Insight into nitrogen transformation during the binary NaOH-Na2CO3 molten salt thermal treatment of waste tires

Qiang Gao,Hongyun Hu,Chan Zou,Yang Hu,Huan Liu,Xian Li,Hong Yao

Molten salt thermal treatment of solid waste is a promising way for energy recovery and pollutant removal. However, the migration of nitrogen during pyrolysis of waste tires poses a challenge for cleaner production. This study investigated nitrogen conversion pathways during waste tires pyrolysis using a binary NaOH-Na 2 CO 3 salt at 425, 500, and 575 °C. The results demonstrated that amine-N and pyrrole-N were the main N-containing species in waste tires. Compared to conventional pyrolysis, the molten salt enhanced the deamination of the amine-N while increasing levels of NH 3 and aniline. Owing to the catalytic benefits of molten salt, the C-N bond cleavage of amine-N was initiated by electron transfer involving the combination of NH 2 -Phenyl structures with Na + . The hydrocracking of Phenyl-NH-Phenyl in amine-N was facilitated by the available H radicals (or H 2 ). Furthermore, molten salt obstructed the contact of C(N) sites on the char surface, causing the suppression of HCN production. The SH radicals might be consumed by OH – ions, which disabled the formation path of benzothiazole from aniline and SH radicals. Comparatively, pyrrole-N showed limited reactivity and remained in char. This work contributes to nitrogen regulation during the molten salt treatment of solid waste.

N-formylation of amines using phenylsilane and CO2 over ZnO catalyst under mild condition

Pengbo Wang,Qian He,Hao Zhang,Qingdi Sun,Yujie Cheng,Tao Gan,Xiaohui He,Hongbing Ji

Several research studies have been conducted on N -formylation of amines using phenylsilane and CO 2 . However, most of these studies involved tedious processes of catalyst preparation or complex procedures. In the present study, we describe the use of a simple and commercially available ZnO catalyst for selective N -formylation of amines under mild condition. High-yielding N -formylation products with good recyclability and wide substrate scope were obtained, which can promote fine chemical synthesis and CO 2 capture.

Construction of electro-neutral surface on dialysis membrane for improved toxin clearance and anti-coagulation/inflammation through saltwater fish inspired trimethylamine N-oxide (TMAO)

Yang Liu,Guiliang Li,Qiu Han,Haibo Lin,Qiang Li,Gang Deng,Fu Liu

Dialysis membranes capable of preventing coagulation and inflammation and achieving high toxin clearances are highly desired for clinical hemodialysis. Here, we constructed an electro-neutral and hydrophilic surface on Polysulfone (PSf) membrane, which both improved the hemocompatibility and toxin clearance. The membrane surface was modified with trimethylamine N-oxide (TMAO) through Michael addition reaction and subsequent crosslinking with glycidyl ether oxypropyl trimethoxysilane (KH560). Saltwater fish inspired zwitterionic monomer TMAO without spacers between positive and negative headgroups is thought to generate a hydrated and net-charged layer that prevents non-specific plasma proteins and platelets adsorption and activation. Coagulation time, serine protease Factor XIIa, bradykinin (BK), kallikrein (KLK) and anaphylatoxins (C3, C5 and SC5b-9) were examined to explore blood compatibility. All results showed that the highly hydrated and electro-neutral surface could efficiently alleviate the activation of the coagulation cascade, kallikrein-kinin system (KKS) and complement system simultaneously. Furthermore, the modified PSf membrane is favorable to toxins clearance (e.g., urea, creatine, lysozyme and vitamin B12) during simulated dialysis process. It is manifested that the TMAO based surface modification is feasible for improving the hemocompatibility and toxin clearance of dialysis membranes.

