energy for infants. OPL is more abundant in Chinese breast milk fat and might be better for Chinese infants’ growth. However, few studies have investigated the effect of OPL on the growth and intestinal health of the organism in early life. OPL-rich oil with 45.77% OPL was prepared by immobilized lipase-catalyzed synthesis and purification. The effects of OPL on the nutritional properties and the regulation of intestinal microbiota in early life were further investigated in vivo (Micropterus salmoides). Dietary OPL-rich oil significantly increased the juvenile fish weight gain rate, protein content, and muscular polyunsaturated fatty acids, which in turn markedly altered the muscle texture in springiness and cohesiveness. Dietary OPL-rich oil could also protect intestinal tissues by significantly increasing fish intestinal fold height, mucosal thickness, and intestinal wall thickness. Furthermore, dietary OPL-rich oil regulated intestinal microbiota. Particularly, OPL significantly increased the probiotics (Cetobacterium_sp014250685, Streptomyces_mutabilis, Saccharopolyspora_spinosa, and Nocardiopsis_kunsanensis) and reduced the potential pathogens (Staphylococcus_nepalensis, Salmonella_enterica, the Candidatus_berkiella). The structured OPL significantly promoted fish growth and improved nutritional composition due to its higher bioavailability relative to tripalmitate (PPP). Moreover, OPL significantly improved the growth, cholesterol metabolism, and intestinal health than the mixed oil (MO), which was attributed to the higher palmitic acid content in the sn-2 position. Overall, the structure of triacylglycerols and its distribution of fatty acids affected early growth and intestinal health, and OPL was more effective in the improvement of juvenile growth and intestinal health.

Clostridium autoethanogenum protein substitution and bile acids addition altered intestinal health and transcriptome profiles of hepatopancreas in Litopenaeus vannamei

Jia Xu,Chaozhong Zheng,Shuyan Chi,Shuang Zhang,Junming Cao,Beiping Tan,Shiwei Xie

Dietary Clostridium autoethanogenum protein (CAP) has the potential to replace fishmeal (FM), but excessive CAP substitutions will affect gut health. Bile acids (BAs) have multiple benefits, but little information is available on the effects of BAs on Pacific white shrimp ( Litopenaeus vannamei ) fed a CAP diet (CAP replacing FM). An 8-week feeding trial were used to investigate the impacts of CAP substitution and BAs addition on structural changes of intestine, apoptosis, inflammatory cytokine and NFκB pathway of intestine, and transcriptomic profiles of hepatopancreas in Pacific white shrimp. Compared to PC (control) treatment, the NC (CAP50, CAP replacing 50% FM) treatment significantly decreased the fold height (FH) and villus length (VL) of midgut, and impaired the mitochondrial cristae. Compared to NC group, the FH was increased in BA1 (CAP50 with 0.03% BAs) and BA2 (CAP50 with 0.06% BAs) groups, the muscle layer thickness (MT) was decreased in BA2 and BA3 (CAP50 with 0.09% BAs) groups, the VL was increased in BA1 group, and the mitochondrial cristae was normal and well-defined in BA1 and BA2 groups (all p  < 0.05). The NC increased the genes expression of apoptosis and NFκB signalling pathway, while the BA2 decreased the genes expression of apoptosis, inflammatory cytokine and NFκB signalling pathway. In transcriptomics analysis, a total of 171 differential expression genes (DEGs) were identified in NC group and 50 DEGs in BA2 group. There was 13 DEGs was observed both in NC and BA2 groups. The 5 KEGG pathways were statistically enriched in all treatments, including “Carbon metabolism”, “One carbon pool by folate”, “Glycine, serine and threonine metabolism”, “Amino sugar and nucleotide sugar metabolism”, and “PPAR signaling pathway”. In conclusion, CAP50 substitution impaired the intestinal health and 0.06% BAs addition improved it, both treatments altered the transcriptomics profiles of hepatopancreas in Pacific white shrimp.

