Preparation and biological properties of BCZT/TiO2 electrokinetic conversion coating on titanium surface in vitro for dental implants

Cong Wu,Chun Zhang,Xianyi Yan,Junhua Cheng,Lei Chen,Yufei Tang,Kang Zhao

Titanium implants have the risk of loosening or even failure, especially the force generated by chewing is not conducive to the use of conventional titanium bioactive coatings in the oral environment. In this work, a BCZT/TiO 2 bio-piezoelectric coating was prepared on titanium surface. The force generated by chewing is cleverly converted into the piezoelectric charge on the coating surface , and the formation of the combination of implant and alveolar bone is accelerated by its promoting effect on bone repair. The formation mechanism of the coating was studied. The BCZT/TiO 2 bio-piezoelectric coating showed good piezoelectric properties . The calcium deposition results of BCZT/TiO 2 coating with static , physiological loading and low-frequency ultrasonic conditions were compared. Interestingly, the effect of polarized BCZT/TiO 2 coating applied with low-intensity pulsed ultrasound on promoting calcium deposition was similar to that applied with physiological loading, suggesting that low-intensity pulsed ultrasound has the potential to stimulate piezoelectric effects. In addition, the BCZT/TiO 2 coating was non-toxic and has good biocompatibility and bioactivity. The polarized BCZT/TiO 2 coating showed the best proliferation results, and the osteoblast adhesion morphology on the coating surface was extended. The results provide data support for the application of titanium bio-piezoelectric coating on dental implants .

Effect of ultrasonic waveforms on gas–liquid mass transfer in microreactors

Qiang Zhang,Zhengya Dong,Zhikai Liu,Guangwen Chen

This study aims to investigate the effect of ultrasonic waveforms on the gas–liquid mass transfer process. For a given load power ( P ), continuous rectangular wave yielded stronger bubble oscillation and higher mass transfer coefficient ( k L a ) than continuous triangular and sinusoidal wave. For pulsed ultrasound, the k L a decreased monotonically with decreasing duty ratio ( D ), resulting in weak enhancement at low D (≤33%). For a given average load power ( P A ), concentrating the P for a shorter period resulted in a higher k L a due to stronger cavitation behavior. For a given P A and D , decreasing the pulse period ( T ) led to an increase in k L a , which reached a constant high level when the T fell below a critical value. By optimizing the D and T , a k L a equivalent to 92% of that under continuous ultrasound was obtained under pulsed ultrasound at a D of 67%, saving 33% in power consumption.

Periosteum-bone inspired hierarchical scaffold with endogenous piezoelectricity for neuro-vascularized bone regeneration

Yao Zhao,Yunfan Cai,Wenkai Wang,Yongkang Bai,Mingyi Liu,Yan Wang,Wen Niu,Zhixiao Luo,Lingyun Xia,Juanfang Zhu,Fei Zhao,Franklin R. Tay,Lina Niu

The development of scaffolds for repairing critical-sized bone defects heavily relies on establishing a neuro-vascularized network for proper penetration of nerves and blood vessels. Despite significant advancements in using artificial bone-like scaffolds infused with various agents, challenges remain. Natural bone tissue consists of a porous bone matrix surrounded by a neuro-vascularized periosteum, with unique piezoelectric properties essential for bone growth. Drawing inspiration from this assembly, we developed a periosteum-bone-mimicking bilayer scaffold with piezoelectric properties for regeneration of critical-sized bone defects. The periosteum-like layer of this scaffold features a double network hydrogel composed of chelated alginate, gelatin methacrylate, and sintered whitlockite nanoparticles, emulating the viscoelastic and piezoelectric properties of the natural periosteum. The bone-like layer is composed of a porous structure of chitosan and bioactive hydroxyapatite created through a biomineralization process. Unlike conventional bone-like scaffolds, this bioinspired bilayer scaffold significantly enhances osteogenesis, angiogenesis, and neurogenesis combined with low-intensity pulsed ultrasound-assisted piezoelectric stimulation. Such a scheme enhances neuro-vascularized bone regeneration in vivo . The results suggest that the bilayer scaffold could serve as an effective self-powered electrical stimulator to expedite bone regeneration under dynamic physical stimulation.

