Brain natriuretic peptide (BNP) is considered a reliable marker of heart failure disease, and its timely detection can provide important pathological information to prevent or treat heart failure. In this article, an electrochemiluminescence (ECL) sensing device based on a boron carbon nitride/gold nanoparticle (BCN/AuNPs) complex is developed to determine BNP. Prominently, the CRISPR/CAS 13a enzyme reverse cleavage mode, the entropy-driven and hammer hybridization chain reaction processes were involved in the entire detection scheme. Ultimately, with multiple reaction methods and amplification reactions of nucleic acids, this ECL sensing device is able to achieve a detection limit as low as of 0.03 pg/mL and linear range from 0.1 pg/mL to 30 ng/mL for BNP. In addition, the ECL sensing device based on BCN/AuNPs complex obtained satisfactory stability and specificity, and can also be extended to the detection of other pathological markers.

The construction of a sensitive electrochemiluminescence (ECL) immunosensor for the detection of N-terminal pro-brain natriuretic peptide (NT-proBNP) is demonstrated in this paper using terbium metal-organic gel (Tb-MOG) as an ECL emitter. Firstly, stable ECL signal was obtained using the easily synthesized Tb-MOG as the emitter and persulfate (S 2 O 8 2− ) as the co-reactant. Secondly, the reduction of S 2 O 8 2− was catalyzed by the introduction of CoMnO 3 nanoflower as a co-reaction promoter, which further enhanced the ECL intensity. When mixed-valence transition metal ions (Co 3+ /Co 2+ and Mn 4+ /Mn 3+ ) were reversibly transformed in CoMnO 3 , the production of sulfate radical anion (SO 4 •– ) was greatly enhanced. Additionally, the unique nanoflower structure of CoMnO 3 possesses a large specific surface area, enhancing its catalytic performance. Thanks to the well-established sensing strategy, the resultant ECL immunosensor exhibited satisfactory performance in accurately detecting NT-proBNP, with a linear concentration range of 10 fg/mL to 100 ng/mL and a detection limit of 2.1 fg/mL, offering great promise for clinical translation and broader heart failure diagnosis.

A triple recognition voltammetric method for the determination of brain natriuretic peptide (BNP) is described. Gold nanoparticles (AuNPs) and magnetic nanoparticles (MagNPs), sized 26 and 310 nm, respectively, were synthesized and characterized by transmission electron microscopy (TEM), FT-IR, dynamic light scattering (DLS), and Z-potential measurements. Antibody-modified MagNPs and methylene blue–labeled aptamer (Apt-MB)–modified AuNPs were used as an identifier, a signal reporter, and an amplifier, respectively. In the presence of BNP, the magnetic gold nanocomposite is formed through cascade conjugation via specific interaction. It then hybridized with complementary DNA (cDNA) on the interface, thereby amplifying the current signal of Apt-MB and increasing the selectivity of the immunoassay. Results obtained demonstrate the development of a highly selective method with a detection limit of 0.56 pg mL −1 and a linear response over the concentration range 1–10,000 pg mL −1 . The standard deviation of the method is < 6% while the recovery ranged from 92.2 to 104.2%.

Early diagnosis of chronic heart failure (CHF) relies on the ultra-sensitive detection assay of its main biomarker, Brain natriuretic peptide (BNP). To meet such demand, we designed a near-infrared immunoassay biosensor, utilizing a fiber-embedded microfluidic chip for surface-enhanced Raman scattering (SERS) detection. A SERS probe, consisting of metal–organic framework and Au-HEPES coupling nanoparticles (AuHPs), was used to enhance the Raman signal of the toluidine blue (TB) dye. The probe anchored with TB is used as a SERS tag, which is linked to BNP through sandwich method. To enhance the Raman signal further, we embedded a SERS substrate at the bottom of the microfluidic channel, which serves an enhancer when the SERS tagged BNP passes through the microfluidic chip. The amplified Raman signal was collected by an optical fiber probe of a simple and portable Raman detection system with an all-fiber connected configuration. Such design resulted in the lowest detection limit of pg/mL, indicating that the proposed platform can be used for rapid and highly sensitive detection of BNP. In addition, our platform also has the advantages of low sample consumption and high throughput rate, providing an effective scheme for the early diagnosis of CHF.

