Fluorinated branched polymer enables Ce6 and IR780 more effective for phototherapy

Chenglong Mu,Xiaodan Sun,Kai Yan

Phototherapy is gradually becoming a primary technique for treating tumors due to increased efficacy and less side effects. It largely depends on the photosensitizers. Nevertheless, most of photosensitizers show poor stability, how to encapsulate or load them, especially multiple ones, for effective delivery is of paramount importance. To address the challenge, a hyperbranched polymer was prepared by a reversible addition-fragmentation chain transfer polymerization, which is composed of 2,2,3,3-Tetrafluoropropyl methacrylate (TFPMA), bis(2-methacryloyl)oxyethyl disulfide (DSDMA) and poly(oligo ethylene glycol) methyl ether methacrylate (PEGMA). Benefiting from hydrophobic performance of fluorinated segments, this hyperbranched polymers can load hydrophobic IR780 (9%) and Ce6 (12%) due to self-assembly and hydrophobic-hydrophobic interaction. The final formed composite micelles showed good water dispersity, well biocompatibility, and strong stability. Remarkably, they had good photodynamic and photothemal abilities, enabling them to synergistically induce tumor cell apoptosis by oxidative damage and thermal ablation. This work offers a promising composites and strategy for loading photosensitizers stably and effectively towards cancer therapy.

Engineering Metal–Organic Frameworks for Photoacoustic Imaging-Guided Chemo-/Photothermal Combinational Tumor Therapy

Ying Zhang,Lei Wang,Liang Liu,Lin Lin,Feng Liu,Zhigang Xie,Huayu Tian,Xuesi Chen

Imaging-guided therapy has considerable potential in tumor treatment. Different treatments have been integrated to realize combinational tumor therapy with improved therapeutic efficiency. Herein, the conventional metal–organic framework (MOF) MIL-100 is utilized to load curcumin with excellent encapsulation capacity. Polydopamine-modified hyaluronic acid (HA-PDA) is coated on the MIL-100 surface to construct engineering MOF nanoparticles (MCH NPs). The HA-PDA coating not only improves the dispersibility and stability of NPs but also introduces a tumor-targeting ability to this nanosystem. A two-stage augmented photothermal conversion capability is introduced to this nanosystem by encapsulating curcumin in MIL-100 pores and then coating HA-PDA on the surface, which confer the MCH NPs with strong photothermal conversional efficiency. After being intravenously injected into xenograft HeLa tumor-bearing mice, MCH NPs prefer to accumulate at the tumor site and achieve photoacoustic imaging-guided chemo-/photothermal combinational tumor therapy, generating nearly complete tumor ablation. Engineering MOFs is an efficient platform for imaging-guided combinational tumor therapy, as confirmed by in vitro and in vivo evaluations.

Red blood cell membrane-derived phototherapeutic nanodiscs: A new platform for enhanced phototherapy

Zhuge Deli,Li Li,Sun Xueying,Liang Hui,Jin Chenjie,Lu Ailing,Gao Wenli,Zhong Yutong,Li Wenlu,Chen Sihao,Wang Fang,Yan Linzhi,Meng Weiyang,Lin Xiaoji,Zhang Xufei,Liu Yong,Li Yuanfeng,Zhao Yingzheng,Zhang Xiang,Chen Mengchun,Chen Yijie

Phototherapy holds great potential for treating cancer and infections but faces limitations related to photosensitizer accumulation, tissue penetration, and diminished photo-conversion efficiency, particularly in deep-seated tumors and infections. Here, a biomimetic phototherapeutic nanodisc platform consisting of erythrocyte membranes and the photosensitizer IR780 is developed. With the advantages of the ultra-small size and exceptional biosafety of cell membrane-derived nanodiscs, this platform facilitates efficient accumulation and deep tissue penetration at disease sites. Upon near-infrared (NIR) irradiation, IR780 delivered via the nanodisc exhibits enhanced photothermal conversion efficiency, markedly inhibiting tumor growth in an orthotopic 4T1 breast cancer model and reducing bacterial load in a methicillin-resistant Staphylococcus aureus (MRSA) skin infection model. Furthermore, the nanodisc platform demonstrates outstanding biocompatibility in mice. In conclusion, this nanodisc system significantly extends the functional potential of cellular nanodiscs, presenting a promising strategy to address the challenges of photosensitizer delivery and biosafety in phototherapeutic applications.