Low-intensity pulsed ultrasound inhibits chondrocyte senescence by inhibiting PI3K/AKT/mTOR signaling

Han Wang,Guan Mengtong,Liao Bo,Han Xiaoyu,Li Kaiting,Chen Qing,Guo Xiya,Niu Yajuan,Zhu Ying,Bai Dingqun

Cellular senescence is an important cause of age-related degenerative diseases, including osteoarthritis (OA). Chondrocyte senescence is crucial in OA onset and progression. As a non-invasive, safe, and widely used physical rehabilitation factor, the effect and mechanism of low intensity pulsed ultrasound (LIPUS) on chondrocyte senescence remain unclear. This study evaluated the inhibitory effect of LIPUS on OA chondrocyte senescence in vitro and in vivo. The effect of LIPUS on chondrocyte senescence was examined by RT-qPCR, enzyme-linked immunosorbent assay (ELISA), and western blotting. Changes in levels of reactive oxygen species (ROS) and γ-h2ax foci in senescent chondrocytes were detected using fluorescent staining. Chondrocyte senescence was evaluated by senescence-associated β-galactosidase (SA-β-gal) staining. The PI3K inhibitor LY294002 and the PI3K agonist 740Y-P were used to investigate whether PI3K/AKT/mTOR signalling was involved in the effect of LIPUS in senescent chondrocytes. Chondrocyte senescence and cartilage degeneration were analyzed in a destabilization of the medial meniscal (DMM) mouse model by immunohistochemistry, hematoxylin and eosin staining, and safranin-O/fast green staining. LIPUS inhibited the expression of the senescence-associated secretory phenotype (SASP) factors CCL4 and CCL2 and the senescence phenotype in doxorubicin-treated chondrocytes by inhibiting the PI3K/AKT/mTOR pathway. LIPUS alleviated chondrocyte senescence and attenuated OA progression in the DMM mice. These results demonstrated a novel role for LIPUS in inhibiting chondrocyte senescence and the SASP by modulating PI3K/AKT/mTOR signalling. Our findings expanded the clinical application of LIPUS and provide a new, non-invasive, and safe treatment approach to prevent and treat age-related degenerative joint disorders.

Fabrication and characterisation of α-chitin nanofibers and highly transparent chitin films by pulsed ultrasonication

Yun Lu,Qingfeng Sun,Xilin She,Yanzhi Xia,Yixing Liu,Jian Li,Dongjiang Yang

α-Chitin nanofibers were fabricated with dried shrimp shells via a simple high-intensity ultrasonic treatment under neutral conditions (60   KHz, 300   W, pH   =   7). The diameter of the obtained chitin nanofibers could be controlled within 20–200   nm by simply adjusting the ultrasonication time. The pulsed ultrasound disassembled natural chitin into high-aspect-ratio nanofibers with a uniform width (19.4   nm after 30   min sonication). The EDS, FTIR, and XRD characterisation results verified that α-chitin crystalline structure and molecular structure were maintained after the chemical purification and ultrasonic treatments. Interestingly, ultrasonication can slightly increase the degree of crystallinity of chitin (from 60.1 to 65.8). Furthermore, highly transparent chitin films (the transmittance was 90.2% at a 600   nm) and flexible ultralight chitin foams were prepared from chitin nanofiber hydrogels.

LIPUS Responsive Dopamine-Modified PVDF Piezoelectric Nanofiber Membrane for Full-Thickness Skin Wound Healing

Zhiying Zhang,Lijun Liu,Huan Wang,Wangni Xie,Wenhao Zhai,Linlin Wen,Boya Zhang,Kexuan Liu,Xue Zhang,Shuchen Liu,Lei Huang,Daowei Li,Yanmin Zhou

