Defect-Boosted Piezoelectric and Nanozymatic Synergetic Catalysis for Deep Bacterial Abscess Therapy

Chao Peng,Wenting Wu,Huixiang Huo,Jing Li,Erkang Wang

Piezocatalytic therapy, distinguished by its superior tissue penetration under ultrasound activation and its low dependence on the oxygen concentration for generating multiple reactive oxygen species (ROS), has emerged as a promising strategy for deep abscess treatment. However, its efficacy remains constrained by a suboptimal bandgap, weak piezoelectric responses, and inadequate surface-active sites. To address these limitations, a facile Ru-atom doping strategy is first proposed using the Bi4O5Br2-based nanosystem (Bi4O5Br2@Ru) to tailor the bandgap and boost the piezo- and enzyme-like catalysis. The Ru dopants induce lattice distortions and elevate oxygen vacancies simultaneously, thereby enhancing piezoelectric responses while conferring peroxidase (POD)- and catalase (CAT)-like enzymatic activities. Under ultrasound excitation, the piezoelectric field optimizes the conduction band alignment of Bi4O5Br2@Ru, enhancing its POD-like activity to generate multiple ROS including ·OH, 1O2, and ·O2–. The mixed Ru (III/IV) valence states in Bi4O5Br2@Ru induce glutathione depletion, thereby further enhancing the oxidative stress capacity to combat biofilms. The enhanced CAT-like activity further alleviates hypoxia within biofilm microenvironments. RNA transcriptomic analysis confirms that Bi4O5Br2@Ru disrupts energy metabolism by interfering with the tricarboxylic acid cycle. The dual-modality therapy leverages doping engineering to seamlessly combine piezoelectric properties with enzyme-like catalytic functions, offering a highly promising therapeutic strategy for the treatment of deep-seated infectious diseases.

Yan Zhao,Jing-Jing Zhao,Jia-Xin Guo,Shuang-Qing Liu,Yang Li,Xiao-Yi Wang,Rong Li,Hui-Qin Tang,Zhen-Yu Li,Hui-Fen Yang,Bing Chen

Graphical Getting under the skin : BSA-CuS nanoparticles were used as photothermal reagents for depth photothermal therapy (PTT) treatment of subcutaneous abscesses. These nanoparticles exhibited remarkable photothermal conversion and low cytotoxicity combined with tissue-penetrating NIR laser (1064 nm) light. PTT based on BSA-CuS and 1064 nm NIR laser irradiation was found to sterilize drug-resistant pathogens in vitro and to eliminate subcutaneous abscesses in vivo effectively. That is OK, thank you! The treatment of subcutaneous abscess caused by drug-resistant bacteria is facing great difficulties and receiving more attention. In this work, we employed BSA-CuS nanoparticles as a photothermal reagent to apply photothermal therapy (PTT) to combat drug-resistant bacteria in vitro and subcutaneous abscess in vivo . The BSA-CuS nanoparticles were found to be stable and biocompatible without cytotoxicity toward NIH3T3 and 4T1 cells. In vitro experiments showed that three species of drug-resistant pathogens, including Escherichia coli , Staphylococcus aureus , and Candida albicans , could be effectively sterilized under co-incubation with BSA-CuS nanoparticles and then irradiation with 1064 nm NIR laser via tissue penetration. BSA-CuS nanoparticles together with 1064 nm NIR laser irradiation could also effectively diminish subcutaneous abscesses caused by drug-resistant bacteria on mice under PTT and depth PTT without causing any serious side effects and organic damage in vivo .That is OK, thank you!

pH-Responsive Metal-Phenolic Network Nanoparticles for Synergistic Chemo-Photodynamic Antibacterial Therapy

Ji-Rou Wu,Yi-Ru Wang,Jia-Yue Du,Ya-Li Meng,Xu-Ying Liu,Xiao-Lei Zhang,Yan-Fei Kang