Dietary supplementation of chenodeoxycholic acid improved the growth performance, immune response and intestinal health of juvenile Penaeus monodon fed a low fish-meal diet

Shiwei Xie,Dan Wei,Lixia Tian,Yongjian Liu

Bile acid metabolism in aquatic animals usually is suppressed when fish meal (FM) is replaced by plant proteins. An eight weeks feeding experiment was conducted to evaluate the effects of dietary chenodeoxycholic acid (CDCA) supplementation on growth, immune response and intestinal health of Penaeus monodon fed a low FM diet. Three diets were formulated: high FM (25 %) diet (HF), low FM (15 %) diet (LF), and low FM diet supplemented with 0.1 % CDCA (LFC). Shrimp fed Diet LF showed lower weight gain and feed efficiency than those fed Diet HF. Low dietary FM up-regulated the mRNA level of xbp1 , and down-regulated the mRNA levels of sod , iap and NF-κB signaling pathway-related genes ( traf6 , tube ). Dietary supplementation of CDCA up-regulated the expression of relish and anti-oxidative-related genes ( sod and cat ) in the hepatopancreas and midgut. Low dietary FM impaired the intestinal fold and induced the endoplasmic reticulum swollen of intestinal epithelial cell, dietary supplementation of CDCA alleviated this problem. The intestinal microbiota diversity was lower in shrimp fed Diet LF than those fed HF. Dietary supplementation of CDCA in a low FM diet increased the intestinal microbiota diversity by decreasing the relative abundance of dominant phylum Proteobacteria. Low dietary FM increased the relative abundance of a dominant pathogenic bacterium ( Vibrio ), which was decreased by the supplementation of CDCA. These results indicated that dietary supplementation of CDCA in a low FM diet improved the growth performance, immune response and intestinal health of shrimp.

Effects of 4-hydroxyphenylacetic acid, a phenolic acid compound from Yucca schidigera extract, on immune function and intestinal health in meat pigeons

Yuxin Shao,Yipu Li,Shuxian Xie,Bo Zhang,Jing Li,Yalei Wang,Yusheng Gao,Jianyuan Ma,Tenghe Ma,Zheng Wang

This study aimed to evaluate the effects of 4-hydroxyphenylacetic acid ( 4-HPAA ), a phenolic acid compound from Yucca schidigera extract, in Caco-2 cells and artificial rearing of pigeons. In vitro experiments showed that 4-HPAA at different levels exhibited higher cell viability than shikimic acid and 3,4-dihydroxybenzoic acid, and with the highest viability observed at 600 μM. Additionally, 600 μM 4-HPAA alleviated LPS-induced upregulation of IL-1β and CASPASE1 expressions ( P < 0.05) . In vivo results demonstrated that supplemental 4-HPAA at 500, 1000, and 2000 mg/kg levels did not affect the body weight of 25-day-old pigeons ( P > 0.05), but all levels increased immunoglobulin G ( IgG ) and IgM levels ( P < 0.05). Compared with control, 2000 mg/kg 4-HPAA significantly reduced serum L-lactic acid, D-lactic acid and lactate dehydrogenase ( LDH ) activity ( P < 0.05); increased spleen and bursa of Fabricius indices and intestinal total antioxidant capacity activity of 25-day-old pigeons ( P < 0.05); and elevated ileal barrier-related genes ( OCCLUDIN, CLAUDIN1 and MUC2) in 7-day-old pigeons, as well as jejunal MUC2 and ileal OCCLUDIN in 25-day-old meat pigeons ( P < 0.05). It also inhibited glycolytic gene hexokinase 2 expression, while upregulating the signaling pathway genes CAMK2G and MKNK1 ( P < 0.05). Supplemental 500 and 1000 mg/kg 4-HPAA significantly increased serum total cholesterol levels ( P < 0.05). Notably, 500 mg/kg 4-HPAA had an adverse effect on intestinal integrity, as indicated by the highest serum diamine oxidase content, and increased glycolytic gene LDH-B expression; However, it enhanced microbial richness and diversity in the intestines of 7-day-old pigeon ( P < 0.05). Besides, 1000 mg/kg 4-HPAA impaired intestinal morphology, as evidenced by a lower ileal villus height-to-crypt depth ratio ( P < 0.05), but resulted in the highest abundance of Lactococcus in 25-day-old pigeons. In conclusion, 600 μM 4-HPAA exhibits anti-inflammatory activity in Caco-2 cells, and 2000 mg/kg 4-HPAA promotes immune function and intestinal barrier integrity while inhibiting glycolytic gene expression and the production of related serum metabolites in meat pigeons.