Objective Large full-thickness skin wounds pose significant challenges, particularly in achieving scar-free healing and the regeneration of skin appendages. This study introduces a portable approach for promoting scarless healing and skin appendage regeneration by utilizing low-intensity pulsed ultrasound (LIPUS) activated piezoelectric electrospun membranes to modulate the local electrical environment. Methods Dopamine-modified polyvinylidene fluoride (DA/PVDF) nanomembranes were fabricated via electrospinning, followed by piezoelectric characterization under varying LIPUS stimulation. Cell adhesion was examined using SEM and laser confocal microscopy to assess surface interactions. Cell proliferation and migration were further analyzed using the CCK-8 assay and Transwell migration assay, respectively. Finally, the effects of DA/PVDF membranes on full-thickness skin defect healing were tested in a mouse model. The healing process was documented with photographs, and functional skin regeneration was evaluated through histological analysis. Results The DA/PVDF nanomembranes had an average diameter of 732 ± 232 nm and generated a voltage of 450 mV under LIPUS stimulation, a 1.28-fold increase compared to PVDF membranes alone. In vitro, LIPUS-activated membranes enhanced cell adhesion and proliferation, resulting in a 1.14-fold increase in cell growth over three days. The transwell migration assays showed 244.67 ± 7.85 migrated cells. In vivo, the DA/PVDF+LIPUS group exhibited significantly higher wound healing rates, with improved epidermal regeneration, collagen fiber deposition and remodeling, and enhanced blood vessel and skin appendage formation. Conclusion DA modification enhances the piezoelectric properties of PVDF membranes, boosting cell adhesion and promoting dermal and vascular regeneration. LIPUS-generated mechanical waves effectively stimulate membrane deformation, producing a localized electrical microenvironment that mimics the natural bioelectric field of skin and accelerates functional wound healing.

Low-intensity pulsed ultrasound enhances delivery of 30 nm Q10 for improving mental and memory disorder in APP/PS1 mice

Luo Qi,Ye Xuanjie,Xu Jinan,Sun Zihui,Wang Panpan,Chen Haishu,Gao Tingting,Deng Qiangfeng,Wang Ziwen,Wang Qin,Chen Xiaoxiao,Zhao Zhuowen,Wu Yiqing,Yang Chuang,Lyv Wanjia,Lyv Xingzhou,Li Ye,Zhao Hang,Jiang Ke,Gu Ziqi,Lin Jing,Sun Yu,Tan Tao,Xu Haiyun,Tong Zhiqian

Patients with Alzheimer's disease (AD) often experience mental and memory disorders with poor outcomes. Coenzyme Q10 can degrade formaldehyde (FA) and improve Alzheimer-related symptoms, but its ability to cross the blood–brain barrier (BBB) is limited. This study investigated whether low-intensity pulsed ultrasound (LIPUS) enhances 30 nm Q10 delivery and improve symptoms in AD model mice. Here, 30 nm Q10 was prepared by encapsulating Q10 in liposomes coupled with PEG, creating PEG-Q10@NPs under 30 nm in diameter. Wild-type mice and APPswe/PS1dE9 mice (a familial AD model) received 30 nm Q10 via intraperitoneal injection, or a combination of 30 nm Q10 and LIPUS (50 or 100 100 mW/cm 2 ). Then the mice’s anxiety-like and depression-like behaviors and biochemical index were evaluated. We found that the combination therapy of LIPUS at 100 mW/cm 2 and 30 nm Q10 was more effective in ameliorating psychosis in AD mice than individual treatments with 30 nm Q10. This effectiveness was linked to higher levels of brain Q10, serotonin (5-HT), and dopamine (DA), along with lower levels of FA and plaques. Especially, excessive FA directly inactivated 5-HT and DA in vitro. The enhanced cellular uptake of Q10 and improved BBB permeability facilitated by LIPUS were confirmed in both cultured cells and wild-type mice. Unexpectedly, LIPUS at the different intensity only partially alleviated anxiety and depression symptoms and memory deficits in AD mice. Hence, this combination therapy of LIPUS and 30 nm Q10 is an innovative strategy for ameliorating mental and cognitive disorders in AD. Graphic

3D bioprinted piezoelectric hydrogel synergized with LIPUS to promote bone regeneration

