Anastomotic leaks are among the most dreaded complications following gastrointestinal (GI) surgery, and contrast-enhanced X-ray gastroenterography is considered the preferred initial diagnostic method for GI leaks. However, from fundamental research to clinical practice, the only oral iodinated contrast agents currently available for GI leaks detection are facing several challenges, including low sensitivity, iodine allergy, and contraindications in patients with thyroid diseases. Herein, we propose a cinematic contrast-enhanced X-ray gastroenterography for the real-time detection of GI leaks with an iodine-free bismuth chelate (Bi-DTPA) for the first time. The Bi-DTPA, synthesized through a straightforward one-pot method, offers distinct advantages such as no need for purification, a nearly 100 % yield, large-scale production capability, and good biocompatibility. The remarkable X-ray attenuation properties of Bi-DTPA enable real-time dynamic visualization of whole GI tract under both X-ray gastroenterography and computed tomography (CT) imaging. More importantly, the leaky site and severity can be both clearly displayed during Bi-DTPA-enhanced gastroenterography in a rat model with esophageal leakage. The proposed movie-like Bi-DTPA-enhanced X-ray imaging approach presents a promising alternative to traditional GI radiography based on iodinated molecules. It demonstrates significant potential in addressing concerns related to iodine-associated adverse effects and offers an alternative method for visually detecting gastrointestinal leaks.

Introduction Nanoparticles are increasingly used as drug carriers for oral administration. The delivery of drug molecules is largely dependent on the interaction of nanocarriers and gastrointestinal (GI) mucus, a critical barrier that regulates drug absorption. It is therefore important to understand the effects of physical and chemical properties of nanocarriers on the interaction with GI mucus. Unfortunately, most of the nanoparticles are unable to be prepared with satisfactory structural monodispersity to comprehensively investigate the interaction. With controlled size, shape, and surface chemistry, copolymers are ideal candidates for such purpose. Materials and methods We synthesized a series of diblock copolymers via the atom transfer radical polymerization method and investigated the GI mucus permeability in vitro and in vivo. Results Our results indicated that uncharged and hydrophobic copolymers exhibited enhanced GI absorption. Conclusion These results provide insights into developing optimal nanocarriers for oral administration.

Considering the controversial safety and risk solutions of ionic liquids (ILs), more essential basic researches are required as their applied fields become broader and applied scale grows larger. Under this background, the permeation behaviors of six common imidazole ILs on porcine gastric and enteric membranes together with the feasibility of their removal by adsorption with montmorillonite were investigated in vitro. Through the single-factor investigation of different cations, anions and ionic liquid concentrations, the current results show that the IL of 1-ethyl-3-methylimidazolium bromide ([Emim]Br) has the strongest permeation capacity on the gastric membrane during the tests. The cumulative permeation amount of Q n can reach 0.72 mg/cm 2 , and the cumulative permeation percentage alsoreaches 95.77 %; the whole permeation process conforms to the Hixcon-Crowell model. As comparison, the maximum cumulative penetration percentage of [Emim]Br on enteric member was determined as 89.11 %, and related models was also compared. In order to remove the potential IL in digestive tract, montmorillonite was screened out from those popularily used antidotes in clinic and the adsorption of [Emim]Br in gastric fluid by montmorillonite has been proved more consistent with the second-order kinetics and Langmuir model. The results of molecular simulation, infrared spectrum (IR), X-ray diffraction (XRD) and thermogravimetric (TG) characterization exhibit that [Emim]Br has been successfully adsorbed into the interlayer of montmorillonite. As a whole, the study provided a scientific and effective way to diminish the hidden danger of ionic liquid entering the blood through the digestive system.

Gastrointestinal disease is the most common health concern that occurs due to environmental, infectious, immunological, psychological, and genetic stress. Among them, the most frequent diseases are gastric ulcer (GU) and ulcerative colitis (UC). DSS-induced UC and ethanol-stimulated GU models resemble the pathophysiology of human gastrointestinal disease. The current study was designed to explore the anti-oxidation, anti-inflammation, anti-cell death properties of terazosin, an α-adrenergic receptor antagonist, in vivo and in vitro. Our results indicate that terazosin dramatically activates Pgk1, and upregulates glycose metabolism, evidenced by the enhanced ATP production and higher LDH enzymatic activity. Also, terazosin significantly enhances p-AKT expression and inhibits NF-κB p65 activation through abrogating the phosphorylation of IKBα, as well as lowers Caspase-1 and GSDMD expression. The findings in this study demonstrate that terazosin exhibits anti-inflammatory effects by downregulating NF-κB-GSDMD signal pathway, along with enhancing glycolysis for gastrointestinal disease treatment. Meanwhile, we also find terazosin ameliorates ethanol-induced gastric mucosal damage in mice. Collectively, as a clinical drug, terazosin should be translated into therapeutics for gastrointestinal disease soon. Keywords:terazosin;gastrointestinal disease;anti-inflammation;Pgk1;cell death