A novel fermented tea FOFT for obesity resistance and gut health restoration via modulation of the gut-fat-liver axis in HFD-induced obese mice

Yan Ma,Zhengnan Jiang,Hai Xie,Jian Wang,Minni Li,Huifang Ge,Wenbin Li,Zhongwen Xie,Daxiang Li,Xueling Gao,Hongyan Wang

Fermented oxidation fresh tea (FOFT) is a unique dark tea produced by a single strain of Eurotium cristatum , and its impact on obesity has not been extensively studied. Here, we investigated the anti-obesity and metabolic benefits of FOFT using a HFD-induced obesity mouse model. The HFD-fed mice or ND-fed mice were orally administered with FOFT for 12 weeks. The results showed that FOFT invention significantly suppressed body weight gain, ameliorated liver and adipocyte morphology, and improved glucolipid metabolism and intestinal homeostasis imbalance in HFD-fed mice. Notably, FOFT administration caused no adverse effects in ND mice, indicating its safety at tested doses. Moreover, FOFT restored intestinal microbiome dysbiosis and elevated the short-chain fatty acid levels. This gut microbiome shift was associated with reduced systemic inflammation and attenuated hepatic lipid accumulation. These findings suggest that FOFT, as a novel functional food, holds great promise for resistance to obesity and its comorbidities.

Effects of hydroxyproline supplementation in low fish meal diet on growth, immunity and intestinal health of Litopenaeus vannamei

Menglin Shi,Liangjuan Sun,Liutong Chen,Kangyuan Qu,Beiping Tan,Shiwei Xie

This experiment investigated the impacts of hydroxyproline (Hyp) supplementation in low fish meal diets on growth, antioxidant capacity, immunity and intestinal health of Litopenaeus vannamei . Six diets were formulated for the experiment, with the high fish meal diet containing 25 % fish meal (HF) and the remaining diets incorporating a mixture of soybean protein concentrate (SPC) and Clostridium autoethanogenum protein (CAP) replacing 15 % fish meal. These diets were supplemented with varying levels of Hyp (0 %, 0.2 %, 0.4 %, 0.6 %, and 0.8 %, designated as LF, H1, H2, H3, and H4, respectively). A total of 960 healthy shrimp (0.32±0.00 g) were divided into 6 groups at random; each group had 4 duplicates, each containing 40 shrimp. The shrimp were fed four times a day for a period of 51 days. The results indicated that dietary Hyp supplementation had no significant effects on growth performance and whole body composition of L. vannamei ( P > 0.05). The antioxidant enzyme activities of hemolymph and hepatopancreas were significantly decreased after fish meal was replaced, and were significantly increased after dietary Hyp supplementation ( P < 0.05). The expression of antioxidant-related genes in intestinal and hepatopancreas was significantly increased after dietary Hyp supplementation ( P < 0.05). In the intestine, the gene expression of penaeidins 3 ( pen3 ), dorsal , anti-lipopolysaccharide factor ( alf ) and crustins ( cru ) in the H2 group is the highest, and the gene expression of B-cell lymphoma-2 ( bcl2 ) in the H3 group is the highest ( P < 0.05). The genes expression of tumor necrosis factor-alpha ( tnf-a ), Bcl-2-assoxicated x protein ( bax ) and cytochrome c ( cyc-t ) in LF group reached the maximum ( P < 0.05). Intestinal H&E stained sections showed that low-fish meal diet reduced the mucosal folds height of shrimp, and H2 and H3 groups significantly increased the mucosal fold height of shrimp ( P < 0.05). Therefore, adding Hyp to the diet can improve the antioxidant capacity, immunity and intestinal health of L. vannamei .

Molecular properties and gut health benefits of enzyme-hydrolyzed konjac glucomannans

Jun-Yi Yin,Lu-Yao Ma,Ming-Yong Xie,Shao-Ping Nie,Jian-Yong Wu

Konjac glucomannan (KGM) with a molecular weight (MW) of 823.4 kDa was partially degraded by endo-1,4-β-mannanase. Two hydrolyzed KGM fractions (KGM-M-1: 147.2 kDa and KGM-M-2: 21.5 kDa) were characterized and applied to the animal tests in comparison with the native KGM. After oral feeding to the mice, KGM-M-1 and KGM-M-2 significantly increased the levels of short chain fatty acids (SCFAs) in the colonic contents and the native KGM increased the SCFAs in the cecum. The more significant effect of the native KGM in the cecum may be attributable to its high viscosity, slowing down the movement of intestinal microflora through the cecum, while the lower MW KGM-M-1 and KGM-M-2 could move more easily through the colon to be fermented by colonic bacteria. This new finding may be useful for future research and development of low-MW KGM polysaccharides through enzyme hydrolysis for the desired gut health benefits.

Dietary leucine supplementation improves intestinal health of mice through intestinal SIgA secretion

Song B.,Zheng C.,Zha C.,Hu S.,Yang X.,Wang L.,Xiao H.

