A luminescent reagent, 3,4,9,10-perylenetetracarboxylic acid (PTCA), was linked to a metal–organic framework (MOF) Fe(III)-MIL-88B-NH 2 through an amide reaction to form a [email protected] (III)-MIL-88B-NH 2 complex, and then gold nanoparticles (AuNPs) were grown in situ. Thus, the [email protected] (III)-MIL-88B-NH 2 @Au electrochemiluminescence (ECL) probe was obtained. The MOF of 2-aminoterephthalic acid with large specific surface area, pore sizes, and structures, which served both as the carrier and a co-reaction promoter, promoted the conversion of more S 2 O 8 2- into SO 4 •- , further enhancing the ECL signal of PTCA. An electrochemiluminescence resonance energy transfer (ECL-RET) platform was designed for the ultrasensitive and highly selective detection of cystatin C (CysC). The developed ECL-RET biosensor presented a linear range from 0.01 pg/mL to 100 ng/mL and a detection limit of 2.2 fg/mL (S/N = 3). Moreover, all the results showed that the developed ECL-RET biosensor broadened the application in the fields of biosensing and medical diagnosis.

Cystatin C (CysC) has been proven to be used to diagnose acute kidney injury (AKI) rapidly and sensitively early. Therefore, it is urgent to develop a sensitive, novel, and rapid method for detecting CysC. In this work, a novel photoelectrochemical (PEC) biosensor was designed for ultrasensitive CysC detection. Ti-MOF@DM-LZU1@Au as a photosensitive material was first modified on the ITO electrode surface. Then, Ab1 and CysC were assembled on the electrode via the specific immunoresponse of an antigen and antibody. Lastly, the conjugate Ab2/l-Cys bilayer/l-Cys-hemin/G-quadruplex with self-catalytic enzyme performance, as a signal amplification approach, could further react with CysC and Ab1, which resulted in a stronger photocurrent. As expected, the constructed PEC sensor realized the ultrasensitive detection of CysC, with a detection range of 10 pg/mL to 16 μg/mL and a lower limit of 8.023 pg/mL. The biosensor had excellent repeatability, selectivity, and stability. Moreover, it can provide a new method for the sensitive and rapid detection of other protein molecules in clinical practice.
Background Contrast-induced acute kidney injury (CI-AKI) is a common complication after exposure to contrast media. Renal ischaemia occurs in the initial stage of CI-AKI, however, there are very few effective measures to improve renal perfusion . Methods A total of 114 patients with an estimated glomerular filtration rate (eGFR) of 60–89 ml/min/1.73m 2 were randomly assigned to two groups: enhanced external counterpulsation (EECP) group ( N = 57) and control group (N = 57). Two hours after contrast exposure, EECP group received EECP treatment for 1 h while no intervention was performed control group. The primary outcome was the incidence of serum cystatin C concentration to 10% above the baseline concentration at 24 h after contrast administration. The secondary outcomes were the incidence of CI-AKI (defined as an increase in serum creatinine concentration to ≥0.5 mg/dl or by 25% compare to the baseline after contrast exposure), contrast clearance and adverse clinical events. Results The primary outcome was observed in 26 patients (6 EECP and 20 control; 11% vs. 35%; P = 0.002). CI-AKI occurred in four patients (0 EECP and 4 control; 0% vs. 7%; P = 0.042). The clearance rate of iopromide in the initial 3 h was significantly different between EECP and control group (59.92 ± 8.84 vs 46.80 ± 9.26 ml/min/1.73 m 2 ; P < 0.001). No adverse clinical events were observed in this study. Conclusions This study demonstrates that EECP increases the contrast clearance and may have an effect in reducing the risk of CI-AKI. The study has been registered in Chinese Clinical Trial Registry (ChiCTR 2,000,039,190).