Recent advances in synthetic micro/nanomotors in numerous biofluids have garnered widespread hobby because of their capability biomedical programs. However, present micro/nanomotor systems for shipping inside the gastrointestinal (GI) tract are inefficient in propulsion. Here, we present a micromotor-tableted tablet that achieves lively retention in the GI tract in vivo. The drug-loaded magnesium-based totally absolutely Janus micromotors, enveloped in an enteric polymer-protected magnetic tablet, stay sturdy within the stomach. The movement of the micromotor tablets (MPs) is facilitated by means of the use of an outside low-frequency gradient magnetic difficulty, directing them to focused regions inside the intestines. A excessive-frequency alternating magnetic concern induces water-powered propulsion of the micromotors via the magnetocaloric effect, attaining lively retention in GI tissue. The integration of this newly evolved MP tool lets in energetic retention of micromotors in vivo, promising more suitable drug transport for GI treatment.

Ovomucin (OVM) exhibits pH-responsive conformational changes, aggregating under acidic conditions and extending under alkaline conditions, making it a promising stabilizer for double emulsions (DEs) designed to protect probiotics in the gastrointestinal environment. This study presents an OVM-based DE system for the efficient encapsulation and delivery of Companilactobacillus crustorum MN047. The primary W 1 /O emulsion was formed by homogenizing olive oil with 4% PGPR and sodium chloride solution (8:2 ratio), followed by homogenization with 2.5% pH 9 OVM solution (3:7 ratio) to create the DE. Particle size, zeta potential, rheology, and interfacial tension analyses confirmed the stabilizing role of OVM under alkaline conditions, whereas CLSM and circular dichroism revealed the structural effects of OVM, resulting in increased α-helix and decreased β-sheet contents under acidic conditions. DE achieved an encapsulation efficiency of 93.55%, with protection rates of 82.26% in gastric fluid and 66.82% in intestinal fluid. In a dextran sulfate sodium-induced ulcerative colitis mouse model, OVM-encapsulated C. crustorum MN047 demonstrated enhanced effects in inflammation alleviation, intestinal barrier integrity, and gut microbiota regulation. This study underscores the potential of OVM-based DE systems for probiotic oral delivery and introduces an effective encapsulation approach for gastrointestinal applications.

Objective: Camellia nitidissima Chi, a Chinese medicine commonly used by ethnic minorities in Guangxi, China, is beneficial for clearing heat, detoxifying, inducing diuresis, and suppressing swelling. It has various pharmacological properties, including antitumor, anti-inflammatory, and antioxidant. However, its potential application in radioprotection remains unclear. In this study, we aimed to determine whether Camellia nitidissima Chi has radioprotective effects against radiation-induced gastrointestinal and hematopoietic damage. Methods: The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) techniques were used to assess the ability of Camellia nitidissima Chi to scavenge free radicals. We conducted a 30-day survival rate experiment to evaluate the radioprotective capabilities of Camellia nitidissima Chi. Additionally, we developed models of radiation-induced intestinal and hematopoietic damage. Alterations in the white blood cell (WBC) count, total superoxide dismutase (T-SOD), glutathione (GSH), and protein expression linked to apoptosis were observed. Results: Camellia nitidissima Chi scavenged 84.72% and 93.47% of DPPH and ABTS, had a certain radiation protection potential, and increased the survival rate of mice to over 90%. Moreover, following exposure, Camellia nitidissima Chi enhanced WBC, T-SOD, and GSH levels. Camellia nitidissima Chi increased B-cell lymphoma-extra large (BCL-XL) expression and suppressed Bcl-2 associated X protein (BAX) expression, providing radioprotection to cells. Conclusions: Camellia nitidissima Chi has a strong antioxidant ability; it can improve the survival rate of mice after lethal dose irradiation and protect against radiation-induced hematopoietic and gastrointestinal damage. These findings can serve as a guide for using Chinese medicines for radioprotection.

In the current study, how pectin retards the digestibility of wheat gluten was investigated using a static in vitro gastric-duodenal model. The degree of protein hydrolysis was estimated using the o-phthaldialdehyde method, while the in vitro digestograms were mathematically fitted using a single first-order kinetics model. Peptides' profile, free amino acids compositions, gluten-pectin interactions and their effects on enzymatic activities of proteolytic enzymes as well as on the gluten secondary structures under digestive conditions were studied using combined techniques. Results showed that pectin could retard gluten digestibility through 1). preferential absorption to insoluble gluten aggregates by electrostatic interactions; 2). increasing the helix and reducing the β-sheet content of the solubilized gluten protein fractions in terms of their secondary molecular structures; 3). reducing pepsin activity by forming negatively charged pectin-gluten mixtures which then interacted with the positively charged pepsin molecules. The deeper insight into gluten-pectin interactions and their influences on gluten digestibility under gastrointestinal conditions provides important clues for developing effective forms of dietary fiber to improve the nutritional benefits of plant protein in individuals.