Meng Li,Xiantong Hu,Xiaofei Liu,Lingzhou Zhao,Wanmin Zhao,Ying Li,Xiaomei Bie,Hua Jiao,Yantao Zhao,Chufan Ma

Bone defects resulting from trauma, tumor resection, non-union of fractures, and infections present enormous challenges in treatment. Although three-dimensional (3D) bioprinting plays an important role in repairing bone tissues, the lack of mechanical properties and osteoinductive ability of the bioinks remains a barrier for the application of the technology. In this study, we used advanced 3D bioprinting technology to create a novel piezoelectric hydrogel scaffold (Gel/PBT@BMSCs) which consisted of bone marrow-derived mesenchymal stem cells (BMSCs), gelatin methacryloyl (GelMA), and polyethylene glycol (PEG)-modified barium titanate (BT) nanoparticles. The piezoelectric hydrogel scaffold provided a stable 3D microenvironment for cell growth and adhesion, enhancing cell viability and osteogenic activity when subjected to low-intensity pulsed ultrasound (LIPUS) stimulation. Furthermore, in vivo experiments demonstrated that the innovative hydrogel scaffold significantly accelerated the healing process of bone defects and exhibited impressive bone regeneration capabilities. These findings highlight the potential of piezoelectric hydrogel for further research and application in the field of bone tissue engineering, and offer new approaches for the treatment of bone defects.

The Therapeutic Potential of Low-Intensity Pulsed Ultrasound in Enhancing Gallbladder Function and Reducing Inflammation in Cholesterol Gallstone Disease

Fang Chen,Run Guo,Tian Chen,Liping Liu,Fan Ding,Gang Zhao,Bo Zhang

Background: Cholesterol gallstone disease (CGS) is often accompanied by gallbladder contraction dysfunction and chronic inflammation, but effective therapeutic options remain limited. This study investigates whether a low-intensity pulsed ultrasound (LIPUS) treatment can improve gallbladder motility and alleviate chronic inflammation while exploring the underlying mechanisms. Methods: Gallbladder motility was assessed through in vitro and in vivo contraction tests, while bile condition was evaluated by observing bile crystal clearance. Tissue analysis and Western blotting were performed to examine the expression of the cholecystokinin A receptor (CCKAR) and α-smooth muscle actin (α-SMA) as markers of gallbladder smooth muscle health and the inflammatory microenvironment. Blood cholesterol levels were measured via biochemical assays. Results: LIPUS treatment obviously enhanced gallbladder contractility in response to CCK-8 stimulation and accelerated bile crystal clearance. It also reduced inflammatory cell infiltration and tissue edema, and promoted new capillary formation in the gallbladder, mitigating the progression of CGS. Furthermore, LIPUS restored CCKAR expression and improved the thickness of the gallbladder smooth muscle layer, providing a structural basis for increased smooth muscle contractility. Conclusion: LIPUS improves gallbladder motility and reduces chronic inflammation in CGS by enhancing CCKAR expression and smooth muscle integrity. These findings highlight the potential of LIPUS as a non-invasive therapeutic approach for managing CGS.

The electrical properties and in vitro osteogenic properties of 3D-printed MgO@BT/HA piezoelectric ceramic disk

Kai Chen,Zhenyao Qin,Mingjun Zhang,Lin Wang,Shikang Zheng,Yingtong Wang,Chuang Chen,Haoyu Tang,Yuhao Zhong,Hongxuan Yang,Guomin Wu

The repair of bone defects from various causes remains a significant challenge in clinical settings. Piezoelectric materials have been demonstrated to promote osteogenic differentiation, among which barium titanate (BT) has been widely reported. Magnesium oxide (MgO) dopants not only enhance the piezoelectric properties but also stimulate the osteogenic and angiogenic properties in stem cells. In this study, a MgO-modified BT/hydroxyapatite (MgO@BT/HA) piezoelectric ceramic scaffold was constructed using three-dimensional (3D)-printed technology. The results indicated that the addition of MgO improved the electrical properties of BT ( d 33 = 68.26 pC/N), and facilitated an increase in the proportion of HA (30%) while maintaining the piezoelectric coefficient ( d 33 = 2.04 pC/N) of the ceramic system. Furthermore, under the induction of low-intensity pulsed ultrasound (LIPUS) stimulation, in vitro biosafety experiments confirmed that MgO@BT/HA exhibits excellent biosafety and promotes the osteogenic differentiation of dental pulp stem cells (DPSCs).

