Co-assembly of curcumin and a cystine bridged peptide to construct tumor-responsive nano-micelles for efficient chemotherapy

Yemei Dai,Zelei Jiang,Jingyi Li,Mengfan Wang,Chang Liu,Wei Qi,Rongxin Su,Zhimin He

The effective uptake and release of hydrophobic antitumor drugs in cancer cells is a practical challenge for tumor chemotherapy. Many methods were developed to conquer it through modifying drug molecules with hydrophilic groups, or fabricating nanodrugs based on hydrophilic materials. In recent years, peptides have attracted significant interest as part of a promising platform for fabricating nanodrugs due to their low cytotoxicity, favorable variability and self-assembly property. In this study, a cystine bridged peptide (CBP) was designed to co-assemble with a hydrophobic antitumor drug curcumin (CCM), to form a tumor-responsive nanodrug. The hydrophilicity of the peptide promotes the water-dispersity of nanodrugs, and the disulfide bond in cystine, which is cleavable by glutathione (GSH), was involved considering the overexpressed GSH in tumor microenvironments. In vitro and in vivo tests on cervical cancer cells revealed that the obtained nanodrug can rapidly dissociate at tumor sites and inhibit the tumor growth with limited side effects on healthy tissues.

Tumor-Responsive Small Molecule Self-Assembled Nanosystem for Simultaneous Fluorescence Imaging and Chemotherapy of Lung Cancer

Yuming Yang,Caixia Yue,Yu Han,Wei Zhang,Aina He,Chunlei Zhang,Ting Yin,Qian Zhang,Jingjing Zhang,Yao Yang,Jian Ni,Jielin Sun,Daxiang Cui

Cancer therapeutic drugs face various transportation barriers in transit to the tumor site, making the delivery of effective drug concentrations problematic. Moreover, these drugs are very difficult to use due to their adverse off-target effects. Thus, it is very essential to develop a drug delivery system that can deliver drugs to achieve effective local concentrations without side effects on healthy tissues. Herein, the authors report a self-assembled nanodrug system in which hydrophobic antitumor drugs are packaged into nanoparticles to improve water solubility, tumor targeting ability, blood retention time, and chemotherapeutic effect. The nanodrugs are degraded into smaller ones when exposed to the tumor microenvironment, extravasated from leaky regions of the tumor vasculature, and displayed matrix metalloproteinase-2 (MMP-2)-induced degradation and antitumor property. To construct this unique system, an amphiphilic multifunctional molecule (Pep-Cy5) is synthesized by attaching a MMP-2-cleavable peptide to a hydrophobic near-infrared dye, Cy5. Two hydrophobic anticancer drugs are conjugated to Pep-Cy5 through hydrophobic interactions to form the self-assembled nanodrug system. The MMP-2-induced degradation and hydrophobic antitumor drug interchangeability features of this nanosystem enable the hydrophobic antitumor drugs to exhibit longer blood-retention times, improved intratumoral accumulation, fewer side effects, and higher anticancer efficacies compared with free drugs.

Tumor-Responsive Upconversion Nanoparticles with Tunable Degradability and Ultrabright Emission for Optical Bioimaging

Wubin Lv,Jin Ye,Qiang Wang,Chunsheng Li,Jun Wang,Zhiyong Zhang,Na Niu,Zhiguo Liu,Jiating Xu,Yujie Fu

Rare earth (RE)-based inorganic upconversion nanoparticles (UCNPs) have huge potency for applications varying from bioimaging to theranostics of tumors. For these applications, it is of great importance that UCNPs be expelled innocuously from a living body during a rational period of time after executing their diagnostic and/or therapeutic tasks. Despite great efforts to modulate biodegradability, a reasonable biodegradation rate for currently available UCNPs in weakly alkaline physiochemical buffers has not yet been realized. Herein, to achieve a tunable biodegradation rate for UCNPs, a class of core–shell-structured nanoparticles is invented by heteroepitaxially growing a CaF2:Yb shell with varying Zr4+ doping content on a biodegradable Na3ZrF7:Yb,Er core. The CaF2:Yb,Zr shell can not only enhance the emission intensity but also ensure the overall biodegradation of the nanocrystals. The degradation rate of core–shell UCNPs can be easily modulated by changing the Zr4+ doping content in the shell, and the degradation rate is pH-responsive; thus, tumor acidity-triggered degradation can be realized. When sodium alginate (SA)-modified UCNPs are used for the bioimaging of tumors, the intracellularly degraded metal ions and the released SA molecules can self-assemble through a coordination cross-linking effect, thereby improving the tumor retention of the nanosystems. These discoveries will unequivocally promote the future clinical applications of RE-based UCNPs.

A GSH-Gated DNA Nanodevice for Tumor-Specific Signal Amplification of microRNA and MR Imaging–Guided Theranostics

Nan Yan,Lin Lin,Caina Xu,Huayu Tian,Xuesi Chen

Developing tumor-responsive diagnosis and therapy strategies for tumor theranostics is still a challenge owing to their high accuracy and specificity. Herein, an AND logic gated–DNA nanodevice, based on the fluorescence nucleic acid probe and polymer-modified MnO 2 nanosheets, for glutathione (GSH)-gated miRNA-21 signal amplification and GSH-activated magnetic resonance (MR) imaging–guided chemodynamic therapy (CDT) is reported. In the presence of overexpressed miRNA and GSH (tumor cells), the nanodevice can be in situ activated and release significantly amplified fluorescence signals and MR signals. Conversely, the fluorescence signal is quenched and MR signal remains at the background level with low miRNA and GSH (normal cells), efficiently reducing the false-positive signals by more than 50%. Under the guide of miRNA profiling and MR imaging, the tumor-responsive hydroxyl radical ( · OH) can effectively kill tumor cells. Furthermore, the nanodevice shows catalase-like activity and glucose oxidase–like activity with the performance of O 2 production and glucose consumption. This is the first time to fabricate a tumor-responsive theranostic DNA nanodevice with tumor-specific signal amplification of microRNA and GSH-activated MR imaging for CDT, potential hypoxia relief and starvation therapy, which provides a new insight for designing smart theranostic strategies.
