Highly sensitive determination of tumor exosomes is significant for early diagnosis of cancers and precision therapy. Herein, a sandwich peptide-based electrochemiluminescence (ECL) biosensor was developed for determination of phosphatidylserine (PS)-positive exosomes, a promising biomarker for early diagnosis of ovarian malignancy. A PS-specific binding peptide with high affinity was immobilized on Au nanoflowers (AuNFs) modified biosensing interface for recognition and capture of exosomes. Meanwhile, g-C 3 N 4 nanosheet loaded with luminol capped AuNPs (Lum-AuNPs@g-C 3 N 4 ) nanocomposite was used as the ECL signal nanoprobe. The g-C 3 N 4 nanosheets with large surface area were not only utilized as the carrier to immobilize more peptides for recognition of exosomes but also used to catalyze co-reactant H 2 O 2 decomposition to achieve the ECL signal amplification of luminol-H 2 O 2 system. Under optimal conditions, the biosensor showed superior performances compared with most currently available methods, including wider linear range across 5 orders of magnitude and a lower detection limit (LOD) down to 39 particles μL −1 . Moreover, the biosensor could be applicable for determination of exosomes in complex biological samples. This study indicates the combination of peptide recognition with nanoprobe as a label for signal amplification in sandwich ECL biosensing is a great promising strategy for sensitive and cost-effective determination of exosomes.

Rationale Cervical cancer is one of the most common malignant tumors in women, and it is essential to explore potential biomarkers such as glycopeptides closely related to cancer in physiological samples of cervical cancer patients. Sample pretreatment is required before direct detection using mass spectrometry because there are certain limitations. Meanwhile, it is still highly desired to promote the functionalization and application of metal–organic framework (MOF)-derived materials. Methods Using a post-synthesis modification method, a novel type of boric acid-functionalized MOF probe (designated as UiO-66@PEI@Au@B(OH) 2 ) is prepared for recognition of glycopeptides. The results are obtained using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and nano-liquid chromarography–tandem mass spectrometry. Results The UiO-66@PEI@Au@B(OH) 2 probe exhibits a low detection limit (0.6 fmol μL −1 ), an excellent recovery rate, comparatively good reusability and selectivity (HRP digests:BSA digests = 1:500). When UiO-66@PEI@Au@B(OH) 2 is used to selectively capture glycopeptides from the serum of a healthy person and a cervical cancer patient, 101 glycopeptides corresponding to 54 glycoproteins and 108 glycopeptides corresponding to 57 glycoproteins are detected, respectively. Conclusions The successful preparation of UiO-66@PEI@Au@B(OH) 2 provides a path for the investigation of the functionalization of MOF-derived materials. The excellent performance of UiO-66@PEI@Au@B(OH) 2 not only demonstrates the huge potential of functionalized MOFs in the glycoproteome, but also opens up new phases of the application of MOF-based materials.

Protein kinases play important regulatory roles in intracellular signal transduction pathways. The aberrant activities of protein kinases are closely associated with the development of various diseases, which necessitates the development of practical and sensitive assays for monitoring protein kinase activities as well as for screening of potential kinase-targeted drugs. We demonstrate here a robust luminescence resonance energy transfer (LRET)-based protein kinase assay by using NaYF4:Yb,Er, one of the most efficient upconversion nanophosphors (UCNPs), as an autofluorescence-free LRET donor and a tetramethylrhodamine (TAMRA)-labeled substrate peptide as the acceptor. Fascinatingly, besides acting as the LRET donor, NaYF4:Yb,Er UCNPs also serve as the phosphopeptide-recognizing matrix because the intrinsic rare earth ions of UCNPs can specifically capture the fluorescent phosphopeptides catalyzed by protein kinases over the unphosphorylated ones. Therefore, a sensitive and generic protein kinase assay is developed in an extremely simple mix-and-read format without any requirement of surface modification, substrate immobilization, separation, or washing steps, showing great potential in protein kinases-related clinical diagnosis and drug discovery. To the best of our knowledge, this is the first report by use of rare earth-doped UCNPs as both the phospho-recognizing and signal reporting elements for protein kinase analysis.

A novel electrochemical biosensing method for protein kinase (PKA) activity was demonstrated by using a reduced graphene oxide-polydopamine-silver nanoparticle-Ti4+ (rGO-PDA-AgNPs-Ti4+) nanocomposite. The obtained nanocomposite possessed an integral capability for phosphopeptide recognition and signal readout. The polydopamine modified reduced graphene oxide (rGO-PDA) was firstly prepared based on a self-polymerization method of dopamine. The silver ions were adsorbed onto polydopamine (PDA) layer and directly reduced into silver nanoparticles (AgNPs), which was used for electrochemical signal reporting. Then, the Ti4+ cations were attached onto the PDA layer for phosphopeptide recognition according to the strong coordination ability of PDA with Ti4+ and phosphate group. The prepared rGO-PDA-AgNPs-Ti4+ nanocomposites were characterized with different methods. The developed rGO-PDA-AgNPs-Ti4+ nanocomposites were then employed for electrochemical analysis of PKA-catalyzed kemptide phosphorylation. The sensitive detection toward PKA activity was realized with an experimental detection limit of about 0.01 U/mL. It may be also extended for the inhibitor evaluation. Thus, it provided a facile and sensitive means for electrochemical analysis of PKA activity and inhibitor screening.

Reversible binding of cell targeting motifs on biomaterial surfaces can be used to mimic the dynamic bioactivities of the extracellular matrix (ECM), thus showing potential to modulate cell behaviours in physiopathologic processes. As the markers of tumour metastasis, circulating tumour cells (CTCs) are especially essential for the diagnosis of cancer. Dynamic display of cancer targeting motifs on biometric interfaces thus enables specific isolation of rare cancer cells and delivery of critical information for cancer therapy. In this work, we report a dynamic biointerface for the reversible binding of cancer-targeting motifs on quartz through molecular recognition between the natural glycopeptide vancomycin (Van) and the D-alanine-D-alanine (AA) dipeptide. This active surface could not only realize specific cancer cell adhesion but also induce nondestructive cell detachment by the competitive addition of the free AA dipeptide. The cell isolation efficiency in the medium could reach 70% within 120 min, showing high specificity and separation efficiency. Furthermore, the dynamic display of cancer-targeting motifs on magnetic beads enabled magnetic cancer cell capture and AA dipeptide-triggered cell release. Therefore, the natural dynamic strategy using noninvasive glycopeptide recognition holds promise for rare cell detection, isolation and cell-based disease diagnosis.
