Coupling process study of lipid production and mercury bioremediation by biomimetic mineralized microalgae

Yang Peng,Aosong Deng,Xun Gong,Xiaomin Li,Yang Zhang

Considering the high concentration of mercury in industrial wastewater, such as coal-fired power plants and gold mining wastewater, this research study investigated the coupling process of lipid production and mercury bioremediation using microalgae cells. Chlorella vulgaris modified by biomimetic mineralization. The cultivation was divided in two stages: a natural cultivation for 7   days and 5   days of Hg 2+ addition (10–100   μg/L) for cultivation at different pH values (4–7) after inoculation. Next, the harvested cells were eluted, and lipid was extracted. The fluorescein diacetate (FDA) dye tests demonstrated that the mineralized layer enhanced the biological activity of microalgae cells in Hg 2+ contaminated media. Hg distribution tests showed that the Hg removal capacity of modified cells was increased from 62.85% to 94.74%, and 88.72% of eluted Hg 2+ concentration was observed in modified cells compared to 48.42% of raw cells, implying that more mercury was transferred from lipid and residuals into elutable forms.

Effects of two common antibiotics on the growth and lipid production of Tetradesmus obliquus cultivated in the effluent of municipal wastewater treatment plant

Du Shuhao,Dou Xiang,Meng Fanping,Peng Xiaoling

Using the effluent of municipal wastewater treatment plant (WWTP) to cultivate lipid-producing microalgae is likely to reduce the production cost of algae-derived biodiesel. However, the toxicity of antibiotics present in the effluent may affect the growth of microalgae and lipid production. This study investigated the effects of erythromycin (ERY) and tetracycline (TC) on the growth and lipid production of Tetradesmus ( Scenedesmus ) obliquus cultivated in the effluent of municipal WWTP. The results indicated that ERY was more toxic to T. obliquus compared to TC. The critical concentrations of ERY resulting in S. obliquus algae growth inhibition and increased lipid accumulation were 100 and 10 μg L −1 , respectively. Maximum lipid accumulation was observed at an ERY concentration of 50 μg L −1 , while lipid accumulation was significantly decreased at ERY concentrations greater than 250 μg L −1 . Interestingly, the contents of polyunsaturated fatty acids in T. obliquus exposed to ERY were significantly increased, while the contents of total saturated fatty acids and monounsaturated fatty acids were significantly decreased. TC had no effect on the growth or lipid production when the concentration reached 500 μg L −1 . Overall, the presence of ERY and TC in the municipal WWTP effluent does not largely affect the growth or lipid production of T. obliquus .

Microbial lipid production by oleaginous yeast in D-xylose solution using a two-stage culture mode

Jintao Lin,Shuangyue Li,Mingzhong Sun,Cuili Zhang,Wenbo Yang,Zehui Zhang,Xiaojie Li,Shao Li

The economical production of biodiesel from lignocellulose hydrolysates requires a robust D-xylose fermentation path. The present work found that Lipomyces starkeyi AS 2.1560 and Rhodosporidium toruloides AS 2.1389 could produce micro-lipid effectively from D-xylose solution by a two-stage culture mode. Results demonstrated that both L. starkeyi and R. toruloides could achieve high lipid production in xylose solution. High cell density fermentation was carried out by a feed-batch fermentation mode. After fermentation for 48 h in a bioreactor using unsterile xylose solution by L. starkeyi, the biomass, lipid and lipid content reached 97.4 g L−1, 63.9 g L−1, and 65.5%, respectively. Accordingly, the lipid productivity reached 1.3 g L−1 h−1 in the second stage. And the overall lipid productivity was 0.55 g L−1 h−1. Therefore, L. starkeyi and R. toruloides are highly effective D-xylose-fermenting oleaginous yeasts for lipid production in xylose solution. The two-stage culture method should provide a new opportunity to make the industrial production of biodiesel possible in the future.