Dual active nanozyme-loaded MXene enables hyperthermia-enhanced tumor nanocatalytic therapy

Minglu Tang,Yangtian Shi,Liang Lu,Jingqi Li,Zhaocong Zhang,Jiatong Ni,Wenxin Wang,Yanhua Zhang,Tiedong Sun,Zhiguang Wu

Nanozymes, which have enzymatic activity and physicochemical properties of nanomaterials, have attracted extensive attention in the field of tumor therapy. However, the low activity of nanozymes in the tumor microenvironment leads to limited therapeutic effects. Here, we report an MXene-integrated nanozyme that exhibits dual catalytic activities, exerting synergistic effects to enhance tumor therapy. The MXene-integrated nanozyme consists of CeO 2 nanozymes with dual enzyme-like activities (simulating catalase and peroxidase) loaded on MXene. In such a construct, the catalase- and peroxidase-like activity of CeO 2 nanozymes alleviates hypoxia and elevates oxidative stress in the tumor microenvironment, and the overexpressed glutathione (GSH) is consumed by the redox reaction of CeO 2 nanozymes. The CeO 2 nanozyme-loaded MXene could couple the photothermal effect and dual enzyme-like activities, resulting in enhanced tumor nanocatalytic therapy with a tumor growth inhibition (TGI) effect of 92%. Dual active nanozyme-loaded MXene is a promising material for the development of nanozyme catalytic therapy in the future.

A Novel Microbubble Capable of Ultrasound-Triggered Release of Drug-Loaded Nanoparticles

Wang Jiayu,Li Pan,Tian Rui,Hu Wenjing,Zhang Yuxia,Yuan Pei,Tang Yalan,Jia Yuntao,Zhang Liangke

Drug-loaded microbubbles have shown attractive potential in disease treatment applications. The present work presents a unique ultrasound (US)-triggered system in which drug-loaded nanoparticles and perfluorocarbon gas are encapsulated within the internal space of microbubbles. The prepared curcumin-loaded albumin nanoparticle payload microbubbles (CcmANP-MB) exhibited a mean diameter of 4895.1 nm ± 421.2 nm and a drug-loading efficiency of 2.23% ± 0.08% (297% increase compared with the drug loading of common drug-loaded microbubbles). US allowed the release of the internal payload. In vitro US-triggered drug release experiments showed that the drug release of CcmANP-MB was delayed by lipid membranes and significantly increased after sonication. In vitro and in vivo US imaging experiments demonstrated that CcmANP-MB evidently enhances US imaging, which indicates that the microbubbles possess good acoustic properties even after encapsulation of nanoparticles. Tumor bearing mice were administered with CcmANP-MB through the tail vein and were then exposed to ultrasound, which resulted in an enhanced drug accumulation in tumor tissues and a significant increase in tumor growth inhibition rate (57.1%) compared with CcmANP-MB alone (28.8%) as well as curcumin-loaded albumin nanoparticle (26.2%). Therefore, the combination of lecithin microbubbles and albumin nanoparticles provides a platform for targeted drug delivery in clinical therapy and disease diagnosis.

Mechanistic study of enhanced drug release in mixed pH-responsive peptide-loaded liposomes

Genpei Zhang,Xilong Zhang,Kai Yue,Weishen Zhong

Liposomes serving as nanocarriers offer significant advantages in drug delivery for tumor treatment. There still exists challenges in controlling drug release by disintegrating the liposome membrane for the improvement of therapeutic efficiency. In this paper, a novel method involving the mixture of short peptides with pH-responsive characteristics into the cargo has been introduced. This approach facilitates the release of doxorubicin (DOX) in the acidic tumor tissue environment. The efficacy of this improvement was elucidated through molecular dynamics simulations and experiments. Liposomes incorporating a 1:1 ratio of peptides–DOX exhibited pronounced pH sensitivity and an enhanced drug release profile. The underlying mechanism is attributed to the peptides entering tumor tissues and undergoing protonation in acidic conditions, which increases the hydrophilicity of the peptide–DOX clusters and the internal surface tension of the liposomes. This alteration disrupts the balance between the inner and outer surface tensions of the nanocarrier, causing the liposomes to structurally disintegrate and thus enhancing drug release. The results from both thermodynamic analysis results and experimental data confirm the augmented drug release efficiency of this method, offering valuable theoretical insights for nanoparticle design and determining the optimal mixing ratio for therapeutic applications.

