High-quality cysteamine-coated CdTe quantum dots (CA-CdTe QDs) were successfully synthesized in aqueous phase by a facile one-pot method. Through hydroxylamine hydrochloride-promoted kinetic growth strategy, water-soluble CA-CdTe QDs could be obtained conveniently in a conical flask by a stepwise addition of raw materials. The photoluminescence quantum yield (PL QY) of the obtained QDs reached 9.2% at the emission peak of 520 nm. The optical property and the morphology of the QDs were characterized by UV–vis absorption spectra, photoluminescence spectra (PL) and transmission electron microscopy (TEM) respectively. Furthermore, the fluorescence of the resultant QDs was quenched by copper (II) (Cu 2+ ) and mercury (II) (Hg 2+ ) meanwhile. It is worthy of note that to separately detect Hg 2+ , cyanide ion could be used to eliminate the interference of Cu 2+ . Under the optimal conditions, the response was linearly proportional to the logarithm of Hg 2+ concentration over the range of 0.08–3.33 μM with a limit of detection (LOD) of 0.07 μM.

Cysteamine (CA) serves as a cystine-depleting agent employed in the management of cystinosis and a range of other medical conditions. Monitoring blood CA levels at the point of care is imperative due to the risk of toxicity associated with elevated CA dosages. An additional significant challenge is presented by the intricate composition of human plasma and the presence of various interfering biological thiols, which possess similar structures or properties. Here, this work proposes an AI-enhanced Lab-on-a-disc system, also termed AI-LOAD, for multiplexed point-of-care testing of cysteamine. The AI-LOAD system incorporates an online whole blood separation mechanism alongside a naked-eye colorimetric detection module, facilitating the rapid and precise visual identification of cysteamine. Remarkably, the system necessitates only 40 μL of whole blood to analyze eight samples within 3-min, achieving a limit of detection as low as 10 μM, which is lower than the physiological toxic concentration of 0.1 mM. By leveraging diverse colorimetric responses generated through interactions between gold nanoparticles of varying sizes and different biological thiols, combined with artificial intelligence methodologies, the system successfully accomplished specific recognition of various biological thiols with 100 % accuracy. The proposed AI-LOAD will drive advancements in centrifugal microfluidics for point-of-care testing, thereby holding potential for broader applications in future biomedical research and in vitro diagnosis.

Exploring new energy donor/acceptor pairs for constructing FRET system has important research significance. In this study, a FRET assembly by using carbon dots (CDs) as energy donor and gold nanorods (Au NRs) as energy acceptor has been proposed through covalent bond interactions by taking advantage of cysteamine as a bridge. The assembly is characterized by UV–vis absorption spectra, fluorescence spectra , FT-IR spectra and TEM images, revealing that Au NRs successfully absorbed on the surface of CDs. As for the occurrence of FRET process from CDs to Au NRs, the fluorescence signal of CDs-cysteamine-Au NRs assembly quenched significantly. Interestingly, Pb 2+ ions could bind completely with cysteamine and disturbed the FRET process, which activated fluorescence signal of the system. Based on these experimental phenomena, CDs-cysteamine-Au NRs assembly was performed as an off-on FRET biosensor for Pb 2+ ions. The linear range obtained is from 0 to 155 μM with the detection limit of 0.05 μM. Moreover, the FRET-based sensor exhibited high selectivity to the analyte Pb 2+ ions against other interference substances. Furthermore, the present approach was successfully applied to detect Pb 2+ ions in real samples, which suggested its potential applications in environmental monitoring.

Membrane fouling induces catastrophic loss of separation performance and seriously restricts the applications of reverse osmosis (RO) membranes. Inspired by the mussel structure, polydopamine (PDA) and cystamine molecules (CA) with excellent anti-fouling properties were used to prepare accessible, biocompatible, and redox-responsive coatings for RO membranes. The PDA/CA-coated RO membranes exhibit a superior water flux of 65 L m−2 h−1 with a favourable NaCl rejection exceeding 99%. The water permeability through the PDA/CA-coated membrane is much higher than that of most membranes with similar rejection rates. Due to the formed protective hydration layers by PDA/CA coatings, anti-fouling properties against proteins, polysaccharides and surfactants were evaluated separately, and ultralow fouling properties were demonstrated. Moreover, the disulfide linkages in CA molecules can cleave in a reducing environment, yielding the degradation of PDA/CA coatings, thereby removing the foulants deposited on the coatings. The degradation endows the coated membranes with satisfying longtime anti-fouling properties, where the flux recovery reaches up to 90%. The construction of redox-responsive smart coatings not only provided a promising route to alleviate membrane fouling but can also be upscaled for use in numerous practical applications like sensors, medical devices, and drug delivery.