Use of elephant grass (Pennisetum purpureum) acid hydrolysate for microbial oil production by Trichosporon cutaneum

Xue-Fang Chen,Chao Huang,Lian Xiong,Bo Wang,Gao-Xiang Qi,Xiao-Qing Lin,Can Wang,Xin-De Chen

Elephant grass (Pennisetum purpureum) dilute acid hydrolysate contains 34.6 g/L total sugars. The potential of lipid production by oleaginous yeast Trichosporon cutaneum grown on elephant grass acid hydrolysate was investigated for the first time. During the fermentation process on the elephant grass acid hydrolysate, glucose, xylose, and arabinose could be well utilized as carbon sources by T. cutaneum. Interestingly, xylose was almost no use before glucose was consumed completely. This illustrated that simultaneous saccharification of xylose and glucose by T. cutaneum did not occur on elephant grass acid hydrolysate. The highest biomass, lipid content, lipid yield, and lipid coefficient of T. cutaneum were measured after the sixth day of fermentation and were 22.76 g/L, 24.0%, 5.46 g/L, and 16.1%, respectively. Therefore, elephant grass is a promising raw material for microbial oil production by T. cutaneum.

Engineering the Oleaginous Yeast Rhodosporidium toruloides for Improved Resistance Against Inhibitors in Biomass Hydrolysates

Lyu Liting,Chu Yadong,Zhang Sufang,Zhang Yue,Huang Qitian,Wang Shuang,Zhao Zongbao K.

Conversion of lignocellulosic biomass into lipids and related chemicals has attracted much attention in the past two decades, and the oleaginous yeast Rhodosporidium toruloides has been widely used in this area. While R. toruloides species naturally have physiological advantages in terms of substrate utilization, lipid accumulation, and inhibitor resistance, reduced lipid production and cell growth are noticed when biomass hydrolysates are used as feedstocks. To improve the robustness of R. toruloides, here, we devised engineered strains by overexpressing genes responsible for phenolic compound degradation. Specifically, gene expression cassettes of the manganese peroxidase gene (MNP) and versatile peroxidase gene (VP) were constructed and integrated into the genome of R. toruloides NP11. A series of engineered strains were evaluated for lipid production in the presence of typical phenolic inhibitors. The results showed that R. toruloides strains with proper expression of MNP or VP indeed grew faster in the presence of vanillin and 5-hydroxymethylfurfural than the parental strain. When cultivated in concentrated mode biomass hydrolysates, the strain VP18 had improved performance as the cell mass and lipid content increased by 30% and 25%, respectively. This study provides more robust oleaginous yeast strains for microbial lipid production from lignocellulosic biomass, and similar efforts may be used to devise more advanced lipid producers.

Co-cultivation of microalga and xylanolytic bacterium by a continuous two-step strategy to enhance algal lipid production

Zhangzhang Xie,Weitie Lin,Jianfei Luo

To enhance microalgal lipid production, canonical two-step cultivation strategy that by transferring the microalgal cells grown in nutrient-replete medium to nutrient-depleted medium is widely used. However, the harvesting step during the transfer raises the production cost. To avoid the harvesting step, this study developed a continuous two-step (CTS) cultivation strategy. In the strategy, Chlorella sacchrarophila was grown in bioreactor while a xylanolytic bacterium Cellvibrio pealriver grown in an inner bag that embedded in the bioreactor; after the first-step co-cultivation, the inner bag is removed which then start the second-step cultivation of C. sacchrarophila . Based on the strategy, the lipid production was determined as 825.34–929.79 mg·L −1 , which were 1.7–1.9 times higher than that of cultivation in canonical two-step strategy using glucose as feedstock. During the CTS strategy, the co-cultivation using xylan as feedstock promotes the microalgal growth and the removal of inner bag produces nutrient-depleted condition for enhancing microalgal lipid production.

Effects of light intensity on oxygen distribution, lipid production and biological community of algal-bacterial granules in photo-sequencing batch reactors

Fansheng Meng,Limeng Xi,Dongfang Liu,Weiwei Huang,Zhongfang Lei,Zhenya Zhang,Wenli Huang

The effects of light intensity (0–225 µmol m −2  s −1 ) on oxygen distribution, lipid production and biological community structure of algal-bacterial granules were investigated in six identical photo-sequencing batch reactors (with a dark/light cycle of 12 h/12 h). Typically green algal-bacterial granules could be developed at a light intensity of ≥135 µmol m −2  s −1 . The lipid content was significantly increased under higher light intensity, while the percentage of saturated fatty acid methyl esters was remarkably decreased. Results showed that light intensity ≥90 µmol m −2  s −1 yielded enough O 2 production from algae, creating aerobic/anoxic zone (0.3–0.6 mg-O 2 /L) in the core of granules and thus efficient algal-bacterial symbiosis system. Enhanced nitrogen and phosphorus removals were achieved in the reactors with stronger light illumination, probably attributable to the enrichment of ammonia oxidizing bacteria (Comamonadaceae and Nitrosomonadaceae) and algae ( Navicula and Stigeoclonium ). Illuminance ≥180 µmol m −2  s −1 was found to be unfavorable for Nitrospiraceae.