The emergence of drug-resistant bacteria and their easy recurrence have been challenging in the clinical treatment of skin abscesses. Therefore, there is an urgent need to develop treatments against bacterial infections. This study developed an antibacterial PDT/CDT dual-mode treatment platform (Ce6/TA-Fe3+ NPs) to treat subcutaneous abscesses and promote wound healing. The Ce6/TA-Fe3+ NPs can quickly generate 1O2 under illumination and ·OH through the Fenton reaction in the microenvironment, leading to the eradication of biofilms. In particular, the Ce6/TA-Fe3+ NPs demonstrated over 90% effectiveness in fighting Staphylococcus aureus (S. aureus) bacteria and successfully eliminated biofilms after being exposed to a 660 nm laser for 10 min. Antibacterial mechanism research showed that the Ce6/TA-Fe3+ NPs induced a speedy rupture of the bacterial cell membranes, membrane lipid peroxidation, protein leakage, inhibition of intracellular ATP, and DNA damage based on the synergistic effect of PDT and CDT. Meanwhile, the Ce6/TA-Fe3+ NPs demonstrated good biosafety to normal cells and can clear the bacteria from the infected wound to promote the healing process of the infected area. The experimental results in vivo demonstrated that Ce6/TA-Fe3+ NPs effectively treated skin abscesses. Therefore, this study suggested that the Ce6/TA-Fe3+ NPs possessed the ability to kill bacteria, eliminate biofilms, and effectively treat subcutaneous abscesses based on the synergistic mechanism of PDT and CDT, which provided the promising strategy for the antibacterial medication and treatment of subcutaneous abscesses.

NIR-Responsive Nanoplatform Based on CuO2 with Self-Supplied H2O2/O2 for Synergistic Photodynamic-Chemodynamic-Photothermal Therapy of Skin Abscesses

Yingnan Zhu,Jiahang Si,Tiantian Xu,Lan Zheng,Xinyu Xiao,Xiaohan Sun,He Ding,Yuze Dong

Chemodynamic therapy (CDT) and photodynamic therapy (PDT) have emerged as promising strategies for treating skin abscesses. However, their therapeutic efficacy is fundamentally constrained by the hypoxic and hydrogen peroxide-deficient microenvironment characteristic of abscesses. Herein, we developed a near-infrared (NIR) light-responsive nanoplatform (ICG/CuO2@PCM) by encapsulating copper peroxide (CuO2) and indocyanine green (ICG) within a phase-change material (PCM), which enables control over self-supplied H2O2/O2 release via an NIR-triggered cascade. Upon irradiation, ICG-mediated photothermal conversion elevates the temperature beyond the PCM melting threshold, triggering a burst release of CuO2 and ICG. CuO2 generates hydroxyl radicals (•OH) through a Fenton-like reaction in the acidic wound microenvironment. Additionally, CuO2 can thermally decompose under photothermal conditions to produce a sustained oxygen supply, which ICG immediately converts into singlet oxygen (1O2), thereby enhancing PDT efficiency. Furthermore, photothermal therapy not only enhances the efficiency of CDT but also provides additional antibacterial support. Remarkably, ICG/CuO2@PCM (NPs), coupled with irradiation, accelerate abscess wound recovery by dispersing biofilm, killing bacteria, and promoting macrophage polarization toward an anti-inflammatory phenotype. Overall, this NIR-responsive nanoplatform highlights the promise in controlled modulation of microenvironment-adaptive antimicrobial strategies to overcome current limitations of hypoxic infections.

Antibacterial activity of the novel oxazolidinone contezolid (MRX-I) against Mycobacterium abscessus