Since understanding the healthy status of gastrointestinal tract (GI tract) is of vital importance, clinical implementation for GI tract-related disease have attracted much more attention along with the rapid development of modern medicine. Here, a multifunctional theranostic system combining X-rays/CT/photothermal/photoacoustic mapping of GI tract and imaging-guided photothermal anti-bacterial treatment is designed and constructed. PEGylated W 18 O 49 nanosheets (PEG-W 18 O 49 ) are created via a facile solvothermal method and an in situ probe-sonication approach. In terms of excellent colloidal stability, low cytotoxicity, and neglectable hemolysis of PEG-W 18 O 49 , we demonstrate the first example of high-performance four-modal imaging of GI tract by using these nanosheets as contrast agents. More importantly, due to their intrinsic absorption of NIR light , glutaraldehyde-modified PEG-W 18 O 49 are successfully applied as fault-free targeted photothermal agents for imaging-guided killing of bacteria on a mouse infection model. Critical to pre-clinical and clinical prospects, long-term toxicity is further investigated after oral administration of these theranostic agents. These kinds of tungsten-based nanomaterials exhibit great potential as multi-modal contrast agents for directed visualization of GI tract and anti-bacterial agents for phothothermal sterilization.

The recent emergence of versatile micro/nanorobots provides new strategies of healthcare and disease treatment, especially for targeted delivery of therapeutic agents to otherwise inaccessible regions. However, for successful translation of laboratory microrobotic tools into clinical outcomes, critical challenges still remain, such as controllable cargo loading/release on site, robust spatiotemporal navigation in real time and reliable biocompatibility/biodegradation inside the body. Herein, we report the loading, transport and release of molecular cargos using cell-based magnetic microswimmers engineered from multicellular Spirulina . The loading is attained by harnessing the dehydration and rehydration of Spirulina cells, inside which model macromolecules are encapsulated as a discrete phase, and for small molecules, as a continuous phase. Driven by low-strength magnetic fields, the molecule-laden swimmer manages to perform navigation in an intestinal tract mimicry, imitating the process of guest-molecule transport in the gastrointestinal tract. The loaded molecules can be released from the swimmer through host degradation and/or concentration gradient-driven diffusion, with the speed of release tailored by the magnetite-coating thickness. Furthermore, evaluation tests in a stem-cell system infer that the bioactivity of the guest molecules remains intact after the loading-release procedure. In summary, we have demonstrated a feasible approach for gastrointestinal delivery of molecular agents using cell-based microswimmers.

The antimicrobial, anticancer, and anti-SARS-CoV-2 activities of cepharanthine (CEP) combined with luteolin (Lut) were compared and analyzed by in vitro bioactivity experiment and molecular docking. CEP shows stronger biological activity than Lut, and their combination shows synergism without antagonism, especially in the fungal inhibition and anticancer activity experiments. It is believed that the antiviral activity is still effective confirmed by the simulation between two drugs and anti-SARS-CoV-2. Further, pepsin (Pep) was chosen for imitative gastroenteric environments to study its protease activity and structure effected by two drugs. The results show that the inhibitory mechanism of Lut/CEP is non-competitive inhibition, the binding mechanism is static quenching driven by enthalpy, the order of drug addition can affect the strength of binding ability, and the strength of binding ability is positively correlated with the inhibitory effect on enzyme activity. The ingestion of CEP before ingestion of Lut is more conducive to the efficient utilization of them. Their combination can not only decrease the dosage and toxicity, but also enhance the anti-inflammatory and anti-virus effects. These results can provide theoretical support for the development and utilization of Lut and CEP, and may supply one of the treatment for deal with the newly appeared viruses.

The role of Pediococcus pentosaceus in the gastrointestinne has received considerable attention in recent decades. This study aimed to investigate the effects of the short-term administration of P. pentosaceus on physiological characteristics, inflammation, and intestinal microecology in mice. In this study, 90 male C57BL/6J mice were divided into 15 groups, with 14 groups treated with a daily intragastric administration of different genotypes of P. pentosaceus. After three weeks of intragastric administration P. pentosaceus had a mild effect on mice. It could be seen that different P. pentosaceus strains had different effects on the gut microbiota and intestinal microecology. P. pentosaceus VCQYC5144M12 possessing an Enterolysin A operon may have been harmful, activating the expression of inflammatory factors, while P. pentosaceus DYNDL69M8 consisting of only a pediocin-like operon increased the abundance of beneficial bacteria and increased the content of acetic acid. The presence of various genotypes of bacteriocin may have been the explanation for variations among strains. This may provide theoretical support for further exploring the probiotic effect and patterns of P. pentosaceus.