Assessment of the effects of nutrients on biomass and lipid accumulation in Dunaliella tertiolecta using a response surface methodology

Xiao-Ying Qv,Yan-Yun Guo,Jian-Guo Jiang

Bio-energy is arousing the common interest of worldwide researchers to investigate it as a substitute for fossil resources. Compared to other bio-energy sources, algae energy has many advantages. This research was aimed to increase the lipid productivity of Dunaliella tertiolecta, an oleaginous microalgae, by optimizing algal culture medium. Plackett–Burman Design (PBD) was adopted to screen out the critical factors from seven nutritional elements. Box–Behnken design (BBD) was used to seek the optimal nutritional conditions for achieving maximum lipid productivity. Results showed that four nutrients, i.e. NaNO3, NaCl, NaHCO3 and NaH2PO4 2H2O, played a critical role in the accumulation of lipid. GC analysis verified that the four critical nutrients mentioned above had different influences on fatty acid methyl esters, moreover, NaCl had the most prominent effects on fatty acid composition of D. tertiolecta. NaCl 87.69 g L−1, NaNO3 1.77 g L−1, NaH2PO4·2H2O 1.92 g L−1, NaHCO3 5.57 g L−1 were proven to be the optimal culture conditions, under which the lipid productivity in the experiment was 12.576 ± 0.12 mg per L per day, which is approaching the theoretically predicted lipid productivity of 12.925 mg per L per day. The lipid productivity was increased by 31.4% under the optimal culture conditions compared to that under the initial medium composition, indicating that optimization of nutrients could effectively improve the lipid production of D. tertiolecta.

Contaminant removal performance and lipid productivity of a cyanobacteria-bacteria consortium containing exogenous phytohormones during the treatment of antibiotic-polluted wastewater

Youshuai Fang,Guannan Lin,Ying Liu,Jian Zhang

In this study, a cyanobacteria-bacteria consortium containing native wastewater bacteria and immobilized Synechococcus sp. was constructed. The cyanobacterial cellular responses (including growth, biomass and lipid productivity) and contaminant removal ability (for TN, TP, COD and antibiotics) in the consortium were evaluated during the advanced treatment of wastewater containing 10–50 μg/L of mixed antibiotics (amoxicillin, tetracycline , erythromycin , sulfadiazine and ciprofloxacin) with the addition of a certain phytohormone (indole-3-acetic acid, gibberellin A3 or 6-benzylaminopurine) at trace level within a period of four days. Each phytohormone promoted the growth of Synechococcus sp. and increased the tolerance of Synechococcus sp. to mixed antibiotics. Indole-3-acetic acid coupled to moderate antibiotic stress could elevate lipid productivity and lipid content of Synechococcus sp. to 33.50 mg/L/day and 43.75%, respectively. Phytohormones increased the pollutant removal performance of the cyanobacteria-bacteria consortium through the stimulation of cyanobacterial growth and the regulation of cyanobacteria-bacteria interaction, which increased the abundances of microalgae-associated bacteria including Flavobacterium , Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium , Bosea , Sphingomonas and Emticicia . Up to 80.83%, 98.06%, 83.26%, 99.84%, 99.50%, 89.41%, 65.61% and 60.65% of TN, TP, COD, amoxicillin , tetracycline , erythromycin , sulfadiazine and ciprofloxacin were removed by the consortium with the addition of phytohormones. In general, indole-3-acetic acid was the optimal phytohormone for enhancing lipid production and contaminant removal performance of the cyanobacteria-bacteria consortium.