AimsLeucine supplementation promotes intestinal health, but the mechanism is largely unknown. This study aimed to elucidate the mechanisms underlying the beneficial effects of leucine on intestinal homeostasis. Methods and ResultsFemale ICR mice (6‐week‐old) were randomly assigned into three groups: (i) mice received a basal diet; (ii) mice received a dietary 0·5% crystalline l‐leucine supplementation; and (iii) mice received a dietary 1·0% crystalline l‐leucine supplementation. Our results showed that leucine supplementation stimulated the secretion of SIgA in mice ileum and expression of cytokines related to SIgA production. Moreover, leucine supplementation improved the expression of mTOR and p70S6K1 expression. Further study showed that leucine supplementation markedly decreased microbiota richness and induced a shift in the Firmicutes : Bacteroidetes ratio in favour of Firmicutes.ConclusionsTherefore, our data suggested that leucine supplementation could enhance intestinal health through the regulation of mTOR pathway and promoting SIgA secretion in the mouse intestine, which might be associated with intestinal microbiota.Significance and Impact of the StudyThe present study found that dietary leucine supplementation of mice could improve intestinal health by enhancing intestinal SIgA secretion via a nonexclusive mechanism, which might include T cell‐dependent pathway, T cell‐independent pathway and gut microbiota.

Effects of pectic oligosaccharide on growth performance, organ indexes and intestinal health in broilers exposed to aflatoxin B1

Zhengqi Ye,Yongzhi Qin,Haijun Zhang,Meitian Xian,Hui Ye,Qingyun Cao,Zemin Dong,Changming Zhang,Jianjun Zuo,Weiwei Wang

This study aimed at investigating the influences of pectic oligosaccharide ( POS) on growth performance, organ indexes and intestinal health in broilers exposed to aflatoxin B 1 ( AFB 1 ). In vitro experiment was conducted to assess reactive oxygen species ( ROS )-scavenging ability of POS. In vivo experiment was then implemented by allocating 320 one-day-old yellow-feathered broilers to 4 groups (8 replicates/group), according to a 2 × 2 factorial arrangement with POS addition (0 or 400 mg/kg) and AFB 1 exposure (with or without) as the two factors. All parameters were determined on d 21. The results revealed an in vitro ability of POS to scavenge ROS. AFB 1 exposure tended to decrease ( P < 0.10) average daily gain ( ADG ) and average daily feed intake ( ADFI ), as well as caused abnormities in liver, thymus and bursal indexes. It also elevated ( P < 0.05) intestinal ROS and malondialdehyde contents but reduced ( P < 0.05) intestinal catalase and glutathione peroxidase activities, as well as caused intestinal injuries including the decreasing trends ( P < 0.10) of ileal villus height:crypt depth ratio and the relative mRNA expression of claudin-1 together with the increasing trend ( P < 0.10) of serum diamine oxidase ( DAO ) activity. POS addition tended to elevate ( P < 0.10) ADG and ADFI, as well as mitigated AFB 1 -induced elevation ( P < 0.10) in spleen index and reduction ( P < 0.05) of bursal index. Moreover, POS addition tended to enhance ( P < 0.10) ileal total antioxidant capacity in AFB 1 -exposed broilers. Besides, POS addition lowered ( P < 0.05) ROS level and increased ( P < 0.05) glutathione content in ileum, as well as showed a tendency ( P < 0.10) to elevate ileal claudin-1 expression and decrease serum DAO activity of broilers regardless of AFB 1 exposure. Collectively, supplemental POS benefited growth performance and organ indexes of broilers exposed to AFB 1 , which could attribute to its ability to ameliorate intestinal redox state and integrity. These findings underscore a potential of POS to attenuate the detriments of AFB 1 contamination in diets.

Dietary fish oil levels modulated lipid metabolism, immune response, intestinal health and salinity stress resistance of juvenile Penaeus monodon fed a low fish-meal diet