Trimethylamine oxide promotes myocardial fibrosis through activating JAK2-STAT3 pathway

Xiaoyun Yang,Yi Wang,Yujin Feng,Feng Wang,Lijuan Gao,Yueheng Wang

Myocardial infarction is often accompanied by symptoms of myocardial fibrosis, causing myocardial systolic and diastolic dysfunction. High levels of TMAO are considered to be strongly connected to cardiovascular diseases occurrence. The objective of this investigation was to determine whether formal TMAO affects the development of cardiac muscle fibers. The left anterior descending (LAD) coronary artery was ligated to construct myocardial infarction mouse model, while the control group mouse was only operated vessel separation. According to whether or not TMAO intervention is given, mice were divided into control, TMAO, myocardial infarction model(model), and myocardial infarction model + TMAO(model + TMAO). TMAO and model + TMAO group received 0.24 % TMAO in drinking water for one month. After modeling, echocardiography assessed cardiac function, and cardiac tissues were collected for analysis. Masson staining observed collagen deposition, while immunohistochemistry was conducted to identify fibronectin, collagen III, collagen I, and JAK/STAT pathway related proteins levels. In vitro, mouse cardiac fibroblasts were cultured and treated with varying TMAO concentrations (0, 300, 600, 900 μM), and Western blot was carried out to analyse changes in fibronectin, collagen III, and collagen I. Subsequently, the JAK2/STAT3 pathway inhibitor AG490 was utilized to treat cells and further examine the changes of fibronectin and collagen. The in vivo data showed TMAO significantly increases fibrosis and upregulates fibronectin, collagen III, and collagen I. In vitro experiments indicated that TMAO promotes fibronectin, collagen III, and collagen I levels through regulating the JAK2/STAT3 pathway, ultimately accelerating fibrosis.

Surface hydration of trimethylamine N–oxide/perovskites hybrid aerogel facilitates anti–fouling solar desalination and seawater electrocatalysis

Hao Zhang,Xueling Xu,Guangyao Zhang,Jie Yang,Yi Lu

Developing active and stable electrocatalysts for oxygen evolution reaction is of great significance to generate oxygen and hydrogen by seawater electrolysis, which however remains some issues that high energy consumption and chloride corrosion. Here we pioneered a polymer/inorganic hybrid engineering strategy for achieving excellent salt resistance solar evaporation and oxygen evolution performance (OER). The La 0.9 Sr 0.1 CoO 3 /nickel foam–cellulosic aerogel (LSC1/NF–CA) was synthesized by engineering Trimethylamine N–oxide (TMAO) polymer brush onto the surface of hybrid aerogels composed of La 0.9 Sr 0.1 CoO 3 /Ni foam (LSC1/NF) and cellulose/polyvinyl alcohol, which has good hydrophilic features, intrinsic solar capture capacity, and stable OER performance in seawater. Benefiting from the synergy of solar desalination with surface hydration effect of TMAO, the LSC1/NF–CA presents a superior catalytic activity , requiring a lower overpotential of 341 mV and a tafel slope of 58.2 mV dec −1 at 10 mA cm −2 under 6.0  sun illumination . Our results establish a new strategy that can guide the design of stable and efficient polymer/inorganic hybrid catalysts, which shows a promising prospect as alternatives to noble metal–based catalysts for seawater splitting.

Protocatechuic acid alleviates TMAO-aggravated atherosclerosis via mitigating inflammation, regulating lipid metabolism, and reshaping gut microbiota

Huafang Ding,Jianhui Liu,Zixing Chen,Shouhe Huang,Chi Yan,Erika Kwek,Zouyan He,Hanyue Zhu,Zhen-Yu Chen

Trimethylamine-N-oxide (TMAO) is a risk factor for atherosclerosis. As a natural phenolic acid, protocatechuic acid (PCA) is abundant in various plant foods. The present study was to investigate the effect of PCA on TMAO-aggravated atherosclerosis in ApoE-/- mice. The mice were randomly divided into five groups and fed one of the following five diets for 12 weeks: namely a low-fat diet (LFD), a western diet (WD), a WD+0.2% TMAO diet (WDT), a WDT+0.5% PCA diet (WDT+LPCA), and a WDT+1.0% PCA diet (WDT+HPCA). Results demonstrated that dietary TMAO exacerbated the development of atherosclerosis by eliciting inflammation and disturbing lipid metabolism. PCA at 1% diet reduced TMAO-induced aortic plaque by 30% and decreased the levels of plasma proinflammatory cytokines. PCA also improved the lipid metabolism by up-regulating hepatic gene expression of peroxisome proliferator-activated receptor alpha (PPARα). In addition, PCA supplementation enhanced fecal excretion of fatty acids and decreased hepatic fat accumulation. PCA supplementation favorably modulated gut microbiota by increasing α-diversity with an increase in the abundance of beneficial genera (Rikenella, Turicibacter, Clostridium_sensu_stricto and Bifidobacterium) and a decrease in the abundance of adverse Helicobacter genus. In summary, PCA could alleviate the TMAO-exacerbated atherosclerosis and inflammation, improve the lipid metabolism, and modulate gut microbiota.

