Background In eudicots, germination begins with water uptake by the quiescent dry seed and is greatly related to the permeability of micropyle enriched callose layers. Once imbibition starts, seeds undergo a cascade of physiological, biochemical, and molecular events to initiate cellular activities. However, the effects of callose on water uptake and following seed metabolic events during germination are largely unknown. Cotton ( Gossypium hirsutum ) is a eudicot plant with natural fiber and edible oil production for humans. Here, we addressed this question by examining the role of GhGLU19 , a gene encoding β-1,3-glucanase, in cotton seed germination. Results GhGLU19 belongs to subfamily B and was expressed predominately in imbibed seeds and early seedlings. Compared to wild type, GhGLU19 -suppressing and GhGLU19 -overexpressing transgenic cotton lines showed the higher and lower seed germination percentage, respectively. Callose was enriched more at inner integument (ii) than that in embryo and seed coat in cotton seeds. In GhGLU19 -suppressing lines, callose at ii of cotton seeds was greatly increased and brought about a prolonged water uptake process during imbibition. Both proteomic and transcriptomic analysis revealed that contrary to GhGLU19 -overexpressing lines, the glycolysis and pyruvate metabolism was decreased, and abscisic acid (ABA) biosynthesis related genes were downregulated in imbibed seeds of GhGLU19 -suppressing lines. Also, endogenous ABA was significantly decreased in GhGLU19 -suppressing line while increased in GhGLU19 -overexpressing line. Conclusions Our results demonstrate that suppression of GhGLU19 improves cotton seed germination via accumulating callose of inner integument, modulating glycolysis and pyruvate metabolism, and decreasing ABA biosynthesis. This study provides a potential way for improving germination percentage in cotton seed production, and other eudicot crops.

Cotton aphids ( Aphis gossypii Glover) are important pests of cucurbit crops. Plant defenses at different cell layers, including the leaf surface, mesophyll cells, and phloem, are employed to defend aphids. Here, we assessed differences in aphid resistance among six watermelon varieties and elucidated the defense mechanisms underlying aphid-resistant/susceptible watermelon varieties. The population abundance, offspring number per female, and meantime of phloem-feeding (E2 phase) of aphids were the highest on XiNong (XN), followed by JingXin (JX), TianWang (TW), ZaoJia (ZJ), and MeiFuLai (MFL), and these parameters were the lowest on JinMeiDu (JMD). Further analyses showed that there was no correlation between the aphid resistance of six watermelon varieties and defenses at the leaf surface. For defenses at mesophyll cells, aphid infestation increased salicylic acid (SA) content at 48 h post-infestation (hpi) and ROS accumulation at 6 and 12 hpi in six watermelon varieties. For phloem defenses, aphid infestation increased callose content in JMD plants but decreased callose content in JX, TW, ZJ, MFL, and XN plants at 6, 12, 24, and 48 hpi. Moreover, callose deposition suppressed by 2-deoxy-d-glucose (2-DDG) neutralized the resistance of JMD plants and exacerbated the susceptibility of XN plants to aphids, which exhibited a higher population abundance and E2 phase time. Collectively, phloem defenses regulated by aphid-induced callose deposition were responsible for differences in aphid resistance among the watermelon varieties.

Paeonia lactiflora is an important garden ornamental flower and has become a new cut flower in the international market in recent years. Stem mechanical strength is an important index of cut flower quality. P. lactiflora cultivars possess varying stem mechanical strength, which directly result in different cut P. lactiflora quality. In our previous study, a significant difference in phloem callose deposition was detected between two P. lactiflora cultivars with varying stem mechanical strength. However, the relationship and interaction mechanism between the phloem callose and mechanical strength in P. lactiflora stems are still unclear. This study aimed to analyze the effects of reduced phloem callose on the mechanical strength in P. lactiflora stems. The results revealed a negative correlation between the callose content and mechanical strength in P. lactiflora stems during early developmental stages. Exogenous 2DDG treatment led to a 13.6% reduction in the callose content, concomitantly enhancing the stem mechanical strength by 14.2% via facilitating the formation and secondary cell wall (SCW) thickening of xylem fiber cells. Further investigations showed that exogenous 2DDG treatment promoted carbon transport in P. lactiflora stems, thereby increasing the carbon content. Additionally, it induced alterations in the metabolism of lipids, amino acids, and organic acids in the stems and spurred the diffusion of lipids from the pith to the xylem. Consequently, reducing the content of callose improved the formation and SCW thickening of xylem fiber cells by modifying material transport and metabolism, thereby enhancing the stem mechanical strength of P. lactiflora . This study offers novel perspectives regarding the regulatory mechanism of P. lactiflora stem mechanical strength.

The root rot mainly caused by Fusarium solani is a bottleneck in the cultivation of Panax notoginseng . In this study, we reported a gene encoding a plant cell wall structural protein, P. notoginseng proline-rich protein (PnPRPL1), whose transcription was upregulated by F. solani and induced by some hormone signals. The PnPRPL1 recombinant protein significantly inhibited the growth and conidial germination of the root rot pathogens. Downregulation of PnPRPL1 by RNA interference (RNAi) in P. notoginseng leaves increased the susceptibility to F. solani , whereas overexpression of PnPRPL1 in tobacco ( Nicotiana tabacum ) enhanced the resistance to F. solani . Compared with wild-type tobacco, the PnPRPL1 -overexpressing transgenic tobacco had higher reactive oxygen species (ROS)-scavenging enzyme activities, lower ROS levels, and more lignin and callose deposition. The opposite results were obtained for the P. notoginseng expressing PnPRPL1 RNAi fragments. Furthermore, the PnPRPL1 promoter transcription activity was induced by several plant hormones and multiple stress stimuli. In addition, the transcription factor PnWRKY27 activated the expression of PnPRPL1 by directly binding to the promoter region. Thus, PnPRPL1 , which is positively regulated by a WRKY transcription factor, encodes an antimicrobial protein that also mediates ROS homoeostasis and callose/lignin deposition during the response to F. solani infection.

Alpha-1,3-glucan is often synthesized on the surface of pathogenic filamentous fungi cell walls to block pathogen-associated molecular patterns (PAMPs) generation by host plant enzymes and the subsequent immune system response of the plant. Here, Botrytis cinerea susceptibility was assessed in tomato fruit to determine whether the fruit could recognize this camouflage and mount an immune response to it. The results showed that local mechanical wounds treated with dextran and laminarin, except amylopectin, could locally and then systemically activate disease resistance against B. cinerea infection in tomato fruit. Dextran treatment effectively elicited fruit callose deposition and phenylpropanoid and flavonoid biosynthesis to a greater extent than α-glucanase activity relative to the mock group surface wounds. Enzymatic hydrolysis of this polysaccharide provided some help in improving host disease resistance. Taken together, these results demonstrate that tomato fruit can perceive α-1,3-glucan as a kind of PAMPs but have limited ability to degrade it.
