Reversible inhibition of trypsin activity with soybean flour in hide bating process for leather quality improvement

Huan Wang,Chao Lei,Yunhang Zeng,Ying Song,Qixian Zhang,Bi Shi

Trypsin is used to remove non-collagenous proteins from cattle hide in leather processing but prone to damage hide surface and reduce leather quality due to its rapid protein hydrolysis action on the hide surface and slow penetration through the whole hide. A novel trypsin bating method was developed using a reversible inhibition of trypsin with high-yield, low-cost and environment-friendly soybean flour (SF). When the mass ratio of trypsin to SF was 1:20, the trypsin activity was decreased by 77 % at a reaction time of 5 min and completely returned in 100 min because of the competitive Kunitz trypsin inhibitor in SF. Bating with trypsin-SF mixture avoided the damage to hide surface caused by trypsin, improved the dispersion uniformity of hide collagen fibers and increased the softness and the mechanical strength of leather. This work exhibits huge potential for improving the leather quality and provides new insights into the soybean application.

An ultrasensitive fluorescent nanosensor for trypsin based on upconversion nanoparticles

Mingmin Wu,Xiaoyong Wang,Kun Wang,Zijian Guo

Trypsin and its inhibitors are relevant to many physiological processes and diseases. In this study, a nanosensor capable of detecting trypsin and its inhibitors was designed based on the fluorescence resonance energy transfer (FRET) between upconversion nanoparticle (UCNP) and gold nanoparticle (AuNP). UCNP and AuNP were linked by a trypsin-sensitive peptide DDDDARC, forming the non-fluorescent UCNP-peptide-AuNP nanosensor. In the presence of trypsin, the peptide was cleaved and the quenched fluorescence was restored; in the presence of trypsin inhibitors , the recovery of the fluorescence was decreased. The nanosensor showed a superb sensitivity and selectivity for trypsin and its inhibitors, with a detection limit of 4.15   ng   mL −1 for trypsin. UCNP-peptide-AuNP could eliminate the interference of background fluorescence and avoid the light toxicity, and potentially be used to diagnose trypsin-related diseases or screen trypsin inhibitors.

Evaluation of the toxicity of ionic liquids on trypsin: A mechanism study

Yunchang Fan,Xing Dong,Lingling Yan,Dandan Li,Shaofeng Hua,Chaobing Hu,Chengcheng Pan

The toxicity of ionic liquids (ILs) was evaluated by using trypsin as biomarker. Experimental results indicated that the trypsin activity was inhibited by ILs and the degree of inhibition highly depended on the chemical structures of ILs. Primary analysis illustrated that hydrophobicity of ILs was one of the driven forces ruling the ILs-trypsin interaction. Thermodynamic parameters, Gibbs free energy change (Δ G ), enthalpy change (Δ H ) and entropy change (Δ S ) were obtained by analyzing the fluorescence behavior of trypsin in the presence of ILs. Both negative Δ H and Δ S suggested hydrogen bonding was the major driven force underlying the IL-trypsin interaction. To assess the toxicity of ILs, it should be considered the combination of the hydrogen bonding ability and hydrophobicity of ILs. A regression based model was established to correlate the relationship of the inhibitory ability, hydrophobicity and hydrogen bonding ability of ILs.

Evaluation of the binding mechanism of iodine with trypsin and pepsin: A spectroscopic and molecular docking

Yanqing Wang,Qianqian Han,Gencheng Zhang,Hongmei Zhang

In this work, the effects of I 2 on the activities and conformational structures of digestive enzymes, trypsin and pepsin were studied. The results indicated that the enzyme activities were decreased to some extent in the presence of I 2 , especially trypsin. Upon gradual addition of I 2 , the intrinsic fluorescence quenching of trypsin and pepsin were observed by mainly static collision and hydrophobic forces . I 2 is more likely to cause the fluorescence quenching of trypsin than that of pepsin. Compared with pepsin, trypsin has a greater ability to bind with I 2 . The synchronous fluorescence spectral results indicated that I 2 induced the quaternary structure changes of trypsin/pepsin and changed the hydrophobicity of Tyr and Trp residues . In addition, molecular docking was used to obtain the binding mode and the various amino acid residues of trypsin and pepsin with I 2 . These investigations may constitute a solid work to further explain the process of migration and transformation of I 2 in digestive system.