Protective effect of vitexin against high fat-induced vascular endothelial inflammation through inhibiting trimethylamine N-oxide-mediated RNA m6A modification

wenwen Li,Zeyuan Deng,Shuang Xiao,qian Du,Mengru Zhang,Hailing Song,Caidong Zhao,Liufeng Zheng

The high-fat diet (HFD) is a major risk factor for cardiovascular disease. However, the specific effects of HFD on vascular inflammation and the protective role of vitexin, a bioactive small-molecule compound derived from food, require further research. This study investigated the protective effects of vitexin intervention against HFD-induced vascular inflammation and its underlying mechanism. The results demonstrated that vitexin intervention significantly reduced body weight, serum total cholesterol, and low-density lipoprotein cholesterol levels in HFD-fed mice. Vitexin also improved vascular pathological changes and inflammatory status in the mice. Furthermore, vitexin intervention reduced serum TMAO levels in HFD-fed mice and achieved this effect by altering gut microbiota composition. HFD significantly increased N6-methyladenosine (m6A) levels in aorta tissues, while vitexin intervention reversed this abnormal m6A level. Vitexin could directly bind to fat mass and obesity-associated protein (FTO), potentially promoting m6A demethylation. The dose-response relationship between TMAO and inflammation/m6A was further validated in HUVEC cell and in vivo mouse experiments. Specifically, TMAO increased m6A levels and inflammation, while vitexin inhibited TMAO-mediated m6A modification, exhibiting anti-inflammatory effects. In conclusion, this study demonstrates the protective role of vitexin against HFD-induced vascular inflammation by inhibiting TMAO-mediated RNA m6A modification and laying the foundation for the development of functional foods.

A Polysiloxane Delivery Vehicle of Cyclic N-Halamine for Biocidal Coating of Cellulose in Supercritical CO2

Leixuan Li,Yan Xin,Fengze Wu,Xiangrong Lyu,Qiyuan Yao,Xiaoting Yin,Qiang Zhang,Wenjuan Shan,Yong Chen,Qiuxia Han

CyclicN-halamines are highly antimicrobial, very stable, and not susceptible to bacterial resistance. A polysiloxane delivery vehicle was synthesized to deliver cyclic imideN-halamine onto cellulose via a benign and universal procedure that does not require a harmful solvent or chemical bonding. In brief, Knoevenagel condensation between barbituric acid and 4-hydroxybenzaldehyde furnished 5-(4-hydroxybenzylidene)pyrimidine-2,4,6-trione, whose phenolic O−H was subsequently reacted with the Si−H of poly(methylhydrosiloxane) (PMHS) via silane alcoholysis. The product of silane alcoholysis was interpenetrated into cellulose in supercritical CO2(scCO2) at 50 °C, to form a continuous modification layer. The thickness of the modification layer positively correlated with interpenetration pressure in the experimental range of 10 to 28 MPa and reached a maximum value of 76.5 nm, which demonstrates the ability for tunable delivery, to control the loading of the imide N−H bond originating from barbituric acid unit. The imide N−H bonds on cellulose with the thickest modifier were then chlorinated into N−Cl counterparts usingtert-butyl hypochlorite, to exert a powerful biocidability, providing ~7 log reductions of bothS. aureusandE. coliin 20 min. The stability and rechargeability of the biocidability were both very promising, suggesting that the polysiloxane modifier has a satisfactory chemical structure and interlocks firmly with cellulose via scCO2interpenetration.Keywords:cyclic imideN-halamines;polysiloxane;delivery vehicle;antibacterial surface;cellulose;scCO2interpenetration