Effect of filamentous algae in a microalgal-bacterial granular sludge system treating saline wastewater: Assessing stability, lipid production and nutrients removal

Jinhua Cao,Fanzhen Chen,Zheng Fang,Yue Gu,Hao Wang,Jingfang Lu,Yanmeng Bi,Shaopo Wang,Wenli Huang,Fansheng Meng

In this study modified microalgal-bacterial granular sludge (MBGS) was constructed and employed to compare the performance for treating 1%-5% saline wastewater with aerobic granular sludge (AGS). Filamentous algae were found to flourish at 1% salinity when nutrients were temporarily restricted to low level (COD 0, N 10 mg/L, P 0.5 mg/L). A significant improvement of granule stability was detected as the integrity coefficients of MBGS was only 0.12–0.24 rather than 0.19–0.48 of AGS under 1%-5% salinities, which reduced the risk of particle disintegration. Filamentous algae including Leptolyngbya and Geitlerinema occupied 91.2% of identified algae, and were beneficial for enhancing the biomass content and lipid production to about 1.27–1.37, 3.1–5.0 times than AGS. The MBGS had best nitrogen and phosphorus removal efficiencies of 93.4% and 64.6% at 1% salinity, and showed higher resistance to 3%-5% salinities. This study could provide meaningful information for using this modified MBGS technology in practice.

A combined light regime and carbon supply regulation strategy for microalgae-based sugar industry wastewater treatment and low-carbon biofuel production to realise a circular economy

Xiang Wang,Zi-Hao Qin,Ting-Bin Hao,Guang-Bin Ye,Jin-Hua Mou,Srinivasan Balamurugan,Xiao-Yun Bin,Joseph Buhagiar,Hong-Mei Wang,Carol Sze Ki Lin,Wei-Dong Yang,Hong-Ye Li

The replacement of fossil fuels with clean and renewable biofuels is of both research and market interest for realising a circular economy. However, microalgae-based biofuels have shown promise as alternative low-carbon biofuels to other crop-based biofuels, some key obstacles in their production remain to be addressed, such as high costs and low lipid productivity. In this study, a Chlorella sp. CSH4 was cultivated using a combined light regime and carbon supply regulation strategy to enhance sugar industrial wastewater bioremediation, biomass accumulation and lipid production. Blue light irradiance of 200 μmol photons m −2 s −1 together with 10 g/L glucose and 9.2 g/L glycerol supply was found to effectively enhance the biomass accumulation and pollutant-removal capacity of Chlorella sp. during the growth phase and its lipid production during the stationary phase. Furthermore, the biodiesel properties of the lipid retrieved from Chlorella sp., as demonstrated by its fatty acid profile, were found to be suitable for commercial application. Possible mechanisms were explored to explain how this combined strategy caused this microalga to exhibit highly efficient biomass and lipid production together with efficient pollutant removal. Moreover, upscaled semi-continuous treatment using both sugar industry wastewater and negligible carbon sources (e.g., food waste hydrolysate and crude glycerol) with a mass balance analysis was conducted to initially validate the feasibility of applying our combined strategy for microalgae-based wastewater treatment. In sum, this study demonstrated the feasibility of cultivating a microalga using a combined strategy comprising a light regime and carbon supply regulation to achieve both wastewater treatment and low-carbon biofuel production.

Improvement on lipid production by Scenedesmus obliquus triggered by low dose exposure to nanoparticles

He Meilin,Yan Yongquan,Pei Feng,Wu Mingzhu,Gebreluel Temesgen,Zou Shanmei,Wang Changhai

Carbon nanotubes (CNTs), α-Fe 2 O 3 nanoparticles (nano Fe 2 O 3 ) and MgO nanoparticles (nano MgO) were evaluated for the effects on algae growth and lipid production. Nano Fe 2 O 3 promoted cell growth in the range of 0–20 mg·L −1 . CNTs, nano Fe 2 O 3 and nano MgO inhibited cell growth of Scenedesmus obliquus at 10, 40 and 0.8 mg·L −1 respectively. Neutral lipid and total lipid content increased with the increasing concentration of all tested nanoparticles. The maximum lipid productivity of cultures exposed to CNTs, nano Fe 2 O 3 and nano MgO was observed at 5 mg·L −1 , 5 mg·L −1 and 40 mg·L −1 , with the improvement by 8.9%, 39.6% and 18.5%. High dose exposure to nanoparticles limited increase in lipid productivity, possibly due to the repression on cell growth caused by nanoparticles-catalyzed reactive oxygen species (ROS) generation, finally leading to reduction in biomass and lipid production. Reduced accumulation of fatty acids of C18:3n3, C18:3n6 and C20:2 was observed in cells exposed to nanoparticles.