Mechanism and Nature of Inhibition of Trypsin by Ligupurpuroside A, a Ku-Ding Tea Extract, Studied by Spectroscopic and Docking Methods

Wu Zhibing,Shen Liangliang,Han Qingguo,Lu Jun,Tang Haifeng,Xu Xu,Xu Hong,Huang Fengwen,Xie Jiangfeng,He Zhendan,Zeng Zheling,Hu Zhangli

Ligupurpuroside A is a glycoside extracted from Ku-Ding tea. As extracts from Ku-Ding tea exhibit anti-inflammatory property, we hypothesize that Ligupurpuroside A may be an active compound which inhibits trypsin activity during the anti-inflammatory process. The mechanism and nature of inhibition of trypsin by Ligupurpuroside A have been studied by multi-spectroscopic method, enzyme-activity assay and molecular docking. Enzyme activity assay reveals that Ligupurpuroside A significantly inhibits the activity of trypsin through a competitive manner with an IC 50 value of 3.08 × 10 −3  mol L −1 . Fluorescence titration together with thermodynamic analysis indicate that a Ligupurpuroside A-trypsin complex is formed, and that hydrophobic force and hydrogen bonding are the main forces stabilizing the complex. UV-vis absorption, synchronous fluorescence and circular dichroism spectra show that the interaction between Ligupurpuroside A and trypsin induces conformational changes of trypsin with a decrease in the contents of α-helix and β-sheet. Finally, molecular docking further suggests that Ligupurpuroside A molecule binds within the active pocket of trypsin via hydrophobic force and hydrogen bond. Results from this study of the interaction of trypsin with its natural inhibitor should be useful to minimize the antinutritional effects and make full use of tea extracts in the food industry, and be also helpful to the design of the drugs for the diseases related to overexpression of trypsin.

Trypsin stabilized copper nanoclusters as a highly sensitive and selective probe for fluorescence sensing of morin and temperature

Xian-song Wang,Shen Zhang

Herein, a platform for sensitive fluorescence sensing of morin was developed based on the copper nanoclusters , which was carried out by using a one-pot method. The copper nanoclusters (Trypsin@Cu NCs) were fabricated through CuCl 2 as a metal precursor, trypsin as a protective agent and hydrazine hydrate as a reducing agent. The Trypsin@Cu NCs exhibited blue fluorescence under the excitation wavelength of 378 nm. Using Trypsin@Cu NCs as the patterned nanosensors , the fluorescent “turn off” phenomenon was developed for the measurement of morin with the linear decline of emissive intensity at 459 nm. The fluorescence quenching reason was subjected to the static quenching and inner filter effect . Under the optimal conditions, an excellent linear response between ln(F 0 /F) of Trypsin@Cu NCs and morin amounts in the light of the range of 0.5–150 μM, and the limit of detection was 0.083 μM. Moreover, this proposed nanosensor could be applied for the sensing of temperature. This sensing method was displayed to be a practical determination system for the measurement of morin in real samples, which could open new horizons to develop novel detection models for morin.

Ascorbic Acid-Loaded Apoferritin-Assisted Carbon Dot-MnO2 Nanocomposites for the Selective and Sensitive Detection of Trypsin

Qingqing Tan,Ruirui Zhang,Weisu Kong,Fengli Qu,Limin Lu

A novel “turn-on” fluorescent trypsin detection platform dependant on carbon dot-MnO2 (CD-MnO2) nanocomposites and ascorbic acid-loaded apoferritin (APOAA) was fabricated. The detection mechanism relied on trypsin-catalyzed enzymolysis of APOAA, which released ascorbic acid (AA) as a reducing agent to disintegrate the MnO2 nanosheets, causing the recovery of the fluorescence of CDs. An excellent performance and high sensitivity of trypsin determination were observed with a detection limit (LOD) of 0.3411 ng/mL. This work provides us with a unique strategy for trypsin detection in human serum samples, which reveals the potential applications in clinical detection.

Mechanism of curcumin-induced trypsin inhibition: Computational and experimental studies

Yan-Qing Wang,Hong-Mei Zhang,Yi-Jun Kang,Yun-Lan Gu,Jian Cao

In the present study, the experimental and theoretical methods were used to analyze the binding interaction of food dye , curcumin with trypsin. The results of fluorescence spectroscopic measurements indicated that curcumin binding resulted in the obviously intrinsic fluorescence quenching with the increase concentration of curcumin. This binding interaction is a spontaneous process with the estimated enthalpy and entropy changes being −15.70 kJ mol −1 and 40.25 J mol −1  K −1 , respectively. Hydrogen bonds and hydrophobic forces played an important role in the complex formation between curcumin and trypsin. Moreover, curcumin could enter into the primary substrate-binding pocket and makes the activity of trypsin decrease remarkably with the increasing concentration of curcumin.