Gut microbiota dependent trimethylamine N-oxide aggravates angiotensin II–induced hypertension

Shan Jiang,Yongjie Shui,Yu Cui,Chun Tang,Xiaohua Wang,Xingyu Qiu,Weipeng Hu,Lingyan Fei,Yun Li,Suping Zhang,Liang Zhao,Nan Xu,Fang Dong,Xiaoqiu Ren,Ruisheng Liu,Pontus B. Persson,Andreas Patzak,En Yin Lai,Qichun Wei,Zhihua Zheng

Gut microbiota produce Trimethylamine N-oxide (TMAO) by metabolizing dietary phosphatidylcholine, choline, l -carnitine and betaine. TMAO is implicated in the pathogenesis of chronic kidney disease (CKD), diabetes, obesity and atherosclerosis. We test, whether TMAO augments angiotensin II (Ang II)-induced vasoconstriction and hence promotes Ang II-induced hypertension. Plasma TMAO levels were indeed elevated in hypertensive patients, thus the potential pathways by which TMAO mediates these effects were explored. Ang II (400 ng/kg −1 min −1 ) was chronically infused for 14 days via osmotic minipumps in C57Bl/6 mice. TMAO (1%) or antibiotics were given via drinking water. Vasoconstriction of renal afferent arterioles and mesenteric arteries were assessed by microperfusion and wire myograph, respectively. In Ang II-induced hypertensive mice, TMAO elevated systolic blood pressure and caused vasoconstriction, which was alleviated by antibiotics. TMAO enhanced the Ang II-induced acute pressor responses (12.2 ± 1.9 versus 20.6 ± 1.4 mmHg; P < 0.05) and vasoconstriction (32.3 ± 2.6 versus 55.9 ± 7.0%, P < 0.001). Ang II-induced intracellular Ca 2+ release in afferent arterioles (147 ± 7 versus 234 ± 26%; P < 0.001 ) and mouse vascular smooth muscle cells (VSMC, 123 ± 3 versus 157 ± 9%; P < 0.001 ) increased by TMAO treatment. Preincubation of VSMC with TMAO activated the PERK/ROS/CaMKII/PLCβ3 pathway. Pharmacological inhibition of PERK, ROS, CaMKII and PLCβ3 impaired the effect of TMAO on Ca 2+ release. Thus, TMAO facilitates Ang II-induced vasoconstriction, thereby promoting Ang II-induced hypertension, which involves the PERK/ROS/CaMKII/PLCβ3 axis.

A diet high in sugar and fat influences neurotransmitter metabolism and then affects brain function by altering the gut microbiota

Guo Yinrui,Zhu Xiangxiang,Zeng Miao,Qi Longkai,Tang Xiaocui,Wang Dongdong,Zhang Mei,Xie Yizhen,Li Hongye,Yang Xin,Chen Diling

Gut microbiota (GM) metabolites can modulate the physiology of the host brain through the gut–brain axis. We wished to discover connections between the GM, neurotransmitters, and brain function using direct and indirect methods. A diet with increased amounts of sugar and fat (high-sugar and high-fat (HSHF) diet) was employed to disturb the host GM. Then, we monitored the effect on pathology, neurotransmitter metabolism, transcription, and brain circularRNAs (circRNAs) profiles in mice. Administration of a HSHF diet-induced dysbacteriosis, damaged the intestinal tract, changed the neurotransmitter metabolism in the intestine and brain, and then caused changes in brain function and circRNA profiles. The GM byproduct trimethylamine-n-oxide could degrade some circRNAs. The basal level of the GM decided the conversion rate of choline to trimethylamine-n-oxide. A change in the abundance of a single bacterial strain could influence neurotransmitter secretion. These findings suggest that a new link between metabolism, brain circRNAs, and GM. Our data could enlarge the “microbiome–transcriptome” linkage library and provide more information on the gut–brain axis. Hence, our findings could provide more information on the interplay between the gut and brain to aid the identification of potential therapeutic markers and mechanistic solutions to complex problems encountered in studies of pathology, toxicology, diet, and nutrition development.