Shan. Gao,Wenjuan. Nie,Lina. Liu,Lei. Su,Yingxia. You,Ruixue. Geng,Naihui Chu

Objective To evaluate contezolid (MRX-I) antibacterial activity against Mycobacterium abscessus in vitro and in vivo and to assess whether MRX-I treatment can prolong survival of infected zebrafish. Methods MRX-I inhibitory activity against M. abscessus in vitro was assessed by injecting MRX-I into zebrafish infected with green fluorescent protein-labelled M. abscessus . Thereafter, infected zebrafish were treated with azithromycin (AZM), linezolid (LZD) or MRX-I then maximum tolerated concentrations (MTCs) of drugs were determined based on M. abscessus growth inhibition using one-way ANOVA. Linear trend analysis of CFU counts and fluorescence intensities (mean ± SE values) was performed to detect linear relationships between MRX-I, AZM and LZD concentrations and these parameters. Results MRX-I anti- M. abscessus minimum inhibitory concentration (MIC) and MTC were 16 μg/mL and 15.6 μg/mL, respectively. MRX-I MTC-treated zebrafish fluorescence intensities were significantly lower than respective LZD group intensities (whole-body: 439040 ± 3647 vs. 509184 ± 23064, p < 0.01); head: 74147 ± 2175 vs. 95996 ± 8054, p < 0.05). As MRX-I concentration was increased from 0.488 μg/mL to 15.6 μg/mL, zebrafish whole-body, head and heart fluorescence intensities decreased. Statistically insignificant differences between the MRX-I MTC group survival rate (78.33%) vs. corresponding rates of the 62.5 μg/mL-treated AZM MTC group (88.33%, p > 0.05) and the 15.6 μg/mL-treated LZD MTC group (76.67%, p > 0.05) were observed. Conclusion MRX-I effectively inhibited M. abscessus growth and prolonged zebrafish survival when administered to M. abscessus -infected zebrafish, thus demonstrating that MRX-I holds promise as a clinical treatment for human M. abscessus infections.

Encapsulation of liquid metal nanoparticles inside metal–organic frameworks for hydrogel-integrated dual functional biotherapy

Juanjuan Li,Zhenzhen Fu,Yong Liu

Liquid metals represent emerging functional materials for many applications, yet their chemistries for surface/interface engineering and composite development have remained elusive. Here, we present a nanocomposite material featured by a liquid metal core encapsulated in a zeolitic-imidazolate framework shell. An initial functionalization step of liquid metal nanoparticles with amphiphilic surfactant molecules allows on-demand manipulation of the interparticle assembly between liquid metals and metal–organic frameworks, yielding a supraparticle-like composite material with multilevel surface structure and synergistic multifunctionality. The resulting supraparticles are employed as light-activable nanofillers in an alginate-based hydrogel, which displays good injectability, physiologically responsive sol–gel transformation, stable photothermal performance, and controllable zinc ion release. A temperature increase of 31.0 °C is achieved with the composite hydrogel after laser irradiation for 10 min (808 nm laser, 1.5 W/cm 2 ). The composite hydrogel then serves as safe, biodegradable (65.2 % degradation 14 days post implantation), dual functional antitumor and antibacterial biomedicine in an epidermal tumor model and an abscess model. Tumor cells/bacteria seeded on mice are completely (∼100 %) eradicated after a single treatment with laser-activated composite hydrogel. This study offers a pioneering road map for the design and fabrication of liquid metal/metal–organic framework composites, which is expected to trigger broad innovations in liquid metal nanotechnologies not limited to nano/biomedicine.

CuS nanoenzyme against bacterial infection by in situ hydroxyl radical generation on bacteria surface

Li Zhao,Xie Cheng-Jia,Ren Xiao-Wei,Zhang Qun,Ma Bao-Jin

Nanoenzyme-mediated antibacterial strategies have been widely exploited to overcome the shortcomings (such as drug resistance and mild-to-severe side effects) of antibiotic therapy. The peroxidase-like activity of nanoenzymes possesses great potential against bacterial infection by the generation of hydroxyl radical (·OH) in the specific microenvironment. However, the lifetime of ·OH is extremely short, and a large amount of the ·OH generated within the infection microenvironment cannot come into contact with bacteria quickly enough, thus resulting in low treatment efficiency. Here, chitosan-oligosaccharide-modified CuS nanoparticles possessing positive charges (PCuS NPs) were prepared using a one-pot method. PCuS NPs exhibited efficient peroxidase-like activity. Importantly, the PCuS NPs can combine with bacteria via electrostatic attraction. The direct contact between the PCuS NPs and bacteria enabled the generation of ·OH in situ on the bacterial surface, ultimately leading to a high antibacterial efficacy at a low concentration in the presence of H 2 O 2 . At an effective antibacterial concentration, the PCuS NPs exhibited high cytocompatibility. Furthermore, in vivo results revealed that PCuS NPs not only decreased the size of abscesses but also reduced inflammation and promoted collagen fiber formation. Therefore, PCuS NPs possess great potential against bacterial infection via in situ ·OH generation based on electrostatic attraction. Graphical abstract

Fe2+/Fe3+ Conversation-Mediated Magnetic Relaxation Switching for Detecting Staphylococcus Aureus in Blood and Abscess via Liposome Assisted Amplification