Thermo-responsive “hairy-rod” polypeptides for smart antitumor drug delivery

Jianxun Ding,Li Zhao,Di Li,Chunsheng Xiao,Xiuli Zhuang,Xuesi Chen

A series of thermo-responsive “hairy-rod” polypeptides was efficiently synthesized by grafting of azide-terminated (co)polymers of 2-(2-methoxyethoxy)ethyl methacrylate (MEO2MA) or 2-(2-(2-methoxyethoxy)ethoxy)ethyl methacrylate (MEO3MA) (i.e., N3-PMEOiMA) onto poly(γ-propargyl-L-glutamate) (PPLG) through a “click” reaction. The thermo-responsiveness and secondary structure of the resultant polypeptides (i.e. PLG-g-PMEOiMA) were shown to be dependent on the MEO2MA : MEO3MA ratio in PMEOiMA side chains. PLG40-g-P(MEO2MA18-co-MEO3MA9) (P2) and PLG40-g-P(MEO2MA7-co-MEO3MA18) (P4) with lower critical solution temperatures at 25.6 and 34.8 °C, and cloud points at 26.9 and 35.6 °C in physiological saline, respectively, were selected to self-assemble into micelles for doxorubicin (DOX) loading and release. In vitro DOX release from DOX-loaded micelles could be accelerated by a decrease of temperature and pH. Confocal laser scanning microscopy and flow cytometry confirmed the efficient internalization and intracellular DOX release of DOX-loaded micelles towards HeLa cells (a human cervical carcinoma cell line). In vitro methyl thiazolyl tetrazolium assays revealed that the polypeptides were cytocompatible, and DOX-loaded micelles showed efficient cellular proliferation inhibition. Hemolysis tests indicated that micelles were hemocompatible, and the encapsulation with polypeptides significantly reduced the hemolysis ratio of DOX. Therefore, the thermo-responsive polypeptide micelles, which are stable in physiological condition while releasing payloads in the acidic intracellular microenvironment, are promising for smart drug delivery.

Carrier-free nanodrugs for in vivo NIR bioimaging and chemo-photothermal synergistic therapy

Juanfang Lin,Chao Li,Yan Guo,Junjie Zou,Pengyu Wu,Yuqin Liao,Bingchen Zhang,Jingqing Le,Ruirui Zhao,Jing-Wei Shao

The combination of chemotherapy and photothermal therapy displays improved anti-cancer effects and lower systematic toxicity of a free drug compared with monotherapy. In this study, we designed innovative, carrier-free nanodrugs (PTX/ICG NDs) composed of the chemotherapeutic agent paclitaxel (PTX) and the photosensitizer indocyanine green (ICG) via self-assembly. The nanodrugs not only incorporated two different modalities into one delivery system for combined chemo-photothermal therapy but also enhanced the solubility of PTX without the need for any carrier. The as-prepared PTX/ICG NDs exhibited the merits of a relatively uniform size of 140 ± 1.4 nm, surface charge of −36 ± 2.2 mV, and high drug loading content of PTX. The combination strategy exerted a synergistic effect on the cytotoxicity of cancer cells in vitro, which could be attributed to the high cellular uptake and sustained release of PTX. Furthermore, an in vivo study indicated that PTX/ICG NDs showed higher accumulation in the tumor site than free ICG and possessed strong synergistic chemo-photothermal therapy efficacy against tumors in H22 tumor-bearing mice. Taken together, our study demonstrates that PTX/ICG NDs hold promise to become an alternative chemo-photothermal therapy agent to treat cancers.

Facile Synthesis of Fluorescent Hyper-Cross-Linked β-Cyclodextrin-Carbon Quantum Dot Hybrid Nanosponges for Tumor Theranostic Application with Enhanced Antitumor Efficacy

Mingliang Pei,Jui-Yu Pai,Pengcheng Du,Peng Liu

Fluorescent hyper-cross-linked β-cyclodextrin-carbon quantum dot (β-CD-CQD) hybrid nanosponges of about 200 nm with excellent biocompatibility and strong bright blue fluorescence excited at 365 nm with a high photoluminescence quantum yield (PLQY) of 38.0% were synthesized for tumor theranostic application by facile condensation polymerization of carbon quantum dots (CQDs) with β-cyclodextrin (β-CD) at a feeding ratio of 1:5. The DOX@β-CD-CQD theranostic nanomedicine, around 300 nm with DOX-loading capacity of 39.5% by loading doxorubicin (DOX) via host–guest complexation, showed a pH responsive controlled release and released DOX in the simulated tumor microenvironment in a sustained release mode, owing to the formation constant in the supramolecular complexation of DOX with the β-CD units in the β-CD-CQD nanosponges. The proposed DOX@β-CD-CQD theranostic nanomedicine could be internalized into HepG2 cells, and the released DOX was accumulated into the cell nuclei, demonstrating an antitumor efficacy more enhanced than that of the free drug.