Trimethylamine N-Oxide–Derived Zwitterion Coating for Polyurethane Ureteral Stents Prevents Encrustation Formation

Kaiguo Xia,Guang Chen,Bingbing Hou,Zhe Wang,Yaqi Zhu,Yuexian Xu,Shanfu Zhang,Qiang Xuan,Yezi You,Zongyao Hao

A ureteral stent with strong resistance to proteins, bacteria, and multivalent ions is crucial for the safe treatment of urologic diseases. Generally, the proteins, bacteria, and multivalent ions present in urine tend to bind to the stent surface, leading to aggregation, nucleation, and subsequent stent encrustation. Stent encrustation can induce or exacerbate urinary tract infections and obstructions, thereby seriously harming kidney function. Although hydrophilic coatings on ureteral stents can reduce the binding of proteins, bacteria, and multivalent ions, encrustation still occurs. To date, preventing stent encrustation formation remains a significant challenge. Here, we grafted dense trimethylamine oxide (TMAO)-derived zwitterionic polymers onto the stent surface via a branched amplification strategy. These zwitterions can strongly bind water molecules, forming a stable hydration shell that repels proteins, bacteria, and multivalent ions from adhering to the surface of the polyurethane ureteral stent, thus rendering the stent anti-encrustation. The results showed that the TMAO-derived zwitterion-coated stents exhibited a significantly reduced encrustation weight (13.8% of the original polyurethane stent) and demonstrated good safety. This approach offers a promising method for enhancing stent encrustation resistance. Statement of Significance This study successfully developed a TMAO-derived zwitterionic coating on the surface of a polyurethane stent, creating a superhydrophilic surface with a minimal contact angle of 5.2 o . This surface effectively shields the stent from interactions with proteins, bacteria, and multivalent ions in urine, demonstrating favorable anti-protein adsorption and antibacterial adhesion properties. The superhydrophilic surface formed by the TMAO-derived zwitterionic coating on the stents (PTMAO-s) provides strong anti-fouling resistance and enhanced anti-encrustation properties. Under identical conditions, the encrustation resistance of PTMAO-s is approximately 7.2-fold greater than that of original polyurethane stents (PU), 3.6-fold greater than Bard commercial stents, and 2.1-fold greater than betaine-coated stents (PSBG-s).

Trimethylamine N-oxide Induced Chronic Kidney Injury by Triggering PANoptosis

D Shao,L Bai,Q Chen,Y Chen,Z Qiu,Y Liu,S Jin,Y Wu,J Dai

Trimethylamine N-oxide (TMAO) is involved in the development of kidney disease. However, the specific mechanism by which it leads to kidney injury is unclear. This study explored the role of regulated cell death in TMAO-induced kidney injury. We constructed a TMAO-induced chronic kidney injury model by intraperitoneal injection of TMAO (100 micromol/kg/day for three months). Plasma creatinine (Cre) and urea nitrogen (BUN) levels were measured to evaluate kidney function. Masson staining was used to evaluate kidney pathological changes. The expression levels of regulated cell death-related proteins were measured using western blotting. Plasma Cre and BUN, the area of kidney fibrosis in the TMAO group significantly increased. The western blotting results showed cleaved-Caspase-8, Caspase-8, Caspase-1, NOD-like receptor protein 3 (NLRP3), interleukin-1beta (IL-1beta), cleaved-gasdermin D (cleaved-GSDMD), Z-DNA binding protein 1 (ZBP1), phosphorylation of receptor-interacting protein kinase 3 (RIP3) and mixed-lineage kinase domain-like pseudokinase (MLKL) significantly elevated in the TMAO group. The transferrin receptor 1 (TFR1), ferritin heavy chain (FTH), ferroportin (FPN), nuclear factor erythroid 2-related factor 2 (NRF2), and glutathione peroxidase 4 (GPX4) protein expression in kidney tissue of the TMAO group significantly up-regulated. However, there was no change in iron and MDA levels. The results suggested that PANoptosis, including pyroptosis, apoptosis, and necroptosis components, might be involved in TMAO-induced chronic kidney injury.