Dissection of binding of trypsin to its natural inhibitor Gensenoside-Rg1 using spectroscopic methods and molecular modeling

Jialiang Lin,Yang Xu,Yuhan Wang,Songyang Huang,Junwei Li,Manjunath D. Meti,Xu Xu,Zhangli Hu,Johnson Liu,Zhendan He,Hong Xu

The interaction of trypsin with Gensenoside-Rg1 (G-Rg1) was studied using fluorescence, ultraviolet–visible (UV–vis), and circular dichroism (CD) spectroscopies along with enzyme activity assay and molecular docking. The enzyme activity assays showed that G-Rg1 inhibited the activity of trypsin effectively. The fluorescence experiments indicated that a complex of G-Rg1–trypsin was formed and that the fluorescence of trypsin was quenched by G-Rg1 via a mixed-quenching mechanism (both static and dynamic quenching). The thermodynamic analysis suggested that hydrophobic interaction and hydrogen bond were the major forces between G-Rg1 and trypsin. According to the theory of Förster’s non-radiation energy transfer, the binding distance between trypsin and G-Rg1 was calculated to be 2.01 nm, which implies that energy transfer occurred within the complex. The experimental results obtained from UV–vis absorption spectra, synchronous fluorescence spectra, and CD spectra indicated that G-Rg1 was mainly located on tryptophan moiety and that the interaction between G-Rg1 and trypsin led to conformational changes of trypsin with some α-helix and unordered coil structures being transformed into β-sheet structures. In addition, docking results supported the above experimental findings and suggested the possible binding location of G-Rg1 on trypsin along with the possible hydrogen bonds and hydrophobic interactions between G-Rg1 and trypsin. The experimental results from this study should be useful to minimize the antinutritional effects and make full use of Genseng extracts in the food industry and also be helpful to the design of the drugs for the diseases related to overexpression of trypsin.Communicated by Ramaswamy H. Interaction of phenolic acids with trypsin: Experimental and molecular modeling studies

Benguo Liu,Huizhi Xiao,Jiaqi Li,Sheng Geng,Hanjun Ma,Guizhao Liang

Trypsin is a kind of protease for digestion and food processing, whose activity can be inhibited by phenolic acids in plant foods. However, most reports explained the inhibitory difference of phenolic acids based on the number and position of substituent groups, which failed to reveal the comprehensive inhibitory mechanism. In this work, the inhibitory effects of 11 common phenolic acids on trypsin were investigated. Amongst the tested cinnamic and benzoic acid derivatives, caffeic acid and gallic acid showed the strongest anti-trypsin activity with a noncompetitive inhibition pattern, respectively. The fluorescence analysis displayed that both the quenching rate constant (K q ) and binding constant (K A ) of caffeic acid were higher than those of gallic acid. Molecular docking illustrated their different binding modes with trypsin. The ONIOM calculations revealed that the binding capacity of caffeic acid was higher than that of gallic acid, which could explain their difference in their inhibitory behaviors.

Nano-encapsulant of ascorbic acid-loaded apoferritin-assisted photoelectrochemical sensor for protease detection

Jiexia Chen,Guang-Chao Zhao

A signal-on photoelectrochemical (PEC) biosensor based on nano-encapsulant of ascorbic acid-loaded apoferritin - assisted for protease detection is described. The loaded ascorbic acid could be released from the central cavity of apoferritin into the buffer solution as sacrificial electron donor to capture the photo-generated holes of CdTe quantum dots when light is turned on. In this system, the biosensor relied on monitoring the photocurrent intensity as a result of enzymatic substrate proteolysis in the homogeneous aqueous solution. A low detection limit of 2.7   ng   mL −1 for trypsin in the linear range from 30 to 450   ng   mL −1 is achieved. We believe that such a sensing system not only holds the potential ability as a probe for trypsin activity assay, but also could be used for the corresponding inhibitor-screening. It might provide a potentially feasible alternative tool for determining trypsin in human serum in a clinical laboratory. The established method could open a different perspective for PEC enzyme detection and provide a new platform for future development of PEC analysis with other clinically important proteins.

Enzymatic response of heparin-protamine complex: Spectroscopic investigation and application for lung adenocarcinoma cells detection

Zichun Qi,Nana Pan,Dongxue Han,Jiahua He,Jin-ao Li,Luqi Yang,Xiaojuan Wang,Fang Huang

Though the heparin-protamine complex (HP complex) is a crucial system utilized in clinical settings, the metabolic pathways of this complex remain inadequately understood. Herein, the enzymatic degradation of the heparin-protamine complex by trypsin and its broader implications were investigated. By utilizing fluorescent gold nanoclusters liganded with the HP complex (AuNCs-HP complex), we observed significant morphological and spectral changes during enzymatic degradation. Experiments showed that AuNCs-HP complex could be degraded and cleaved into small fragments by trypsin. Moreover, the AuNCs-HP complex demonstrated its potential as a highly sensitive spectral sensing platform, enabling precise measurement of trypsin activity with an outstanding detection limit (0.34 ng mL −1 ). Additionally, we explored its utility for specific tumor cell detection, focusing on lung adenocarcinoma cells, and successfully identified their presence through distinctive fluorescence changes. These remarkable findings not only contribute valuable insights into targeted degradation systems but also offer promising opportunities for cancer biomarker detection. The AuNCs-HP complex serves as an innovative tool for real-time trypsin activity monitoring, paving the way for advanced biomedical applications.