Guo Xin,Deng Xin-Chun,Zhang Ya-Qing,Luo Qin,Zhu Xiao-Kang,Song Yang,Song Er-Qun

Rapid and accurate detection of pathogenic bacteria of Staphylococcus aureus ( S. aureus ) is of importance in the field of clinical research and practical application. Herein, we report a magnetic relaxation switching (MRS)-based strategy for detecting S. aureus in complex samples via the assistance of signal amplification system of liposome. In this design, vancomycin (Van) modified 200 nm magnetic beads (MB 200 -Van) and biotinylated immunoglobulin (Biotin-IgG) were employed as capture probes for S. aureus which following labelled by biotinylated liposomes loading with glucose molecule (Biotin-liposome@Glu, BLGs) based on the affinity of biotin with streptavidin. The released glucose molecules combining with glucose oxidase generated Fe 3+ , causing transverse relaxation time ( T 2 ) signal change. The combination of dual recognition of S. aureus , liposome assisted signal amplification, and the MRS-based assay achieved direct and specific detection of S. aureus in blood and abscess samples. The as-proposed method for S. aureus assay in this study shows potential application for infectious diagnosis in clinical.

Neutrophil-inspired photothermo-responsive drug delivery system for targeted treatment of bacterial infection and endotoxins neutralization

Li Chengnan,Gan Yingying,Li Zongshao,Fu Mengjing,Li Yuzhen,Peng Xinran,Yang Yongqiang,Tian Guo-bao,Yang Yi Yan,Yuan Peiyan,Ding Xin

Background P. aeruginosa , a highly virulent Gram-negative bacterium, can cause severe nosocomial infections, and it has developed resistance against most antibiotics. New therapeutic strategies are urgently needed to treat such bacterial infection and reduce its toxicity caused by endotoxin (lipopolysaccharide, LPS). Neutrophils have been proven to be able to target inflammation site and neutrophil membrane receptors such as Toll-like receptor-4 (TLR4) and CD14, and exhibit specific affinity to LPS. However, antibacterial delivery system based on the unique properties of neutrophils has not been reported. Methods A neutrophil-inspired antibacterial delivery system for targeted photothermal treatment, stimuli-responsive antibiotic release and endotoxin neutralization is reported in this study. Specifically, the photothermal reagent indocyanine green (ICG) and antibiotic rifampicin (RIF) are co-loaded into poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NP-ICG/RIF), followed by coating with neutrophil membrane to obtain antibacterial delivery system (NM-NP-ICG/RIF). The inflammation targeting properties, synergistic antibacterial activity of photothermal therapy and antibiotic treatment, and endotoxin neutralization have been studied in vitro. A P. aeruginosa -induced murine skin abscess infection model has been used to evaluate the therapeutic efficacy of the NM-NP-ICG/RIF. Results Once irradiated by near-infrared lasers, the heat generated by NP-ICG/RIF triggers the release of RIF and ICG, resulting in a synergistic chemo-photothermal antibacterial effect against P. aeruginosa (~ 99.99% killing efficiency in 5 min). After coating with neutrophil-like cell membrane vesicles (NMVs), the nanoparticles (NM-NP-ICG/RIF) specifically bind to inflammatory vascular endothelial cells in infectious site, endowing the nanoparticles with an infection microenvironment targeting function to enhance retention time. Importantly, it is discovered for the first time that NMVs-coated nanoparticles are able to neutralize endotoxins. The P. aeruginosa murine skin abscess infection model further demonstrates the in vivo therapeutic efficacy of NM-NP-ICG/RIF. Conclusion The neutrophil-inspired antibacterial delivery system (NM-NP-ICG/RIF) is capable of targeting infection microenvironment, neutralizing endotoxin, and eradicating bacteria through a synergistic effect of photothermal therapy and antibiotic treatment. This drug delivery system made from FDA-approved compounds provides a promising approach to fighting against hard-to-treat bacterial infections.