Here we demonstrate a type of pH and redox dual-responsive micelles, which were self-assembled in aqueous solution by an amphiphilic polymer, methoxypoly(ethylene glycol)-cystamine-poly(L-glutamic acid)-imidazole (mPEG-SS-PGA-IM). Considering tumor cells or tissues exhibiting low pH values and high glutathione (GSH) concentration, mPEG-SS-PGA-IM micelles possessed the charge conversion at pH of tumor tissues, which can facilitate cellular uptake of tumor cells. Furthermore, mPEG-SS-PGA-IM micelles can escape from endo/lysosomes based on the proton sponge effect, following degraded by higher concentration of GSH in cytoplasm. CLSM images of HCT116 cells indicated that mPEG-SS-PGA-IM micelles can escape from endo/lysosomes and enter cytoplasm. MTT assay showed that (paclitaxel) PTX-loaded mPEG-SS-PGA-IM micelles had higher cytotoxicity against HCT116 cells compared with PTX-loaded mPEG-PBLG and mPEG-SS-PBLG micelles. These results indicated that these mPEG-SS-PGA-IM micelles, as novel and effective pH- and redox-responsive nanocarriers, have great potential to both improve drug targeting efficiency while also enhancing the antitumor efficacy of PTX.

An amphiphilic cationic graft polymer polyethylenimine-cystamine-poly(ε-caprolactone) (PSSP) with intracellular redox potential-responsive cleavable ability was synthesized for the first time by coupling poly(ε-caprolactone) (PCL) to polyethylenimine (PEI) via a bio-cleavable disulfide linkage. PSSP based micelles, which can degrade in the intracellular reducing environment, were developed as smart non-viral gene vectors for enhanced miR-34a delivery. The PSSP/miR-34a complexes showed effective and stable miRNA condensation with a narrow particle size of around 120 nm. The gel retardation assay showed that PSSP micelles could protect miRNA from RNase and achieve intracellular release of miRNA in the reducing environment. Compared with PEI-25k, PSSP revealed comparable transfection efficiencies in the B16F10 cell with less cytotoxicity. The B16F10 cell treated with PSSP/miR-34a complexes showed a significantly higher cell apoptosis than the PEI-25k/miR-34a complexes. These encouraging results indicated that PSSP may possess promising potential as a smart non-viral gene vector with low cytotoxicity, intracellular redox potential to trigger miRNA release and high transfection efficiency.

Porous graphene-based aerogels (GAs) hold great promise for water purification due to their fascinating properties. However, weak connections between the graphene oxide nanosheets cause instability and incompressibility in GAs during practical applications. Herein, the small organic molecule of cysteamine was selected as a covalent cross-linker to fabricate GAs through a facile one-step synthesis method. The porosity and mechanical stability of GAs were simultaneously improved due to strong covalent cross-linking between the graphene oxide nanosheets via cysteamine as the linkage. The resultant graphene/cysteamine aerogels (GCAs) were compressible with excellent structural stability under different acid, base, and organic solution conditions. The maximum oil absorption capacity of the GCAs reached as high as 251 g/g and did not obviously decrease even after regeneration by squeezing. In addition, the GCAs exhibited a high adsorption capacity for common dyes, such as methylene blue (207.8 mg/g) and methyl orange (70.2 mg/g). As expected, the as-prepared aerogels with high adsorption capacities, structural stability and excellent compressibility, have great potential as recyclable and durable materials for water purification.

An efficient Surface-enhanced Raman scattering (SERS) method for the detection of cysteamine hydrochloride (CSH) was developed by synthesizing a composite substrate comprising silver nanoparticles (AgNPs) functionalized with MoS 2 and β -cyclodextrin ( β -CD). The enhanced Raman signals of CSH by β -CD/MoS 2 /AgNPs substrate were the contribution of electromagnetic enhancement (EM) as well as chemical enhancement (CM), and the enhancement factor (EF) can reach up to 3.11 × 10 6 (peak at 633 cm −1 ). Various instrumental techniques were used to characterize the substrate, such as X-ray diffraction (XRD), thermogravimetric analysis (TGA), transmission electron microscopy (TEM), high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM) and ultraviolet visible (UV–vis). The binding of β -CD/MoS 2 /AgNPs and CSH was confirmed by UV–vis and Fourier transform infrared (FT-IR). The optimal experimental conditions were determined by single factor experiments as well as response surface model. The influences of different metal ions and analogous drugs on the detection of CSH were investigated. Under optimum conditions, a good linear correlation (R = 0.9997) was established for CSH in the range of 10.00–1000.00 nmol/L, and the limit of detection (LOD) was as low as 0.78 nmol/L (S/N = 3). The contents of CSH in meat samples were detected. The recovery was 96.6–103.1 %, and the relative standard deviation (RSD) of the measurement was 0.7–3.9 % ( n = 7).