Doxorubicin (DOX) is a commonly used anthracycline in cancer chemotherapy. The clinical application of DOX is constrained by its cardiotoxicity. Myricetin (MYR) is a natural flavonoid widely present in many plants with antioxidant and anti-inflammatory properties. However, MYR's beneficial effects and mechanisms in alleviating DOX-induced cardiotoxicity (DIC) remain unknown. C57BL/6 mice were injected with 15 mg/kg of DOX to establish the DIC, and MYR solutions were administrated by gavage to investigate its cardioprotective potentials. Histopathological analysis, physiological indicators assessment, transcriptomics analysis, and RT-qPCR were used to elucidate the potential mechanism of MYR in DIC treatment. MYR reduced cardiac injury produced by DOX, decreased levels of cTnI, AST, LDH, and BNP, and improved myocardial injury and fibrosis. MYR effectively prevented DOX-induced oxidative stress, such as lowered MDA levels and elevated SOD, CAT, and GSH activities. MYR effectively suppressed NLRP3 and ASC gene expression levels to inhibit pyroptosis while regulating Caspase1 and Bax levels to reduce cardiac cell apoptosis. According to the transcriptomic analysis, glucose and fatty acid metabolism were associated with differential gene expression. KEGG pathway analysis revealed differential gene enrichment in PPAR and AMPK pathways, among others. Following validation, MYR was found to alleviate DIC by regulating glycolipid metabolism and AMPK pathway-related genes. Our findings demonstrated that MYR could mitigate DIC by regulating the processes of oxidative stress, apoptosis, and pyroptosis. MYR is critical in improving DOX-induced myocardial energy metabolism abnormalities mediated by the AMPK signaling pathway. In conclusion, MYR holds promise as a therapeutic strategy for DIC.

Introduction: Black ginseng (BG) was processed by “steaming and drying” (generally nine times) repeatedly to produce “rare saponins” and secondary ginsenosides. Both ginseng (GS) and red ginseng (RG) were commonly used in treating heart failure (HF), and the latter was confirmed to be more potent, implying the presence of rare ginsenosides that contribute positively to the treatment of heart failure. Previous research indicated that rare ginsenosides are more abundant in BG than in RG. Consequently, this study aims to investigate the effects of BG and its components on HF to elucidate the active substances and their underlying mechanisms in the treatment of HF.Methods: The effects of BG and its fractions (water-eluted fraction (WEF), total saponin fraction (TSF), and alcohol-eluted fraction (AEF)) on rats with isoproterenol (ISO)-induced HF were explored, and steroids belonging to the hypothalamic–pituitary–adrenal (HPA) and hypothalamic–pituitary–gonadal (HPG) axes were determined quantitatively using the ultra-performance liquid chromatography–triple quadrupole tandem mass spectrometry (UPLC-QqQ-MS/MS) method. In addition, 16S rDNA sequencing was performed on the gut microbiota, followed by GC–MS analysis of short-chain fatty acids (SCFAs), and the biochemical indexes related to energy metabolism and the serum cyclic nucleotide system were also analyzed by ELISA.Results: Based on a thorough evaluation of energy metabolism and the endocrine system, it was observed that the effects of BG components on the hypothalamic–pituitary–thyroid (HPT) and HPA axes were more pronounced. Notably, the treatment efficacy of the low dose of the total saponin fraction (TSFL), water decoction (WD), and high dose of the polysaccharide fraction (PSFH) was superior based on pharmacodynamic indicators such as brain natriuretic peptide (BNP), creatine kinase (CK), and estradiol (E2)/T). Furthermore, the WD and BG components exhibited significant effects on androgens (T and androstenedione (A4)). The TSFL group exerts an anti-inflammatory effect by regulating Lactobacillus/Erysipelotrichales. The WD, PSFH, and TSFL may impact inflammatory cytokines through the gut microbiota (Lactobacillus/Erysipelotrichales) and their metabolites (acetate and butyrate), exerting an anti-inflammatory effect.Discussion: The BG and all its split components demonstrated varying levels of efficacy in alleviating HF, and TSF and PSF exhibited a significant protective effect on HF. The main active components in TSF were revealed to be ginsenosides Rk1, Rk3, 20-(S)-Rg3, and 20-(S)-Rh2 by the H9C2 cell experiment. The decoction of BG and its components exhibited a potent impact on androgen hormones, with an elevation trend. This phenomenon may be attributed to the activation of the eNOS-NO pathway through androgen regulation, thereby contributing to its anti-HF activities. The WD, PSFH, and TSFL may exert anti-inflammatory effects through the intestinal flora (Lactobacillaceae/Erysipelotrichaceae) and its metabolites (acetic acid and butyric acid), which affect the inflammatory factors. The different mechanisms of action of each component of HF also reflect the significance and necessity of the overall role of traditional Chinese medicine (TCM). Our research was the first to report that the E2/T is related to HF and can be used as an indicator to evaluate HF.