Identification of a potential anti-nontuberculous mycobacterial drug candidate targeting a mycothiol disulfide reductase

Tianyu Liu,Yong Nian,Ying Liu,Simin Xu,Chujie Lu,Wei Yang,Wei Chen,Wei Lin

The resistance of nontuberculous mycobacteria (NTM) to conventional anti-tuberculosis drugs and its growing infection rate year by year urgently require new treatment strategies. Structure-based virtual screening, which can greatly improve efficiency and reduce costs in the early stage of drug development, is an indispensable part of modern drug discovery. In this study, the crystal structure of the mycothiol disulfide reductase from Mycobacterium abscessus ( Mab Mtr) was determined. Through virtual screening, compound AK-968/11492032 was identified as a promising candidate capable of fitting well into the potential MSSM-binding pocket of Mab Mtr. It was discovered that AK-968/11492032 and its derivatives ( Y6B and Y6C ) could produce antimicrobial effects on the Mycobacterial type strain Mycobacterium smegmatis . Moreover, microscale thermophoresis analysis was employed to evaluate the high binding affinity of the compounds to Mab Mtr. Furthermore, the key residues (S14, I47, H451) of Mab Mtr involved in the interaction with AK-968/11492032 were predicted and confirmed through molecular docking and mutational analysis, Mab Mtr was verified as the target for it to exert antibacterial effects through in vitro enzyme activity and in vivo gene knockout, complementation, and overexpression. These findings provide a potential development target to develop effective and specific anti-NTM drugs.

Multifunctional ZnFeO4-Based Antibiotic Cross-Linked Nanoplatform for Magnetically Targeted Treatment of Microbial Biofilms

Xingyue Yang,He Ren,Jiexin Li,Boyang Sun,Di Lu,Shuying Li,Yumiao Zhang

During the infections of wounds or medical device surfaces, bacteria such as Pseudomonas aeruginosa are prone to forming biofilms which prevent the penetration of antimicrobial agents. Herein, a combined therapy using bactericidal nano-micelles Ce6-ZnFeO4@ABCsi-micelles was developed that employed colistin cross-linked Pluronic micelles as a carrier, with the magnetic metal oxide ZnFeO4 and photosensitizer dihydroporphyrin (Ce6) encapsulated in the core of micelles. Driven by an external magnetic field, the nano-micelles could penetrate the external barrier of the biofilm. Reactive oxygen species (ROS) generated by the photosensitizer Ce6 disrupted the barrier of the biofilm, which facilitated the distribution of the Ce6 and colistin antibiotics into the biofilm, realizing a 99.5% clearance rate of bacteria. Using such a combination of chemotherapy and photodynamic therapy, Ce6-ZnFeO4@ABCsi-micelles could clear wound infection in a murine abscess model, owing to the targeted driving effect of the external magnetic field, along with the chemotherapeutic effect of colistin and the photodynamic effect of Ce6. This design provides an effective targeted therapeutic strategy to address biofilm infections.

Specific Chemiluminescence Imaging and Enhanced Photodynamic Therapy of Bacterial Infections by Hemin-Modified Carbon Dots

Ke Cheng,Henggang Wang,Shan Sun,Mingyu Wu,Hongzhe Shen,Ke Chen,Zhiyuan Zhang,Si Li,Hengwei Lin

Antibacterial photodynamic therapy (aPDT) is a promising antibiotics-alternative strategy for bacterial infectious diseases, which features broad-spectrum antibacterial activity with a low risk of inducing bacterial resistance. However, clinical applications of aPDT are still hindered by the hydrophobicity-caused inadequate photodynamic activity of conventional photosensitizers and the hypoxic microenvironment of bacterial infections. To address these problems, herein, a promising strategy is developed to achieve specific chemiluminescence (CL) imaging and enhanced PDT of bacterial infections using hemin-modified carbon dots (H-CDs). The H-CDs can be facilely prepared and exhibit favorable water solubility, augmented photodynamic activity, and unique peroxidase-mimicking capacity. Compared with the free CDs, the photodynamic efficacy of H-CDs is significantly augmented due to the increased electron–hole separation efficiency. Moreover, the peroxidase catalytic performance of H-CDs enables not only infection identification via bacterial infection microenvironment-responsive CL imaging but also oxygen self-supplied aPDT with hypoxia-relief-enhanced bacteria inactivation effects. Finally, the enhanced aPDT efficiencies of H-CDs are validated in both in vivo abscess and infected wound models. This work may provide an effective antibacterial platform for the selective imaging-guided treatment of bacterial infections.