Photoenhanced gene transfection by a curcumin loaded CS-g-PZLL micelle

Jian-Tao Lin,Wen-Jia Pan,Jun-Ai Zhang,Wei Wang,Jia Zhong,Jia-Min Su,Tong Li,Ying Zou,Guan-Hai Wang

The codelivery of drug and gene is a promising method for cancer treatment. In our previous works, we prepared a cationic micelles based on chitosan and poly-( N -3-carbobenzyloxylysine) (CS-g-PZLL), but transfection ratio of CS-g-PZLL to Hela cell was low. Herein, to improve the transfection efficiency of CS-g-PZLL, curcumin was loaded in the CS-g-PZLL micelle. After irradiation, the obtained curcumin loaded micelle showed a better transfection, and the p53 protein expression in Hela cells was higher. The apoptosis assay showed that the complex could induce a more significant apoptosis to Hela cells than that of curcumin or p53 used alone, and the curcumin loaded micelle inducing apoptosis was best after irradiation. Therefore, CS-g-PZLL is a safe and effective carrier for the codelivery of drug/gene, and curcumin could be used as a photosensitizer to induce a photoenhanced gene transfection, which should be encouraged in improving transfection and tumor therapy.

Boosting Type-I and Type-II ROS Production of Water-Soluble Porphyrin for Efficient Hypoxic Tumor Therapy

Han Sun,Ruihua Guo,Yanhui Guo,Jitao Song,Zhiliang Li,Fengling Song

As the most successful clinically approved photosensitizers, porphyrins have been extensively employed in the photodynamic therapy (PDT) of cancers. However, their poor water solubility, aggregation-induced self-quenching on ROS generation, and a low tolerance for a hypoxic condition usually result in unsatisfied therapeutic outcomes. Therefore, great efforts have been dedicated to improving the PDT efficacy of porphyrin-type photosensitizers in treating hypoxic tumors, including combination with additional active components or therapies, which can significantly complicate the therapeutic process. Herein, we report a novel water-soluble porphyrin with O-linked cationic side chains, which exhibits good water solubility, high photostability, and significantly enhanced ROS generation efficacy in both type-I and type-II photodynamic pathways. We have also found that the end charges of side chains can dramatically affect the ROS generation of the porphyrin. The cationic porphyrin exhibited high in vitro PDT efficacy with low IC50 values both in normoxia and hypoxia. Hence, during in vivo PDT study, the cationic porphyrin displayed highly effective tumor ablation capability. This study demonstrates the power of side-chain chemistry in tuning the photodynamic property of porphyrin, which offers a new effective strategy to enhance the anticancer performance of photosensitizers for fulfilling the increasing demands for cancer therapy in clinics.

Customized multi-stimuli nanovehicles with dissociable ‘bomblets’ for photothermal-enhanced synergetic tumor therapy

Jiayin Lin,Gang Li,Ke Jiang,Tao Xu,Chang Liu,Lipin Wang,Xu Zhang,Defu Cai,Chao Wu,Xianshi Meng,Wenquan Zhu

Recently, the therapeutic effect of chemotherapy has been obviously impaired due to premature drug release, low tumor penetration, and multidrug resistance of nanoplatforms. In this paper, a novel multiple-sensitive drug delivery system (MC-ss-CDs) was developed by gating long-wavelength emitting carbon dots (CDs) on the openings of mesoporous carbon nanoparticles (MC) through disulfide bonds. The MC with excellent photothermal transition efficiency and high drug storage capacity for doxorubicin (DOX) was used as the delivery carrier. The CDs had multiple functions, including intelligent switching to hinder unwanted release, photothermal therapy (PTT) agents to improve the heat generation effect of MCs and bioimaging trackers to monitor drug delivery. The disulfide bonds, as the linkers between MC carriers and CDs, are stable under normal physical conditions and relatively labile under high GSH concentrations in the cytoplasm of tumor cells. After arriving at the tumor microenvironment, DOX/MC-ss-CDs can rapidly break into DOX/MC and CDs under high GSH concentrations. DOX/MC could realize efficient integration of PTT and chemotherapy on the surface of the tumor by stimuli-responsive DOX release and synergetic heating of MC and CDs. The small-sized CDs with excellent penetrating ability could effectively enter the deep tumor and realize NIR-triggered photothermal ablation. The DOX/MC-ss-CDs showed a chemophotothermal effect with a combination index of 0.38 in vitro and in vivo . Therefore, the DOX/MC-ss-CDs could be employed as a trackable nanovehicle for synergistic chemotherapy and PTT at different depths.