Shiwei Xie,Dan Wei,Yongjian Liu,Lixia Tian,Jin Niu

Dietary fish meal (FM) and fish oil (FO) replacement are crucial for the sustainable development of aquaculture. An eight weeks feeding trial was conducted to evaluate the potential replacing FM with soy protein concentrate , and the suitable FO level for a low FM diet of Penaeus monodon . A diet containing 25% FM was used as the control (C diet), and three other diets were formulated to contain 15% FM, and supplemented with 3%, 2%, 1% FO [high FO (HF), medium FO (MF) and low FO (LF), respectively]. The experiment was conducted in quadruplicates (30 shrimp per replicate, average weight 1.00 ± 0.01 g). The results indicated that besides the final body weight, there were no differences in weight gain, feed efficiency and survival rate observed among the four groups. The results of the acute salinity stress showed that shrimp fed the C and MF diets showed a similar survival rate, which was significantly higher than those fed a LF diet. Shrimp that were fed HF, MF and LF diets showed a lower level of fatty acid transport-related mRNA expression in the hepatopancreas , compared with those fed the C diet. Shrimp fed HF, MF and LF diets showed lower expression levels of superoxide dismutase , endoplasmic reticulum protein 57 and immune deficiency (IMD) in the hepatopancreas than those fed the C diet. Shrimp that were fed a LF diet showed the lowest expression levels of inhibitor of apoptosis proteins, Toll and tumor necrosis factor receptor associated factor 6 (TRAF6) in the hepatopancreas, as well as the highest expression levels of Toll, extracellular signal-regulated kinase, Relish and TRAF6 in the intestine among four groups. Intestinal microstructure damage was observed in shrimp that were fed a LF diet, and intestinal endoplasmic reticulum (ER) swelling was observed in shrimp fed MF and LF diets. In conclusion, these results indicated that the growth performance of shrimp was not affected when the FM and FO contents decreased, whereas a low content of FM negatively influenced the immune response in shrimp by modulating IMD and ER stress related gene expression. Under low FM conditions, a suitable dietary FO level would benefit to the lipid transport, hepatopancreas and intestine health, as well as the immune response of shrimp.

Effects of Dietary Aflatoxin B1 on Hybrid Grouper (Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂) Growth, Intestinal Health, and Muscle Quality

Hao Liu,Ruitao Xie,Weibin Huang,Yuanzhi Yang,Menglong Zhou,Baiquan Lu,Biao Li,Beiping Tan,Xiaohui Dong

This study investigated the effects of varying doses of dietary aflatoxin B1 (AFB1) on the growth, intestinal health, and muscle quality of hybrid grouper. Four diets with varying AFB1 concentrations (0, 30, 445, and 2,230 μg kg−1) were used. Elevating AFB1 concentrations led to a decline in growth indexes, specifically the weight gain rate and the specific growth rate, although the survival rate remained unchanged. Morphological indicators showed a dose-dependent decline with AFB1 exposure. Intestinal MDA content and hindgut reactive oxygen species (ROS) levels increased, while antioxidant indexes and digestive enzymes decreased with higher AFB1 levels. AFB1 negatively influenced hindgut tight junction protein and antioxidant-related gene expression while promoting inflammation-related gene expression. The presence of AFB1 in the experiment led to a decrease in beneficial intestinal bacteria, such as Prevotella, and an increase in harmful intestinal bacteria, such as Prevotellaceae_NK3B31_group. Muscle lipid and unsaturated fatty acid content significantly decreased, while muscle protein and liver AFB1 content increased dramatically with higher AFB1 concentrations. AFB1 caused myofibrillar cleavage and myofilament damage, leading to increased spaces between muscle fibers. In conclusion, diets with AFB1 levels exceeding 30 μg kg−1 inhibited hybrid grouper growth, while levels surpassing 445 μg kg−1 resulted in hindgut ROS accumulation, inflammation, elevated intestinal permeability, reduced digestive enzyme activity, and compromised muscle quality.

Characteristic Analysis of Peptide Fraction Extracted from Dendrobium aphyllum After In Vitro Gastrointestinal Digestion and Fermentation by Human Fecal Microbiota

Liu Huifan,Ma Juanjuan,Yin Zhina,Wu Hui

To characterize the digestive properties of an antioxidative peptide fraction (DA-P peptide) extracted from Dendrobium aphyllum and its effects following fermentation, DA-P peptide was used as a substrate for artificially stimulated in vitro gastrointestinal digestion and fermentation by human fecal microbiota. After in vitro digestion, the reducing power and DPPH- and ABTS- free radical-scavenging capabilities of DA-P peptide were significantly improved. During fermentation, changes in pH, peptide contents, and enzyme activities were evaluated and the results were well-correlated with each other; DA-P peptide lowered the pH value of the fermentation environment and stimulated the proliferation and metabolism of intestinal flora, indicating that DA-P peptide enhanced colon health. Thus, DA-P peptide showed antioxidative activities after gastrointestinal digestion and beneficial effects on intestinal flora when fermented by gut microbiota. Our results provide a foundation for studies of D. aphyllum peptide, which has not been widely examined.