The discrimination of cysteamine (CA) from mercapto amino acids (including glutathione, cysteine and homocysteine, GSH/Cys/Hcy) with high efficiency is essential to detect biothiols in a physiological environment. Herein, a nano-assembly was constructed by combining one-pot synthesized β-cyclodextrin-modified AuNPs and encapsulated rhodamine 6G (R6G-β-CD@AuNPs) without covalent modification. Interestingly, when the pH value of the tested solution was approximately the isoelectric point (pI) of the biothiol, the quenched fluorescence of R6G-β-CD@AuNPs was obviously restored by the introduction of the biothiol, which was due to the replacement of the R6G-β-CD composite by the biothiol on the surfaces of AuNPs via a stronger S–Au bond. The pI-mediated surface ligand exchange process was designed for the first time for the discrimination and detection of CA and GSH/Cys/Hcy at the pH values of 9.5 and 5.5, respectively. Under the optimized conditions, the enhanced fluorescence intensity displayed a good linear response to the concentration of each biothiol, with the detection limits of CA, GSH, Cys and Hcy calculated as 66.8, 417.6, 652.7 and 603.4 nM, respectively. The ingenious fabrication of the R6G-β-CD@AuNPs sensing platform eliminated the interferences from cholesterol by host–guest interactions and enabled high selectivity for the detection of biothiols in human serum samples. The pI-mediated sensing platform exhibits great potential for biothiol-related therapeutic drug monitoring.

Over the past decades, bacterial infections resulting from the misuse of antibiotics have garnered significant attention. Among the alternative antibacterial strategies, photodynamic therapy (PDT) has emerged as a promising non-antibiotic approach. However, persistent bacterial biofilms, particularly those composed of gram-negative bacteria with their protective outer membranes, have exhibited remarkable resilience to PDT. To address this challenge, we have developed an antibacterial composite material, silver nanoparticles coated-copper cysteamine (Cu-Cy-PEG@AgNPs), which demonstrates remarkable antibacterial activity against both gram-positive and gram-negative bacteria. Specifically, under UV irradiation, Cu-Cy-PEG@AgNPs achieves a sterilization efficiency of approximately 100% at a low concentration of 25 µg/mL. The incorporation of silver nanoparticles significantly enhances the antibacterial performance of Cu-Cy, effectively eradicating persistent bacterial biofilm infections. Moreover, Cu-Cy-PEG@AgNPs exhibits excellent biocompatibility with L929 cells, indicating its potential for use in relevant applications to combat bacterial infections.

In this study, cysteamine-functionalized molybdenum disulfide quantum dots (MoS 2 QDs) were synthesized by a one-pot hydrothermal method . A range of techniques including of Thioflavin T and 8-Anilino-1-naphthalenesulfonic acid fluorescence assays , circular dichroism , and transmission electron microscope have been employed to determination the efficacy of MoS 2 QDs on the inhibition/reversion of fibrillation and hindering cytotoxicity induced by protofibrils and amyloid fibrils of bovine serum albumin (BSA). Results demonstrated that MoS 2 QDs could effectively inhibit the fibrillogenesis and destabilize preformed fibrils of BSA in a concentration-dependent manner. Cytotoxicity protection and imagine on Hela cells was investigated using the methyl thiazolyl tetrazolium (MTT) assay. It was found that MoS 2 QDs not only has good biocompatibility, low toxicity and good cell penetration, but also could effectively decrease the cytotoxicity caused by the formed fibrils of BSA. The results obtained in this work suggested the potential biological application of MoS 2 QDs in therapeutics and provided new insight into the design of multifunctional nanomaterials for amyloid-related diseases.

To effectively cut off the transmission of Brucella to humans through milk, a novel biosensor for detection of Brucella was constructed using cysteamine functionalized nanogold (Cys-Au) and 4-Mercaptobenzoic acid (4-MBA) modified gold electrode as the platform. The morphology of the prepared nanomaterials was characterized by laser particle sizer, XPS and SEM, and the results showed the 4-MBA film was modified on the gold electrode surface, and the diameter of successfully prepared nanogold and Cys-Au are about 55 and 65 nm, respectively. The electrochemical performance of the biosensor was tested in Brucella antigen solution by square wave voltammetry (SWV). The results showed the reproducibility and stability of the biosensor were good, the spiked recovery in milk was in the range of 95.12–105.27%, and the minimum detection limit (S/N = 3) was 5.12 × 10 2 cfu/mL with a linear range from 1.6 × 10 2 to 1.6 × 10 8 cfu/mL. In addition, the minimum detection limit was reduced from 5.12 × 10 2 cfu/mL to 7.0 × 10 1 cfu/mL after processing the SWV data by second-order derivative transformation, which showed Second-order derivative transformation is very helpful to further optimize the performance of the sensor. Compared with the previously reported test methods, the prepared biosensors have a wider linear range and lower limit of detection (LOD). Therefore, the constructed biosensor can be used as a promising tool for the trace detection of Brucella in milk samples.