Enhancing function, stability, and intracellular uptake of kaempferol in zein-pectin nanoemulsions by a novel dual-frequency pulsed ultrasound system

Wenjuan Qu,Xinyue Deng,Yuhan Li,Cunshan Zhou,Haile Ma

In order to enhance the stability and bioavailability of fat-soluble kaempferol (Kae), the zein-pectin nanoemulsions were synthesized by a novel dual-frequency pulsed ultrasound (DFPU) technology to improve the loading effect of Kae. The effects of ultrasonic operating parameters on the nanoemulsions were examined, along with the influence of DFPU treatment on the structure, stability, digestion, and intracellular uptake of the nanoemulsions in this study. The results indicated that both emulsion encapsulation and DFPU treatment significantly enhanced the protection and delivery of Kae. Compared to the unsonicated emulsion, the emulsifying activity index, stability index, Kae embedding rate, and hydroxyl radical scavenging rate increased by 133.81%, 4.18%, 19.20%, and 19.73%, respectively, while the particle size decreased by 94.43% after DFPU treatment. DFPU treatment notably improved the stability of the nanoemulsions during long-term storage, high temperature, and salt concentration conditions, as well as the retention of Kae by 15.59% in intestinal digestives. Furthermore, DFPU treatment significantly enhanced the Caco-2 intracellular absorption rate, transport rate and bioavailability of Kae by 7.67%, 9.96% and 14.67%, respectively at a dosage of 0.6 mg/mL, which was attributed to the significant downregulation of mRNA expression levels of tight junction protein Occludin and efflux proteins MDR1 and BCRP by 21.27%, 51.05%, and 62.26%, respectively. This downregulation enhanced the intracellular transport capacity of Kae while reducing its exocytosis, thereby improving cellular utilization. The mechanism of improving the intracellular bioavailability of kaempferol was elucidated. This study offers a promising nanoemulsion for the efficient oral delivery of fat-soluble active ingredients, fully facilitating its antioxidant activity.

Low-intensity pulsed ultrasound alleviating myelosuppression of Sprague-Dawley rats after combined treating by paclitaxel and carboplatin

Dong Luo,Wenzhi Chen,Wei Wang,Junlin Chen,Haopeng Xu,Jinyun Chen,Yan Wang

Background Our team has previously reported that low intensity pulsed ultrasound (LIPUS) can alleviate myelosuppression in rats induced by single chemotherapy drugs. But in clinics, chemotherapy is often performed with multiple drugs simultaneously. To be closer to the clinical status quo, this experiment was designed to show whether it was the same effect of LIPUS on myelosuppression caused by combination therapy of chemotherapy drugs. Methods The rat model of myelosuppression was established by continuous injection of paclitaxel and carboplatin for 4 days. These myelosuppressive rats were randomly divided into LIPUS group (n=40) and control group (n=40). The LIPUS group was given continuous LIPUS irradiation for 7 days, while the control group was given sham irradiation (no energy output). The evaluation of blood cells counts, Hematoxylin-Eosin staining (H&E staining), scanning electron microscopy, enzyme-linked immunosorbent assay (ELISA) and real-time quantitative PCR (qPCR) was then performed. Results The results showed in the LIPUS group the blood cells count, hematopoietic tissue of bone marrow, the colonies formed from adhering of bone marrow stromal cells, levels of hematopoietic regulators and adhesion molecules all increased (LIPUS group vs. control group, P<0.05). Conclusions The results indicated that LIPUS can relieve myelosuppression induced by combined treatment of paclitaxel and carboplatin. The mechanisms may be LIPUS can increase the levels of hematopoietic regulators and adhesion molecules.
