Different kinds of proteins interact with the digestible lipids in various ways, affecting the adsorption behavior of proteins and digestion. The ordered porous layer interferometry (OPLI) system was constructed by the silica colloidal crystal (SCC) films used to monitor the real-time binding assessment between bovine serum albumin (BSA), casein, fibrinogen, and triolein. The OPLI system reflected the changes in protein mass on the SCC films in real time through the migration of the interference spectrum of the SCC films, which was converted into the changes in optical thickness (ΔOT) that can be monitored. OPLI systematically studied the degradation of proteins by trypsin and found that the lipid layer enhanced the hydrolysis of trypsin to BSA and fibrinogen but weakened the degradation of trypsin to casein. The high OT of the lipid layer (10.45 nm) indicated that trypsin was more likely to adhere to the casein surface on the lipid layer than to hydrolyze casein. In contrast, the reduced OT (−6.40 nm) on the SCC films indicated that trypsin was more inclined to hydrolyze casein than to attach to casein. The OPLI system provided the technical basis for real-time tracking of protein hydrolysis behavior in complex food systems.

A method for real-time monitoring of lipid digestion based on photonic crystals formed from silica was developed. As an effective “net”, the highly ordered silica colloidal crystal (SCC) film provides structural support for lipid payload. This method based on optical interferometric film kinetics was used to record the whole kinetics progress of olive oil hydrolysis by lipase in real time and calculate the kinetic Michaelis constant. The kinetic parameters were compared with the results determined by the titration method. The effects of bile salt content on lipase and olive oil layer were studied. This method provides a potential evaluation system for real-time digestion and degradation of edible oil in the food field. It also provides a basis for further real-time evaluation of lipid bioavailability in food systems by real-time recording the release and degradation of lipids in the food nano-matrix.

As a functional ingredient, dietary fibers can be neither degraded by digestive enzymes in the human gastrointestinal tract nor absorbed by individuals. However, they play a significant role in controlling healthy lipid digestion and bioavailability. The real-time impact of dietary fibers on lipid digestion cannot be tracked in situ. The optical interference lipid layer was prepared by loading triolein into an ordered porous layer composed of a silica colloidal crystal (SCC) array. Ordered porous layer interferometry (OPLI) system was constructed for real-time tracking the adsorption of water-soluble dietary fibers such as pectin and polydextrose, insoluble dietary fiber (ethylcellulose, EC), and modified dietary fiber (nanocrystalline cellulose , CNC) at the aqueous/lipid interface, respectively. The process of mass change of the lipid layer caused by the lipase degradation of triolein was transformed into real-time monitoring optical thickness change (ΔOT). The effect of dietary fibers as a functional component on the release and degradation of the lipid layer was analyzed by studying digestion rate, digestion time, and real-time OT curve. Different molecular structures, solubility, and chemically modified dietary fibers had different retardation on lipid digestion. The real-time investigation of the reaction at the interface deepens the understanding of the interaction between lipids and dietary fibers in complex food matrices. Meanwhile, the method provides a basis for designing functional foods with specific health benefits.

Helicobacter pylori ( H. pylori) infection remains the leading cause of gastric adenocarcinoma, and its eradication primarily relies on the prolonged and intensive use of two antibiotics. However, antibiotic resistance has become a compelling health issue, leading to H. pylori eradication treatment failure worldwide. Additionally, the powerlessness of antibiotics against biofilms, as well as intracellular H. pylori and the long-term damage of antibiotics to the intestinal microbiota, have also created an urgent demand for antibiotic-free approaches. Herein, we describe an antibiotic-free, multifunctional copper-organic framework (HKUST-1) platform encased in a lipid layer comprising phosphatidic acid (PA), rhamnolipid (RHL), and cholesterol (CHOL), enveloped in chitosan (CS), and loaded in an ascorbyl palmitate (AP) hydrogel: AP@CS@Lip@HKUST-1. This platform targets inflammatory sites where H. pylori aggregates through electrostatic attraction. Then, hydrolysis by matrix metalloproteinases (MMPs) releases CS-encased nanoparticles, disrupting bacterial urease activity and membrane integrity. Additionally, RHL disperses biofilms, while PA promotes lysosomal acidification and activates host autophagy, enabling clearance of intracellular H. pylori . Furthermore, AP@CS@Lip@HKUST-1 alleviates inflammation and enhances mucosal repair through delayed Cu 2+ release while preserving the intestinal microbiota. Collectively, this platform presents an advanced therapeutic strategy for eradicating persistent H. pylori infection without inducing drug resistance.
