Size-tunable and biodegradable thrombin-functionalized carboxymethyl chitin microspheres for endovascular embolization

Fan Leng,Sai Lei,Bin Luo,Siyao Lv,Long Huang,Xulin Jiang

As a minimally invasive method, endovascular embolization has been an effective strategy for controlling bleeding and tumor treatment. Herein, carboxymethyl chitin embolic microspheres were prepared with the aqueous two-phase carboxymethyl chitin/polyethylene glycol system without using any crosslinking agents and thrombin-functionalized embolic microsphere named as [email protected] was made after covalent introduction of thrombin. The size of the microspheres can be adjusted from 5 to 500 μm. The data of in vitro and in vivo tests indicated that these microspheres possessed good degradability and biocompatibility. Meanwhile, [email protected] can significantly promote blood clotting and enhance the strength of the blood clots . More importantly, [email protected] displayed better embolization effect than that of the commercial available Gelfoam Alicon® and polyvinyl alcohol-based embolic microspheres CalliSpheres® in rat femoral vein and rabbit ear artery embolization models. Therefore the size-tunable and biodegradable thrombin-functionalized carboxymethyl chitin microspheres [email protected] possess great potential for effective hemostasis and endovascular embolization.

NIR Light-Fuse Drug-Free Photothermal Armor-Piercing Microcapsule for Femoral Vein Thrombosis Therapy

Ning Yang,Zhicheng Qian,Renqiang Yuan,Weikun Li,Xin Tan,Zonghao Liu,Qianli Zhang,Liqin Ge,Ling Liu

Acute thrombosis and its complications are leading global causes of disability and death. Existing thrombolytic drugs, such as alteplase and urokinase (UK), carry a significant bleeding risk during clinical treatments. Thus, the development of a novel thrombolysis strategy is of utmost urgency. Based on the previous work, the hollow structure of microcapsules (MC) is fabricated. Subsequently, armor-piercing MC, known as Fucoidan/S-Nitrosoglutathione/Melanin@MC (FGM@MC) is obtained, using a layer-by-layer (LBL) self-assembly method. Utilizing near-infrared (NIR) light as a trigger, the FGM@MC demonstrated photothermal thrombolysis at the site of thrombus due to its stable and outstanding photothermal properties. Simultaneously, photothermal stimulation leads to the release of a significant amount of nitric oxide from the FGM@MC, resulting in cavitation effects for mechanical thrombolysis. In vivo experiments confirmed the stable release of nitric oxide under NIR light irradiation. Treatment of femoral vein thrombosis in rats revealed that the thrombolytic effectiveness of FGM@MC+NIR (53.71%) is comparable to that of UK (59.70%). Notably, FGM@MC does not interfere with the coagulation function of rats and exhibits a favorable safety profile. In conclusion, this study demonstrates that the drug-free armor-piercing microcapsule has significant potential in the treatment of thrombosis, offering a safe and effective alternative to traditional thrombolytic therapies.

Kaolin-loaded carboxymethyl chitosan/sodium alginate composite sponges for rapid hemostasis

Yannan Song,Shuo Li,Huifang Chen,Xinyi Han,Gregory J. Duns,Wubliker Dessie,Wufei Tang,Yimin Tan,Zuodong Qin,Xiaofang Luo

There are several factors that contribute to the mortality of people who suffer from unmanageable bleeding. Therefore, the development of rapid hemostatic materials is necessary. Herein, novel rapid hemostatic composite sponges were developed by incorporation of kaolin (K) into carboxymethyl chitosan (CMCS)/sodium alginate (SA) via a combination of methods that includes ionic crosslinking, polyelectrolyte action, and freeze-drying. The CMCS/SA-K composite sponges were cross-linked with calcium ions provided by a sustained-release system consisting of D-gluconolactone (GDL) and Ca-EDTA, and the hemostatic ability of the sponges was enhanced by loading the inorganic hemostatic agent-kaolin (K). It was demonstrated that the CMCS/SA-K composite sponges had a good porous structure and water absorption properties, excellent mechanical properties, outstanding biodegradability , and biocompatibility. Simultaneously, they exhibited rapid hemostatic properties, both in vitro and in vivo . Significantly, the hemostatic time of the CMCS/SA-K 60 sponge was improved by 82.76 %, 191.82 %, and 153.05 %, compared with those of commercially available gelatin sponges in the rat tail amputation, femoral vein, and liver injury hemorrhage models respectively, indicating that its hemostatic ability was superior to that of commercially available hemostatic materials. Therefore, CMCS/SA-K composite sponges show great promise for rapid hemostasis .
